Histochemical Characteristics in Relation to Meat Quality Traits of Eight Major Muscles from Hanwoo Steers

  • The objective of this study was to determine the relationship between composition of muscle fiber types and meat quality traits of eight major muscles from Hanwoo steers. Longissimus lumborum (LL), psoas major (PM), semimembranosus (SM), semitendinosus (ST), gluteus medius (GM), triceps brachii (TB), rectus abdominis (RA) and superficialis flexor (SF) muscles were obtained from 9 Hanwoo steers and subjected to histochemical analysis. There were significant (p<0.05) differences in fiber number percentage (FNP) and fiber area percentage (FAP) of fiber types among these 8 major muscles. SF had the highest FNP of type I (55.9%), followed by PM (46.4%), TB (45.4%), RA (38.5%), LD (36.8%), GM (36.0%), SM (22.2%), and ST (18.8%). FAP of type IIB ranged from 9.9% in SF to 58.7% in ST. Meat quality traits, including fat content, myoglobin content, collagen content, CIE $L^*$ and $a^*$, drip and cooking loss, sarcomere length and Warner-Bratzler shear force, were all significantly (p<0.05) different among these muscles. Due to such diversities among these 8 muscles, lack of correlations were found between fiber type composition and meat quality traits. These results suggest that correlation for each individual muscle should be used to improve meat quality and profitability of retail beef cuts.

Regional bond strength of dentin bonding systems to pulp chamber dentin (치수강 상아질에 대한 상아질 접착제의 결합 강도에 관한 연구)

  • 본 연구는 치수강 상아질 부위 및 상아질 접착제 종류에 따른 결합 강도를 측정하고 이들 사이의 상관관계를 구명하고자 시행되었다 45개의 대구치를 포매 후, 대조군에서는 교합면 법랑질 제거 후 #600 SiC paper까지 순차연마하여 상아질을 노출시켰고, 실험군에서는 치수강 개방 후 1시간동안 NaOCl에 보관 후 axial wall과 pulpal floor를 노출시켰다. 노출된 상아질 면에 상아질 접착제를 적용한 후 Z-100을 충전한 다음 40초간 광중합하였다. 사용된 상아질 접착제는 Scotchbond Multi-Purpose와 Single Bond, Clearfil SE Bond였다. $37^{\circ}C$ 증류수에 24시간 보관 후, 저속 diamond saw를 이용하여 0.7mm 두께로 수직절단하고 고속 diamond point(#104)로 단면적 $1{mm}^2$가 되도록 시편을 제작하고, Universal testing machine에서 미세인장 결합강도를 측정하였다. 1. 모든 상아질 접착제의 미세인장강도는 대조군, axial wall군, pulpal floor군 순으로 감소하는 경향을 보였다. 2. 대조군에서 SM과 BB는 SE에 비해 유의성 있게 높은 결합강도를 나타내었다(p<0.05). 3. SM과 SB는 대조군에 비해 axial wall군과 pulpal floor군에서 유의성있게 낮은 결합강도를 보였으나, SE에서는 pulpal floor군만이 유의성 있게 낮은 결합강도를 보였다(p<0.05). 4. Axial wall군과 pulpal floor군에서는 상아질 접착제의 종류에 따른 유의차를 보이지 않았다. 5. 전자현미경 소견에서는 대조군에 비해 axial wall군과 pulpal floor군이 더 부드러운 접착 계면을 나타내었다. 혼성층의 두께는 결합강도의 감소와는 관련이 없었다.

Effect of rc Mutation on Semen Characteristics, Spermatogenic Tissues and Testosterone Profile in Blind Rhode Island Red Cockerels

  • Seven rc mutant and seven normal male birds (Rhode Island Red suie, RIR) were used in this study to determine the effects of rc mutation on semen characteristics, testosterone profile and spermatogenic tissues. All birds were randomly selected at week 12 of age and housed in individual cages and were fed and watered ad libitum. The birds were exposed to a 14L:10D light cycle during experiment. Semen were collected at weeks 22 to 23 from each bird twice a week and evaluated for semen volume (SV), sperm concentration (SC), total sperm count (TSC), percent of sperm motility (%SM), dead sperm (%DS), and sperm metabolic activity (SMA). To determine the testosterone concentration (TC) in plasma, blood was collected at weeks 12, 16 and 18. Testicular tissue were collected, processed and evaluated for semineferous tubule diameter (STD), round spermatid number (RSN), percent elongated sperm (%ES) and semineferous tubules length (STL). Body weight (BW), comb weight (CW) and testes weight (TW) were weighted at the end of experiment (week 23). The SV, TSC and %SM were significantly higher in normal birds but the %DS was higher in blind birds (p<0.05). The SC did not differ significantly between the two groups but its value was higher in normal birds. The sperm metabolic activity in the first h of collection did not differ significantly between the two groups but after 24 h, the level of SMA in normal group was significantly higher (p<0.05). The level of TC did not differ significantly between the two genotype groups but normal birds had higher TC in all collections except the last one. The STD, RSN, %ES and STL in normal birds were higher when compared to blind birds but the differences were insignificant except for ES percent. The BW, CW and TW between the two groups did not differ significantly but the weights were higher in normal group compared to blind birds. Statistical analysis of semen characteristics, testosterone profile and histological factors were indicated detrimental effects of rc mutation in prepubertal RIR blind male birds due to lack of light.

A Study on the Evaluation of Tension-Compression Fatigue Characteristics of Glass Fiber/Epoxy 4-Harness Satin Woven Laminate Composite for the Railway Bogie Application (철도차량 대차 적용 유리섬유/에폭시 4-매 주자직 적층 복합재의 인장-압축 피로특성 평가 연구)

  • This paper describes the evaluations of tension-compression fatigue characteristics and life for glass fiber/epoxy laminate composite applied to railway bogie to reduce weight. Test samples of tension-compression fatigue were composed of glass fiber/epoxy 4-harness woven laminate composites with different stacking sequence of warp-direction, fill-direction and ${\pm}45^{\circ}$-direction. The tension-compression fatigue test was conducted with stress ratio (R) of -1 and frequency of 5Hz. Goodman diagram were used to evaluate the fatigue characteristics and life of glass fiber/epoxy 4-harness satin woven laminate composite. Anti-buckling jig was designed to prevent buckling of specimen under compression load. The test results showed that the fatigue characteristics of glass fiber/epoxy 4-harness satin woven laminate composite with stacking sequence of warp-direction had a good performance in comparison with that of SM490 used to conventional metal railway bogie.

The Compressive Strength of Longitudinally Stiffened Plates Undergoing Local and Distortional Buckling (국부좌굴과 뒤틀림좌굴이 발생하는 종방향 보강재로 보강된 강판의 압축강도)

  • This paper describes an experimental research on the structural behavior and the ultimate strength of longitudinally stiffened plates subjected to local, distortional, or mixed-mode buckling under compression. The stiffened plate undergoes local, distortional, or interactive local-distortional buckling according to the flexural rigidity of the plate's longitudinal stiffeners and the width-thickness ratios of the sub-panels of the stiffened plate. A significant post-buckling strength in the local and distortional modes affects the ultimate strength of the longitudinally stiffened plate. Compression tests were conducted on stiffened plates that were fabricated from 4mm-thick SM400 steel plates with a nominal yield stress of 235MPa. A simple strength formula for the Direct Strength Method based on the test results was proposed. This paper proves that the Direct Strength Method can properly predict the ultimate strength of stiffened plates when the local buckling and distortional buckling occur simultaneously or nearly simultaneously.

Neutralization of Human Papillomavirus by Specific Nanobodies Against Major Capsid Protein L1

  • The human papillomavirus (HPV) is the main cause of cervical cancer in developing countries. Rapid diagnosis and initiation of treatment of the HPV infection are critical. Various methods have been employed to reduce the immunogenicity of antibodies targeting HPV serotypes. Nanobodies are the smallest fragments of naturally occurring single-domain antibodies with their antigen-binding site compromised into a single domain. Nanobodies have remarkable properties such as high stability, solubility, and high homology to the human VH3 domain. In this study, a phagemid library was employed to enrich for nanobodies against the L1 protein of the human papilloma virus. Binding reactivity of the selected clones was evaluated using phage enzyme-linked immunosorbent assay (phage-ELISA). Finally, two nanobodies (sm5 and sm8) with the best reactivity against the Gardasil vaccine and the purified HPV-16 L1 protein were expressed and purified using a $Ni^+$-NTA column. The accuracy of expression and purification of the nanobodies was confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting assays. In vitro studies demonstrated that neutralization was achieved by the selected nanobodies. The ease of generation and unique features of these molecules make nanobodies promising molecules for the new generation of HPV diagnosis and therapy.

Efficacy Test of Polycan, a Beta-Glucan Originated from Aureobasidium pullulans SM-2001, on Anterior Cruciate Ligament Transection and Partial Medial Meniscectomy-Induced-Osteoarthritis Rats

  • The object of this study was to assess the efficacy of Polycan from Aureobasidium pullulans SM-2001, which is composed mostly of beta-1,3-1,6-glucan, on osteoarthritis (OA)-induced by anterior cruciate ligament transection and partial medial meniscectomy (ACLT&PMM). Three different dosages of Polycan (85, 42.5, and 21.25 mg/kg) were orally administered once a day for 84 days to male rats a week after ACLT&PMM surgery. Changes in the circumference and maximum extension angle of each knee, and in cartilage histopathology were assessed using Mankin scores 12 weeks after Polycan administration. In addition, cartilage proliferation was evaluated using bromodeoxyuridine (BrdU). As the result of ACLT&PMM, classic OA was induced with increases in maximum extension angles, edematous knees changes, and capsule thickness, as well as decreases in chondrocyte proliferation, cartilages degenerative changes, and loss of articular cartilage. However, these changes (except for capsule thickness) were markedly inhibited in all Polycan- and diclofenac sodium-treated groups compared with OA control. Although diclofenac sodium did not influence BrdU uptake, BrdU-immunoreactive cells were increased with all dosages of Polycan, which means that Polycan treatment induced proliferation of chondrocytes in the surface articular cartilage of the tibia and femur. The results obtained in this study suggest that 84 days of continuous oral treatment of three different dosages of Polycan led to lesser degrees of articular stiffness and histological cartilage damage compared with OA controls 91 days after OA inducement, suggesting that the optimal Polycan dosage to treat OA is 42.5 mg/kg based on the present study.

THE EFFECT OF ADHESIVE PROPERTY ON MICROTENSILE BOND STRENGTH TO HUMAN DENTIN (상아질 접착제의 성상이 미세인장결합강도에 미치는 영향)

  • The purposes of this study were to evaluate the effect of adhesive property on microtensile bond strength and to determine the failure mode. Flat occlusal dentin surfaces were prepared using low-speed diamond saw. The dentin was etched with 37% phosphoric acid. The following adhesives were applied to the etched dentin to manufacturer's directions: Scotchbond Multi-Purpose in group SM, Prime&Bond NT in group NT, Scotchbond Multi-Purpose followed by Tetric-flow in group TR. After adhesive application, a cylinder of resin-based composite was built up on the occlusal surface. Each tooth was sectioned vertically to obtain the $1{\;}{\times}{\;}1\textrm{mm}^2$ "sticks". Microtensile bond strength were determined. Each specimen was observed under stereomicroscope and scanning electron microscope (SEM) to examine the failure mode. Data were analyzed using one way ANOVA. The results of this study were as follows:1. The microtensile bond strength value were:group SM ($18.98{\pm}3.01MPa$). group NT ($16.01{\pm}4.82MPa$) and group TR ($17.56{\pm}3.22MPa$). No significant statistical differences were observed among the groups (P>0.05). 2. Most of specimens showed mixed failure. In group TR, there was a higher number of specimens showing areas of cohesive failure in resin.

Hyaluronic acid and proteoglycan link protein 1 suppresses platelet-derived growth factor-BB-induced proliferation, migration, and phenotypic switching of vascular smooth muscle cells

  • The development of atherosclerotic cardiovascular disease is associated with the phenotypic switching of vascular smooth muscle cells (SMCs) from a contractile to a synthetic state, leading to cell migration and proliferation. Platelet-derived growth factor-BB (PDGF-BB) modulates this de-differentiation by initiating a number of biological processes. In this study, we show that gene expression of hyaluronic acid (HA) and proteoglycan link protein 1 (HAPLN1) was upregulated during differentiation of human aortic SMCs (HASMCs) into a contractile state, but downregulated upon during PDGF-BB-induced dedifferentiation. This is the first study showing that the treatment of HASMCs with full-length recombinant human HAPLN1 (rhHAPLN1) significantly reversed PDGF-BB-induced decrease in the protein levels of contractile markers (SM22α, α-SMA, calponin, and SM-MHC), and inhibited the proliferation and migration of HASMCs induced by PDGF-BB. Furthermore, our results show that rhHAPLN1 significantly inhibited the phosphorylation of FAK, AKT, STAT3, p38 MAPK and Raf mediated by the binding of PDGF-BB to PDGFRβ. Together, these results indicated that rhHAPLN1 can suppress the PDGF-BB-stimulated phenotypic switching and subsequent de-differentiation of HASMCs, highlighting its potential as a novel therapeutic target for atherosclerosis and other vascular diseases.

Response Spectrum Analysis-Induced Limit Acceleration of Soil Pile Systems (지반-기초말뚝 상호작용을 고려한 응답스팩트럼의 적용 한계가속도)

  • In this study, the limit range of input acceleration was investigated based on time domain and response spectrum analyses by considering the relative density, groundwater depth, and soil type. Special attention was paid to the input acceleration and shear modulus of soil, which affect pile behavior. The surrounding soil was identified as an elastoplastic material and subjected to FLAC3D analysis using the Mohr-Coulomb and Finn models as well as FB-Multiplier analysis using a nonlinear p-y curve for soil spring. Based on the analyses, the limit range of acceleration on the pile is much higher for SP soil than for SM soil, and the groundwater level tends to reduce the limit range of input acceleration, irrespective of soil conditions. The limit range of acceleration was mainly affected by the shear modulus. The limit range of acceleration with nonlinear soil behavior is proportional to the relative density of the surrounding soil.
