Isolation and Characterization of a Marine Derived Bacterium Glaciecola sp. SL-12 Producing β-agarase (한천분해효소를 생산하는 해양유래 세균 Glaciecola sp. SL-12의 분리 및 특성)

  • A novel agar-degrading bacterium SL-12 was isolated from seashore of Kijang at Busan, Korea, and cultured in marine broth 2216 media. Isolated bacterium SL-12 was identified as Glaciecola genus by 16S rDNA sequencing with 98% identity. The optimum pH of the enzyme activity was 7.0 and the optimum temperature for the reaction was $30^{\circ}C$. The enzyme hydrolyzed neoagarohexaose to yield neoagarobiose as the main product, indicating that the enzyme is ${\beta}$-agarase. Thus, isolated bacterium and the enzyme would be useful for the industrial production of neoagarobiose.

  • A novel agar-degrading bacterium SL-5 was isolated from seashore of Homigot at Kyung-Buk province, and cultured in marine broth 2216 media. The bacterium SL-5 was identified as Thalassomonas genus by 16S rDNA sequencing with 96% identity. Growth rate was faster at $27^{\circ}C$ than at $37^{\circ}C$ and agarase was produced as growth-related. The optimum pH of the enzyme activity was 7.0 and the optimum temperature for the reaction was $40^{\circ}C$. Although the enzyme had no thermostability, the enzyme activity was remained over 80% at $60^{\circ}C$. The enzyme hydrolyzed neoagarohexaose to yield neoagarobiose as the main product, indicating that the enzyme is $\beta-agarase$. Thus, the enzyme would be useful for the industrial production of neoagarobiose.

  • The purpose of this study was to develop a new connection method between steel beams and PC columns known as SL connectors. Composite moment frames consisting of PC columns (or composite columns) and steel beams make the best use of advantages of both concrete and steel materials. However, the connection between two members of different materials can be complex and/or increase the fabrication costs significantly. The concept of SL connectors is based on using wedges and the emphasis is on a self-locking (SL) feature. SL connectors are easy to install and provide better seismic performance compared to conventional connections. To evaluate the seismic performance of the steel beam-to-PC column joints with SL connectors, cyclic load tests were conducted. Test result showed that steel beam-to-concrete column joint with SL connectors was able to provide sufficient performance for use in seismic resistant moment frames.

Associations between Brain Perfusion and Sleep Disturbance in Patients with Alzheimer's Disease

  • Background and Purpose Although sleep disturbances are common and considered a major burden for patients with Alzheimer's disease (AD), the fundamental mechanisms underlying the development and maintenance of sleep disturbance in AD patients have yet to be elucidated. The aim of this study was to examine the correlation between regional cerebral blood flow (rCBF) and sleep disturbance in AD patients using technetium-99m hexamethylpropylene amine oxime single-photon emission computed tomography (SPECT). Methods A total of 140 AD patients were included in this cross-sectional study. Seventy patients were assigned to the AD with sleep loss (SL) group and the rest were assigned to the AD without SL group. SL was measured using the sleep subscale of the Neuropsychiatric Inventory. A whole-brain voxel-wise analysis of brain SPECT data was conducted to compare the rCBF between the two groups. Results The two groups did not differ in demographic characteristics, severity of dementia, general cognitive function, and neuropsychiatric symptoms, with the exception of sleep disturbances. The SPECT imaging analysis displayed decreased perfusion in the bilateral inferior frontal gyrus, bilateral temporal pole, and right precentral gyrus in the AD patients with SL group compared with the AD patients without SL group. It also revealed increased perfusion in the right precuneus, right occipital pole, and left middle occipital gyrus in the AD with SL group compared with the AD without SL group. Conclusions The AD patients who experienced sleep disturbance had notably decreased perfusion in the frontal and temporal lobes and increased rCBF in the parietal and occipital regions. The findings of this study suggest that functional alterations in these brain areas may be the underlying neural correlates of sleep disturbance in AD patients.

Effects of Isometric Exercise on Muscle Activity and Body Balance Ability in Asymmetrical Walkers

  • Purpose: This study was to investigate the effects of isometric exercise on muscle activity and body balance ability in asymmetrical walkers. Research design, data, and methodology: Twenty gait asymmetry people were divided to unilateral exercise group (UG, n=10) and bilateral exercise group (BG, n=10). UG were performed unilateral exercise for 60 minutes, three times a week, and 16 weeks, and BG were performed one side and then the other side alternately for 60 minutes, three times a week, and 16 weeks. Muscle activity and body balance ability were measured before, after 4 and 16 weeks isometric exercise. Moreover, SI (symmetry index; SI) was calculated from the measured value of SL (step length). Statistical analyses were conducted using one-way ANOVA and two-way ANOVA with repeated measures, a paired t-test, and multiple comparisons according to Scheffe. Results: In the muscle activity, ST decreased significantly in short step length(S-SL) and BG of LS compared to before isometric exercise(p<.05), and GCM decreased significantly in BG of S-SL(p<.05). As for body balance ability, the mSEBT-A difference between L-SL and S-SL was decreased significantly in UG(p<.05). And the respective total scores of L-SL and S-SL, mSEBT-PM and mSEBT-PL were increased significantly in BG(p<.05). Conclusions: As a result, in this study above, it was confirmed that isometric exercise improved muscle activity and body balance ability in asymmetrical walkers.

Ameliorative Effects of Soybean Leaf Extract on Dexamethasone-Induced Muscle Atrophy in C2C12 Myotubes and a C57BL/6 Mouse Model (콩잎 추출물의 근위축 개선 효과)

  • Sarcopenia, a condition characterized by the insidious loss of skeletal muscle mass and strength, represents a significant and growing healthcare challenge, impacting the mobility and quality of life of aging populations worldwide. This study investigated the therapeutic potential of soybean leaf extract (SL) for dexamethasone (Dexa)-induced muscle atrophy in vitro and in an in vivo model. In vitro experiments showed that SL significantly alleviated Dexa-induced atrophy in C2C12 myotube cells, as evidenced by preserved myotube morphology, density, and size. Moreover, SL treatment significantly reduced the mRNA and protein levels of muscle RING-finger protein-1 (MuRF1) and muscle atrophy F-box (MAFbx), key factors regulating muscle atrophy. In a Dexa-induced atrophy mouse model, SL administration significantly inhibited Dexa-induced weight loss and muscle wasting, preserving the mass of the gastrocnemius and tibialis anterior muscles. Furthermore, mice treated with SL exhibited significant improvements in muscle function compared to their counterparts suffering from Dexa-induced muscle atrophy, as evidenced by a notable increase in grip strength and extended endurance on treadmill tests. Moreover, SL suppressed the expression of muscle atrophy-related proteins in skeletal muscle, highlighting its protective role against Dexa-induced muscle atrophy. These results suggest that SL has potential as a natural treatment for muscle-wasting conditions, such as sarcopenia.

A Study on the Growth and Spawning of Korean Scallop (Chlamys farreri) around Wando, Korea (한국 완도연안 비단가리비(Chlamys farreri)의 성장과 산란)

  • The population habitat and density of Korean scallops, Chlamys farreri, were investigated to estimate population ecological characteristics from samples randomly collected around Wando from November, 1998 to October, 1999. Age and growth of the Korean scallops were determined from their ring radii. Maturation and spawning were studied using data of ovary maturity stage, gonadosomatic index, and fecundity. Seawater temperature and specific gravity ranged from 7.6 to $25.9^{\circ}C$ and from 1.0188 to 1.0260, respectively. Also dissolved oxygen and pH ranged from 6.48 to 9.50 ppm and from 8.17 to 8.80. Rocky and gravel bottom had a maximum habitat density of $$28.83 inds/100m^2$$ , which accounted for 82.4 % of the overall habitat area. The relationship between shell length (SL, mm) and shell height (SH, mm) of the Korean scallops was fitted : SH=1.021 SL+2.211 $(R^2=0.989)$. The shell length-total weight (TW, g) relationship was $TW=0.0003; SL^{2.837};(R^2=0.980)$. Then von Bertalanffy growth parameters were estimated from a nonlinear regression method, and the values were as follows : $SH_{\infty}=117.4 mm$K=0.61/year,; and; t_0=-0.017 year., The 50 % maturity at age was 0.21 year with the shell height of 18.3 mm, and spawning occurred twice a year, that is, June/July and October. The relationship between fecundity (Fc) and shell length was$Fc=697.03 SL^{2.683}(R^2=0.984)$, and the fecundity-gonad weight (GW, g) relationship was Fc=10,076,090 GW+15,608,781 $(R^2=0.990)$.

Mathematical Analysis on TTI's Estimation Accuracy of Food Shelf Life Depending on its Discrepancy in Temperature Dependence (상호 온도의존성의 차이에 따른 TTI의 식품 shelf life 예측 정확성에 대한 수리적 분석)

  • TTI is a small label of which the color changes by time-temperature history during food storage. The food shelf life (SL) was compared with that of TTI, the time for TTI to reach the end-point of its color change, for the various discrepancies in two Arrhenius activation energies (Ea), an important parameter of temperature dependence. The SL of TTI and food were mathematically simulated, based on zero-order and first-order kinetics, respectively. In the case Ea of food was smaller than that of TTI, the SL of food was larger than that of TTI, meaning TTI reaches the end-point of color change earlier even though food is still fresh. In the case of Ea of food > Ea of TTI, the food reaches the SL earlier than the TTI. In addition, the magnitude of ${\Delta}Ea$ between food and TTI led to the bigger ${\Delta}SL$. To be safe, $SL_{Food}$ > $SL_{TTI}$ would be practical although $SL_{Food}{\fallingdotseq}SL_{TTI}$ is ideal.

Mass Production of Artificial Seedlings in Hard Clam Meretrix petechialis (Lamarck) (말백합 Meretrix petechialis (Lamarck) 인공종묘의 대량생산)

  • Mass production method on artificial seedling production of hard clam Meretrix petechialis was developed indoor culture system. Spawning of adult clam (SL $65.8{\pm}8.4mm$) was induced using the combined method of air exposure and water temperature raising. The fertilized eggs were developed to D-shaped larvae after 17.7 hours at $27^{\circ}C$ and hatching rate was 6.1%. Shell length (SL) of D-shaped larvae was measured to be $131.4{\pm}2.6{\mu}m$ and thereafter the larvae grew to the settled spats with SL $190.2{\pm}7.5{\mu}m$ in 4 days. Estimated survival rate of settled spats was 48.1%. Spat collection on 130,000 spats with SL $0.19{\pm}0.01mm$ performed conducted by sand bottom circulation filtering method. Collected spats grew up to $3.1{\pm}0.8mm$ in 46 days, $6.6{\pm}1.8mm$ in 87 days, and $10.5{\pm}0.9mm$ in 114 days. The relative growth between SL and shell height (SH) was calculated to be SH = 0.8501SL + 0.0196 ($R^2=0.9987$) during the whole spat period. During spats rearing, they were suffered from one time of mass mortality at SL 3.1 mm, but 51,000 spats were finally survived with the rate of 39.2% at 114 days of spat rearing in indoor tank system.

  • In the early stage of sacroilitis, it is'difficult to detect sacroiliac(Sl) abnormalities by conventional plain X-ray even though there are characteristic symptoms of ankylosing spondylitis. 3 dimensional volume rendering from the CT image was performed to make an early de tection of the structural changes of Sl joint. 2 cases who had clinical impression of ankylosing spondylitis without sacroilitis in plane X-ray and 1 case of typical ankylosing spondylitis as well as 1 case of normal control were studied. The Sl Joints were separated and each joint surface of sacrum and ilium was independently reconstructed by a special 3D manipulation program. All 2 patiant who complained of inflammatory lower back pain with no abnormal findings in the plain X-ray showed structural changes in 3 dimensionally reconstructed surface Image of the Sl joint compared to the normal control. Authors tried several parameters, such as fourler analysis of each surface and the mean and variance of Sl joint gap. We couldn't tell the statistical significance because of the limited number of cases. However, the parameters showed difference according to the progression of disease.
