Guaiane Sesquiterpenoids from Torilis japonica and Their Cytotoxic Effects on Human Cancer Cell Lines

  • A new compound 2 and two known guaiane-type sesquiterpenoids were isolated from the methylene chloride-soluble fraction of the methanolic extract of the fruits of Torilis japonica (Umbelliferae) through repeated silica gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as torilin (1), 11-acetoxy-8-angeloyloxy-$1{\beta}$-hydroxy-4-guaien-3-one ($1{\beta}$-hydroxytorilin, 2), and 11-acetoxy-8-angeloyloxy-$1{\alpha}$-hydroxy-4-guaien-3-one ($1{\alpha}$-hydroxytorilin, 3) by spectroscopic analysis. Compounds 1-3 exhibited cytotoxicity against human A549, SK-OV-3, SK-MEL-2, and HCT15 tumor cells.

Hydrolysable Tannins and Related Compound having Cytotoxic Activity from the Fruits of Terminalia chebula

  • The cytotoxicity-directed fractionation of MeOH extract of Terminalia chebula fruits led to the isolation of three hydrolyzed tannins and a related compound, gallic acid(1), $1,2,3,4,6-penta-O-galloy-{\beta}-_D/-glucopyranose(II)$,. chebulagic acid (III) and chebulinic acid(IV), as active principles. They were shoen to exhibit moderate cytotoxicity against cultured human tumor cell lines including A-549, SK-OV-3, Sk-MEL-2, XF-498 and HCT-15 in vitro.

  • A bioassay-guided fractionation of the whole extract of Viscum album (a parasitic plant : Loranthaceae) led to the isolation of two triterpenoidal components, oleanolic acid (1), ${\beta}$-amyrin acetate (2), homoflavoyadorinin B (3) as well as large quantity of free fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition.   • Phytochemical works on the aerial parts of Aster oharai (Compositae) led to the isolation of a new sesquiterpene hydroperoxide, 7$\alpha$-hydroperoxy-3, 11-eudesmadiene (2) and seven known compounds, teucdiol B (1), $\alpha$-spinasterol (3), oleanolic acid (4), $\alpha$-spinasterol 3-Ο-$\beta$-D-glucopyranoside (5), methyl 3,5-di-Ο-caffeoyl quinate (6), 3,5-di-Ο-caffeoylquinic acid (7), 3,4-di-Ο-caffeoylquinic acid (8). The chemical structures of 1-8 were established by chemical and spectroscopic methods. Compound 2 showed cytotoxicity against cultured human tumor cell lines in vitro, SK-OV-3 (ovarian), SK-MEL-2 (skin melanoma), and HCT15 (colon) with $ED_{50}$ values ranging from 3.86-17.21 $\mu$g/mL.

  • Activity-guided fractionation and repeated column chromatography afforded two cytotoxic compounds R-3 and R-4 from the root of Rumex japonicus HOUTT. Compou nds were identified as musizin and emodin, respectively, by the physicochemical and spectral data. Besides R-3 and R-4, two compounds R-1 and R-2, chrysophanol and physcion, respectively, were also isolated. The compound R-3 and R-4 exhibited cytotoxicity against cultured human tumor cell lines, A-549, SK-OV-3, SK-MEL-2, XF498 and HCT15 with $ED_{50}$ values ranging from 2.68 to $10.06{\mu}g/ml$.

  • The activity-oriented fractionation of Psoralea corylifolia led to an isolation of a (+) bakuchio 1 as an activ eprinciple of its antitumoral property in vitro 1 was observed to exhibit a mild cytotoxicity against five kinds of cultured human cancer cell lines, i. e. the A549, SK-OV-3, SK-MEL-2, XF-498 and HCT15. The synthesized 2, 3-epoxide of (+)-bakuchiol 3 showed the similar activity as the (+) bakuchiol 1, whereas the other oxidation derivatives 4 and 5 including the acetyl (+) bakuchiol 2 showed a decreased activity.

  • Through bioassay-guided separation of the chemical constituents from the heartwood of Dalbergia odorifera, an 2'-O-methoxyisoliquiritigenin(1) was isolated as cytotoxic principle. 1 showed potent cytotoxic activity against the three kinds of human cancer cell lines (A-549, SK-MEL-2 and SK-OV-3) with similar activity to 5-Fluorouracil.

  • Methanol extracts of five Berberidaceae species were examined against tissue factor inhibitory and tumour cell growth inhibitory activity. Methanol extracts of Berberis koreana Palibin showed a strong cytotoxicity activity against SK-MEL-2 (Melanoma) tumour cell lines with more than 90% in $25{\mu}g/m\ell$ and against A549 (Lung carcinoma), SK-OV-3 (Ovarian cancer), XF498 (CNS cancer) and HCTl5 (Colon cancer), other Berberidaceae species except B. koreana species have no effect on the tumour cells. Biologically active compound, therefore, was isolated through the activity guided fractionation and purification. The structure was confirmed by NMR. FT-IR and MS to 2-(3,4-dihydroxybenzyl)-ethyl alcohol. It showed cytotoxicity activity against SNU-C4 tumour cell lines with 50.7% in $50{\mu}g/m\ell$. Methanol extracts of 5 Berberidacae species have no effect on the tissue factor inhibitory activity.

  • Bioassay-guided fractionation of the aerial parts of Pilea mongolica(Urticaceae) afforded two cytotoxic triterpenoids, epi-oleanolic acid (I) and oxo-oleanolic acid (II). The structures of the compounds were confirmed by spectral and synthetic evidences. Compound I and compound II exhibited cytotoxicity against cultured human tumor cell lines, A549 (non small cell lung adenocarcinoma), SK-OV-3 (ovarian), SK-MEL-2 (skin melanoma), XF498 (CNS) and HCT15 (colon) with $ED_{50}$ values of $3.2-8.1{\mu}g/ml$ and $0.7-6.8 {\mu}g/ml$, respectively.

  • To evaluate the antitumor activity and antimetastatic effects of Bujeongyangeumtang(BJYET), studies were done experimentally. The results were obtained as follows: 1. BJYET extracts exhibited a significant cytotoxicity against A549, SK-MEL-2, SK-OV-3 and B16-BL6 cell lines. 2. The T/C% was 118.2% in BJYET treated group in S-180 bearing ICR mice. 3. BJYET extracts exhibited inefficient adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 4. BJYET extracts showed a significant inhibition of lung metastasis of B16-BL6 cells in C57BL/6. 5. In vitro neovascularization assays, angiogenesis was significantly inhibited in BJYET treated group than control group. These results suggested that BJYET extracts might be usefully applied for prevention and treatment of cancer.
