A Pattern Recognition Receptor, SIGN-R1, Mediates ROS Generation against Polysaccharide Dextran, Resulting in Increase of Peroxiredoxin-1 and Its Interaction to SIGN-R1

  • Streptococcus pneumoniae is the major pathogen that frequently causes serious infections in children, the elderly and immunocompromised patients. S. pneumoniae is known to produce reactive oxygen species (ROS) and S. pneumoniae-produced ROS is considered to play a role in pneumococci pathogenesis. SIGN-R1 is the principal receptor of capsular polysaccharides (CPSs) of S. pneumoniae. However, there is a considerable lack of knowledge about the protective role of SIGN-R1 against S. pneumoniae-produced ROS in SIGN-$R1^+$ macrophages. While investigating the protective role of SIGN-R1 against ROS, we found that SIGN-R1 intimately bound to peroxiredoxin-1 (Prx-1), one of small antioxidant proteins in vitro and in vivo. This interaction was increased with ROS generation which was produced by stimulating SIGN-R1 with dextran, a polysaccharide ligand of SIGN-R1. Also, SIGN-R1 crosslinking with 22D1 anti-SIGN-R1 antibody increased Prx-1 in vitro or in vivo. These results suggested that SIGN-R1 stimulation with CPSs of S. pneumoniae increase the expression level of Prx-1 through ROS and its subsequent interaction to SIGN-R1, providing an important antioxidant role for the host protection against S. pneumoniae.

A Novel Complement Fixation Pathway Initiated by SIGN-R1 Interacting with C1q in Innate Immunity

  • Serum complement proteins comprise an important system that is responsible for several innate and adaptive immune defence mechanisms. There were three well described pathways known to lead to the generation of a C3 convertase, which catalyses the proteolysis of complement component C3, and leads to the formation of C3 opsonins (C3b, iC3b and C3d) that fix to bacteria. A pivotal step in the complement pathway is the assembly of a C3 convertase, which digests the C3 complement component to form microbial-binding C3 fragments recognized by leukocytes. The spleen clears microorganisms from the blood. Individuals lacking this organ are more susceptible to Streptococcus pneumoniae. Innate resistance to S. pneumoniae has previously been shown to involve complement components C3 and C4, however this resistance has only a partial requirement for mediators of these three pathways, such as immunoglobulin, factor B and mannose-binding lectin. Therefore it was likely that spleen and complement system provide resistance against blood-borne S. pneumoniae infection through unknown mechanism. To better understand the mechanisms involved, we studied Specific intracellular adhesion molecule-grabbing nonintegrin (SIGN)-R1. SIGN-R1, is a C-type lectin that is expressed at high levels by spleen marginal-zone macrophages and lymph-node macrophages. SIGN-R1 has previously been shown to be the main receptor for bacterial dextrans, as well as for the capsular pneumococcal polysaccharide (CPS) of S. pneumoniae. We examined the specific role of this receptor in the activation of complement. Using a monoclonal antibody that selectively downregulates SIGN-R1 expression in vivo, we show that in response to S. pneumoniae or CPS, SIGN-R1 mediates the immediate proteolysis of C3 and fixation of C3 opsonins to S. pneumoniae or to marginal-zone macrophages that had taken up CPS. These data indicate that SIGN-R1 is largely responsible for the rapid C3 convertase formation induced by S. pneumoniae in the spleen of mice. Also, we found that SIGN-R1 directly binds C1q and that C3 fixation by SIGN-R1 requires C1q and C4 but not factor B or immunoglobulin. Traditionally C3 convertase can be formed by the classical C1q- and immunoglobulin-dependent pathway, the alternative factor-B-dependent pathway and the soluble mannose-binding lectin pathway. Furthermore Conditional SIGN-R1 knockout mice developed deficits in C3 catabolism when given S. pneumoniae or its capsular polysaccharide intravenously. There were marked reductions in proteolysis of serum C3, deposition of C3 on organisms within SIGN-$R1^+$ spleen macrophages, and formation of C3 ligands. The transmembrane lectin SIGN-R1 therefore contributes to innate resistance by an unusual C3 activation pathway. We propose that in the SIGN-R1 mediated complement activation pathway, after binding to polysaccharide, SIGN-R1 captures C1q. SIGN-R1 can then, in association with several other complement proteins including C4, lead to the formation of a C3 convertase and fixation of C3. Therefore, this new pathway for C3 fixation by SIGN-R1, which is unusual as it is a classical C1q-dependent pathway that does not require immuno globulin, contributes to innate immune resistance to certain encapsulated microorganisms.

  • In this paper we prove that if AX=b is a partial sign-solvable linear system with A being sign non-singular matrix and if ${\alpha}=\{j:\;x_j\;is\;sign-determined\;by\; Ax=b\}, then $A_{\alpha}X_{\alpha}=b_{\alpha}$ is a sign-solvable linear system, where $A_{\alpha}$ denotes the submatrix of A occupying rows and columns in o and xo and be are subvectors of x and b whose components lie in ${\alpha}$. For a sign non-singular matrix A, let $A_l,\;...,A_{\kappa}$ be the fully indecomposable components of A and let ${\alpha}_i$ denote the set of row numbers of $A_r,\;r=1,\;...,\;k$. We also show that if $A_x=b$ is a partial sign-solvable linear system, then, for $r=1,\;...,\;k$, if one of the components of xor is a fixed zero solution of Ax=b, then so are all the components of x_{{\alpha}r}$.

GROUND STATE SIGN-CHANGING SOLUTIONS FOR A CLASS OF SCHRÖDINGER-POISSON-KIRCHHOFF TYPEPROBLEMS WITH A CRITICAL NONLINEARITY IN ℝ3

  • In the present paper, we are concerned with the existence of ground state sign-changing solutions for the following Schrödinger-Poisson-Kirchhoff system $$\;\{\begin{array}{lll}-(1+b{\normalsize\displaystyle\smashmargin{2}{\int\nolimits_{{\mathbb{R}}^3}}}{\mid}{\nabla}u{\mid}^2dx){\Delta}u+V(x)u+k(x){\phi}u={\lambda}f(x)u+{\mid}u{\mid}^4u,&&\text{in }{\mathbb{R}}^3,\\-{\Delta}{\phi}=k(x)u^2,&&\text{in }{\mathbb{R}}^3,\end{array}$$ where b > 0, V (x), k(x) and f(x) are positive continuous smooth functions; 0 < λ < λ1 and λ1 is the first eigenvalue of the problem -∆u + V(x)u = λf(x)u in H. With the help of the constraint variational method, we obtain that the Schrödinger-Poisson-Kirchhoff type system possesses at least one ground state sign-changing solution for all b > 0 and 0 < λ < λ1. Moreover, we prove that its energy is strictly larger than twice that of the ground state solutions of Nehari type.

THE EXISTENCE OF TWO POSITIVE SOLUTIONS FOR $m$-POINT BOUNDARY VALUE PROBLEM WITH SIGN CHANGING NONLINEARITY

  • In this paper, the existence theorem of two positive solutions is established for nonlinear m-point boundary value problem by using an inequality for the following third-order differential equations $$({\phi}(u^{\prime\prime}))^{\prime}+a(t)f(t,u(t))=0,\;t{\in}(0,1)$$, $${\phi}(u^{\prime\prime}(0))=\sum^{m-2}_{i=1}a_i{\phi}(u^{\prime\prime}({\xi}_i)),\;u^{\prime}(1)=0,\;u(0)=\sum^{m-2}_{i=1}b_iu({\xi}_i)$$, where ${\phi}:R{\rightarrow}R$ is an increasing homeomorphism and homomorphism and $\phi(0)=0$. The nonlinear term f may change sign, as an application, an example to demonstrate our results is given.

  • The purpose of this study is to understand the feature and phenomena of women's costume in Choson dynasty by the approach of semiotics. In order to do so, F Saussure's linguistic symbol theory, C. Peirce's conception on semiology theory and cultural semiology of R. Barthes ware used as tool to analyse traditional women's costumes in $Chos\v{o}n$ dynasty. And I choose basic conceptions based on their semiotic theory. which were langue & parole. dennotation & connotation, paradigm & syntagm, code, and analysed. structure of sign, communication though the non-language. The fashion of the $Chos\v{o}n$ dynasty contains various meaning as sign symbol system and makes the communication possible as it is mentioned above. We nay understand the sign symbol system described in the fashion sign by analysing the structure and meaning operation of sign on the basis of social, political, and idealistic background of the times, to understand the polysemy quality of the fashion.

A Nonlinear Elliptic Equation of Emden Fowler Type with Convection Term

  • In this paper we give conditions for the existence of, and describe the asymtotic behavior of, radial positive solutions of the nonlinear elliptic equation of Emden-Fowler type with convection term ∆p u + 𝛼|u|q-1u + 𝛽x.∇(|u|q-1u) = 0 for x ∈ ℝN, where p > 2, q > 1, N ≥ 1, 𝛼 > 0, 𝛽 > 0 and ∆p is the p-Laplacian operator. In particular, we determine ${\lim}_{r{\rightarrow}}{\infty}\,r^{\frac{p}{q+1-p}}\,u(r)$ when $\frac{{\alpha}}{{\beta}}$ > N > p and $q\,{\geq}\,{\frac{N(p-1)+p}{N-p}}$.

지향각, 시선각 정보를 이용한 이동표적의 충돌각 제어 (Impact Angle Control for Non-maneuvering Target with Look Angle Measurements and Line of Sight)

  • 본 논문은 이동표적에 대해서 고정형 탐색기(Strapdown Seeker)가 장착된 유도탄의 지향각 제한을 고려하여 충돌각(Impact Angle)을 제어하는 유도법칙을 제안하고 있다. 제안한 유도법칙은 슬라이딩 모드를 기반으로 하고 있으며, 탐색기에서 제공하는 지향각과 항법 정보를 통해 얻을 수 있는 시선각 정보와 발사 초기 표적 획득장비 등을 통해 제공 받는 표적 속도와 표적 이동경로각을 이용하여 가속도 명령을 생성한다. 제안한 슬라이딩 surface의 수렴을 통해 표적 요격과 충돌각 제어가 가능하다. 또한, 최대, 최소 지향각에서 지향각의 미분결과의 부호가 지향각의 부호와 반대가 된다는 것을 보임으로써 지향각 제한을 넘지 않는다는 것을 확인할 수 있다.

POSITIVE SOLUTIONS OF NONLINEAR m-POINT BVP FOR AN INCREASING HOMEOMORPHISM AND POSITIVE HOMOMORPHISM ON TIME SCALES

  • In this paper, by using fixed point theorems in cones, the existence of positive solutions is considered for nonlinear m-point boundary value problem for the following second-order dynamic equations on time scales $({\phi}(u^{\Delta}))^{\nabla}+a(t)f(t,\;u(t))=0$, t $\in$ (0, T), $u(0)=\sum\limits^{m-2}_{i=1}a_iu(\xi_i)$, $\phi(u^{\Delta}(T))=\sum\limits^{m-2}_{i=1}b_i{\phi}(u^{\Delta}(\xi_i))$, where $\phi$ : R $\rightarrow$ R is an increasing homeomorphism and positive homomorphism and ${\phi}(0)=0$. In [27], we obtained the existence results of the above problem for an increasing homeomorphism and positive homomorphism with sign changing nonlinearity. The purpose of this paper is to supplement with a result in the case when the nonlinear term f is nonnegative, and the most point we must point out for readers is that there is only the p-Laplacian case for increasing homeomorphism and positive homomorphism due to the sign restriction. As an application, one example to demonstrate our results are given.
