p13 from group II baculoviruses is a killing-associated gene

  • p13 gene was first described in Leucania separata multinuclear polyhedrosis virus (Ls-p13) several years ago, but the function of P13 protein has not been experimentally investigated to date. In this article, we indicated that the expression of p13 from Heliothis armigera single nucleocapsid nucleopolyhedrovirus (Ha-p13) was regulated by both early and late promoter. Luciferase assay demonstrated that the activity of Ha-p13 promoter with hr4 enhancer was more than 100 times in heterologous Sf9 cells than that in nature host Hz-AM1 cells. Both Ls-P13 and Ha-P13 are transmembrane proteins. Confocal microscopic analysis showed that both mainly located in the cytoplasm membrane at 48 h. Results of RNA interference indicated that Ha-p13 was a killing-associated gene for host insects H. armigera. The AcMNPV acquired the mentioned killing activity and markedly accelerate the killing rate when expressing Ls-p13. In conclusion, p13 is a killing associated gene in both homologous and heterologous nucleopolyhedrovirus.

Health-Related Quality of Life among Women Caregivers of Older Adults with Dementia (치매노인을 부양하는 여성의 건강관련 삶의 질)

  • Purpose: The objective of this study was to define the predictors of health related quality of life(HRQoL) of women caregivers of the demented elderly patients. Methods: The subjects of this study were 140 pairs of patients with dementia and their caregivers. The characteristics of dementia patients and caregivers, Barthel index and SF-36 were measured in this study. T-test, ANOVA, Pearson correlation coefficient, and stepwise multiple regression were used for data analysis. Results: The health related quality of life(HRQoL) score of women caregivers was $288.35{\pm}66.10$ for norm based scoring. HRQoL of women caregivers was correlated with patients' ADL, severity of dementia, caregivers' age, burden, and family support. The major factors that affected the physical components of women caregivers of patients with dementia was the age of the caregiver, burden, and ADL which explained 36.0% of HRQoL. The main predictors of women caregivers' emotional state was the caregiver's burden. Conclusion: Patients' support systems must be implemented to improve the physical HRQoL of caregivers. A caregivers' burden relief program needs to be prepared to increase their emotional HRQoL and further studies and efforts will be needed to evaluate those effects.

  • Measurement of quality of life among prostate cancer patients helps the health care providers to understand the impact of the disease in the patients' own perspective. The main aim of this study is to measure the quality of life among prostate cancer patients at University Malaya Medical Center (UMMC) and Universiti Kebangsaan Malaysia Medical Centre (UKMMC) and to ascertain the association factors for physical coefficient summary (PCS) and mental coefficient summary (MCS). A hospital based, cross sectional study using the Short Form-36 (SF-36) questionnaire was conducted over a period of 6 months. A total of 193 respondents were recruited. Their total quality of life score was $70.1{\pm}14.7$ and the PCS score was lower compared to MCS. The factors associated for PCS were: age, living partner, renal problem, urinary problem of intermittency, dysuria and hematuria. Factors associated for MCS were: age, living partner, renal problem, presenting prostatic specific antigen and urinary problem of intermittency and dysuria. Our prostate cancer patients had moderate quality of life in the physical health components but their mental health was less affected.

Quantification of Total Mercury in Antarctic Surface Snow using ICP-SF-MS: Spatial Variation from the Coast to Dome Fuji

  • The total mercury concentration ($Hg_T$) of surface snow samples collected along a ~1500 km transect in east Queen Maud Land was determined using inductively coupled plasma sector field mass spectrometry to address the behavior of Hg on the Antarctic Plateau. Due to the volatile nature of mercury, measures were taken against Hg loss from standard solutions by choosing appropriate container material and stabilizing agents. Glass bottles with Teflon-lined caps were superior to Teflon and polyethylene containers in protecting against Hg loss, but addition of gold chloride ($AuCl_3$) or bromine chloride (BrCl) was necessary to ensure preservation of Hg. As Hg loss was also observed in snowmelt samples, our analysis may underestimate the actual amount of HgT in the snow. Even so, the measured HgT was still very low (< 0.4-10.8 pg $g^{-1}$, n = 44) without a signal of depositional enhancement accompanying photo-oxidation of atmospheric elemental mercury in austral midsummer. Moreover, the dynamic variation along the traverse implies spatial and temporal heterogeneity in its source processes.

Estimation of Mass Discrimination Factor for a Wide Range of m/z by Argon Artificial Isotope Mixtures and NF3 Gas

  • Absolute isotope ratio is a critical constituent in determination of atomic weight. To measure the absolute isotope ratio using a mass spectrometer, mass discrimination factor, $f_{MD}$, is needed to convert measured isotope ratio to real isotope ratio of gas molecules. If the $f_{MD}$ could be predicted, absolute isotope ratio of a chemical species would be measureable in absence of its enriched isotope pure materials or isotope references. This work employed gravimetrically prepared isotope mixtures of argon (Ar) to obtain $f_{MD}$ at m/z of 40 in the magnetic sector type gas mass spectrometer (gas/MS). Besides, we compare the nitrogen isotope ratio of nitrogen trifluoride ($NF_3$) with that of nitrogen molecule ($N_2$) decomposed from the same $NF_3$ thermally in order to identify the difference of $f_{MD}$ values in extensive m/z region from 28 to 71. Our result shows that $f_{MD}$ at m/z 40 was $-0.044%{\pm}0.017%$ (k = 1) from measurement of Ar artificial isotope mixtures. The $f_{MD}$ difference in the range of m/z from 28 to 71 is observed $-0.12%{\pm}0.14%$ from $NF_3$ and $N_2$. From combination of this work and reported $f_{MD}$ values by another team, IRMM, if $f_{MD}$ of $-0.16%{\pm}0.14%$ is applied to isotope ratio measurement from $N_2$ to $SF_6$, we can determine absolute isotope ratio within relative uncertainty of 0.2 %.

Characterization of Recombinant Baculovirus Ewpressing Polyhedrin Gene of Bombyx mori and Autographa californica Nuclear Polyhedrois Virus (두 종류의 다각체 단백질 유전자를 발현하는 유전자 재조합 핵다각체병 바이러스의 특성)

  • Recombinant baculoviruses having expanded host range were selected by coinfection of Autographa california NPV and Bombyx mori NPV into Sf-9 and BmN-4 insect cell lines. In order to determine the polyhedra morhplogy of RecS-A6, one of a recombinant baculovirus, polyhedra of RecS-A6 produced in insect cells were observed by phase contrast microscope and scanning electron microscope. The results revealed that the recombinant baculovirus had a various polyhedra morphology which was different from its parental viruses, suggesting that the various morhpology of recombinant baculovirus with an expanded host range was due to the genetic recombination of viral genome. To analyze the genomic recombinantion of the recombinant baculoviruses, genomic DNAs of two parent viruses and RecS-A6 were digested with restriction endonuclease and subjected to agarose gel electrophoresis. Southern blot analysis revealed that RecS-A6 has two polyhedrin gene of AcNPV and BmNPV in a viral genome. Polyhedral protein of recombinant baculovirus was analysed by SDS-PAGE. The result showed that molecular weight of polyhedral protein of RecS-A6 containing two polyhedrin gene of AcNPV and BmNPV was as the 31 kDa band of AcNPV and 30 kDa band of BmNPV.

  • Process characterization of the chemical mechanical polishing (CMP) process for undensified phosphosilicate glass (PSG) film is reported using design of experiments (DOE). DOE has been addressed to experimenters to understand the relationship between input variables and responses of interest in a simple and efficient way. It is typically beneficial for determining the adequate size of experiments with multiple process variables and making statistical inferences for the responses of interests. Equipment controllable parameters to operate the machine include the down force (DF) of the wafer carrier, pressure on the backside of the wafer, table and spindle speed (SS), slurry flow rate, and pad condition. None of them is independent; thus, the interaction between parameters also needs to be indicated to improve process characterization in CMP. In this paper, we have selected the five controllable equipment parameters, such as DF, back pressure (BP), table speed (TS), SS, and slurry flow (SF), most process engineers recommend to characterize the CMP process with respect to material removal rate (RR) and film uniformity as a percentage. The polished material is undensified PSG. PSG is widely used for the plananization in multi-layered metal interconnects. We identify the main effect of DF, BP, and TS on both RR and film uniformity, as expected, by the statistical modeling and analysis on the metrology data acquired from a series of $2^{5-1}$ fractional factorial design with two center points. This revealed the film uniformity of the polished PSG film contains two and three-way interactions. Therefore, one can easily infer that the process control based on better understanding of the process is the key to success in semiconductor manufacturing, typically when the wafer size reaches 300 mm and is continuously scheduled to expand up to 450 mm in or little after 2012.

On the prediction of unconfined compressive strength of silty soil stabilized with bottom ash, jute and steel fibers via artificial intelligence

  • The determination of the mixture parameters of stabilization has become a great concern in geotechnical applications. This paper presents an effort about the application of artificial intelligence (AI) techniques including radial basis neural network (RBNN), multi-layer perceptrons (MLP), generalized regression neural network (GRNN) and adaptive neuro-fuzzy inference system (ANFIS) in order to predict the unconfined compressive strength (UCS) of silty soil stabilized with bottom ash (BA), jute fiber (JF) and steel fiber (SF) under different freeze-thaw cycles (FTC). The dosages of the stabilizers and number of freeze-thaw cycles were employed as input (predictor) variables and the UCS values as output variable. For understanding the dominant parameter of the predictor variables on the UCS of stabilized soil, a sensitivity analysis has also been performed. The performance measures of root mean square error (RMSE), mean absolute error (MAE) and determination coefficient ($R^2$) were used for the evaluations of the prediction accuracy and applicability of the employed models. The results indicate that the predictions due to all AI techniques employed are significantly correlated with the measured UCS ($p{\leq}0.05$). They also perform better predictions than nonlinear regression (NLR) in terms of the performance measures. It is found from the model performances that RBNN approach within AI techniques yields the highest satisfactory results (RMSE = 55.4 kPa, MAE = 45.1 kPa, and $R^2=0.988$). The sensitivity analysis demonstrates that the JF inclusion within the input predictors is the most effective parameter on the UCS responses, followed by FTC.

Immobilization of ATP on Bovine $\beta$- Caseins by Using Transglutaminase (효소법에 의한 ATP의 Bovine $\beta$-Casein에의 고정화)

  • ATP analogs were immobilized or bovine caseins by the action of transglutaminase. The ATP analogs immobilized on the caseins were enzymatically active and interconverten by kinases. The immobilized ATP was dephosphorylated to the corresponding ADP by hexokinase and rephosphorylated to the ATP in solid form by acetate kinase. Under the conditions chosen, about 55% of the immobilized ATP was dephosphorylated and about 80% of the resulted ADP was rephosphorylated. Bovine $\beta$-casein was more useful than $\alpha$sf-casein as a carrier and C8-substituted ATP analognwas more effective than N6-substituted one in immobilization. Michaelis constant of C8-substituted ATP analog immobilized on $\beta$-casein was similar to that of free form of ATP and that of ATP analog. The immobilized ATP was much more stable than free ATP and its analog, while maximum velocity was reduced to 37% of the free ATP analog. The immobilized ATP was recovered almost completely by calcium precipitation.

Kinematic Analysis of a Scoop Motion in Elite Male Hockey Players (남자 우수하키 선수들의 스쿱 동작에 관한 운동학적 분석)

  • The purpose of this study was to investigate the hokey scoop motion of elite male hockey players. To accomplish the goal of this study, eight male hockey players participated and were divided into two groups (superior group Vs. inferior group). To find differences between groups, a three-dimensional motion analysis was performed with seven infrared cameras (SF: 200Hz). After analyzed their scoop motion, followings were found. 1) The non-significant(p>.05) increase in anterior CG displacement and velocities were found in superior group compare with inferior group) 2) There were no significance found in anterior-posterior stick velocities between groups. However, significant (p<.05) increase in vertical stick velocities were found in superior group than inferior group indicating the superior group has more skilled in scooping. 3) The significant(p<.05) increase in adductional and internal rotational stick released velocities were found in superior group than inferior group.
