Synthesis of Magneli Phases and Application to the Photoelectrochemical Electrode (마그넬리상 합성과 광전기화학셀 전극 응용)

  • Hydrothermal synthesis of highly crystalline $TiO_2$ nanorods is a well-developed technique and the nanorods have been widely used as the template for growth of various core-shell nanorod structures. Magneli/CdS core-shell nanorod structures are fabricated for the photoelectrochemical cell (PEC) electrode to achieve enhanced carrier transport along the metallic magneli phase nanorod template. However, the long and thin $TiO_2$ nanorods may form a high resistance path to the electrons transferred from the CdS layer. $TiO_2$ nanorods synthesized are reduced to magneli phases, $TixO_{2x-1}$, by heat treatment in a hydrogen environment. Two types of magneli phase nanorods of $Ti_4O_7$ and $Ti_3O_5$ are synthesized. Structural morphology and X-ray diffraction analyses are carried out. CdS nano-films are deposited on the magneli nanorods for the main light absorption layer to form a photoanode, and the PEC performance is measured under simulated sunlight irradiation and compared with the conventional $TiO_2/CdS$ core-shell nanorod electrode. A higher photocurrent is observed from the stand-alone $Ti_3O_5/CdS$ core-shell nanorod structure in which the nanorods are grown on both sides of the seed layer.

  • Microscopic structures of epidermis and palynology and isozyme analysis were examined to find out the intergeneric and interspecific relationships and consequently to confirm the position of Hanabusaya aiatica as an endemic genus among 4 genera and 9 species belonging to the Hanabusaya asiatica, Korean endemic, and its allied groups. In the examination of microscopic structures of epidermis, cell patterns of perianth and ultrastructure of seed coat were found to be useful characters for the identification of the 4 genera and 9 species. Palynological characters such as microscopic structure and overall morphology of pollen grains were, however, not enough to distinguish them because of the great variabilities in these traits. Isozyme analysis showed that H. asiatica was very closly related to Campanula punctata and C. takesimana, though there were variations among populations and collected areas in some classified groups, depending on classified groups. Based on these results, the position of H. asiatica as an endemic genus was well confirmed.

Effect of Colchicine on Chromosome Doubling in Codonopsis lanceolata

  • The present study was performed to investigate the effects of the colchicine concentrations on chromosome doubling for producing of tetraploid plants of Codonopsis lanceolata, and its effect on plant morphology. A total of 180 individuals germinated from 16 treatment groups, were exposed to various concentrations (0.05-1.0% w/v) of colchicine for different soaking duration (3-24 hour). The highest numbers of tetraploid plants (3) were observed from the lowest concentration of colchicine (0.05%), and one (1) tetraploid plant was obtained from the 0.5% concentration group with a 6 hour treatment. However, no tetraploid individual was observed in any other treatment groups. The plant height of the diploid (18.1 ㎝) was slightly shorter than that of the tetraploid (13.4 ㎝). The fresh weight of the main root in the diploid (0.5 g) was four-fold higher than the tetraploid (2.2 g). The colchicine-treated plant regeneration rate in C. lanceolata was decreased when the plants were subjected to high concentration of colchicine. In particular, the highest number of tetraploid plants (5 and 3) was obtained from the lower concentration (0.05% and 0.1%) of colchicine for 6-hour treatment, which were a higher rate (29.4% and 30%) of regenerated tetraploid plants than other regenerated plants. As in the seed treatment result, the plant height of the diploid was significantly higher (10.4 ㎝) than tetraploid. The higher morphological changes were observed comparatively from tetraploid plants than the diploid.

Comparison of the morphology and distribution of the genus Megaleranthis Ohwi with those of its relative genera (Ranunculaceae) (모데미풀속과 근연속들의 형태 및 분포 비교 (미나리아재비과))

  • To elucidate the taxonomic status of the genus Megaleranthis Ohwi, a monotype in Korea, its distribution and morphological characteristics were studied and a cluster analysis was performed. The cluster analysis showed that Megaleranthis was separately clustered from Trollius and Calathodes. Megaleranthis is morphologically different from Calathodes Hook.f. et Thomson in having petals, and from Trollius L. in having an involucre instead of a caulescent leaf under the flower and follicles with simple veins. The three genera above are distributed independently within Asia, although they can be found together at the boundaries where their distributions overlap: Trollius and Calathodes occur together between the Sichuan and Yunnan provinces in China, and Megaleranthis and Trollius can both be found on the northern edge of the Korean Peninsula. From both a morphological and a distributional viewpoint, Megaleranthis should be recognized as an independent genus different from both Trollius and Calathodes.

Physiological Character of Juvenility in Higher Plant (고등식물체에서 유년기의 생리적 특성)

  • Common usage of the concept of juvenility implies that there is one physiological phase, the juvenile phase, which manifests itself in the various morphological and physiological phenomena observed in juvenile higher plants. The juvenile phase is often defined as that time from seed germination until the plant attains the ability to flower regulating such behaviour. This definition precludes plants from flowering in the juvenile phase. It is of major interest, therefore, to identify the physiological controls(Bluehreife) regulating such behavior. The length of the juvenile period in higher plants ranges from one year to over 60 years in different species. The long juvenile period of seedling is the main cause of the long duration of the breeding process. I determined the length of the juvenile period in various plants and its control of phase changes in natural system in relation to factors such as plant size and age, shoot morphology, apex size, root system and phytohormonal and nutritional status is reviewed. From the own experimental and observational evidence available it appears that both hormonal and nutritional factors can be involved in control of juvenility but that a specific juvenile or flowering hormone is not involved. Grafting, ringing, scoring, root pruning and fertilization have been used to accelerate flowering, but in most cases these cultured treatments are only successful on plants that were passed the juvenile phase. It is suggested that there are intrinsic difference between the meristematic cells of the apieces of juvenile and adult shoot, which are thus determined with respect to there development potentialities. The problems associated with the maintenance of the determined state through mitosis are discussed. The properties of transitional forms of Ribes nigrum L. intermediate between the juvenile and adult phase, are descrived and there implications discussed. Analogies are drawn between juvenile phenomena in woody perennials and in herbaceous species.

Variation of Tocopherol Composition and Morphology in Soybean [Glycine max (L.) Merr.] Germplasms

  • In order to get information on tocopherol content and composition in fifty-six soybean germplasms were evaluated by HPLC. From the principle component analysis, the first three components accounted for 71.6% of the total variance of tocopherol content in the germplasms. Principal component 1 showed significant correlations with all the morphological markers except 100-seed weight. Soybean germplasms were divided into three groups by the first two principal components. The highest content of ${\alpha}$-tocopherol was $38.6{\mu}g$ per g in Tanner, while that of ${\gamma}$-tocopherol was $195.6{\mu}g$ in PI 91073. In case of ${\delta}$-tocopherol, IT 105622 showed the highest value as $29.8{\mu}g$. The contents of tocopherol were gradually increased from the late August to late September, which was 10 days before maturity, in Alchankong and PI 96322. The tocopherol content was higher in seeds from the plants sown early than those sown late. Total tocopherol contents of PI 96188, Geomjeongkong 2, and Suwon 183 grown in Gimje were higher than those grown in Iksan, but the difference was not observed in PI 96322. These results suggest that the contents of tocopherol in soybean were affected not only by the genotypes but also by environment.

Highly Photocatalytic Performance of flexible 3 Dimensional (3D) ZnO nanocomposite

  • Zinc oxide (ZnO) is one of the most powerful materials for purifying organic pollutants using photocatalytic activity. In this study, we have introduced a novel method to design highly photoreactive flexible 3 dimensional (3D) ZnO nanocomposite [F-ZnO-m (m: reaction time, min)] by electrospinning and simple-step ZnO growth processing (one-step ZnO seed coating/growth processing). Significantly, the F-ZnO-m could be a new platform (or candidate) as a photocatalytic technology for both morphology control and largearea production. The highest photocatalytic degradation rate ([k]) was observed for F-ZnO-m at 2.552 h-1, which was 8.1 times higher than that of ZnO nanoparticles (NPs; [k] = 0.316 h-1). The enhanced photocatalytic activity of F-ZnO-m may be attributed to factors such as large surface area. The F-ZnO-m is highly recyclable and retained 98.6% of the initial decolorization rate after fifteen cycles. Interestingly, the F-ZnO-m samples show very strong antibacterial properties against both Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) after exposure to UV-light for 30 min. The antibacterial properties of F-ZnO-m samples are more effective than those of ZnO NPs. More than 96.6% of the E. coli is sterilized after ten cycles. These results indicate that F-ZnO-m samples might have utility in several promising applications such as highly efficient water/air treatment and inactivation of pathogenic microorganisms.

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed.   • The differentiation process of male germ cells and sperm morphology of the abalone Haliotis discus hannai were described in ultrastructure. The differentiation process of sperm was divided into four stages: spermatogonium, spermatocyte, spermatid and sperm. The process of differentiation from spermatogonium to spermatocyte did not show significant morphological changes. However, during the spermiogenesis there were distinct morphological changes such as chromatin condensation, morphological changes of the nucleus, and formation of acrosome, midpiece and flagellum. The sperm of the abalone consisted of head, midpiece and tail. The head of approximately 5.3 ㎛ in length was composed of a nucleus of high electron dense and bullet-shaped acrosome. The midpiece was composed of the basal body and mitochondria, and five mitochondria were arranged in single layer around the basal body. The cross section of the tail showed a "9+2" axonemal structure. These morphological and structural features are the result of showing that the sperm of H. discus hannai is a primitive type.

Characterization of a New High-lysine Mutant in Barley (Hordeum vulgare L.)

  • A chemical, MNU-induced hulless barley mutant line designated as 'Mutant 98 (M98)' was developed from a Korean hulless waxy barley cultivar, 'Chalssalbori'. The objective of the study was to determine the genetic basis of 'M98' and the possibility of using 'M98' as breeding parent to improve lysine level. Compared to 'Chalssalbori', 'M98' had large embryo and higher lysine content in both the embryo and endosperm. Significantly different lysine content in 'M98' and the other high-lysine barley mutant stocks was observed for two years. However, the genotype by year interaction was not significant. 'M98' was higher than the other high-lysine barley mutant stocks in the percentage of lysine of total amino acid composition (0.75%). The trait of shrunken endosperm of 'M98', which was typical in the high-lysine mutants, was inherited by a single recessive gene. Based on seed morphology and lysine content of $F_1$ seeds, 'M98' had a genetically different gene from the other high-lysine mutants for shrunken endosperm. Segregation of $F_2$ for plump/shrunken endosperm did not fit the expected ratio of Mendelian inheritance except for only one cross combination (GSHO1784 (lys1)/M98). The amino acid analysis of $F_5$ and $F_6$ progenies from the cross between 'M98' and 'Chalssalbori' revealed that the attempt to increase the range of lysine content of plump lines did not go beyond the limit of the average high-lysine barley germplasm.
