Establishment of Incubational Conditions for Rat Testicular Cells (랫드 고환세포의 배양조건 설정에 관한 연구)

  • This study of culturing testicular cell types in vitro has potential to be an invaluable tool for assessing the mechanisms of testicular toxicity, especially those of intragonadal interaction and spermatogenesis. Combined with the Sertoli/germ cell cultures, Leydig cells provide comprehensive and detailed information on the action of testicular toxicants at the level of the testis. Sertoli/germ cell were isolated and incubated well in vitro from 20~30 g rats and Leydig cells from 250~300 g rats. The Sertoli cells isolated from the testis of the SD rats grew into monolayer on about the 2nd~3rd day of culture, an appreciable cell increment being observed between the 4th~5th day. The Leydig cells isolated from the testis of the SD rats grew into a monolayer on about the 3rd-4th day of culture, an appreciable cell increment being observed between the 5th-7th day. These results suggest that Sertoli and Leydig cells can be cultured as a male fertility evaluation method alternative to the in vivo/conventional fertility test method and further study for the physio-chemical determination is needed.

Case Series of Advanced Non-small Cell Lung Cancer Patients Treated with Hang-Am-Plus (항암플러스 투여로 호전된 진행성 비소세포성 폐암 연속환자증례)

  • Objective : To investigate the therapeutic effects of Hang-Am-Plus (HAP) on advanced non-small cell lung cancer (NSCLC) patients. Method : We prescribed HAP three times a day (3,000-6,000 mg/ day) during the treatment period (8 - 24 months). Computed tomography (CT) was performed to evaluate the therapeutic efficacy. Results : Four patients who were diagnosed with NSCLC were recommended chemotherap y, but refused it because of old age, side effects, or treatment failure, and instead sought oriental medicinal therapy. They were treated around 12 months. The patients showed stable disease (SD) state for 5 months, 3 months, 19 months and 3 months, respectively. Conclusion : These four cases may give us the possibility that HAP offers potential benefits for non-small cell lung cancer patients.

  • These studies were conducted to investigate the antiinflammatory effects and the mechanism of action of natural products. We used the methods of "carrageenan induced foot edema" , "PAF (platelet activating factor) induced foot edema" , "inhibition test of vascular permeability" , "inhibition test of white blood cell migration" , "formation of granuloma" and "adjuvant induced arthritis" to examine the antiinflammatory erects of Angelica gigas, Ledebouriella seseloides, Ginkgo biloba and Bamboo salt (Jukyom). The oral administration of the water extract of Angelica gigantis radix, the methanolic extract of Ginkgo folium and the aqueous solution of Bamboo salt showed antiinflammatory effect on carrageenan and PAF induced foot edema in SD rat at a dose of 1 g/kg. The same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in mice. The aqueous solution of Bamboo salt inhibited the formation of ganuloma in SD rats at a oral dose of 1 g/kg. Angelica gigantis radix seems to have antiinflammatory effect by inhibition of PAF.

Blood Picture of Race Horses in Korea (경주마(競走馬)의 혈액상(血液像)에 관(關)하여)

  • The blood picture of 85 healthy race horses in Korea was investigated. The ranges and mean values of erythrocyte, hemoglobin, hematocrit value, mean corpuscular volume, mean corpuscular hemoglobin concentration, and total white blood cell count in the blood picture were determine. The respective mean value and standard deviation and age differences were as follows: 1. The erythrobyte count was shown as range of 6.20 to $11.32{\times}10^6/mm^3$ with mean of $8.61{\pm}1.92{\times}10^6/mm^3$(SD). The leucocyte count was shown as range 5.0 to $18.0{\times}10^3/mm^3$ with mean of $8.25{\pm}1.51{\times}10^3/mm^3$(SD). There were not significant. differences in age, 2. The mean value of hemoglobin was shown $13.9{\pm}1.7g/100ml(SD)$ ranging 9.8 to 16.8g/100ml. The mean value of hematocrit was shown $40.9{\pm}3.94ml/100ml(SD)$ ranging 26 to 54. There were not significant differences in age. 3. The mean corpuscular hemoglobin was shown as range of 11.8 to 22.2pg with mean of $16.9{\pm}4.69$(SD). The mean corpuscular volume was shown as range of 34.5 to $71.3cu{\mu}$ with mean of $49.0{\pm}7.32cu{\mu}$(SD). The mean corpuscular hemoglobin concentration was shown as range of 30.6 to 39.4 g/100 ml with mean of $34.6{\pm}2.36$(SD). There were not significant differences in age. 4. The correlation among erythrocyte count, hemoglobin and hematocrit value were observed as follows: Erythrocyte count and hemoglobin (+0.328), rythrocyte count and hematocrit vague (+0.319). A linear regression equation was shown as follows: Erythrocyte count and hemoglobin (Y=0.336x+10.977), erythrocyte count and hematocrit value (Y=0.655x+35.274). 5. The high correlation between hemoglobin and hematocrit vague was observed (r= +0.836). A linear regression equation was shown: (Y=1.948x+13.895).

High Prevalence of Listeria monocytogenes in Smoked Duck: Antibiotic and Heat Resistance, Virulence, and Genetics of the Isolates

  • This study aimed at determining the genetic and virulence characteristics of the Listeria monocytogenes from smoked ducks. L. monocytogenes was isolated by plating, and the isolated colonies were identified by PCR. All the obtained seven L. monocytogenes isolates possessed the virulence genes (inlA, inlB, plcB, and hlyA) and a 385 bp actA amplicon. The L. monocytogenes isolates (SMFM2018 SD 1-1, SMFM 2018 SD 4-1, SMFM 2018 SD 4-2, SMFM 2018 SD 5-2, SMFM 2018 SD 5-3, SMFM 2018 SD 6-2, and SMFM 2018 SD 7-1) were inoculated in tryptic soy broth (TSB) containing 0.6% yeast extract at 60℃, followed by cell counting on tryptic soy agar (TSA) containing 0.6% yeast extract at 0, 2, 5, 8, and 10 min. We identified five heat resistant isolates compared to the standard strain (L. monocytogenes ATCC13932), among which three exhibited the serotype 1/2b and D-values of 5.41, 6.48, and 6.71, respectively at 60℃. The optical densities of the cultures were regulated to a 0.5 McFarland standard to assess resistance against nine antibiotics after an incubation at 30℃ for 24 h. All isolates were penicillin G resistant, possessing the virulence genes (inlA, inlB, plcB, and hlyA) and the 385-bp actA amplicon, moreover, three isolates showed clindamycin resistance. In conclusion, this study allowed us to characterize L. monocytogenes isolates from smoked ducks, exhibiting clindamycin and penicillin G resistance, along with the 385-bp actA amplicon, representing higher invasion efficiency than the 268-bp actA, and the higher heat resistance serotype 1/2b.

Anticancer Effects of Black Soybean Doenjang in HT-29 Human Colon Cancer Cells (HT-29 인체 대장암 세포에서 검정콩 된장의 in vitro 항암 효과)

  • In vitro anticancer effects of black soybean doenjang on HT-29 human colon cancer cells were studied. SD (soybean doenjang prepared with nine-time baked bamboo salt) and BD (black soybean doenjang prepared with nine-time baked bamboo salt) were compared with CD (commercial doenjang). There were no significant differences between experimental groups in terms of pH, amino-type nitrogen, and ammonia-type nitrogen levels of the doenjang samples. BD showed the highest antioxidative effect, followed by SD and CD in that order. BD also showed the highest total polyphenol concentration of all samples. CD, SD, and BD extracts showed no toxic effects on normal RAW 264.7 cells at a concentration ranging from 0.1 to 0.5 mg/mL. BD exhibited anticancer effect on HT-29 cells by MTT assay. Also, BD manipulated mRNA expressions in certain factors; it suppressed pro-inflammatory cytokines such as $TNF-{\alpha}$, IL-6, and COX-2, promoted cell-cycle-related genes of p21, and p53, suppressed expression of cyclin D1, and suppressed anti-apoptotic Bcl-2; such manipulation by BD was the strongest, followed by SD and CD in order. From the results above, BD exhibited the highest anticancer effects by inhibiting growth of HT-29 cells, probably by regulating pro-inflammatory cytokines, cell cycling related genes, etc. These results might be due to using black soybeans containing high levels of polyphenol, including anthocyanins.

  • Stem cells have been the subject of increasing scientific interest because of their utility in numerous biomedical applications. Stem cells are capable of renewing themselves; that is, they can be continuously cultured in an undifferentiated state, giving rise to more specialized cells of the human body. Therefore, stem cells are an important new tools for developing unique, in vitro model systems to test drugs and chemicals and a potential to predict or anticipate toxicity in humans. In the present study, in vitro cultured F3 immortalized human neural stem cell line and in vivo adult Sprague Dawley rats was used to evaluate the cytotoxicity of anticancer drug paclitaxel. In vitro apoptotic activity of paclitaxel was evaluated in F3 cell line by a MTT assay and DAPI test. The cell death was induced with the treatment of 20 nM paclitaxel and chromatin degradation was detected by DAPI staining, which was analyzed by fluorescent microscope. In vivo studies, we also observed nestin immunoreactivity on subventricular zone, which is stem cell rich region in the adult brain of the SD rat. Immunofluorescent staining result shows that pixel intensities of nestin were decreased in a dose dependent manner. These results suggest that paclitaxel is able to induce cytotoxic activity both in F3 neural stem cell line and neural stem cell in SD rat brain.

Clinical Investigation of Efficacy of Albumin Bound Paclitaxel plus Platinum Compounds as First-line Chemotherapy for Stage III/IV Squamous Non-small Cell Lung Cancer

  • Objective: To observe the efficacy and toxicity of nanoparticle albumin bound paclitaxel (nab-paclitaxel) plus platinum agent (cisplatin or carboplatin) as first line treatment for stage III/IV squamous non-small-cell lung cancer (NSCLC). Methods: Forty chemotherapy naive patients with stage III/IV squamous NSCLC received nab-paclitaxel $125mg/m^2$ on day 1 and day 8, cisplatin $75mg/m^2$ on day 1, carboplatin area under the concentration-time curve of 5 (AUC=5) on day 1. One cycle of treatment was 3 weeks, and at least two were completed in each case. Results: Of the 40 patients who participated in the study, 25 achieved partial responses (PR), 12 reached a stage of stable disease (SD), and 3 suffered progressive disease (PD). The overall response rate (ORR) was 62.5% and the disease control rate (DCR) was 92.5%. Of the 20 patients without surgery or radiotherapy, 10 achieved PR, 7 reached a stage of SD, and 3 PD. The ORR was 50.0% and the DCR was 85.0%. The median progression-free survival time (PFS) of patients without surgery or radiotherapy was 5.0 months. Of the 20 patients receiving surgery or radiotherapy, 15 had PR and 5 p had SD, with an ORR of 75.0% and a DCR of 85.0%. Specifically, the DDP arm demonstrated a significantly higher ORR than the CBP arm (100%vs 54.5%, P<0.05). Common treatment related adverse events were myelosuppression, gastrointestinal response, baldness and neurotoxicity, most of which were grade 1 to 2. Conclusion: Nab-paclitaxel plus platinum agent (cisplatin or carboplatin) is effective as a first-line chemotheraphy for stage III/IV squamous NSCLC, and its adverse effects are tolerable.

  • Objectives : A case series was conducted to investigate the therapeutic effects of Hang-Am Plus (HAP) on the tumor response and HRQoL (Health Related Quality of Life) in advanced non-small cell lung cancer (NSCLC) patients. Methods : Three patients were given 1,000-2,000 mg of HAP, three times a day (daily total dosage of 3,000-6,000 mg/day) for 12 weeks. Results : After the 3 month administration with HAP, three patients showed stable disease (SD) condition according to the chest computed tomography (CT), and two of the patients reported a decrease in pain levels. Conclusions : The observed NSCLC cases suggest treatment with HAP may be related to the observed tumor growth inhibition and pain reduction.

  • e-Navigation strategy IMO promotes is defined as it is necessary to network to provide various maritime safety information to in land and on board users, and it is expected to provide a large amount and diverse kinds of maritime spatial information services to them frequently. However, as there are some limits to transmit that by current mobile maritime communication technologies, it is required to simplify and optimize the information. In this study, tree node and convex hull method is applied to S-100 SD spatial information to generalize and we arranged the efficiency and effect of generalization by storing in XML form which can be used in general.
