Cell Autonomous Circadian Systems and Their Relation to Inflammation

  • All living beings on earth have an important mechanism of 24-h periodicity, which controls their physiology, metabolism, and behavior. In humans, 24-h periodicity is regulated by the superchiasmatic nucleus (SCN) through external and environmental cues. Peripheral organs demonstrate circadian rhythms and circadian clock functions, and these are also observed in cultured cell lines. Every cell contains a CLOCK: BMAL1 loop for the generation of circadian rhythms. In this review, we focused on cell autonomous circadian rhythms in immune cells, the inflammatory diseases caused by disruption of circadian rhythms in hormones, and the role of clock genes in inflammatory diseases.

Syntheses and Spectroscopic properties of Palladium(II) Complexes with Bidentate Aminophosphine of N,N-Dialkyl-N'-diphenylphosphinodiaminoethane

  • Several new palladium(II) complexes of aminophosphines, $(Pd(L)X_2)$, (L = $Ph_2PNH$ ++ $NR_2$; R = $CH_3(L_1)$, $C_2H_5(L_2)$ : X = Cl, Br, I, and NCS) that contain two different donor atoms of nitrogen and phosphorus as ${\pi}$-electron acceptor, were synthesized and characterized by conductivity measurement, ir, and UV/Vis-spectra. For the dithiocyanatopalladium(II) complexes with aminophosphines, it was confirmed that the thiocyanate group trans to phosphorus is coordinated as Pd-NCS mode and the one trans to nitrogen as Pd-SCN mode, and the aminophosphines form six-membered chelate ring. The spectra of dihalogenopalladium(II) complexes with aminophosphines show that the band maxima are shifted to the short wavelengths as the concentration is decreased.

Spread of Cyst Nematodes in Highland Chinese Cabbage Field in Gangwon-do (강원도 고랭지배추 재배지에서 씨스트선충의 분포 확산)

  • The sugar beet cyst nematode (SBCN), Heterodera schachtii first detected in Taebaek, Gangwon-do in 2011, is one of the major plant parasitic nematodes that cause economic damage to the Chinese cabbage in highland regions. In addition, the distribution of clover cyst nematode (CCN), H. trifolii was confirmed in the highland Chinese cabbage cultivated regions in 2017. In order to investigate the spread of cyst nematodes, this study has been conducted since 2013 in the highland Chinese cabbage cultivation area. In addition, in 2017, the Real-Time PCR technique with the species-specific primer was used to investigate those two cyst nematodes and the soybean cyst nematode (SCN), H. glycines which is known for its distribution in Korea, focusing on the main production regions of highland Chinese cabbage cultivation. The number of infected fields in the Chinese cabbage plantation in highland increased every year to confirm distribution in Taebaek, Samcheok, Jeongseon and Gangneung in 2017, and the cumulative number of infection fields reached 245 by 2017. Of the 41 possible cyst nematode samples for PCR analysis, 61% were CCN, only 9.8% of the SBCN and 29.3% of the SCN were identified. Therefore, some of the previously known SBCN or CCN discoveries are likely to have been infected with SCN. It is believed that the CCN needs to be controlled in the future as CCN have been found to be dominant species in the highland Chinese cabbage plantation regions.

Hydrogen ion-selective membrane electrodes based on tetrabenzylalkylenediamine (Tetrabenzylalkylenediamine을 이용한 수소이온 선택성 막전극)

  • This is on hydrogen ion-selective memebrane electrodes which were made of tetrabenzylmethylenediamine (TBMDA), tetrabenzylethylenediamine (TBEDA), tetrabenzylpropylenediamine(TBPDA) and tetrabenzylhexylenediamine(TBHDA) as neutral carriers. Their response potentials to carbon number between amino groups showed linear selectivities to hydrogen ion in the range of pH 1~pH 9, pH 2~pH 9, pH 3~pH 9 and pH 4~pH 9 and slopes were 48mV/pH, 52mV/pH, 64mV/pH, 59mV/pH respectively. The interferences effect on the cations were measured to alkali metal ions($Li^+$, $Na^+$, $K^+$), alkaline earth metal ions ($Mg^{2+}$, $Ca^{2+}$, $Sr^{2+}$, $Ba^{2+}$), transition metals ions($Cu^{2+}$, $Ni^{2+}$, $Co^{2+}$) and anions($I^-$, $Br^-$, ${NO_3}^-$, $SCN^-$), and selectivity coefficients were measured by separate-solution method. The membrane electrode made of TBMDA among the electrodes showed the best selectivity in acidic solution.

An Arg1239His mutation of the CACNL1A3 gene in a Korean family with hypokalemic periodic paralysis (가족성 저칼륨성 주기성 마비 1예)

  • Familial hypokalemic periodic paralysis (hypoPP) is a rare inherited channelopathy that often presents with episodic weakness accompanied by hypokalemia. Thus far, mutations in the gene encoding two ion channels (CACNL1A3, L-type calcium channel alpha-1 subunit and SCN4A, a sodium channel type IV alpha subunit) have been identified. Several cases of familial hypoPP in children have been reported in Koreans, but there are only a few cases with identified mutations. We report a 12-year-old boy and his affected mother with hypoPP who has a heterozygous G to A substitution at codon 1239 in exon 30 of the CACNL1A3 gene that causes a change from arginine to histidine (Arg1239His, CACNL1A3). This mutation is common among Caucasians; however, it has not yet been reported in Koreans. The patients were treated with oral acetazolamide and potassium replacement and were instructed to avoid precipitating factors. After the medication and lifestyle modification, the paralytic attacks significantly decreased.

Perchlorate Ion-Selective PVC Membrane Electrode Based on the Quaternary Ammonium Salts (제4급 암모늄염을 이용한 과염소산 이온선택성 PVC막 전극)

  • Perchlorate ion-selective PVC membrane electrode responsive to $10^{-6}M$ was developed by incorporating the ion-pair complex of perchlorate with the quaternary ammonium salts as a active material. The effect of chemical structure, the content of active material, the kinds of plasticizers, and the membrane thickness on the electrode characteristics such as the linear response range and Nernstian slope of the electrode were studied. With the results, the useful pH range and the selectivity coefficients to various interfering anions were compared and investigated. It was obtained that the effect of the chemical structure of an active material on the electrode characteristics was improved with increasing the alkyl chain length of the quarternary ammonium salts in the ascending order of Aliquat 336P, TOAP, TDAP, and TDDAP. The electrode characteristics was improved with the decrease of the active material content below the optimum membrane composition, and DBP was the best as a plasticizer. The optimum membrane composition was 9.09wt% of TDDAP, 30.3wt% of PVC, and 60.6wt% of ptasticizer(DBP). And the optimum membrane thickness was0.45mm at this composition. Under the above condition, thelinear response ranger was $10^{-1}~1.2 {\times} 10^{-6}M$, and the detection limit was $5.1{\times}10^{-7}M$ with the Nernstian slope of 57mV/decade of activity of perchlorate ion. The electrode potential was stable within the pH range from 4 to 11. The selectivity coefficient was as shown below : $SCN^->I^->NO_3^->Br^->ClO_3^->F^->Cl^->SO_4^{2-}$

Influences of Peroxidase on Lysozyme Activity (Peroxidase가 Lysozyme 활성에 미치는 영향)

  • It is well known that many antimicrobial proteins in saliva interact with each other. The purpose of the present study was to investigate the interactions of lysozyme with peroxidase in the aspects of enzymatic activity in vitro. The interactions of lysozyme with peroxidase were examined by incubating hen egg-white lysozyme(HEWL) with bovine lactoperoxidase(bLP). The influence of peroxidase system on lysozyme was examined by subsequent addition of potassium thiocyanate and hydrogen peroxide. Lysozyme activity was determined by turbidity measurement of a Micrococcus lysodeikticus substrate suspension. Peroxidase activity was determined with an NbsSCN assay. The Wilcoxon signed rank test was used to analyze the changes of enzymatic activities compared with their controls. bLP at physiological concentrations enhanced the enzymatic activity of HEWL(P < 0.05) and its effect was dependent on the concentration of peroxidase. However, HEWL did not affect the enzymatic activity of bLP. Thiocyanate did not affect the enzymatic activity of HEWL, either. The addition of potassium thiocyanate and hydrogen peroxide did not lead to additional enhancement of the enzymatic activity of HEWL. The changes of hydrogen peroxide concentration in the peroxidase system did not affect the enzymatic activity of HEWL. Collectively, despite an in vitro nature of our study, the results of the present study provide valuable information on the interactions of lysozyme and peroxidase in the aspects of enzymatic activity in oral health care products and possibly in the oral cavity.

Effect of Salts and Temperature upon the Rate and Extent of Aggregation of Casein during Acidification of Milk (산에 의한 우유단백질의 응고속도에 염과 온도가 미치는 영향)

  • The rate and extent of coagulation of milk using fast acidification with 0.1 N HCl were monitored by changes in viscosity and turbidity at various temperatures and pH. Also the gelation rate of milk using slow acidification with D-glucono-${\delta}$-lactone was measured in a small strain rheological scanner. Coagulation of milk casein occurred in a specific pH range and was accompanied by an abrupt increase in viscosity at pH 5.0. Acid coagulation rate was enhanced by increasing temperature from $20^{\circ}C{\sim}50^{\circ}C$, and the maximum rate was shown around pH 5.0. The addition of salt ($CaCl_{2}$) reduced the maximum coagulation rate at all temperature ranges and shifted the pH ranges for maximum coagulation rate and the onset pH of coagulation. The onset of gelation and the rate of network formation during slow acidification were facilitated by Cl ion, but suppressed by SCN-ion, as indicated by the rate of rigidity development. The susceptibility to syneresis was greater in the gel made at lower temperature and around pH 4.6, while preheated milk at $90^{\circ}C$ for 5 min prior to acidification showed the same syneresis profile at all heating temperatures ($60{\sim}90^{\circ}C$).

Relation between Various Body Fluid Volumes and Body Weight or Lean Body Mass in the Rats (흰쥐의 체액량과 체중 및 무지방 체중 사이의 관계)

  • Relationships between red ceil volume $(^{51}Cr-cell)$, total blood volume (red cell volume divided by hematocrit ratio), and extracellular fluid volume (SCN distribution space) and body weight (ranging between 73 and 384 grams) or lean body mass were studied in 59 nembutalized rats. Lean body mass was determined by means of underwater weighing method on rats clipped and eviscerated. There were positive correlations between body weight or lean body mass and the absolute values (in milliliters) of body fluid volumes. Body fluid volumes expressed on the body weight or lean body mass basis, however, showed negative correlations between body weight (grams) or lean body weight (grams) with one exception. Red cell volume expressed as % lean body mass showed a positive correlation with lean body mass. The other results are summarized as follows: 1. Body density of rats was 1.0561 $(range:\;1.0123{\sim}1.0781)$ and 19.8% body weight of total body fat was obtained. The mean value of lean body mass was 80.2% body weight 2. The correlation between body weight and lean body mass was high, namely, coefficient of correlation was r=.99. 3. The correlation between the absolute value of red cell volume (ml) and body weight showed a high correlation, namely, r= 92 and between the lean body mass coefficient of correlation was r=.93. On a weight basis, red cell volume was 2.67 ml/100 gm body weight or 3.48 ml/100 gm lean body mass. The coefficient of correlation between body weight (grams) and red cell volume (% body weight) was r=-. 30. The coefficient of correlation between lean body mass (grams) and red cell volume (% lean body mass) was r=. 50. Thus, the following regression equation was obtained. Red cell volume (% lean body mass)=. 00243 Lean body mass (gm)+3. 12. 4. Total blood volume was 6.06% body weight or 7.83% lean body mass. The correlation between these blood volume values and body weight or lean body mass were negative, namely, r= -.43 and r=-.42 respectively. 5. Extracellular volume (SCN space) was 30.0% body weight or 37.2% lean body mass. These percentage values showed negative correlations between body weight or lean body mass and coefficients of correlation were r=-.40 and r=-.54 respectively. 6. The rate of increase in body weight or lean body mass is accompanied by a smaller rate of increase in blood volume and extracellular fluid volume. The rate of increase in red ceil volume paralled that of lean body mass.

Differential Pulse Voltammetric Determination of Iron(III) Ion with a Sodium Dodecyl Sulfate Modified Glassy Carbon Electrode (시차펄스전압전류법에서 도데실황산나트륨이 수식된 유리탄소전극에 의한 선택성 있는 철(III) 이온의 정량)

  • A selective method for the determination of iron(III) ion with a sodium dodecyl sulfate(SDS) modified glassy carbon electrode was proposed. It was based on the electrostatic attraction and complexation of the SDS modifier, $(DS^-)_n-Fe^{3+}$. The determination of iron(III) ion was performed by a differential pulse voltammetry(DPV), and the reduction peak potential of $(DS^-)_n-Fe^{3+}$ was +0.466(${\pm}0.002$)V vs. Ag/AgCl. For the determination of iron(III) ion, a linear calibration curve was obtained within the iron(III) ion concentration range of $0.50{\times}10^{-5}{\sim}10{\times}10^{-5}mol/L$, and the detection limit was $0.14{\times}10^{-5}mol/L$. $Cu^{2+}$, $Ni^{2+}$, $Co^{2+}$, $Pb^{2+}$, $Zn^{2+}$, and $Mn^{2+}$ showed little or no effect on the determination of iron(III) ion, respectively. But, ion such as each $CN^- $ and $SCN^-$ interfered seriously.
