Photocatalayst and Decomposition Properties of TiO2 and TiO2-CdS Powders Prepared by Supercritical Fluid Method (초임계 유체법으로 제조한 TiO2 및 TiO2-CdS계 광촉매의 분해물성 연구)

  • TiO$_2$ and TiO$_2$-CdS powders which were expected to be highly activated photocatalysts were prepared using supercritical fluid method (SCF). The prepared photocatalyst TiO$_2$ powders were crystalline of anatase and ultrafine spherical powders with large specific surface area. When photodecompositoion reaction was done with TiO$_2$ powders prepared by SCF as a photocatalyst in DCA (Dichloroactic Acid) solution, a hazardous organic compound, the photocatlyst, properties of TiO$_2$ powders prepared by SCF were better than that of commercial TiO$_2$ powders.

  • The ground state SCF IMO theory was applied to the Diels-Alder reactions of cyclopentadiene with 2-substituted acrylonitriles. The CNDO/2 MO of the separate systems, including both $\sigma$ and $\pi$ electrons, was used as starting point. The qualitative prediction of the relative reactivity was made with the calculated primary interaction energies. Here the calculated activation energies appeared to be realistic. The stereoselectivity determined by the calculated secondary interaction energies represented the endo-selectivity for all the substituents. The reason for the slightly unsymmetrical ring closure at the transition state was discussed in terms of valence inactive electron densities of the reacting atoms.

  • Protein disulfide isomerase (PDI) catalyzes the oxidation of disulfides and the isomerizatiob of incorrect disulfides in new polypeptides during folding in the oxidizing environment of the endoplasmic reticulum (ER). To increase recombinant protein hSCF (human stem cell factor) production, we have developed expression system using the Bombyx mori PDI (bPDI) as a fusion partner. bPDI gene fusion was found to improve the production of recombinant hSCFs. Thus, we conclude that bPDI gene fusion will be very useful for the large-scale production of biologically active recombinant proteins.

Effects of Juakium-derivative on Experimental Aplastic Anemia (좌귀음가미방이 실험적 재생불량성빈혈에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs have been used to improve the function of the hematopoietic system. The purpose of this study is to examine the hematopoietic effects of Juakium-derivative on aplastic anemia. Methods : In vitro, after bone marrow cells of mice with aplastic anemia induced by benzene were cultured with Juakium-derivative, we measured the gene expressions of hematopoietic cytokines, the intracellular TPO and SCF expression and the colony number of CFU-GEMM and BFU-E. Results : The results are summarized as follows : 1. The gene expressions of hematopoietic cytokines (TPO, c-mpl, SCF, c-kit, IL-3, EPO, EpoR, GM-CSF) and the TPO and SCF expression of the group treated with Juakium-derivative increased significantly more than those of the control group. 2. The colony number of CFU-GEMM and BFU-E in the rEPO plus rIL-3 plus Juakium-derivative group increased significantly compared with the only rEPO plus rIL-3 group. Conclusion : It was acknowledged that Juakium-derivative has hemato-potentiating effects on aplastic anemia induced by benzene and it is expected that Juakium-derivative can be used clinically for the patient with hematopoietic system disorder. This study could also be used further as important research data for Korean oriental medicine about hemato-potentiating. The exact mechanism explaining our study would need to be studied further.

Research on stress distributions around welds of three-planar tubular Y-joints under out-of-plane bending moment

  • Marine structures including offshore wind turbines (OWTs) always work under cyclic loads, which arouses much attention on the fatigue design. The tripod substructure is one of the typical foundation forms for fixed OWTs. The three-planar tubular Y-joint (3Y joint) is one of the important components in fatigue design as it is most likely to have cracks. With the existence of the multiplanar interaction effect, calculating the hot spot stress (HSS) of 3Y joints is complicated. To assist with fatigue design, the distributions of stress concentration factor (SCF) and multiplanar interaction factor (MIF) along weld toe curves induced by the out-of-plane bending moment are explored in this study. An FE analysis method was first developed and verified against experimental results. This method was applied to build a numerical database including 1920 FE models covering common ranges of geometric parameters. A parametric study has been carried out to reveal the distribution patterns of SCF and MIF. After multidimensional nonlinear fittings, SCF and MIF distribution formulas have been proposed. Accuracy and reliability checking prove that the proposed formulas are suitable for calculating the HSS of 3Y joints.

In vitro growth of mouse preantral follicles: effect of animal age and stem cell factor/insulin-like growth factor supplementation

  • Objective: To determine whether animal age impacts in vitro preantral follicle growth. Effects of hCG, stem cell factor (SCF), and/or insulin-like growth factor (IGF) supplementation in growth medium were also investigated. Methods: Intact preantral follicles were mechanically isolated from fresh ovaries of BDF1 mice and cultured in growth medium for 9 to 11 days. Surviving follicles with antrum formation were transferred to maturation medium for 14 to 18 hours. Follicle survival, antrum formation, and retrieval of metaphase II (MII) oocytes were compared among three age categories (4-5, 7-8, and 10-11 week-old). By using 7- to 8-week-old mice, preantral follicles were cultured in growth medium supplemented with hCG (0, 5, or 10 mIU/mL), SCF (50 ng/mL), IGF-1 (50 ng/mL), and SCF+IGF-1. Results: Seven- to eight-week-old mice showed a higher follicle survival and antrum formation and produced more MII oocytes compared to other groups. In the 7- to 8-week-old mice, supplementation of 5 mIU/mL hCG significantly enhanced the antrum formation but the percentage of MII oocytes was similar to that of the control. Supplementation of SCF+IGF-1 did not enhance follicle survival or antrum formation but the percentage of MII oocytes increased modestly (39.1%) than in the control (28.6%, p>0.05, statistically not significant). Conclusion: Seven- to eight-week-old mice showed better outcomes in growth of preantral follicles in vitro than 4- to 5- or 10- to 11-week-old mice. Supplementation of hCG enhanced antrum formation and supplementation of SCF+IGF-1 yielded more mature oocytes; hence, these should be considered in the growth of preantral follicles in vitro.

  • Ex vivo hematopoietic cells can treat patients suffering from hematopoietic malignancies using bone marrow transplantation therapies. A initial cell density of $1.5{\times}10^6cells/ml$ and a growth factors of IL-3(5ng/ml), SCF(5ng/ml) and FL(25ng/ml) result in a 3.6-fold expansion of LTC- IC but a unexpansion of total cells.

Stress Analysis and Life Evaluation of Rotor and Retaining Ring of Generator for fossil power plant (화력용 발전기 회전자 및 리테인 링의 응력해석 및 수명평가)

  • This paper represents that parts of a large generator operating in 1000 MW are affected by centrifugal forces due to high-speed rotation in 3600 rpm and 3D FEM Analyses are required to obtained the structural reliability of the generator. From these results, one would know the weakest locations and the stress distributions. The fatigue life is calculated in order to grasp the remaining life of generator. 2D and 3D analyses are performed to calculate stresses of the generator rotor and the retaining ring. From 2D results, we find the SCF at the slot and sub-slot of the rotor. 3D analysis is applied at the end part of generator rotor, which represents complex geometry, and rotor and retaining ring shrink thermally. With these results, designers of rotor and retaining ring can compare with the results of design code and verify the stress distributions of generator rotor and retaining ring, and then calculate the remaining life from the low-cycle fatigue data.
