Phytochemical Constituents of Amomum xanthioides

  • Seven monoterpenes, three sesquiterpenes, three phenolics and one flavonoid were isolated from the MeOH extract of Amomum xanthioides. Their structures were determined by spectroscopic methods to be caryophyllene oxide (1), bornyl acetate (2), nerolidol (3), spathulenol (4), (-)-borneol (5), (+)-5-endohydroxycamphor (6), vanillic acid (7), protocatechuic acid methyl ester (8), betulabuside A (9), (1R,4S,6R)-6-hydroxyfenchan-2-one-6-O-$\beta$-D-glucopyranoside (10), (1S,4R,6S)-6-hydroxybornan-2-one-6-O-$\beta$-D-glucopyranoside (11), (1R,2S,4S,5R)-angelicoidenol 2-O-$\beta$-D-glucopyranoside (12), 1-O-vanilloyl-$\beta$-D-glucopyranoside (13), and quercetin-3-rhamnopyranoside (14). Compounds 6-14 were isolated for the first time from this plant source. Compounds 3 and 4 exhibited moderate cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

Effects of Different Numbers of Feeding Days and Feeding Rate on Growth of Growing-out Flounder Paralichthys olivaceus (사료 공급일수 및 공급율이 육성기 넙치(Paralichthys olivaceus)의 성장에 미치는 영향)

  • This study evaluated the effects of the number of feeding days and feeding rate on the growth of growing-out flounder Paralichthys olivaceus. Three replicated groups of fish ($141{\pm}1.9g$) were hand-fed a commercial feed under different feeding regimes for 9 weeks. Fish in group 6F-1S were fed to satiation twice daily for 6 consecutive days and starved for 1 day. Fish in groups 4F-1S and 3F-1S were fed to satiation twice daily for 4 and 3 days, respectively, and starved for 1 day. Fish in group 6F-90% were fed a diet of 90% of 6F-1S twice daily for 6 consecutive days and starved for 1 day. The weight gain, feed efficiency and daily feed intake of the fish groups in 4F-1S and 3F-1S were not significantly different from those of group 6F-1S. The weight gain and feed efficiency of fish in group 6F-90% were not significantly different from those of group 6F-1S. The results of this study suggest that the growth of growing-out flounder fed to satiation twice daily for 4 or 3 days after starving 1 day was similar to that of fish fed to satiation for 6 days, and that the proper feeding rate for growth could be lowered to 90% of satiation without growth suppression.

QoS Guaranteed Handover Scheme for Global Roaming in Heterogeneous Proxy Mobile IPv6 Networks (이기종 프록시 이동(모바일) IPv6 네트워크에서 QoS가 보장된 글로벌 로밍 핸:드오버 방식)

  • Mobility and quality of service (QoS) are becoming the more important issues in wireless communications. The traditional Internet service is expanding into new access media and applications. Since wireless communication services are accompanied by frequent handovers at remote sites, scalable and fast handover has become a prerequisite for ubiquitous communication. In this paper, the differentiated service (Diffserv) model is deployed in heterogeneous proxy mobile IPv6 (PMIPv6) networks to satisfy the QoS guaranteed service and fast handover requirements. The operational procedures for QoS guaranteed global roaming are presented. In addition, QoS management and handover cost evaluation schemes based on a mobile host's movement scope are proposed. This paper analyzes the reduction in handover delay in a network-based localized mobility management framework. We propose and analyze a PMIPv6 optimized with a global mobile access gateway (G-MAG), which is a network-based entity, to further improve the handover performance in terms of handover delay while maintaining minimal signaling overhead in the air interface among converged heterogeneous wireless networks. The handover signaling procedures with host-based MIPv6 are compared with network-based proxy MIPv6 (PMIPv6) and fast PMIPv6 assisted by G-MAG to show how much handover delay reduction can be achieved. Analytical results show that the handover delay is significantly reduced.

Significance of Expression of CD44s and CD44v6 in Colorectal Cancer

  • The expression of CD44v is known as a marker of cancer progression and its metastasis in colorectal cancer. It has been known that CD44 variant containing sequences encoded by exon 11 (v6) confer metastatic potential to human colorectal cancer cells. The role of CD44 standard (CD44s) and CD44v6 in colorectal cancer was investigated in this study by immunohistochemical staining of the primary tumors obtained from the colorectal cancer patients. Immunohistochemical staining was performed in 40 patients with colorectal cancer who underwent curative surgery at Keimyung University hospital. The expression CD44s and CD44v6 was observed in 24/40 (60%) and 13/40 (32.5%) respectively. The expression of CD44v6 had correlation with TNM stage (P<0.05), however CD44s had not any correlation with clinicopathological parameters. These results suggest that CD44v6 expression may give an information for tumor progression than decreased expression of CD44s in colorectal cancer cells.

  • The WL6 strain isolated from Kimchi could not be made scientific name because it was identified as three species, i.e., Leuconostoc mesenternides ssp cremoris, Leu. mesenteroides ssp. dextranicum or Lactobacillus bifermentans when it was tested by API kit or Biolog system methods. The unidentifiable WL6 strain was finally reclassified as Lactobacillus bifermentans by genetic identification using two PCR-based specific sequence primer sets which were originated from homologous pepN and 16S rRNA genes.

DEPTOR Expression Negatively Correlates with mTORC1 Activity and Tumor Progression in Colorectal Cancer

  • The mammalian target of rapamycin (mTOR) signaling pathway is upregulated in the pathogenesis of many cancers, including colorectal cancer (CRC). DEPTOR is an mTOR inhibitor whose expression is negatively regulated by mTOR. However, the role of DEPTOR in the development of CRC is not known. The aim of this study was to investigate the expression of DEPTOR and mTORC1 activity (P-S6) in a subset of CRC patients and determine their relation to tumor differentiation, invasion, nodal metastasis and disease-free survival. Here, Immunohistochemical expression of P-S6 (S235/236) and DEPTOR were evaluated in 1.5 mm tumor cores from 90 CRC patients and in 90 samples of adjacent normal mucosa by tissue microarray. The expression of P-S6 (S235/236) was upregulated in CRC, with the positive rate of P-S6 (S235/236) in CRC (63.3%) significantly higher than that in control tissues (36.7%, 30%) (p<0.05). P-S6 (S235/236) also correlated with high tumor histologic grade (p=0.002), and positive nodal metastasis (p=0.002). In contrast, the expression level of DEPTOR was correlated with low tumor histological grade (p=0.006), and negative nodal metastasis (p=0.001). Interestingly, P-S6 (S235/236) expression showed a significant negative association with the expression of DEPTOR in CRC (p=0.011, R= -0.279). However, upregulation of P-S6 (S235/236) (p=0.693) and downregulation of DEPTOR (p=0.331) in CRC were not significantly associated with overall survival. Thus, we conclude that expression of DEPTOR negatively correlates with mTORC1 activity and tumor progression in CRC. DEPTOR is a potential marker for prognostic evaluation and a target for the treatment of CRC.

Anticancer Mechanisms of 3-Heptylamino-6-Allylthiopyridazine and 3-Dipentylamino-6-Allylthiopyridazine in Human Colon Carcinoma RKO Cells (RKO 대장암세포에서 3-헵틸아미노-6-알릴티오피리다진과 3-디펜틸아미노-6-알릴티오피리다진의 항암기전)

  • Allylthiopyridazine derivatives were synthesized and evaluated for anti-proliferative activities in the previous study. In this study, selected two allylthiopyridazine derivatives (compound I, 3-heptylamino-6-allylthiopyridazine and compound II, 3-dipentylamino-6-allylthiopyridazine) were assessed for cytotoxicity and chronic proliferation in human colon carcinoma RKO cells. Two derivatives dose-dependently inhibited cell viability and proliferation. To elucidate the anticancer mechanism of two derivatives, we investigated the expression level of apoptosis-related proteins in RKO cells. Compound I induced the activation of JNK and expression of p53 and p21. On the other hand, compound II showed no change of p53 level. Interestingly, compound II inhibited the nuclear translocation of NF-${\kappa}B$. This result suggested that compound II suppressed cell proliferation. These different mechanisms of these compounds might have occurred through different steric conformation.
