Anhydrous Crosslinked Polymer Electrolyte Membranes Based On ABA Triblock Copolymer (ABA 트리블록 공중합체를 이용한 무가습 가교형 고분자 전해질막)

  • ABA type triblock copolymer of poly(hydroxyl ethyl acrylate )-b-polystyrene-b-poly(hydroxyl ethyl acrylate), i.e. PHEA-b-PS-b-PHEA, was synthesized throughatom transfer radical polymerization (ATRP). This block copolymer was thermally crosslinked with 4,5-imidazole dicarboxylic acid (IDA) via the esterification between the -OH groups of PHEA in block copolymer and the -COOH groups of IDA. Upon doping with ${H_3}{PO_4}$ to form imidazole-${H_3}{PO_4}$ complexes, the proton conductivity of membranes continuously increased with increasing ${H_3}{PO_4}$ content. The PHEA-b-PS-b-PHEA/IDA/${H_3}{PO_4}$ polymer membrane with [HEA]:[IDA]:[${H_3}{PO_4}$]=3:4:4 exhibited a maximum proton conductivity of 0.01 S/cm at $100^{\circ}C$ under anhydrous conditions. Thermal gravimetric analysis (TGA) shows that the PHEA-b-PS-b-PHEA/IDA/${H_3}{PO_4}$ complex membranes were thermally stable up to $350^{\circ}C$, indicating their applicability in fuel cells.

Nutritional Compositions of Three Traditional Actinidia (Actinidia arguta) Cultivars Improved in Korea (국내에서 개량된 3가지 토종 다래 품종의 영양성분 분석)

  • Nutritional compositions of Korean traditional actinidia (Actinidia arguta) cultivars, such as Otumsense, Chiak, and Skinny green, were investigated as high value-added food substances. Among minerals, K content of three cultivars was the highest, and P, Ca, and Na contents were relatively higher than those of other minerals. Contents of essential amino acids were analyzed as follows: Otumsense (198.48 mg/100 g), Chiak (413.50 mg/100 g), and Skinny green (270.07 mg/100 g). Total amino acids of Chiak were the highest among the three cultivars, and major amino acids of the three cultivars were glutamic acid and aspartic acid. Analysis of fatty acids showed that major fatty acids were palmitic acid as a saturated fatted acid and ${\alpha}$-linoleic acid as an unsaturated fatty acid in the three cultivars. Glucose and fructose were major free sugar constituents in the three cultivars. Total free sugar content of Otumsense was relatively higher than others. Finally, niacin and vitamin $B_6$ of Skinny green (1.55 mg/100 g, 1.92 mg/100 g) were the highest among the cultivars. However, ${\beta}$-carotene as a vitamin A precursor ($3.82{\mu}g/100g$) and vitamin C as a natural antioxidant substance (47.18 mg/100 g) of Otumsense cultivar were the highest.

  • This paper presents 6bit 100MHz Interpolation Flash Analog-to-Digital Converter, which can be applied to the Receiver of Wireless Tele-communication System. The 6bit 100MHz Flash Analog-to-Digital Converter simplifies and integrates S-R latch which multiplies as the resolution increases. Whereas the conventional NAND based S-R latch needed eight MOS transistors, this Converter was designed with only six, which makes the Dynamic Power Dissipation of the A/D Converter reduced up to 12.5%. The designed A/D Converter went through $0.18{\mu}m$ CMOS n-well 1-poly 6-metal process to be a final product, and the final product has shown 282mW of power dissipation with 1.8V of Supply Voltage, 100MHz of conversion rate. And 35.027dBc, 31.253dB SFDR and 4.8bits, 4.2bits ENOB with 12.5MHz, 50MHz of each input frequency.

Purification and Characterization of the Bacteriocin Produced by Lactococcus sp. KD 28 Isolated from Kimchi (김치에서 분리한 Lactococcus lactis가 생산하는 박테리오신의 정제 및 특성)

  • The bacterial strain isolated from Kimchi showed antibacterial activity against Micrococcus luteus IAM 1056. The selected strain was identified as Lactococcus lactis by 16S rRNA nucleotide sequence analysis and named as Lactococcus sp. KD 28. The treatment of culture supernatant with proteinase K removed antibacterial activity, indicating its proteinaceous nature, a bacteriocin. This bacteriocin was sensitive to hydrolytic enzymes such as ${\alpha}$-chymotrypsion, trypsin, proteinase K, lipase, ${\alpha}$-amylase and subtilisin A. The bacteriocin was highly thermostable and resistant to heating at $80^{\circ}C$ for up to an hour but 50 % of the total activity was remained at $100^{\circ}C$ for 30 min. The pH range from 2.0 to 8.0 had no effect on bacteriocin activity and it was not affected by solvents such as acetonitrile, isopropanol, methanol, chloroform and acetone up to 50% concentration. The bacteriocin showed antibacterial activity against M. luteus IAM 1056, Lactobacillus delbrueckii subsp. lactis KCTC 1058, Enterococcus faecium KCTC 3095, Bacillus cereus KCTC 1013, B. subtilis KCTC 1023, Listeria ivanovii subsp. ivanovii KCTC 3444, Staphylococcus aureus subsp. aureus KCTC 1916, B. megaterium KCTC 1098 and B. sphaericus KCTC 1184. The bacteriocin was purified through ammonium sulfate concentration, SP-Sepharose chromatography and RP-HPLC. The molecular weight was estimated to be about 3.4 kDa by tricine-SDS-PAGE analysis.

Affinity Maturation of an Anti-Hepatitis B Virus PreS1 Humanized Antibody by Phage Display

  • In a previous study we generated an anti-Hepatitis B Virus (HBV) preS1 humanized antibody (HzKR127) that showed in vivo HBV-neutralizing activity in chimpanzees. However, the antigen-binding affinity of the humanized antibody may not be sufficient for clinical use and thus affinity maturation is required for better therapeutic efficacy. In this study, phage display technique was employed to increase the affinity of HzKR127. All six amino acid residues (Glu95-Tyr96-Asp97-Glu98-Ala99-Tyr100) in the heavy (H) chain complementary-determining region 3 (HCDR3) of HzKR127 were randomized and phage-displayed single chain Fv (scFv) library was constructed. After three rounds of panning, 12 different clones exhibiting higher antigen-binding activity than the wild type ScFv were selected and their antigen-binding specificity for the preS1 confirmed. Subsequently, five ScFv clones were converted to whole IgG and subjected to affinity determination. The results showed that two clones (B3 and A19) exhibited an approximately 6 fold higher affinities than that of HzKR127. The affinity-matured humanized antibodies may be useful in anti-HBV immunotherapy.

Effects of Dietary Bong-ip(Morus alba L.), Gam-chei(Glycyrrhizae glabra), Sol-ip(Pinus densiflora) and Dang-gi(Angelica gigas) on Serum Composition in Rats (뽕잎, 감초, 솔잎 및 당귀분말이 흰쥐의 혈청조성에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary Morus alba L.(Bong-ip, B), Glycyrrhizae glabra(Gam-chei, C), Pinus densiflora(Sol-lp, S) and Angelica gigas(Dang-gi, D)powder on serum composition in rats(Sprague-Dawley male rats, 100-110g). Serum TG(triglyceride, p<0.01), total cholesterol, glucose, total protein, albumin, GGT$({\gamma}-glutamyl$ transferase, p<0.05) were significantly increased D group than that of nomal and other groups, but UA(uric acid, p<0.05) was significantly decreased, and C group(p<0.05) was significantly increased. but C group of urine(p<0.05) was significantly decreased. Also, B and S groups(p<0.05) of BUN(blood urea nitrogen), S group(p<0.05) of ALP(alkaline phosphokinase, Band C(p<0.05) of CPK(creatinine phosphokinsae, p<0.05) were significantly increased. B, S and C groups were better than D group for lipid metabolism, and pretection to liver. Also, B and C groups of glucose were same as normal diet, so Morus alba L. was good food for lipid metabolism and hypoglycemic effect.

Final Diffraction Patterns of the Beam Splitters used in the Soft XRay Interferometer by a He-Ne Laser

  • The soft x-ray(10nm-100nm) interferometer is a modified Mach-Zehnder type interferometer and it consists of two beam-splitters and four totally reflecting mirrors. The beam-splitters used here are 50% transmission and 50% reflection grating type. The diffraction patterns of beam splitters(1st B.S.) were investigated with a He-Ne laser. The diffraction patterns produced by the soft x-ray interferometer (2nd B.S.) were also investigated in intensities positions. The diffraction patterns of 20 degree grazing incidence on the beam splitters(1st B.S.) show a circular array of spots. Both the reflected and the transmitted beams show the same patterns but symmetric circles on the screen. The maximum intensity appears roughly when n is in the zeroth and odd orders and the suppressed peak(missing order) appears when n is in even orders. Intensities of 3 center fringes(n = 0, $\pm$1) are stronger than others. These results confirm the reduced grating equation and make agree with the intensity distribution function. It was found that the final patterns produced by the soft x-ray interferometer (2nd B.S.) consisted of fine fringes which were caused by two of three diffraction beams that were arrived at the second beam-splitter.

Effects of Differents types of Clothing and Colours on Clothing Microclimate in the Subjects wearing Sports Wear under Sunlight (일광하에서 운동시의 스포츠웨어 색상과 의복형태가 의복기후에 미치는 영향)

  • In this study, We endeavored to revaluate the effects of different types of clothing and colors on clothing microclimate in the subjects wearing sports wear at sunlight environment. This study was conducted 4 different kinds (cotton 100%) of clothing ensembles, that was W-1(long trousers and shirt of white color), B-1 (long trousers and shirt of black color), W-s (short trousers and shirt white color), B-s (short trousers and shirt black color) and were done in a climate chamber under sunlight ambient temperature ($33.67{\pm}1.8^{\circ}C$, $46.0{\pm}8.5%RH$) by three males subject who are in good healthy. Start a 20-min rest period, 20-min bouts of exercise and final 20-min recovery period were performed. The kinetic load was given for 20 minutes under the condition of 6.0 km/hr walking speed on the treadmill. The results is as followed In case of same type of garment, temperature within clothing which is based on difference of color the white ensemble keeps higher temperature than black one. According to distribution chart of temperature within clothing in case of chest, white one shows higher temperature than black one, in case of back, black one shows higher temperature than white one. Difference of heart rate was so clear and sequence is W-1>B-1>W-s>B-s, so we could find same tendency with temperature within clothing.

Influence of Temperature and Salinity on the Growth and Size of the Rotifer Brachionus plicatilis and B. rotundiformis (온도와 염분이 Rotifer Brachionus plicatilis와 B. rotundiformis의 성장과 크기에 미치는 영향)

  • Rotifers of the genus Brachionus are commonly used as a live food for larval fish, and rotifers of different sizes are preferred according the mouth size of the fish. Rotifer species vary in size, and individual size can depend on the temperature and salinity of the rearing environment. We investigated the effects of temperature and salinity for two species, B. plicatilis (250-300 ${\mu}m$) and B. rotundiformis (100-220 ${\mu}m$). Two strains of B. plicatilis (CCUMP 36 and 48) and two strains of B. rotundiformis (CCUMP 51 and 56) were received from the Culture Collection of Useful Marine Plankton (CCUMP) at Pukyong National University and cultured with the green alga, Nannochloris oculata (KMMCC 16) from the Korea Marine Microalgal Culture Center (KMMCC). The growth and size of rotifers were examined at three water temperatures ($16^{\circ}C$, $24^{\circ}C$, $32^{\circ}C$) and four salinities (20 psu, 25 psu, 30 psu, 35 psu) under continuous light (40 ${\mu}molm^{-2}s^{-1}$). The maximum density and growth rate of B. rotundiformis were greater than those of B. plicatilis. The lorica length of B. plicatilis ranged from 215.4 to 269.7 ${\mu}m$ and from 154.9 to 206.6 ${\mu}m$ for B. rotundiformis, depending on strain, temperature and salinity. Rotifers were smaller when cultured at high temperatures, regardless of salinity. B. rotundiformis preferred higher salinity than B. plicatilis. The results demonstrated that the size of rotifers could be controlled to some extent by temperature and salinity.

Optimized Processing Condition of Production of Nannochloropsis oculata under Light-emitting Diode (LED) Condition (LED배양조건에서 미세조류 Nannochloropsis oculata의 생산 효율성을 높이는 공정 최적화)

  • The 100 l culture system was made on the basis of LED light, and Nannochloropsis oculata was cultured in f/2 medium at light intensity ($100{\mu}mol/m^2/s$), culture temperature ($20^{\circ}C{\pm}1^{\circ}C$) and LD cycle (12hr). As a result, the maximum biomass of 1.07 g/l was cultured as a result of 100 l mass culture at $100{\mu}mol/m^2/s$ and 24 mg/l nitrate concentration in LED blue (475 nm). The extraction was carried out using sonicator, homogenizer and chemical method 0.5M HCl shredding method. The contents of chlorophyll a, chlorophyll b and carotenoid were 1.6, 0.5 and 0.3 mg/g cell. When using homogenizer, it was measured at 1.0, 0.6 and 0.2 mg/g cell. The chemical breakdown method of 0.5M HCl, chlorophyll a, b, and carotenoid contents were measured as 0.9, 0.8, 0 mg/g cell. The highest amount of biomass during the distruption time was measured at 3.6 mg/g cell at 15 min disintegration and acetone, 3.6 mg/g cell of acetone, methanol, and ethanol were measured as effective solvents. Concentration was measured by using microfilter, disk type continuous centrifuge and tubular type continuous centrifuge were 16.0, 1.1 and 0.5 g/l, respectively. Four kinds of equipment such as hot air dryer, vacuum dryer, spray dryer and freeze dryer were tested to optimize the drying process. As a result, the recovery rates of spray dryer and freeze dryer were 80% and 60%.
