최근 20개월간 분리된 이질균의 고찰과 과거 20년간의 항균제에 대한 감수성의 변천 (Studies on Shigella Isolated during Recent Twenty Months and Changes of Those Antibiotics Susceptibility Patterns for Last Twenty Years)

  • Shigella is one of the most prevalent pathogens for the diarrhoeal diseases in the developing countries. One hundered and six strains of shigella were isolated from January 1980 to August 1981 at the dept. of clinical pathology, Han Yang Medical Center. Subgroups of these strains were identified as one strain of S. dysenteriae, 98 strains of S. flexneri and 7 strains of S. sonnei. None of S. boydii was observed. Sex ratio, male to female was 48 to 58. Age distribution disclosed 6 cases under one year, 11 cases one to under 2 years and 21 cases(19.8%) two to under 3 years. Subtotal of 0 to 9 years showed 64 cases(60.4%). Susceptibility for antibiotics of these strains revealed dibekacin 100%, sisomicin 100%, amikacin 98.1%, cefazolin 97.2%, tobramycin 97.1%, gentamicin 95.2%, colistin 93.0%, minocycline 89.6%, kanamycin 83.0%, carbenicillin 18.9%, streptomycin 18.9%, tmp-smz 8.6%, ampicillin 2.8% and chloramphenicol 1.9%. Patterns of resistance to sulfa, streptomycin, chloramphenical and tetracycline have already started at the early part of 1960 decade. Although ampicillin was highly sensitive to shigella at the end of 1960 to the early part of 1970 decade, this study has disclosed high resistance to the strains. New antibiotics such as amikacin, cefazolin, dibekacin, gentamicin, and tobramycin have revealed highly sensitive to these strains, however, multiresistance for those antibiotics will be shown to be prevalent in this country within several years, where it is probably related to the unrestricted sale and use of antibiotics in man.

백작약 추출물이 식중독 유발 세균의 증식에 미치는 영향 (Effect of Extracts from Paeonia japonica on the Growth of Food-borne Pathogens)

  • 본 연구에서는 과거부터 민간이나 한방에서 널리 사용되어져 온 백작약의 식중독 유발 세균에 대한 항균 활성을 조사해 보고자하였다. 백작약을petroleum ether. chloroform, ethylacetate 및 methanol의 4가지 유기용매 및 열수에 추출하고, Gram(+)세균인 Staphylococcus. aureus 및 Staphylococcus. epidermidis와Gram(-)세균인 Salmonella typhimirium, Salmonella paratyphimirium, Shigella dysenteriae, Shigella flexneri, Shigella sonnei 및 Pseudomonas aeruginosa에 대한 성장 억제 실험을 해 본 바, 백작약의 methanol 추출물이 10 mg/mL 농도에서 Salmonella typhimurium에 대해 가장 높은 항균력을 나타내었다. Staphylococcus aureus에 대한 항균력은 백작약의 ethylacetate 추출물이 5 mg/mL에서 가장 큰 항균 효과를 보였으며, 액체 배지에 배양한 경우 5~10 mg/mL 농도에서 18시간까지 성장 억제 효과가 나타났다.

  • 1978년(年)에 분리(分離)한 9주(株)의 Shigella, 70주(株)의 Salmonella paratyphi A 및 230주(株)의 S. typhi의 약제내성(藥劑耐性), 내성화방지(耐性化防止) 및 제거(除去)에 대(對)하여 실험(實驗)하여 다음과 같은 성적(成績)을 얻었다. Shigella는 79주(株)가 Sh. flexneri, 16주(株)가 Sh. sonnei였는데 1주(株)를 제외(除外)한 94주(株)가 chloramphenicol tetracycline, streptomycin, sulfisomidine에 다약제내성(多藥劑耐性)이었으며 그중(中) 70주(株)는 ampicillin과 carbenicillin에, 80주(株)는 trimethoprim-sulfamethoxazole에, 22주(株)는 nalidixic acid에, 1주(株)는kanamycin에도 내성(耐性)이었다. Gentamicin, amikacin, cephaloridine, rifampin에 내성(耐性)인 균주(菌株)를 없었다. S. paratyphi A와 S. typhi는 공시약제(供試藥劑)에 감수성(感受性)이었으나 다만 rifampin, 또는 sulfisomidine에 약(弱)한 내성(耐性)을 가진 것이 있었다. 다약제내성(多藥劑耐性)인 shigella의 약(約) 80%가 전달성(傳達性) R plasmid를 가지고 있어서 그 내성(耐性)을 E. coli에 전달(傳達)시킬 수 있었다. 내성전달빈도(耐性傳達頻度)는 공시균주(供試菌株) 및 피전달균(被傳達菌)에 따라 차이(差異)가 있었다. 보존(保存)한 내성균주(耐性菌株)는 그 비율(比率)은 균주(菌株)에 따라 차이(差異)가 있으나 내성균(耐性菌)과 감수성균(感受性菌)으로 구성(構成)되어 있었으며 계대배양(繼代培養)에 의하여 내성(耐性)이 쉽게 탈락(脫落)되지는 안하였다. Acriflavine은 내성(耐性)을 탈락(脫落)시키는 효과(效果)는 있었으며 균주(菌株)에 따라 그 차이(差異)가 심(甚)하였고 atabrine은 효과(效果)가 없었다. 약제(藥劑)의 병용(倂用)은 Shigella에 대(對)한 작용(作用)을 증강(增强)시키는 경우(境遇)가 많으며 대체(大體)로 상승작용(相乘作用)을 나타냈고 상가작용(相加作用)을 나타내는 경우(境遇)가 있었으나 길항작용(拮抗作用)은 볼 수 없었다.

성주군 S 초등학교 및 중학교에서 집단 발생한 세균성 이질에 관한 역학조사 (Epidemiologic Investigation on an Outbreak of Shigellosis in Seongju-gun, Korea, 2003)

  • Objectives: An outbreak of shigellosis occurred among students and staff of S primary and middle school, Seongju-gun, in 2003. This investigation was carried out to institute an effective counterplan, and study the infection source and transmission of the shigellosis. Methods: The authors conducted a questionnaire survey among 235 students and staff from S preschool, primary and middle school relating to the ingestion of school lunch and the manifestation of symptoms. Also, the author investigated the drinking water, feeding facility and reconstructed cooking process of the food presumed to be the cause of the shigellosis. The diarrhea cases were defined as confirmed cases and those cases who had had diarrhea more than one time, accompanied with symptoms such as fever, vomiting and tenesmus. Results: From rectal swabs 20 people, between June 28 and July 4, 2003, were confirmed with shigellosis. The diarrhea attack rate was 40.0%. Those who had ingested tomatoes and cubed radish kimchi had significantly higher diarrhea attack rates (p<0.05), with the relative risk of tomatoes being 2.69 (95% CI: 0.98-7.42). The major cause of shigellosis was presumed to be from contaminated tomatoes due to cooking with rubber gloves containing holes. Conclusion: The cooks in charge of school lunches must make doubly sure to not only attend to their sanitation, but also to manage the table wear and items used in providing school lunches. The health care authority should introduce higher-leveled criteria for health care among cooks, so that they cannot cook when the have a case of any infectious disease.

돈(豚)에서 분리(分離)한 Shigella균유래(菌由來) R plasmid의 유전적(遺傳的) 특성(特性)에 관한 연구(硏究) (Genetic properties of R plasmids in Shigella isolates of swine origin in Korea)

  • This paper dealt with the distribution of Shigella spp. on 5 piggeries in Taegu and Kyungpook during the period from August to October 1987. Isolated Shigella were examined for serogrouping, antimicrobial drug resistance and detection of R plasmid. Genetic properties of R plasmid in Shigella have examined to fertility inhibition (Fi) and gel electrophoresis was performed for the isolation of plasmid DNA. The results obtained were summarised as followings; 1. Of total 2,978 samples from 5 piggeries, 82 strains (2.8%) of Shigella spp. were isolated from 82 samples. The isolated strains were identified as S dysenteriae (60 strains), S flexneri (20 strains) and S sonnei (2 strains). 2. Of the 82 strains examined 67 (95.1%) were resistant to one or more antibiotics, such as ampicillin (Am), chloramphenicol (Cm), kanamycin (Km), nalidixic acid (Na), rifampicin (Rf), streptomycin (Sm), sulfademethoxine (Su), and tetracycline (Tc) and higher resistant to Su (90.2%), Sm (63.4%) and Tc (63.4%). 3. Of the 78 resistant Shigella strains 26 (33.3%) harbored conjugative R plasmids and the transfer frequency of Sm (50.0%), Cm(33.3%) resistance was much higher than that of the other drug resistance. 4. The most common resistant patterns were SmSuTc, Su and AmSmSuTc. 5. Out of the 26 Shigella R plasm ids examined for Fi, 14(53.8%) were $Fi^+$ and the remainder were $Fi^-$. 6. The plasmid DNA profiles in Shigella spp. (9 strains) isolated from pigs were confirmed as being 2 to 9 fragments by the gel electrophoresis. Their molecular size ranged 2.17 to 87.62 kilobase (Kb). All strains of Shigella spp. consisted in 15.4 Kb plasmids.

Shigella spp.의 연차적 추이에 관한 조사연구 (A Study on Changing Patterns of Shigella spp. by Years)

  • A total of 3,879 cases of feces and rectal swab from patient was collected in H. hospital from January 1974 to December 1986 in Seoul. Among the materials, the number of cases of Shigella spp. isolated were 197 strains of 139 patients. Infectous pattern and antibiotic sensitivity of Shigella spp. were as follows; The range of percentage of an identified Shigella spp. among total feces and rectal swabs was $1.5{\sim}12.5%$ yearly. The isolation ratios of Shigella spp. per each patient 1.35 for male and 1.19 for female. The isolation ratio of male to female was 1.28:1 in whole group. The isolated Shigella species was 81.0% in S. flexneri, 1.1% in S. boydii and 17% in S. sonnei. The highest number of Shigella spp. was found in August and September according to monthly isolation, on the other hand the lowest number of Shigella spp. was obserbed in March. The seasonal isolation rate of Shigella spp. was 31.7% in Fall, 27.3% in Summer, 21.6% in Winter and 19.3% in Spring. The age specific frequency of Shigellosis was 46.8% in $0{\sim}9$ year group, 8.6% in $10{\sim}19$, 7.2% in $40{\sim}49$ and 6.5% in $50{\sim}59$. The antibiotics showing over 80% susceptibility against Shigella spp. were gentamicin, kanamycin, amikacin, tobramycin, cefoperazone, cefoxitin, cefamandole nafate, cefotaxine and sisomycin.

라이소자임 활성을 보유한 Lactobacillus rhamnosus 배양물의 병원성 미생물에 대한 항균 효과 (Anti-Bacterial Effect of Lactobacillus rhamnosus Cell-Free Supernatant Possessing Lysozyme Activity Against Pathogenic Bacteria)

  • Recently, there has been a growing demand for natural preservatives because of increased consumer interest in health. In this study, we produced Lactobacillus rhamnosus cell-free supernatant (LCFS) and evaluated and compared its antimicrobial activity with existing natural preservatives against pathogenic microorganisms and in chicken breast meat contaminated with Escherichia coli and Staphylococcus aureus. Lactobacillus rhamnosus cell-free supernatant possessed 30 units of lysozyme activity and contained 18,835 mg/L of lactic acid, 2,051 mg/L of citric acid and 5,060 mg/L of acetic acid. Additionally, LCFS inhibited the growth of fourteen pathogenic bacteria, S. aureus, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Listeria innocua, S. epidermidis, L. ivanovii, E. coli, Pseudomonas aeruginosa, Shigella sonnei, Shi. flexneri, Proteus vulgaris, Pseudomonas fluorescens, and Klebsiella pneumoniae. The antibacterial activity of LCFS was stronger than that of egg white lysozyme (EWL), Durafresh (DF) and grapefruit seed extract (GSE). Additionally, LCFS maintained its antimicrobial activity after heat treatment at $50^{\circ}C{\sim}95^{\circ}C$ and at pH values of 3~9. Moreover, LCFS inhibited the growth of E. coli and S. aureus in chicken breast meat. In conclusion, it is expected that LCFS, which contains both lysozyme and three organic acids, will be useful as a good natural preservative in the food industry.

라벤더와 로즈마리 에센셜 오일 나노에멀션의 항균 활성 (Antimicrobial Activity of Lavander and Rosemary Essential Oil Nanoemulsions)

  • Purpose: Essential oils are secondary metabolites of herbs and have antibacterial activities against foodborne pathogens. However, their applications for food protection are limited due to the hydrophobic and volatile natures of essential oils. Methods: In this study, essential oil nanoemulsions of rosemary and lavender were formulated with non-ionic surfactant Tween 80 and water using ultrasonic emulsification, and their antibacterial effects were determined. Results: The antibacterial activities of nanoemulsions were evaluated against 12 strains of 10 bacterial species, and significant antibacterial effects were observed against four Gram-positive and four Gram-negative bacteria but not against Streptococcus mutans and Shigella sonnei. In the disc diffusion test, the diameter of the inhibition zone proportionally increased with the concentration of nanoemulsions. Using cell turbidity measurement, minimum bactericidal concentration (MBC) of the nanoemulsions, which is the lowest concentration reducing viability of the initial bacterial inoculum by ${\geq}99.9%$, was significantly higher than the minimum inhibitory concentration (MIC) of the nanoemulsions. The largest bactericidal effects of lavender and rosemary essential oil nanoemulsions were observed against S. enterica and S. aureus, respectively. Conclusion: Nanoemulsion technique could improve antibacterial activity of essential oil nanoemulsions by increasing the solubility and stability of essential oils. Our findings shed light on the potential use of essential oil nanoemulsions as an alternative to chemical sanitizers in food protection.

Multiplex Polymerase Chain Reaction(PCR)법을 이용한 Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus의 다중동시검출 (Simultaneous Detection of Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus by Multiplex Polymerase Chain Reaction)

  • 본 연구는 국내 주요 식중독 원인균인 Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus를 동시에 검출 및 동정할 수 있는 simultaneous multiplex PCR방법을 개발하고자 하였다. S. aureus의 23s rRNA 유전자(482 bp), V. Parahaemolyticus의 toxR 유전자(368 bp), S. enterica subsp.의 invA 유전자(284 bp)를 특이적으로 검출 및 동정할 수 있는 3개 primer set 즉, STA-5F/STA-5R, ToxR-F/ToxR-R, 139/141을 구축하였으며, 그 결과 정제되어진 각 식중독 원인균의 genomic DNA를 template로 하여 세 균주 모두 10 pg까지 다중동시검출이 가능하였다. 생균수(CFU)와 상응되는 검출한계 결과로써 $10^1\sim10^2$ CFU/reaction의 검출한계를 보였으며 이는 즉, S. aureus $6.0\times10^4$ CFU/mL, S. enterica subsp. $9.5\times10^4$ CFU/mL, V. parahaemolyticus $6.1\times10^5$ CFU/mL의 검출한계를 나타내었다. 균체회수부터 agarose gel 상에서 검출 및 동정까지 3~4 hr의 시간 소요로 single tube 반응으로 세 식중독 원인균의 다중동시검출이 가능하였다. 또한 추가적인 연구를 통하여 세 식중독 원인균주의 검출을 위한 향상된 민감도를 가지는 multiplex PCR법 및 real time PCR을 이용한 다중동시검출법 개발을 위한 기초자료로서 활용 가능할 것이라 사료된다.

황금(Cutellaria baicalensis George) 추출물의 식중독성 미생물에 대한 항균효과 (Antimicrobial Effect of Cutellaria baicalensis George Extracts on Food-Borne Pathogens)

  • 본 연구에서는 식중독 유발세균에 대한 항균활성이 우수한 천연 항균성 물질을 검색하기 위해 예로부터 민간과 한방에서 널리 이용되어 온 황금을 각종 유기용매로 추출하여 식중독 유발세균에 대한 항균활성을 조사해 보았다. 황금을 methanol로 추출한 후, petroleum ether, chloroform, ethyl acetate를 이용하여 실온에서 각각 용매별로 계통 분획하고, 열수추출물을 얻은 후, 9종의 식중독 유발세균(Staphylococcus aureus, Bacillus cereus, Salmonella enteritidis, Shigella flexneri, Escherichia coli, Salmonella typhimurium, Shigella dysenteriae, Pseudomonas aruginosa, Shigella sonnei) 에 대하여 항균효과를 조사하였다. 황금 추i물의 농도별 항균 활성 검색에서는 황금의 ethyl acetate추출물이 가장 큰 항균 효과를 보였으며 S. aureus와 S. dysenteriae가 가장 민감하게 반응하는 균주였다. 황금의 ethyl acetate추출물과 사설초의 ethyl acetate 추출물을 혼합하여 항균력을 측정해 본 결과 두 추출물을 섞어 첨가했을 경우가 단독으로 사용했을 때 보다 상승효과를 나타내었다 또한 황금의 ethyl acetate추출물이 식중독 유발세균의 성장에 미치는 효과를 검정하기 위해 S. aureus및 S. dysenteriae의 배양백에 황금의 ethyl acetate 추물을 각각 5,000 ppm 농도로 첨가했을 때, S. aureus의 생육이 24시간 이상까지 억제됨을 관찰할 수 있었고, S. dysenteriae의 생육도 36시간까지 지연시킬 수 있었다. 이상의 결과 황금의 ethyl acetate추출물은 S. aureus와 S. dysenteriae의 성장을 억제하는 것을 알 수 있었다.
