Effects of antioxidant enzymes and bioaccumulation in eels (Anguilla japonica) by acute exposure of waterborne cadmium

  • This study was conducted to evaluate the acute effects of waterborne cadmium exposure on bioaccumulation and antioxidant enzymes in eels (Anguilla japonica) and to determine the median lethal concentration (LC50). Fish were exposed to different cadmium concentrations (0, 0.15, 0.30, 0.61, 1.83, 3.08, 3.67, 4.29, and 5.51 mg L-1) for 96 h. The LC50 of A. japonica to cadmium was 3.61 mg L-1. Cadmium accumulation generally increased in tissues with increasing waterborne cadmium concentrations. At ≥ 1.83 mg L-1 exposure, all tissues accumulated significant cadmium concentrations compared with the control group, in the order of kidney > liver > gill > spleen > muscle. Measurements of variation in actual cadmium concentrations showed that a reduction of the metal in experimental water was related to cadmium accumulation in tissues. As activity alteration of antioxidant enzymes for reactive oxygen species, superoxide dismutase and catalase activities increased at ≥ 0.61 mg L-1 significantly, glutathione peroxidase and glutathione S-transferase activities were not significantly changed. The results of this study suggest that acute exposure to waterborne cadmium is potentially fatal to A. japonica due to the metal's major accumulation in various tissues and the effect of antioxidant enzyme activity.

군위군 화산습지의 식생: 오리나무림을 중심으로 (Hwasan Wetland Vegetation in Gunwi, South Korea: with a Phytosociological Focus on Alder (Alnus japonica (Thunb.) Steud.) Forests)

  • 화산습지는 경북 군위와 영천 경계의 산정 분지 (표고 644~780 m)에 위치한다. Z.-M. 학파의 식물사회학적 방법으로 현장 식생조사가 이루어졌고, 현존식생도가 제작되었다. 단위식생의 출현종의 행동양식은 피도와 상대기여도를 이용하여 분석하였다. 버드나무하위군락, 전형하위군락, 산돌배하위군락으로 이루어진 오리나무-백당나무군락이 처음으로 기재되었다. 오리나무 우점림에 대한 선행 기재단위식생 (오리나무-진퍼리새군락, 오리나무-일본갈매나무군집, 버드나무-신나무군집 등)과의 군락분류학적, 군락동태학적 대응성이 논의되었다. 본 군락은 오리나무군강(Alnetea japonicae)에 귀속되고 산간 계류 선상지에서의 분지상(分地狀) 범람원 습생림으로 규정되었다. 화산습지의 잠재자연식생이고 증거 식생형으로서 식생보존등급 [I]의 자연식생으로 평가되었다. 입지의 수리수문 환경조건의 극적인 변화(건생화 및 부영양화)로부터 국가적 보호 전략이 요구되었다.

노루발풀(Pyrola japonica) 추출물의 Bacillus subtilis에 대한 항균활성 (Antimicrobial Activity of the Extract from Pyrola japonica against Bacillus subtilis)

  • 우리나라 전국 산지에서 쉽게 구할 수 있는 노루발풀을 건조시켜 80% ethanol 추출물과 여러 가지 용매 분획물의 미생물 증식 억제 효과에 대해서 검색하였다. 노루발풀의 80% ethanol 추출물은 그람 양성균 B. subtilis에서 농도 의존적으로 항균활성을 나타내는 반면 그람 음성균 E. coli에서는 활성이 나타나지 않았다. 노루발풀의 80% ethanol 추출물을 용매 분획하여 얻은 분획물 중 hexane층에서 항균활성이 나타나는 것을 확인하였다. 노루발풀의 항균활성 물질을 검색하기 위한 목적으로 hexane 분획물을 silica gel column chromatography, prep. TLC 및 HPLC를 실시하였으며 그 결과, peak I의 단일 물질이 노루발풀의 B. subtilis에 대한 항균활성을 나타내는 물질임을 확인할 수 있었다.

  • 2014년 9월부터 2015년 8월까지 국내 9개 지점에서 채집된 Anguilla japonica 454개체의 생물학적 특성을 조사한 결과, silvering index (SI) 구분과 GSI의 차이가 큰 pseudo-silver eel이 소양호에서 2개체(PS1~PS2)와 금강하구에서 1개체(PS3)가 확인되었다. Pseudo-silver eel은 외부형태 기준으로는 S2 단계였으나, GSI, 소화관지수 (Gut index) 및 안경지수 (Eye index)에서는 모두 yellow eel 단계인 Y1 및 Y2였고, 생식소발달단계와 난경에서도 Y2 단계였다.

황련 열수추출물을 처치한 인간 대장암 세포 SNU-81에서의 단백질 발현 변화 (Monitoring the Change of Protein Expression in Human Colon Cancer Cell SNU-81 treated with the Water-Extract of Coptis japonica)

  • Background : Anticancer effects of herbal medicine have been reported in various types of cancer, but the systematic approaches to explain molecular mechanism(s) are not established yet. Objective : To find the anticancer-effect and mechanism(s) of Water Extract of Coptis japonica (WECJ) colon cancer cell (SNU-81). Methods : We first selected 11 herbals, and anti-cancer effects of water-extracts from those herbals have been tested in human colon cancer cell line, SNU-81. Among the tested herbals, the WECJ significantly reduced proliferation of SNU-81. To establish a basis of understanding for anti-cancer mechanism, whole proteins have been obtained from SNU-81 harvested at 48 and 96 hrs after the treatment of WECJ, protein expression has been profiled by 2DE-based proteomic approach. Results : Various changes of the protein expression have been monitored, and most frequent dysregulation was found in the molecular chaperons including heat shock protein 90-alpha (Hsp90-alpha), 14-3-3 protein epsilon, T-complex protein 1 subunit alpha, protein disulfide-isomerase A3, and calreticulin. Interestingly, proliferation-associated protein 2G4 has been up-regulated, and it suggests the possible effect of Coptis japonica on ErbB3-regulated signal transduction pathway and growth control of human colon cancer cells. Conclusion : Based upon the present findings, the further study will focus on monitoring various cancer survival factors after artificial regulation of the proteins identified, and it would be the basis for the understanding of the Coptis japonica anti-cancer effect(s) at the molecular level.

GIS를 이용한 송지호 재첩서식의 적지선정 (Suitability Selection on Habitable Environments of Corbicula Japonica in the Songji Lagoon by GIS)

  • 본 논문은 송지호 재첩서식의 적지선정을 위한 방법에 대해 살펴본다. 수차례에 걸쳐 현장탐사와 시료채취를 통한 수질검사로 수집된 자료를 지리정보시스템(GIS)을 이용하여 분석하였다. 송지호의 재첩 서식환경의 특성을 파악하기 위해 수온, 염분, 용존산소 농도, PO4-P 농도를 활용하였다. 본 연구는 서식환경이 유사한 여타 석호 등에서 재첩 증식의 기초자료로 활용할 수 있을 것으로 기대된다.

금은화에서 분리한 Caffeic Acid의 신경세포보호 활성 (Neuroprotective Activity of Caffeic Acid Isolated from Lonicera japonica)

  • We previously reported that caffeic acid isolated from Lonicera japonica showed potent neuroprotective activities against glutamate injured neuronal cell death in primary cortical cells. In this study, we tried to confirm the neuroprotective activity in glutamate injured HT22 cells and elucidate mechanisms of neuroprotective action of caffeic acid. We used glutamate induced HT22 cell death as a bioassay system. The compound decreased reactive oxygen species increased by high concentration of glutamate treatment in HT22 cells. Also, Ca2+ concentration was decreased by this compound. This compound made mitochondrial membrane potential maintain to normal condition. This also affected anti-oxidative enzymes and glutathione contents. Treatment of this compound increased not only glutathione reductase and peroxidase to the control level and also amount of glutathione, an endogeneous antioxidant. These experimental results showed that caffeic acid isolated from L. japonica exerted potent neuroprotective activity through the anti-oxidative pathway.

  • Leaf blight symptoms and abnormal defoliation were found on Stachys riederi var. japonica, a medicinal plant, grown at Hongcheon, Gangwon province, Korea in 2007. A foliar nematode, Aphelenchoides fragariae was isolated from infected leaf tissues. The symptoms observed on host plant were leaf blotching, discoloration and browning of tissues, tan-colored interveinal necrotic lesions and large dead patches of necrotic tissues. This is the first report of A. fragariae occurring on S. riederi var. japonica.

The point of maximum weight off 1000 seeds and 100 seedling, germination speed and germination percentage were reached at 30 day. after anthesis. it would be considered as physiological maturity of Zoysia japonica seed. 2. The seeds moistures contents at physiological maturity was decreased in 18%. 3. Germination vigor deteriorated prior to viability. 4. The seeds were capable of germination in 3:4% at 10 days after anthesis. 5. Field deterioration of seeds occurer from 40 (lays after anthesis to 60 days and sprouted seedilng vigor was very poor.

호라비꽃대 chloroanthus japonica sieb. 의 성분에 관한 연구(I) 결정성분의 분리 및 항균작용 (Studies on Chemical Components of Chloranthus japonica Sieb.(I) Isolation of crystal substance and its antibacterial activity)

  • It has been believed that "Kup kee" (Chloranthus japonica) is good for boils, or dermatopathia, and enteric fever by the Korean folks. However, there is no scientific report on its chemical ingredients and its antibacterial activities so far. The roots of Chloranthus japonica were dried and pulverized. The pulverized powder was then extracted with methanol. Then an extract was obtained by following Stas Otto's method. finally a crystal fraction was obtained through Column Chromatography. Recrystalization from methanol gave white needle crystals, m.p. $198^{\circ}C.$ Generally this compound has shown strong antibacterial effects on Gram negative bacteria. It exerts most powerful effect on Salmonella typhi. on the other hand it shows weak effect on Inaba cholera.
