Molecular Cloning of the Sec61p ${\gamma}$ Subunit Homologue Gene from the Mole Cricket, Gryllotalpa orientalis

  • The Sec61 trimeric complex ($\alpha$,$\beta$, and ${\gamma}$ subunits) is one of the Sec-complex responsible for post-translational protein translocation across the endoplasmic reticulum membrane in diverse organisms. In this study, a cDNA encoding the Sec61p ${\gamma}$ subunit homologue was isolated from the cDNA library of the mole cricket, Gryllotalpa orientalis. Sequence analysis of a 442-bp cDNA clone showed it to contain an open reading frame of 68 amino acid residues consisted of 204-bp. The homologues of the gene were found in the GenBank database in a diverse organism including insect, mammals, fungi, and plants. The deduced amino acid sequence of Sec61p ${\gamma}$ subunit homologue of the mole cricket showed the highest homology to the gene of the singly known insect, Drosophila melanogester (93% identity), and the least homology to that of the baker's yeast, Saccharomyces cerevisiae (37.2%). Phylogenetic analysis also confirmed a close relationship between the insect Sec61p ${\gamma}$ subunit homologues of G. orientalis and D. melanogester. Hydropathy analysis of the cricket mole and published other data suggested that the hydrophobic segment close to C-terminus is predicted to be the putative membrane anchor, Multiple alignment of the Sec61p ${\gamma}$ subunit homologue among several organisms showed the presence of several conserved domains including the conserved proline at position 28.

The assessment of conservation value for agricultural pond wetland using the weighted function of modified RAM (modified RAM의 기능별 가중치 부여를 통한 농촌지역 연못형습지의 보전가치 평가)

  • The pond wetlands in agricultural landscape are important natural resources that carry out the function of bio-diversity conservation. However, recently, those have been gradually embedded as their utility value were disappeared. And, the assessment methods used for pond wetlands are insufficient. Therefore, this study was conducted to examine the conservation value of pond wetlands by using the modified RAM, and present the improvement of assesment methods. The study sites, a total of 32 pond of 4 types by land use, were selected on the basis of Ramsar Convention. Through the analysis of precedent studies, the weighted 8 functions were adjusted. According to the assessment results, pond wetlands made the largest contribution to Fishery and Herpetile Habitat function. In addition, it also made large contribution to Floral Diversity, Wildlife Habitat, and Water Quality Protection function. On the other hand, it made a small contribution to Aesthetics and Recreation, Runoff Attenuation, Shoreline /Stream Bank Protection, and Flood/Storm Water Storage function due to the characteristics of small-scale pond wetlands. In the assessment of 8 functions, house type showed the worst assessment result, and mountain type showed the best assessment result. It is thought that those are due to land use type in terms of vicinity. 10 items among 52 of the modified RAM showed the same assessment results in all land use types. Accordingly, it is required to be deleted and modified the assessment method. On the other hand, it is required to add age, interference, and water use to the assessment method. It is thought that these results can be utilized for the development and modification of assessment methods focused on pond wetlands in rural area.

Analysis of Phylogenetic Relationship of 30 Cultivars of Korean Mulberry (Rosales: Moraceae) in Korea

  • This study was carried out to understand phylogenetic relationships of the 30 mulberry cultivars converved in Korea based on the ITS rDNA region, and they were compared to 40 reference sequences from GenBank. The size and the G+C content of the ITS rDNA gene regions from the 30 Korean mulberry cultivars and 40 reference sequences varied from 612-630 bp and 58.19-61.62%, respectively. Based on the results of the comparative phylogenetic analysis of the ITS rDNA regions of the 30 Korean mulberry cultivars and 40 reference sequences, they were divided into three groups (Group 1, 2, and 3) and two subgroups (Group 1A and 1B within Group 1). The sequence lengths of the Korean mulberry cultivar numbers 1-26 and 27-30 were 615 bp and 616 bp, respectively. At 205 bp location of ITS1 rDNA region, the cultivar numbers 1-26 contain the nucleotide thymine but the cultivar numbers 27-30 contain the nucleotide adenine. In addition, the insertion of the nucleotide adenine at 206 bp location was found only in the four Korean mulberry cultivars (numbers 27-30). Based on these sequence information and phylogenetic result, the 30 Korean mulberry cultivars were identified as M. alba and M. australis. This study will contribute to the construction of genetic database constructions and accurate variety identifications for unidentified mulberry varieties in Korea.

Analysis of Farmland Price Determinants in Parcel-level Using Real Transaction Price of Farmland (농지실거래가격을 활용한 필지 단위 농지가격 결정요인 분석)

  • The primary purpose of this study is to identify various factors that affect farmland prices according to changes in the actual transaction price of farmland over the past decade, and to use this to derive policy implications for price stabilization. To this end, the farmland price model are constructed at the parcel level in the case area (Namwon-si, Jinju-si). The analysis method is based on the Hedonic price function, and the OLS and the quantile regression are used for the parcel level model. As a result of estimating the parcel level farmland price model in the case area, the larger the parcel area, the lower the farmland price, and the higher the farmland price outside the agricultural promotion area. It was found that there was a price difference according to the type of special purpose areas, and the location characteristics showed some differences across the cities. The farmland price models presented in this study are suitable for identifying the factors affecting farmland prices, and are expected to be highly utilized in that it is possible to construct flexible variables suitable for regional characteristics.

A Comparison of Social Capital Tools Developed by International Institutes and Nations (국제기구 및 국가 개발 사회자본 측정도구 비교)

  • Objective: The purpose of this study was to identify and compare social capital measurement tools for the convenient use in public health studies. Method and result: This study examined and compared social capital tools developed individually by the World Bank, the OECD, the United States, United Kingdom, Canada, Australia, Ireland, and Korea. A comprehensive framework was constructed with six conceptual dimensions and sixteen indices. The six dimensions included Membership, Network, Trust, Information and communication, Social and civic participation, and Social cohesion and exclusion, which connoted the structural, cognitive, bonding, bridging, operative, and output elements. The indices of each tool were respectively matched to the indices of the comprehensive framework. The comprehensive tools were Integrated Questionnaire for the Measurement of Social Capital (SC-IQ) of the World Bank with 27 questions and the European Social Survey (ESS) of the OECD with 80 questions. Conclusion: The SC-IQ should be utilized in public health studies due to its simplicity yet comprehensiveness as a social capital tool. The ESS should also be considered as a comprehensive tool.

Secondary Metabolites and Morphological Diversity in the Leaves of Perilla Landrace from Korea

  • Screening and identification of genetic resources based on their phytoconstituents and morphological characters potentially provide baseline data for researchers, breeders, and nutraceutical companies who wish to formulate a nutrient-dense diet and health beneficial supplement. Thus, we evaluated the amount of total phenolic content and major phenolic compounds; examined if phenolic compounds could be used as distinguishing factors for perilla genetic resources; and investigated the relation between some quantitative and qualitative morphological characters with the contents of phenolic compounds in 360 accessions obtained from National Agrobiodiversity Center gene bank, Jeonju, Korea. Total phenolic content (TPC) was estimated using Folin-Ciocalteu colorimetric assay. Individual phenolic compounds were determined using an Ultra-High Performance Liquid Chromatography system equipped with Photodiode Array detector. Considerable variations were observed in TPC (7.99 to 117.47 mg GAE/g DE), rosmarinic acid (RA) (ND to 19.19 mg/g DE), caffeic acid (CA) (ND to 0.72 mg/g DE), apigenin-7-O-diglucuronide (ADG) (ND to 1.24 mg luteolin equivalent (LUE)/g DE), scutellarein-7-O-glucuronide (SG) (ND to 4.32 mg LUE/g DE), and apigenin-7-O-glucuronide (AG) (ND to 1.60 mg LUE/g DE). RA was the most dominant phenolic compound in most accessions (95.3%) followed by SG. The adaxial leaf color was light green, green and dark green in 13.8%, 65.0%, and 21.1 % of the accessions, respectively. 78.8% of the accessions had light green color at the abaxial side with the remaining being described as green. Most of the accessions (96.9%) were cordate shape, the remaining being eclipse. Intensities of green pigment at abaxial and adaxial leaf surfaces were correlated with contents of individual phenolic compounds and TPC whereas leaf length and width had no correlation with TPC, CA and RA, and negatively correlated with ADG, AG, and SG. Leaf shape was not related with content of phenolic compounds, color of leaves, or the length or width of leaves. Accessions IT57426, IT157434, IT267710, and IT267712 which contained relatively high contents of TPC and major phenolic compounds (RA and SG) could be used for further research in breeding and bioassay test. Our study result showed the contents of total phenolics and individual phenolic compounds along with the morphological characters could be useful distinguishing factors for perilla genetic resources.

A radio-tracking study of home-range of the Korean raccoon dog(Nyctereutes procyonoides koreensis) (원격무선추적을 통한 한국너구리(Nyctereutes procyonoides koreensis)의 행동권 연구)

  • The main aim of this study is to estimate home-range of the Korean raccoon dog(Nyctereutes procyonoides koreensis) at a rural area of Gurye in the southern part of South Korea. A radio-tracking was regularly carried out on 4 raccoon dogs for 2 days every 2 months in 2006. During the days, the radio-tracking was usually conducted every 1-3 hours through day-time to night-time. Among the 4 raccoon dogs, 2 individuals(a permanent breeding pair) could be extensively tracked for 5 to 7 months, including all 4 seasons. The result showed that total home-range sizes of the pair were 0.732 $km^2$ and 0.373 $km^2$ for 100% minimum convex polygons(MCP) and 100% kernel(K), respectively, during the monitoring period. Mean home-range sizes of the 2 raccoon dogs were 0.035-0,688 $km^2$ and 0.012-0.341 $km^2$ for MCP and K, respectively. Yearly home-range sizes of the male and female were similar to each other. However, home-range sizes of the raccoon dogs between day-time and night-time were quite different. Furthermore, the raccoon dogs showed a much broader home-range size in spring, summer and fall than in winter season. Finally, the pair had a broad overlapping home-range(about 70-95%), and 1 core area and 4 different feeding areas.

Predicting the Suitable Habitat of Invasive Alien Plant Conyza bonariensis based on Climate Change Scenarios (기후변화 시나리오에 의한 외래식물 실망초(Conyza bonariensis)의 서식지 분포 예측)

  • This study was conducted to predict the changes of potential distribution for invasive alien plant, Conyza bonariensis in Korea. C. bonariensis was found in southern Korea (Jeju, south coast, southwest coast). The habitats of C. bonariensis were roadside, bare ground, farm area, and pasture, where the interference by human was severe. Due to the seed characteristics of Compositae, C. bonariensis take long scattering distance and it will easily spread by movement of wind, vehicles and people. C. canadensis in same Conyza genus has already spread on a national scale and it is difficult to manage. We used maximum entropy modeling (MaxEnt) for analyzing the environmental influences on C. bonariensis distribution and projecting on two different RCP scenarios, RCP 4.5 and RCP 8.5. The results of our study indicated annual mean temperature, elevation and temperature seasonality had higher contribution for C. bonariensis potential distribution. Area under curve (AUC) values of the model was 0.9. Under future climate scenario, the constructed model predicted that potential distribution of C. bonariensis will be increased by 338% on RCP 4.5 and 769% on RCP 8.5 in 2100s.

Calculation of the Yield of Bank Filtration by Using the Horizontal Collector Wells (방사형 집수정에 의한 강변여과수 산출량 산정에 관한 연구)

  • The horizontal collector well is used to treat some weak points of the vertical well in the bank filtration site. In this study two empirical formulas(Milojevic and Petrovic) are selected to examine the applicability for calculating the yield of the horizontal collector. And they are compared with the compute simulation results for multiple wells. Milojevic empirical formula which considers the conditions such as aquifer, well location, the diameter of screen etc. is more applicable than Petrovic formula. Draw-down characteristics of horizontal collector was well simulated by using the computer simulation for multiple wells. The results are well agreement with Milojevic formula, and the draw-down and the retention time of the horizontal collector can be controlled by adjusting the angle of lateral screens.

Assessment of Immune Quality and Pathogen Contamination of Colostrums Collected from Colostrum Banks in Korea (초유은행에서 수거한 초유의 병원체 오염과 면역수준의 평가)

  • Because colostrum is considered to be the sole source of passively acquired maternal antibodies for calves, newborn calves must consume colostrum to gain disease resistance during their early years of life. Storage of surplus colostrum from dairy cows right after calving and feeding newborn calves in deficiency of colostrum to assure adequate uptake of IgG for protection of the calf has been a common practice in the bovine production. In the current study, 35 colostrums were randomly collected from 3 colostrum banks located in different regions of Korea and monitored for general bacterial contamination and major bovine pathogens. Immunoglobulin concentrations and BVDV-specific antibodies were also determined to evaluate the immune quality of the colostrums. Moderate to severe bacterial contamination (up to 72,000,000 CFU/ml) was observed in most of the colostrums collected from colostrum banks. General immune quality of the colostrums was under the satisfactory level since most of the colostrums contained less than 50 g/L of IgG, which is the minimum concentration for good quality colostrums. Therefore, colostrum for colostrum bank should be collected at the first 2-3 post-partum milkings according to proper harvesting and handling procedures to guarantee the safety and quality of colostrum. In addition, it was recommended that colostrum should be heat-treated before frozen and stored in the bank because pasteurization at $63^{\circ}C$ for 30 min was very effective reducing the risk of disease transmission without causing significant degradation of immunoglobulins.
