  • The scope of e-Commerce process becomes wider as the emphasis on the enterprise collaboration grows. It has expanded from its initial settings of order management or billing processes to cover various collaborative processes in the company's value-chain. In order for those collaborative e-Commerce processes to be successful, corresponding business processes should be fully supported by standard bodies. The RosettaNet consortium, one of the most representative international B2B standard bodies, has steadily provided new PIPs to support those expansions. Since individual RosettaNet PIPs correspond to unit tasks that are executed separately in or between companies, multiple PIPs have to be integrally used to properly handle larger business cases. RosettaNet implementation, however, has suffered from the lack of standard guidelines or deliverables to refer under this multi-PIP environment. In this research, a contents-based document routing system is implemented. By applying this routing system to the RosettaNet e-Logistics program where multi-PIP environment is inevitable, we verified our contents-based document routing system is effective to support multi-PIP environment flexibly.

  • Mathematical models have become increasingly popular in both research and management problems involving flow and transport processes in the subsurface. Rosetta is a program to estimate unsaturated hydraulic properties from surrogate soil data such as soil texture data and bulk density. Models of this type are called pedotransfer functions (PTFs) as an alternative measurements since they translate basic soil data into hydraulic properties. These functions may be either measured directly or estimated indirectly through prediction from more easily measured data based using quasi-empirical models.

  • Hydraulic conductivity is the rate of water flux on hydraulic gradient. The van Genuchten Mualem (VGM) model is frequently used for describing unsaturated state of soils, that is composed with the function of soil water potential and soil water content and requests various parameters. This study is to get the value of VGM parameters used Rosetta computer program based on neural network analysis method and to calculate VGM parameters. VGM parameters included Ko(effective saturated hydraulic conductivity), ${\theta}r$(residual soil water content), ${\theta}s$(saturated soil water content), L, n and m. The unsaturated hydraulic conductivity at 10 kPa was calculated by using Rosetta program. Unsaturated hydraulic conductivities of 17 soil series at 1, 3, 5, 7 kPa were also obtained by applying saturated hydraulic conductivity by disk tension infiltrometer based on Gardner and Wooding's equation. Water flow at the water potential of 3 kPa was very low except Namgye, Hagog, Baegsan, Sangju, Seogcheon, Yesan soil series. Unsaturated hydraulic conductivity at 1 kPa showed the highest value for Samgag soil series and was in order of Yesan, Hwabong, Hagog and Baegsan soil series. Those of Gacheon, Seocheon and Ugog soil series were very low. When the value by VGM was compared with the value by disc tension infiltrometer, there was a tendency with exponential function to soils without gravel but there was no tendency to soils including gravel. Conclusively, it would be limited that VGM model for unsaturated hydraulic conductivity analysis applies to Korean agricultural land including gravel and having steep slope, shallow soil depth.

Gene Cloning and Enzymatic Properties of Thermostable Laccase from Thermus thermophilus HJ6 (Thermus thermophilus HJ6 유래 내열성 laccase의 유전자 클로닝 및 효소학적 특성)

  • The gene encoding Thermus thermophilus HJ6 laccase (Tt-laccase) was cloned, sequenced, and comprised of 1,389 nucleotides encoding a protein (462 amino acids) with a predicted molecular mass of 51,049 Da. The deduced amino acid sequence of Tt-laccase showed 99.7% and 44.3% identities to the Thermus thermophilus HB27 laccase and Synechococcus sp. RS9917 laccase, respectively. Tt-laccase gene was expressed as a fusion protein with six histidine residues in E. coli Rosetta-gami (DE3) cells, and the recombinant protein was purified to homogeneity. UV-Vis spectrum analysis revealed that the enzyme has copper atoms, a type I Cu(II) and a type III binuclear Cu(II). The optimum pH for the oxidation of guaiacol was 5.0 and the optimum temperature was $90^{\circ}C$ The half-life of heat inactivation was about 120 min at $90^{\circ}C$ The enzyme reaction was inhibited by sodium azide, L-cystein, EDTA, dithiothreitol, tropolone, and kojic acid. The enzyme oxidized various known laccase substrates, its lowest $K_m$ value being for 4-hydroxyindole, highest $k_{cat}$ value for syringaldazine, and highest $k_{cat}/K_m$ for guaiacol.

Functional Characterization of the ${\alpha}$- and ${\beta}$-Subunits of a Group II Chaperonin from Aeropyrum pernix K1

  • We isolated and functionally characterized the ${\alpha}$- and ${\beta}$-subunits (ApCpnA and ApCpnB) of a chaperonin from Aeropyrum pernix K1. The constructed vectors pET3d-ApCpnA and pET21a-ApCpnB were transformed into E. coli Rosetta (DE3), BL21 (DE3), or CodonPlus (DE3) cells. The expression of ApCpnA (60.7 kDa) and ApCpnB (61.2 kDa) was confirmed by SDS-PAGE analysis. Recombinant ApCpnA and ApCpnB were purified by heat-shock treatment and anion-exchange chromatography. ApCpnA and ApCpnB were able to hydrolyze not only ATP, but also CTP, GTP, and UTP, albeit with different efficacies. Purified ApCpnA and ApCpnB showed the highest ATPase, CTPase, UTPase, and GTPase activities at $80^{\circ}C$. Furthermore, the addition of ApCpnA and ApCpnB effectively protected citrate synthase (CS) and alcohol dehydrogenase (ADH) from thermal aggregation and inactivation at $43^{\circ}C$ and $50^{\circ}C$, respectively. In particular, the addition of ATP or CTP to ApCpnA and ApCpnB resulted in the most effective prevention of thermal aggregation and inactivation of CS and ADH. The ATPase activity of the two chaperonin subunits was dependent on the salt concentration. Among the ions we examined, potassium ions were the most effective at enhancing the ATP hydrolysis activity of ApCpnA and ApCpnB.

Effect of machining precision of single ceramic restorations on the marginal and internal fit (단일 도재 수복물의 가공 정밀도가 변연 및 내면 적합도에 미치는 영향)

  • Purpose: The purpose of this study was to evaluate the machining precision and the marginal and internal fit of single restorations fabricated with three types of lithium disilicate ceramic blocks and to evaluate the correlation. Materials and methods: Single restorations were designed using a CAD software program. The crown designed model file was extracted from the CAD software program. Three types of lithium disilicate blocks (Rosetta; HASS, IPS e.max CAD; Ivoclar vivadent, VITA Suprinity; VITA) were milled using a milling machine. For the fabrication of the crown scanned model file, the intaglio surface of the restoration was digitized using a contact scanner. Then, using the three-dimensional inspection software (Geomagic control X; 3D Systems), the process of the overlap of the crown designed model and the scanned model and 3-dimensional analysis was conducted. In addition, the marginal and internal fit of the crowns was evaluated by a silicone replication method. The difference among three types of single ceramic crown was analyzed using a Kruskal-Wallis H test, and Spearman correlation analysis was performed to analyze the correlation between machining precision and fitness (α=.05). Results: There was a significant difference in the machining precision and the marginal and internal fit according to the type of ceramic block (P<.001). In addition, the machining precision and the marginal and internal fit were positively correlated (P<.001). Conclusion: The marginal fit of crowns fabricated according to the types of ceramic blocks was within the clinically acceptable range (< 120 ㎛), so it can be regarded as appropriate machining precision applicable to all clinical as aspects in terms of the marginal fit.
