Enhanced Phytoremediation of Trichloroethylene - Contaminated Soil by Poplar-Colonizing Recombinants

  • Indigenous bacteria from poplar roots (Populus mnadensis var. eugenei, 'Imperial Carolina') and Southern Californian shrub rhizospheres as well as two tree-colonizing Rhizobium strains (ATCC 10320 and 35645) were genetically engineered to express constitutively and stably toluene o-monooxygenase (TOM) from Burkholderia cepacia G4 by integrating the torn locus into the chromosome. The poplar and Rhizobium recombinants degraded trichloroethylene (TCE) at 0.8-2.1 nmol/min.mg protein (initial TCE concentration, 10u M) and competitive against the unengineered hosts in wheat and barley rhizospheres for one month (colonization at 1-23 $\times$ 10$^{5}$ CFU/cm root). In addition, six of these recombinants colonized poplar roots stably and competitively with populations as high as 79 $\pm$ 12% of all rhizosphere bacteria after 28 days (0.2-31 $\times$ 10$^{5}$ CFU/cm root). Furthermore, five of the most-competitive poplar recombinants (e.g., Pb3-1 and Pb5-1 which were identified as Pseudomonas PsK) retained the ability to express TOM for 29 days as 100 $\pm$ 0% of the recombinants detected in the poplar rhizosphere had constitutive expression of TOM.

  • Generally, the digital controller has many advantages such as high precision, robustness to electrical noise, capability of flexible programming and fast response to the load variation. In this study, we have established proper mathematical equivalent model of Brushless DC (BLDC) motor and estimated the motor parameter by means of the back-emf measurement as being the step input to the controlled target BLDC motor. And the validity of proposed estimation method is confirmed by the test result of step response. As well, we have designed the reasonable digital controller as a consequence of the root locus method which is obtained from the open-loop transfer function of BLDC motor with hall sensor, and the determination of control gain for variable speed control. Here, revised Ziegler-Nichols tuning method is applied for the proper digital gain establishment, and the system stability is verified by the frequency domain analysis with Bode-plot and experimentation.

  • This paper deals with the modeling and experiment of a robot system for force/impact control performance. The basic model is composed of a direct drive motor, servo amplifier, link, force sensor and environments. Based on the developed model, the stability of the whole system was analyzed via root locus method. For the force control, integral force compensation with velocity feedback method shows the best performance of all the explicit force control strategies. In dealing with impact, PID position control and the explicit force control method were implemented. Instead of add more damping to the robot system by velocity feedback, we developed a new passive damping method and it was also applied to enhance the damping characteristic of the system.

  • In this paper, we analyze the effects of scaling factors on the performance of a fuzzy logic controller(FLC). The quantitative relation between input and output variables of FLC is obtained by using a qualsi-linear fuzzy model, and an approximate transfer function of FLC is dervied from the comparison of it with the conventional PID controller. Then we analyze in detail the effects of scaling factor using this approximate transfer function and root locus method. Also we suggest an on-line tuning method for scaling factors which employs an sample performance function and a variable reference for tuning index.

  • The end-point position control of a flexible manipulator is a non-minimum phase system. The PD feedback of the end-point position is not stable in contrast with that of the hub jangle. However, the system can be stabilized conditionally by the feedback of both the hub rate angle and the end-point position. Even in the non-minimum system, the LQG/LTR control law is more systmatic controller design method than the classical control law which uses a root-locus technique.

  • This paper presents a comprehensive study on the stability of several control schemes for the induction motor driven by current source inverters. A stator voltage-controlled current source inverter drive system without a speed sensor is investigated in order to find appropriate control schemes, which are primarily based on direct or, alternatively, indirect frequency control scheme. The overall control systems with either voltage control loop or current and voltage control loops provided in addition to each frequency control scheme are analyzed by utilizing the root locus method and simulated by computer to illustrate the validity of this analysis.

  • The paper describes the parameter tuning of power system stabilizer (PSS) for a power plant based on hybrid system modeling. The existing tuning method based on bode plot and root locus is well applied to keep power system stable. However, due to linearization of power system and an assumption that the parameter ratio of the lead-lag compensator in PSS is fixed, the results cannot guarantee the optimal performances to damp out low-frequency oscillation. Therefore, in this paper, hybrid system modeling, which has a DAIS (differential-algebraic-impusive-switched) structure, is applied to conduct nonlinear modeling for power system and find optimal parameter set of the PSS. The performances of the proposed method are carried out by time domain simulation with a single machine connected to infinite bus (SMIB) system.
