Hydroysis of Rice Straw with Trichoder-ma viride TO4 Cellulase

  • Rice Straw was delignified by autoclaving with 1% NaOH solution at $121^{\circ}C$ for one hour and was disintegrated by a Wiley mill to 60 mesh. This substrate was saccharified with cellulase produced by Trichoderma viride T04 in solid culture me-dium. The rate and extent of hydrolysis were both increased when high enzyme concentration and low substrate concentration were employed. The original cellulose was treated with 0.19 FPA unit for three hours and followed by the second treatment for the same period with the same concentration of enzyme after washing. By doing this the hydrolysis rate at the second stage could increase four folds of that unwashed. The same experiment with 0.32 FPA unit yielded two folds suggesting an end-product inhibition on the recaction system. The extent of hydrolysis however, could not be in-creased by this process.

  • As a process to utilize agricultural residues, simultaneous hydrolysis-fermentation (SSF) was compared with fermentation of enzymic hydrolyzate using koji cultures of Trichoderma sp. KI 7-2 and a thermotolerant yeast Saccharomyces cerevisiae NCYC 716. Cellobiose was not detected in SSF broth whilst 15 mg/$m\ell$ of the disaccharide was found in enzymic hydrolysate of rice straw using the same enzyme source. It was found that converting glucose to ethanol in SSF process reactivated the cellobiase activity, which is inhibited by the accumulation of glucose in enzymic hydrolysis process. Cutting milled rice straw was fermented as effectively as ball milled one in SSF process. From tile results discussions are made on the product inhibition mechanism of cllulolytic enzyme system.

Physiological Functionality of Gugija Products and an In Vivo Examination on Anti-hypertension Effects (구기자 가공품의 생리기능성 및 항고혈압 활성 검증)

  • To develop a new Gugija(Lycium chinensis Mill) product having increased value, the physiological functionality of various commercial Gugija products were investigated. In addition, an in vivo study was performed using spontaneous hypertensive rats(SHR) to examine the anti-hypertension effects of products. The results showed that antioxidant activity was highest in the methanol extract of Gugija leaf pickle(97.7%), and anti-hypertensive angiotensin I -converting enzyme(ACE) inhibitory activity was 80.4% in Gugija doenjang(soybean paste) water extract. Anti-cholesterolemia HMG-CoA reductase inhibitory activity was highest in the methanol extract of Gugija rice cake(66.1 %). However, SOD-like activity was below 30% in most products; and fibrinolytic activity was not detected or was very weak. Ultimately, we selected Gugija tea and Gugija wine as superior anti-hypertensive Gugija products, and subsequent in vivo testing was performed using SHR, comparing the tea and wine to Gugija fruit. Among them, the Gugija fruit demonstrated the best anti-hypertension effects in SHR.

Physicochemical Properties of Endosperm Starch and Breadmaking Quality of Rice Cultivars (쌀 품종의 배유 이화학적 특성에 따른 제빵 적성 비교)

  • The physicochemical properties of rice endosperm from five rice varieties and effects of milling on baking bread properties of rice flour were investigated. Five rice varieties exhibited different level of amylose content. The ratio of longer amylopectin chain length to the distribution of endosperm starches was the highest in Goami3. According to the RVA measurement of rice flours, the pasting temperatures of Seolgaengbyeo and Goamibyeo were lower than those of the other rice varieties. There were differences in the changes of swelling power of rice flour under increasing temperature. Each rice flour sample for bread-making was processed into two different particle size by using an air-classification mill, and significant differences were observed among cultivars in the average particle size and damaged starch content of rice flours. Rice flour of Seolgaengbyeo with fine particle size showed the highest value in specific volume after baking.

  • This study was aimed at the development of the rice product by fermentation of saccharified rice solution with Bifidobacterium. To optimize the preparation of saccharified rice solution for Bifidobacterium fermentation, various pretreatment conditions were established. Grinding for 30seconds by an impact mill was more efficient than any other grinding schemes tested. The preheating before gelatinization showed a positive effect for efficient saccharification, and its optimal conditions were at $60^{\circ}C$ for 45 min. The optimum gelatinization conditions were at $100^{\circ}C$ for 40 min. The optimum levels of enzymes for saccharification of rice were 0.135 unit/g rice powder for ${\alpha}-amylase$ and 3.375 unit/g rice powder for glucoamylase, respectively. The physico-chemical properties of the fermented product by a fastidious Bifidobacterium showed a great potential for a functional rice product. However, an improvement on its flavor was required, which might be achieved by the addition of various fruits and vegetables.

Improving Grain Quality by Controlling Top Dressing of Nitrogen Application Ratio (일품벼 도정 및 쌀 품질 향상을 위한 분시비율 개선연구)

  • This study was carried out to improve the rice grain quality of Ilpum cultivar, especially to reduce the chalky rice rate by controlling top dressing of nitrogen application ratio at the tillering and panicle initiation stage. In the rice plant growth, the number of tiller and leaf area index of maximum tillering stage were influenced by basal dressing and top dressing at tillering stage; however those of heading stage were mainly influenced by top dressing at panicle initiation stage. In yield and yield components, they were influence by the top dressing at panicle initiation stage and the rice yield was increased by percentage of the top dressing. The protein content of rice grain was increased as the rate of top dressing at panicle initiation stage was increased. The rate of chalky and damaged rice was remarkably increased as the top dressing ratio was increased, resulting in reducing the rate of head rice rate. As the results, the characteristics of milling and rice quality was improved when the amount of top dress was reduced as 50% at the panicle initiation stage, and the yield of head rice also increased.

  • The purpose of this study is to examine the Western perspective on the food and food culture of Modern Times in Korea (from the late of Joseon Dynasty until Japanese colonial era). Literature and written records were analyzed. This analysis revealed that the heart of the mill in this period involved rice, and that a common beverage was sungnyung made from boiled scorched rice (in contrast to tea as the common beverage in Japan or China). The most important subsidiary food in Joseon was vegetables, especially Kimchi. Westerners viewed Kimchi as a smell symbolizing Joseon and their meal times. Even though both Kimchi and cheese are fermented food, just like Westerners could not stand the smell of Kimchi, Koreans viewed the smell of cheese unpleasant. Westerners viewed German sauerkraut as Western food counterpart to Kimchi, as sauerkraut is also fermented food made of cabbage. Regarding the eating of dog meat in Joseon, most Westerners viewed it as brutal; however some interpreted it as a difference in food culture. In addition, the eating of raw fish and its intestines felt crude to Westerners. The biggest difference between Joseon's food and Western food was that Joseon had no dairy products and no sugar. The most highly preferred fruit for Westerners was the persimmon, and ginseng was already widely recognized and recorded as a medicinal plant. Joseon's desserts were also favorably evaluated. In contrast, the excessive gluttony, heavy drinking, and unsanitary conditions in Joseon were problems pointed out in many records.

Extraction of Micro Filler from Bio-waste Material (Bio waste 소재로부터의 마이크로 필러 추출)

  • This paper explain about the development of environmental friendly, low cost and stable supply material i.e., rice husk and shell were used as micro incorporating bio waste filler. Those were processed by ball mill and analyzed through micro observation by FE-SEM, EDS and particle size distribution. The obtained filler was mixed with epoxy resin for the manufacturing of CFRP composite and study tensile properties. In EDS analysis main contents of rice husk and rice husk ash are C, O and Si. When rice husk was burned C and Si ration were increased. Shell powder has C, O and Ca. It caused $CaCO_3$ from shell. Surface weighted mean of rice husk powder is $6.19{\mu}m$ and volume weighted mean is $14.77{\mu}m$. And it has rod type particles which caused hair and husk structure parts. Surface weighted mean of rice husk ash powder is $1.55{\mu}m$ and volume weighted means is $8.20{\mu}m$. Surface weighted mean of shell powder is $2.53{\mu}m$ and volume weighted mean is $5.79{\mu}m$. The tensile decreased with increasing the content of micro filler in CFRP composites. In case of rice husk, the significant decrement of tensile strength was observed. and in case of shell powder, there is no effect of changes take place in tensile strength.

A Historical Study on the Achitectural Cooking and Storing Spaces in Traditional Korean Houses (한국(韓國)의 전통적(傳統的) 식생활공간(食生活空間))

  • In the life style of the neolithic age, cooking and sleeping space was in one room dugout without differentiation of spaces, so to say one room system. Ro(a kind of primitive fire place) was used for both cooking and heating. However, in the early part of the Iron Age, the uses of Ro were separated into two major uses of cooking and heating. Especially, L-shaped Kudle(an unique under floor heating structure of Korea) was invented for the new system of heating, extending to Koguryo Period. The life style of Koguryo Dynasty could be seen through the mural paintings of tombs. For these mural paintings contain of cooking space(Kitchen), meat storage, and mill house drawing, we can recognize that houses were specialized many quaters according to their function. Also a kitchen fuel hole for preparing food was built without relation to L-shaped Kudle. But during Koryo Period, Kudle could be set up all over the room, the so-called Ondol(the unique Korean panel heating system) settled down. From this development of heating system, room could be adjacent to kitchen, and kitchen fuel hole and heating fuel hole be onething. This system was developed with variety, extending to Chosun Dynasty. In the period of Chosun Dynasty, a kitchen was made close to an Anbang(Woman's living room), and Anbangs Ondol was heated by the warmth of a cooking fireplace. Therefore Handae Puok, outer kitchen was used in summer. As for its storage space, it was seen that there were a pantry near the kitchen and a store house constructed as an independant building. In the latter, it was devided into a firewood storage, a Kimchi storage, and a rice storage, etc. Especially it is a unique feature that 'Handae-Duyju', an outer rice chest which keeps rice, was constructed as an isolated small building.

Effect of Different Milling Methods of Rice Flour on Quality Characteristics of Jeungpyun (쌀가루의 제분방법이 증편의 품질 특성에 미치는 영향)

  • The characteristics of Jeungpyun were investigated with different milling methods of rice flour. The moisture content of Jeungpyun was similar to that of the rice flours. In the color of Jeungpyun, L-value was increasing but a and b-value were decreasing as its moisture content was increasing. The analysis on the texture of Jeungpyun showed that Jeungpyun by dry milling indicated the higher value in hardness and the lower value in springiness, cohesiveness and chewiness than Jeungpyun by wet milling. The test of the sensory characteristics for Jeungpyun showed that whiteness, moistness, springiness and chewiness was higher for Jeungpyun by wet milling. But the softness was lower. Overallquality of Jeungpyun by wet milling was higher than Jeungpyun by dry milling. Generally, Jeungpyun prepared by Pin-mill was the best in quality. In particular, Jeungpyun by wet milling was better for the added 80% of water and Jeungpyun by dry milling was better for the added 100% of water.
