Biodegradation of Diesel by Rhodococcus fascians in Sand Column (Rhodococcus fascians를 이용한 모래 컬럼내 디젤유 분해)

  • Contamination of soils, groundwater, air and marine environment with hazardous and toxic chemicals is major side effect by the industrialization. Bioremediation, the application of microorganism or microbial processes to degrade environmental contaminant, is one of the new environmental technologies. Because of low water solubility and volatility of diesel, bioremediation is more efficient than physical and chemical methods. The purpose of this study is biodegradation of diesel in sand by using Rhodococcus fascians, a microorganism isolated from petroleum contaminated soil. This study was performed in the column containing sand obtained from sea sides. Changes in biodegradability of diesel with various flow rates, inoculum sizes, diesel concentrations, and pH were investigated in sand column. The optimal condition for biodegradation of diesel by R. fascians in sand column system was initial pH 8 and air flow rate of 30 mL/min. Higher diesel degradation was achieved at larger inoculum size and the diesel degradation by R. fascians was not inhibited by diesel concentration up to 5%.

  • To diagnose lily fasciation, lily bulbs showing fasciation were collected from several greenhouses in Jeju Island, South Korea. Bacteria were isolated from the lily bulbs and amplified with both primers for fasA in plasmid and for putative glycosyltransferase epsH gene in chromosome of Rhodococcus fascians. Three bacterial isolates were detected with the P450 primer set and identified as R. fascians by NCBI blast analysis. Twelve bacterial isolates were identified as R. fascians using RS02785 primer set, including the three bacterial isolates identified as the same pathogen using the P450 primer set. Pathogenicity of these bacterial strains identified as R. fascians was demonstrated. Apparent symptoms were observed on wounded lily leaves after inoculation with each bacterial suspension whereas no symptom was found on lily leaves treated with H2O. Furthermore, bacteria re-isolated from wounded sites were identified as R. fascians. Based on the results, these two sets of primers are recommended for quarantine of R. fascians.

Bioremediation is the application of microorganism or microbial processes to degrade environmental contaminant. Because of low water solubility and volatility of diesel, bioremediation is more efficient than physical and chemical methods. The objective of this study is biodegradation of diesel in sea water by using Rhodococcus fascians which is isolated petroleum-contaminated soil. R. fascians was cultured on sea water containing diesel to determine the diesel degradability. Changes in biodegradability of diesel with various inoculum sizes, diesel concentrations, initial pH, and culture temperature were analyzed by TPH analysis using gas chromatography. The inoculum size 2% was effective for biodegrdation of diesel in sea water by R. fascians. When diesel concentration was 5%, the growth of cell was inhibited by the toxicity of diesel. The optimal temperature and initial pH for degradation of diesel in sea water were $27^{\circ}C$ and pH 8.

  • Microorganisms isolated from petroleum contaminated site were tested for their ability to grow on JP-8 by culturing them on the culture medium that contains JP-8 as a carbon source. The microorganism which grew on JP-8 containing minimal salt medium was separated and identified as Rhodococcus fascians. Changes in JP-8 biodegradation of R. fascians that was isolated from petroleum contaminated site was investigated with various inoculums sizes, JP-8 concentrations, medium pHs, and culture temperatures. The amount of JP-8 was analyzed by TPH using Gas Chromatography.

  • The environmental contamination by organic pollutants is a widespread problem. The most widely distributed pollution can be attributed to oil contamination. Bioremediation, the use of microorganism or microbial processes to degrade environmental contaminant, is one of the new technologies. The objective of the present study is to study the degradation of JP-8 in soil by microorganism. The degradation of JP-8 was analysed by TPH using gas chromatography. Rhodococcus fascians isolated from the petroleum contaminated site was applied for the degradation of JP-8 in the soil column system. Air flow rate of 30 ml/min was sufficient to degrade JP-8 in the soil column as much as 70% of JP-8 in the soil column. The addition of nitrogen source resulted in the increase in JP-8 degradability to 75% of JP-8 and the C:N ratio for JP-8 degradation was 100:10.

Partial Purification and Characterization of Limonoate Dehydrogenase from Rhodococcus fascians for the Degradation of Limonin

  • An extracellular limonoate dehydrogenase was purified 10-fold from a cell-free extract of Rhodococcus fascians by ammonium sulfate precipitation, dialysis, and ultrafiltration. This purified dehydrogenase catalyzed the conversion of limonoate to 17-dehydrolimonoate. The enzyme showed optimum activity at pH 8.0 and $40^{\circ}C$, with $K_m$ value of 0.9$\muM$, and requires Zn ions and sulfhydryl groups for catalytic action. The enzyme activity was inhibited by $Hg^{2+}\;and\;NaN_3$ ions. The degradation of limonin (66%) in Kinnow mandarin juice was successfully demonstrated with partially purified limonoate dehydrogenase. With scale-up preparation of limonoate dehydrogenase, a successful debittering operation of fruit juices appears feasible.

Environmental Factors Affecting on Shrimp Cultivation and Bacterial Examination in Shrimp Aquaculture

  • Cultivation environment for effective production of shrimp in shrimp aquaculture, Institute of Marine & Fishery, Soonchunhyang University located at Tae-An, Chung-Nam Province, with the ultimate aim of probiotics development, was monitored. Several environmental factors including dissolved oxygen, water body temperature, pH, salinity, $NH_4-N$, $NO_2-N$, $NO_3-N$, chlorophyll a affecting on the growth of shrimp were examined. Concentrations of $NH_4-N$, $NO_2-N$, and $NO_3-N$ dissolved in water samples were determined to 0.024-0.034 mg/L, 0.03-0.02, and 0.004-0.009 mg/L, respectively. Chlorophyll a content was examined in the range of 0.002-0.118 $ug/m^3$. In order to understand the distribution of different bacteria in water samples collected in shrimp aquaculture, bacteria were isolated and enumerated on the marine agar plates. Total number of bacteria were increased to approximately $6.5\;{\times}\;10^4$. Thirteen predomonant bacteria were isolated and identified. As the results of BIOLOG test of the isolates, these bacteria were identified as Corynebacterium nirilophilus, Clavibacter agropyri, Sphingomonas adhaesiva, Brevundimonas vesicularis, Vibrio parahaemolyticus, Pseudomonas bathycetes, Vibrio tubiashii, Sphingomonas macrogoltabidus, Rhodococcus, rhodochrous, Burkholderia glumae, Corynebacterium urealyticus, Rhodococcus fascians, Psychrobacter immobilis, respectively. Further work will stimulate the elucidation of pathogenicity, corresponding bacteria related to environment and probiotics, providing good information for effective production of shrimp.
