Scientific Analysis of Formulation Theory of Chungpesagan-tang; The purgative Action of Chungpesagan-tang

  • To analyze scientifically the fundamental formulation theory and drug interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke, water extracts of herbal medicines with Rhei Rhizoma were administered into mice and the purgative action was measured. Water extracts of Raphani Semen or Cimicifugae Rhizoma with Rhei Rhizoma had more potent purgative activity than Rhei Rhizoma alone, although Raphani Semen alone displayed no purgative action. However, Platycodi Radix and Puerariae Radix inhibited the purgative action of Rhei Rhizoma. When Rhei Rhizoma with Raphani Semen or Cimicifugae Radix was extracted with water, sennoside A content in the water extract was increased. However, Puerariae Radix and Platycodi Radix decreased the content of sennoside A in the extracted water. The transforming rate of sennoside A of Rhei Rhizoma was inhibited by Puerariae Radix and Platycodi Radix. The transforming rate of sennoside A of Chungpesagan-tang to rheinan-throne by human intestinal microflora and its purgative activity were similar to those of Rhei Rhizoma. Finally we thought that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

Review of Pharmacological Effects of Coptidis Rhizoma and its Bioactive Compounds (황련(黃連)과 구성 생화합물의 약리작용에 대한 고찰)

  • Objectives: Coptidis Rhizoma is one of the most popular and multi-purpose herbs in traditional medicine. The purpose of this study was to contribute to research and applications of Coptidis Rhizoma in clinic, by analyzing and reviewing international studies on its chemical compositions and pharmacological effects. Methods: This study analyzed 344 articles published from 2000 to 2010 in PubMed, Refworks, Riss, and KTKP. The search keywords were "Coptis chinensis", "Coptis japonica", "Coptidis Rhizoma", "huanglian" and "huanglian in Chinese". From them, we selected 114 articles which met our inclusion criteria. Results: This study reviewed 114 articles on Coptidis Rhizoma and its active components in terms of 'Active components', 'Experimental studies', 'Clinical studies', 'Industrial use' and 'Side Effects/Toxicity'. Conclusions: The active components of Coptidis Rhizoma are berberine, coptisine, epiberberine, palmatine, jateorrhizine, magnoflorine, worenine, etc. It is reported that Coptidis Rhizoma and its active components have anti-inflammatory, antibacterial, antitumor, and antioxidant activity, and cardiovascular, hepatoprotective, antidiabetic, neuroprotective, gastrointestinal, pain relieving, discharge phlegm and metrocyte proliferation effects. Moreover, we found that Coptidis Rhizoma can be used for bath preparation, cosmetic products and as a natural antimicrobial substance.

A Study on the Standardization of Ligustici Rhizoma and Angelicae Tenuissimae Radix including the Comparison for the Biological Activity on the Active Ingredients (Ligustici Rhizoma(고본(藁本))와 Angelica tenuissimae Radix(한국고본(韓國藁本)뿌리)의 규격화(規格化) 및 유효성분(有效成分)의 진통효과(鎭痛效果) 비교(比較)에 관한 연구(硏究))

  • Major separation for the active ingredients and characterization of chemical properties in conjunction with screening test on animal were performed in order to analyze and standardize Ligustici Rhizoma or Angelicae Tenuissimae Radix as an important oriental herbal medicine for antiphlogistic or an important oriental herbal medicine for antiphlogistic or an anodyne. Furthermore the structure, composition and contents of ingredients for essential oil in Angelicae Tenuissimae Radix(Suckpo, Korea) were determined by means of Ge/MS followed by screening test on Z-ligustilide(82%) known as major ingredient as well as butylidenephthalide collected by HPLC with normal phase semiprep-column. The total active ingredient in Ligustici Rhizoma from China or Angelicae Tenuissimae harvested at Choonyang(Kyungnam, Korea), Jungsun(Kangwon, Korea), Suckpo(Kyungnam Korea), Youngchun(Kyungnam, Korea) have been determined showing higher abundant for three times on the product in Korea compared to that in China. In addition, the major component in Ahgelicae Tebyussunae Radux extract was found to be Z-ligustilide(70-80%) which is very different from that in Ligustici Rhizoma senkyunolide(39%) as major species. For screening test of Ligustici Rhizoma or Angelicae Tenuissimae Radix extracts toward the target animal, the efficiency has been shown the similarity on both extracts. Taking into account the level of ingredient, the total efficiency may be three times higher on Angelicae Tenuissimae Radix in Korea compared to Ligustici Rhizoma in China. As a result of present study, it is preferable to distinguish between Ligustici Rhizoma and Angelicae Tenuissimae Radix for better usage of oriental herbal medicine because of very different composition and abundant in spite of their similar screening effect.

  • In oriental medicine, Rhei Radix et Rhizoma & Gardeniae Fructus are frequently used materials for anti-inflammatory effect especially urinary tract disease and for neuropsychosis. Bang-Yak-Hap-Pyun(方藥合編) is widely used for clinical herbal prescription book. In Bang-Yak-Hap-Pyun(방약합편), Rhei Radix et Rhizoma is used 0.8g~16g. and Gardeniae Fructus is used 1.2g~6g. The most important fact is that the prescriptions used Rhei Radix et Rhizoma & Gardeniae Fructus at the same time are 4(Yang-Gyuk-San, Pal-Jung-San, Sa-Chung-Hwan, Suk-Gyul-Myung-San. The ratio of Rhei Radix et Rhizoma & Gardeniae Fructus are 2:1, 1:1, 1:1, 1:2 ). The number of prescription Rhei Radix et Rhizoma & Gardeniae Fructus in Bang-Yak-Hap-Pyun is total 45. 1 in sang-tong(上統), 5 in jung-tong(中統), 39 in ha-tong(下統). The group of mostly used disease is about urinary tract. The ratio of Rhei Radix et Rhizoma in Bang-Yak-Hap-Pyun is 3.33%~57.14%. And the ratio of Gardeniae Fructus is 3.47%~98%. The mostly quoted medical literature what prescription included Rhei Radix et Rhizoma & Gardeniae Fructus in Bang-Yak-Hap-Pyun are Dong-Yi-Bo-Gam and Man-byung-hoe-chun.

Effect of Drynariae Rhizoma in RANKL-induced Osteoclast Differentiation (골쇄보가 RANKL에 의해 유도되는 파골세포의 분화에 미치는 영향)

  • Bone homeostasis is regulated by the balance between bone-resorbing osteoclasts and bone-forming osteoblasts. Osteoporosis, rheumatoid arthritis and periodontal disease are related with up-regulated osteoclast formation and its activity. Gol-Swae-Bo(Drynariae Rhizoma) is widely used on skeletal disease. In this study, we sought to examine the effect of Drynariae Rhizoma in RANKL-induced osteoclast differentiation. The extract of Drynariae Rhizoma inhibited RANKL-induced osteoclast differentiation in a dose dependent manner without cytotoxicity. receptor activator of nuclear factor-${\kappa}B$ ligand(RANKL) mediated $I{\kappa}B$ degradation in bone marrow macrophages(BMMs). However, the extract of Drynariae Rhizoma inhibited RANKL induced $I{\kappa}B$ degradation in BMMs. And mRNA expression of OSCAR, TRAP, c-Fos and NFATc1 was suppressed by the extract of Drynariae Rhizoma. Moreover, the extract of Drynariae Rhizoma inhibited the protein expression of NFATc1 and c-Fos induced by RANKL. After all the analysis, these results suggest that Drynariae Rhizoma may be good candidate of medicine in the treatment of bone-related disease.

Study on the Applications of Rhizoma dioscoreae Mainly Blended Prescription in Dongeuibogam (『동의보감(東醫寶鑑)』 중(中) 산약(山藥)이 주약(主藥)으로 배오(配伍)된 방제연구(方劑硏究))

  • Objectives : This report describes 45 studies related to the use of Rhizoma dioscoreae main blended prescription from Donguibogam. Methods : The following conclusion were reached through investigations on the prescriptions that use Rhizoma dioscoreae as a key ingredient. Prescriptions that Rhizoma dioscoreae was taken as a monarch drug are utilized for 18 therapeutic purposes, for example, internal injury, stool disease, pediatric disease. In paticular, 24% of prescriptions appear in the chapter of internal injury, and 18% of those appear in the chapter of stool. Results : Prescriptions that utilize Rhizoma dioscoreae as the main ingredient are used in the treatment of indigestion, diarrhea and paralysis. Rhizoma dioscoreae is used in pathogenic factors such as water, water with damp, and used in pathology related to stomach system. Conclusions : The dosage of Rhizoma dioscoreae is 8li(about 0.29 gram) to 3jeon(about 11.25 gram), however 1jeon(about 3.75 gram) has been taken the most for clinical application. Sagunjatang and Siljangsan is the most useful base prescription which use the Rhizoma dioscoreae as the main ingredient.

The Protective Effects of Dioscoreae Rhizoma on the Exposure to UVA of MEF cells (산약의 Mouse embryonic fibroblast cell에 대한 자외선 손상 방어효과)

  • Purpose: This study was to determine the protective effects of Dioscoreae Rhizoma on the Mouse Embrio Fibroblast (MEF) cells exposed to the ultraviolet rays(UVA). Methods: The samples were assigned randomly to five groups; control group without any treatments, UVA group exposed only to UVA, DR group exposed only to the Dioscoreae Rhizoma, UVA-DR group exposed to UVA before being treated with the Dioscoreae Rhizoma, and DR-UVA group treated with the Dioscoreae Rhizoma before being exposed to UVA. The survival rate of cells, metabolic rate of cells, transformation of nucleus within cells, alteration of cell cycle, effects on the apoptosis, the change of the amount of protein related to cell cycle were measured in order to determine the cell protective effects of the Dioscoreae Rhizoma on each group. Results: 1. DR-UVA group has more cell protective effects compared to the UVA group in all experiments, indicating that the Dioscoreae Rhizoma protects skin from UVA physically and chemically. 2. UVA-DR group shows more efficiency compared to UVA group in rapid recovery of damaged cell and leading highly damaged cells to apoptosis, preventing the expression of abnormal cells. Conclusions: Dioscoreae Rhizoma has effects of protecting MEF cells from UVA, of recovering cells damaged by UVA, and of prohibiting the expression of abnormal cells.

  • Herbal medicinal interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke was investigated. Chungpesagan-tang had more potent purgative activity than Rhei Rhizoma alone. When each Chungpesagan-tang composing herbal medicine with Rhei Rhizoma was extracted with water, water extract of Raphani Semen with Rhei Rhizoma had the most potent purgative activity. However, Raphani Semen had not purgative activity. When Rhei Rhizoma with Raphani Semen was extracted, sennoside A content in the water extract was increased. The transformation of sennoside A of Rhei Rhizoma to rheinanthrone by human intestinal microflora was weakly induced by Raphani Semen. These results suggest that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

  • Angioedema is a localized transient swelling of sudden onset in the face, lips, tongue, hand, feet, respiratory system and gasteointestinal system. Angioedema is classified as allergy, hereditary, idiopathic or induced by medication as like aspirin, nonsteroidal anti-inflammatory agent, opiates, antibioics and angiotensin-coverting enzyme. Arisaematis Rhizoma Ingestion grows up every place in Korea, dries humidity and eliminates engorgement. so Arisaematis Rhizoma Ingestion was used to paralytic, hemiplegia, bell's palsy, tetanus. but Arisaematis Rhizoma Ingestion is toxic, can not be used pregnant women. We experienced a case of angioedema induced by Arisaematis Rhizoma Ingestion in a 52-years-old male patient who bited a Arisaematis Rhizoma Ingestion. We treated the patient based on traditional oriental medicine case report. As a result, the symtoms disappeard rapidly.

The Effect of Cnidii Rhizoma Herbal-acupuncture at $GB_{34}$(Yangleungchean) on Hyperlipidemia in Rats (양릉천 천궁약침이 흰쥐의 고지혈증에 미치는 영향)

  • Objectives & Methods : The purpose of this study is to observe the effects of Cnidii Rhizoma Herbal solution at $GB_{34}$(Yangleungchean) on hyperlipidemia in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological change of liver and aorta. Results : 1. In the Cndii Rhizoma Herbal-acupuncture group, The levels of total cholesterol were significantly decreased, and the ratio of HDL to Total cholesterol, Phospholipid/total cholesterol were significantly increased as compared with those of the control group. 2. In the Cndii Rhizoma Herbal-acupuncture group, HMG-CoA reductase activity was significantly decreased than those of the control group NP(needle prick) group and saline group. 3. In the Cndii Rhizoma Herbal-acupuncture group, phathohistological change of liver was suppressed as compared with the control group. Conclusions : From the above results, it is suggested that Cndii Rhizoma Herbal-acupuncture at $GB_{34}$ has a therapeutic effect on hyperlipidemia.
