Protective Effect of R. palmatum on 1-Methyl-4-phenyl-l,2,3,6-tetrahydropyridine (MPTP)-induced Neurotoxicity in Mice (생쥐의 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-유도 신경독성에 대한 대황의 보호효과)

  • The protective efficacy of Rheum palmatum water extract on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism was studied in C57BL/6 mice. In order to demonstrate neuroprotective effect of R. palmatum extract, animals were administered intraperitoneally with the water extract (100 or 200 mg/kg/day) for 14 days, and MPTP (10 mg/kg/day) was injected subcutaneously into the mice for the first 6 consecutive days from the beginning 1 hr before R. palmatum extract treatment. All animals were measured the several neurobiochemical markers such as dopamine level and monoamine oxidase B (MAO-B) activity in various regions of brain. The treatment of mice with R. palmatum extract was confirmed recovery effect on MAO-B activity in the cerebellum and the cerebral cortex. R. palmatum extract was attenuated the MPTP-induced depletion of substantia nigra dopamine. The contents of MDA, a marker of lipid peroxidation, in brain tissues (cerebellum and cerebral cortex mitochondria) were decreased significantly by R. palmatum extract. These results suggest that R. palmatum water extract plays an effective role in attenuating MPTP-induced neurotoxicity in mice. This protective effect of R. palmatum might be estimated the result from the inhibitory activity on monoamine oxidase B and the enhancement of antioxidant activity.

  • Objectives: To examine the relaxational response to the water extract of Rheum palmatum L. and Rheum undulatum L. on rat thoracic aorta and abdominal aorta. Methods: Segments of thoracic aorta and abdominal aorta obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. Results : We found that the thoracic aorta segments responded to the water extract of genus Rheum with a dose-dependent vasorelaxation. At $10^{-4}$ M 5-HT, the maximal contraction force was 93.5% of the maximum KCI-response. The 5-HT induced contractions at $10^{-4}$ M were inhibited by 86.4% and 62.1 % after addition of the high concentrations of R. palmatum root (RPR) and leaf (RPL) and R. undulatum root (RUR) and leaf (RUL). At 10 mg/ml RPR and RUR, the relaxational response at thoracic aorta and abdominal aorta with and without endothelium were 86.4%, 83.2%, 85.8%, and 62.1% of the maximum 5-HT induced contraction. Conclusion: Our result showed that RPL and RUL induced dose-dependent vasorelaxation on rat thoracic aorta and abdominal aorta, and that RPL and RUL roots have more potent effects than the leaves.

Vasodilation Effect of the Water Extract of Alcohol Processed Rheum palmatum L. in Rat Thoracic Aorta (주침 장엽대황이 백서의 흉부대동맥 혈관이완에 미치는 영향)

  • We have examined the relaxational response to the water extract of Rheum palmatum L.(ERP) and water extrc alcohol processed Rheum palmatum L.(ARP) in isolated thoracic aorta from sprague dawley (SD) rat. Rat thoracic aort investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to ERP and ARP investigated in vessels precontracted with 5-hydroxytryptamine(5-HT). We found that the thoracic aorta segments respo to ERP and ARP with a dose-dependent vasorelaxation. We found that ; The thoracic aorta segments responded to I and ARP with a dose-dependent vasodilation. The 5-HT induced contraction at 10/sup -4/M were inhibited by 71.7% and a: after addition of the 0.01 g/mL water extract of ERP and ARP. The 5-HT induced contraction at 10/sup -4/M were inhibite 100% after 10/sup -3/M emodin. The concentration of emodin was 0.027% and 0.098% in ERP and ARP. In condusion, ERP ARP induced relaxation in the isolated rat thoracic aorta were composed of dose-dependent relaxation. and it has pi vasodilation.

  • Rheum palmatum L. has been used for treatment of hypertension, lipemia, and paramenia in the oriental gerbal medicines for a long time. We have examined the relaxational response to the water extract of Rheum palmaum L. in isolated thoracic aorta from sprague dawley (SD) rat in the presence and absence of endothelium. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to Rhizoma Rhei were investigated in vessels precontracted with 5-hydroxytryptamine. We found that the ghoracic aorta segments responded to the water extract of Rheum palmatum L. (ERP) with a dose-dependent vasorelaxation. We found that 1.The thoracic aorta sehments responded to ERP with a dose-dependent vasodiliation. 2.The 5-HT induced contraction at 10$\^$-4/M were inhibited by 85.8% after addition of the 0.1 g/mL water extract of ERP. 3. The 5-HT induced contraction at 10$\^$-4/ M with and without endothelium were inhibited by 86.4% and 85.8% after addition of the 0.1g/mL ERP. 4. After pre-treatment of the thoracic aorta with 10$\^$-4/M N$\^$G/-monomethyl-L-arginine(L-NMMA), inducible niric oxide synthase inhibitor, the vessels has not response to the contraction. In conclusion, ERP induced reaxation in the isolated rat thoracic aorta were composed of dose-dependent relaxation. and it has potent vasodilation.

Isolation and Characterization of an Antibacterial Substance from Rheum palmatum for Treatment of Bacterial Vaginosis (대황으로부터 세균성 질염 치료를 위한 항균성 물질의 분리 및 특성)

  • Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.

Packagng of Fresh Curled Lettuce and Cucumber by Using Low Density Polyethylene Films Impregnated with Antimicrobial Agents (항균소재를 함유시킨 저밀도폴리에틸렌 필름에 의한 상추와 오이의 포장)

  • Low density polyethlene(LDPE0 films of 50${\mu}{\textrm}{m}$ thickness were fabricated with addition of antimicrobial agents of Rheum palmatum extract, Coptis chinensis extract, sorbic acid and Ag-substitude inorganic zirconium matrix in 1% concentration. The films were compared in physical properties, tested in antimicrobial activity against some selected microorganisms on the agar plate medium and then applied for packaging fresh curled lettuce and cucumber to preserve their qualities. The films with Rheum palmatum extract, Coptis chinensis extract, and Ag-substituted inorganic zirconium matrix did not show any antimicrobial activity on the disk test against Escherichia coli, Staphylococcus aureus, Leuconostoc mesenteroides, Saccharomyces cerevisiae, Aspergillus niger, Aspergillus oryzae, Penicilluium chrysogenum, while film with sorbic acid did against E. coli, S. aureus and L. mesenteroides. The added antimicrobial agents changed the color and light transmittance of the films, but did not affect their mechanical tensile strength, heat shrinkage and wettability. For the packaged curled lettuce and cucumber stored at 5$^{\circ}C$ and 1$0^{\circ}C$, all the LDPE films impregnated with antimicrobial agents showed the reduced growth of total aerobic bacteria in the vegetables compared with control film without any additive until it reached the level around 108/g. They did not give any negative effect on other quality attributes during storage.

  • This experiment was designed to investigate the effect of Rheum palmatum(RHP) on the Alzheimer's disease. The effects of RHP extract on amyloid precursor proteins(APP), acetylcholinesterase(AChE), glial fibrillary acidic protein(GFAP) mRNA of PC-12 cell treated by $A{\beta}\;and\;IL-1{\beta},\;IL-6,\;TNF-{\alpha}$ mRNA of THP-1 cell treated by LPS and AChE activity of PC-12 cell lysate treated by $A {\beta}$and behavior of memory deficit rats induced by scopolamine and mice glucose, uric acid, AChE activity of memory deficit rats induced by scopolamine were investigated, respectively. The results were summarized as follows ; 1. RHP extract suppressed APP, AChE, GFAP mRNA in PC-12 cell treated by $A{\beta} $. 2. RHP extract suppressed $IL-1{\beta} $, IL-6 $TNF-{\alpha}$ mRNA in THP-1 cell treated by LPS. 3. RHP extract suppressed AChE activity in cell lysate of PC-12 cell treated by $A{\beta}$. 4. HP extract increased glucose, decreased uric acid and AChE significantly in the serum of the memory deficit rats induced by scopolamine. 5. RHP extract group showed significantly inhibitory effect on the memory deficit of mice induced by scopolamine in the experiment of Morris water maze. According to the above results, it is suggested that RHP extract might be usefully applied for prevention and treatment of Alzheimer’s disease and memory deficit symptom.

  • The antimicrobial extracts of rheum palmatum and coptis chinensis root as well as grapefruit seed extract(GFSE) were applied to dipping treatment for keeping qualities of cucumbers, respectively, which were then packed in low density polyethylene(LDPE) films incorporated with 1 % antimicrobial extracts and stored $10^{\circ}C$. Dipping and packaging in the antimicrobial agents suppressed the growth of putrefactive microorgani는 and the decay ratio of cucumbers. In addition, the loss ratio of ascortic acid content and their weight was decreased during the sotrage of cucumbers. Consequently, the combined method of dippinf and packaging in antimicrobial agents turned out to be superior to dipping treatment or film-packaging in the view point of decay ratio and the keeping qualities of cucumbers.

A Study on the Antigastritic Effects of Rheum Species Extracts and Their Active Components (대황 추출물의 항위염 작용과 유효성분에 관한 연구)

  • Rheum tanguticum has long been used in oriental medicine as antipyretics, analgesics, anti-inflammation, aperient, hypertension medicine and medicine for skin disease. This study has investigated the effectiveness of defense for gastritis making use of Rheum tanguticum and its similar plants, Rumex cripus, Rheum officinale, Rhem palmatum and Rheum undulatum. Chysophanol, chrysophanol-8-O-glc, Desoxyrhaponticin desoxyrhaponti-genin, emodin, isorhaponticin, 2-methoxy-4-hydroxyanthraquinone-5-O-glc, physcion, pirace-tannol-3'-O-glc, resveratrol, rhaponticin and rhapontigenin are used as the components of Rheum tanguticum. In HCl ethanol-induced gastritis in rat, the most effective extract was 70 percent ethanol which is of Rheum tanguticum, showing the inhibition of 91.8 percent to the gastric lesions. 70% ethanol extract of Rhem palmatum and Rumex cripus shown inhibition of 75.6 percent and 73.2 percent, respectively. This effectiveness is considered as acid-neutralizing capacity, anti-H.pylori and anti-oxidant activity. 70% ethanol extract of Rheum tanguticum and its component, piracetannol-3'-O-glc exhibited higher free radical scavenging activity than others. These results suggest that Rheum species extracts and their active components could be utilized for the treatment of gastritis. Furthermore, these results are expected to contribute the standardization with objectivity and reliability for Rheum species, medicinal herbs. In addition, it can contribute to the prevention of indiscriminate distribution of imitation, and the rising rate of dependence on imports of medicinal herbs, and mixing prevention of low-quality goods.

Evaluation on Toxicity of Habitual dose of Extract of Rheum Palmatum through Single Blinded Pilot Study (단일맹검시험을 통한 대황(大黃) 상용량(常用量)의 독성작용 평가)

  • This study is aimed to test physiological and hematological actions of Rheum palmatum through clinical pilot study optimized for usual oriental medicine prescription. Thirty-one cases were finally collected and the sample extract 100 $m{\ell}$ of Rheum palmatum (ERP) was administered two times in a day during 3 days and checked with blood CBC test, urinalysis, liver function test, abdominal X-ray as well as general diagnostic process of oriental medicine; pattern identification, assessment of shapes and constitution. The total toxic effective rate of ERP was 9.68% in 3 cases of the whole in case of consistent uneasy state in the subjective symptomatic assessment and aggravation of blood and urine examination. The relevant diagnostic factors were so-eum constitution and essence type subject, carapaces species, lung type in the aspect of Jisan shape theory. On the other hand, the positive effective rate of ERP was 29.03% manifesting improvement of vital signs or present illness and no abnormal changes of blood chemistry and urinalysis and simple abdomen radiology. The relevant diagnostic factors were tae-eum constitution, energy type subject, aves species, heart and liver type in Jisan's shape theory. And the other cases were manifesting no specific change through the administration of ERP. The ratio of the numbers of decrease and increase was 44:5 in the tenderness and hardness of abdominal palpation. But if the pattern identification doesn't comply with the aim of rhubarb application despite of constipation and abdominal pain, the tendeness and biochemical report was shown in abnormal change. The mean number of diarrhea by ERP was $16.77{\pm}6.95$ during 3 days after administration, and the frequent areas of abdominal pain were lower>middle>upper in order, and it meant the target site of ERP too. Besides, the toxic reaction against ERP was expressed highly in case of decrease in blood cell count and hemoglobin, hematocrit having blood deficiency syndrome. Likewise, the toxicity of ERP was influenced by pattern identification manifesting present disease condition and diagnostic factors of four constitutions, Jisan's shape theory collaterally. In conclusion, evaluation of herbal toxicity in order for using as a clinical guideline, various diagnostic pattern information and shape features like the above should be studied together with other pharmacologic toxicology test for the future.
