Multiple Gonadotropin-Releasing Hormone Neuronal Systems in Vertebrates

  • Gonadotropin-releasing hormone (GnRH) was originally isolated as a hypothalamic peptide that regulates reproduction by stimulating the release of gonadotropins. Using comparative animal models has led to the discovery that GnRH has a more ancient evolutionary origin. Durinq evolution GnRH peptide underwent gene duplication and structural changes to give rise to multiple molecular forms of GnRHs. Mammalian GnRH initially considered to be the sole molecular form, is now grouped as a family of peptides along with GnRH variants determined from representatives in all classes of vertebrates. Vertebrate species including primates and humanshave more than one GnRH variant in individual brains; a unique GnRH form in the forebrain and chicken IIGnRH in the midbrain. Furthermore, several species of bony fish have three molecular variants of GnRH: salmon GnRH sea-bream GnRH and chicken II GnRH. Also, it has been shown that in addition to the olfactory placodes and the midbrain, there is a third embryonic source of GnRH neurons from the basal diencephalon in birds and fish, which might be true for other vertebrates. Therefore, comparative animal models like fish with discrete sites of expression of three molecular variants of GnRH in individual brains, could provide insight into novel functions of GnRH variants, conservation of gene regulation, and mechanisms governing reproduction in vertebrates.

Rhizopus속의 염색체에 관한 연구 1 (Chromosomal Studies on the genus of Rhizopus I)

  • This experiment was designed to elucidate the life cycle of 7 species (Rh.nigricans, Rh. delemar, Rh.oryzae, Rh.acidus, Rh.tritici, Rh. formosaensis and Rh. japonicus) in genus Rhizopus isolated from Korean soil, so as to seize the appropriate stage for detecting their chromosomal number and nuclear size under the method of thin layer slide culture using modified Rogers(1965a) medium. The results are summarized as the folowings ; 1. The haploid chromosome number are found 16 in Rh. japonicus are 8, respectively. 2. Comparing the 7 species of Rhizopus with each other, it may be concluded that the basic haploid chromosome number of genus Rhizopus distributed in Korean soil are 8 and that Rh. nigricans is double of the basic hapolid chromosome number (n = 16). Besides them, the other two species (Rh. tritici and Rh. formosaensis) are believed aneuploids.

  • 인삼은 우수한 약리 활성 작용을 보이는 전통적인 약초이다. 본 연구에는 프로바이오틱스 Lactobacillus helveticus KII13과 Pediococcus pentosaceus KID7 균주를 진세노사이드(ginsenoside) 함량을 증가시키기 위해 조 인삼 추출물을 발효시켜 진세노사이드를 형질전환 시키는데 사용되었다. 발효삼 추출물의 TLC(Thin-layer chromatography) 분석 결과, 5일간의 발효 후 주요 사포닌인 진세노사이드 Rg3, Rh1 및 Rh2로 변환되는 것으로 나타났다. HPLC 분석을 수행하여 주요 및 미량 진 세노사이드를 정량화하였다. 3일째에는 Rg3가 나타나고, 5일째에는 Rh2 및 Rh1이 나타난다. L. helveticus KII13과 P. pentosaceus KID7의 공동 배양은 주요 진세노사이드(Rb1과 Rg1)를 미량 진세노사이드(Rg3, Rh2, Rh1)로 전환시키는 것을 학인하였다.

우의 난포낭종에 대한 HCG 및 GnRH 제제의 치료효과 (Therapeutic Effect of Human Chorionic Gonadotrophin(HCG) and Gonadotrophin Releasing Hormone(GnRH) on Cows with Ovarian Follicular Cyst)

  • A total of 600 Holstein cows in Chonnam province was examined to make a diagnosis on the ovarian follicular cyst. By clinical signs and rectal examination, 57 cows were found to have ovarian follicular cyst. Attempts were made to treat the cows which had ovarian follicular cyst with GnRH, HCG respectively. The results obtained were summarized as follows : 1. The rates of estreous induction with GnRH or HCG were 91.4%, 77.2%, respectively. The GnRH treated group was showed significantly higher than HCG treated group. The mean days from the GnRH or HCG treated to estrum were 25.1 and 23.5 days, respectively. 2. The Conception rates with GnRH or HCG treatment were 78.2% and 76.5%, respectively. 3. Services per conception with GnRH or HCG treatment were 1.5 and 2.1 respectively. 4, Days from GnRH or HCG treatment to concept were 38.2 and 45.8 days, respectively. 5. Intramuscular injection with GnRH and intraovarian injection with HCG were revealed the most effective routes in all the other routes.

  • NaY 제올라이트에 Pt/Rh비를 달리하여 이온교환법으로 제조한 Pt-Rh촉매의 특성을$^{129}Xe$-NMR과 EXAFS 방법으로 특성조사를 하였다. $^{129}Xe$-NMR과 EXAFS 자료는 PtRh 이원금속 클러스터의 표면으로 Rh 원자들이 모이는 것으로 나타났다. 모사 자동차 배기가스를 사용하여 희박, 과농 및 양론조건에서 CO, HC 및 $NO_x$ 전환율을 측정하였으며, Pt-Rh/NaY(Pt/Rh=1) 이원금속 촉매가 다른 촉매들보다 세가지 오염물질에 대한 반응활성이 높게 나타남을 알 수 있었다.

한우에 Ov-synch 처리시 배란시기와 수태율 구명에 관한 연구 (Synchronization Rate, Size of the Ovulatory Follicle, and Pregnancy Rate after Synchronization of Ovulation Method in Hanwoo)

  • 본 연구는 한우에 GnRH + PGF$_2$$\alpha$+GnRH (Ov-synch)를 처리하여 배란동기화를 시켰으며, 2차 GnRH 투여후 배란시간, 수태율, 계절별 수태율, 산차별 수태율을 조사하였으며, 시험축은 2산 이상의 개체를 무작위로 선발하여 실험에 공시하였으며, 배란동기화 처리후 1회 인공수정을 실시하고 수태율을 환산하였다. 호르몬 처리방법으로는 GnRH + PGF$_2$$\alpha$ + Gn-RH(Ov-synch)법을 이용하였으며, 배란시간의 조사는 초음파를 이용하여 2차 GnRH 투여 후 24시간부터 31시간까지 2시간 간격으로 난소를 촬영하여 배란 여부를 조사하였다. 1. 배란동기화 처리후 24시간부터 31시간까지 2시간 간격으로 난소의 상태를 확인한 결과 28∼30시간 사이에 80%(20/25)로 가장 많이 배란된 것으로 나타났다. 2. 수태율에 있어서는 계통별 1회 수정 수태율은 고급육 계통이 48.1%(38/79)로 다소 높게 나타났다. 3. 산차에 따른 수태율은 1∼2, 3∼4산차에서 각각 44.3, 55%로 나타났다. 4. 계절별로는 여름보다는 봄과 가을에서 47.3%로 다소 높은 경향이였다.

성성숙 호르몬 처리에 의한 쏘가리의 배란 유도 (Induced Ovulation in the Mandarin Fish, Siniperca scherzeri by Sex-Maturation Hormones)

  • 인간의 태반성 성선자극 호르몬(HCG) 또는 성선자극 호르몬-방출 호르몬 유도체(GnRH-a) 단독 주사와 HCG+GnRH-a, GnRH-a+prostaglandin E sub(2) (PGF sub(2)), GnRH-a+pimozide를 복합적으로 주사하여 성숙한 암컷 쏘가리에서 배란 유도 실험을 하였다. 호르몬과 유도체의 반응 효과는 배란후 인공 수정하여 수정율과 배체형성율을 측정하여 결정하였다. 일반적으로 GnRH-a 실험군이 HCG 실험군에 비해서 수정율과 배체형성율 및 부화율이 높았다. HCG(5,000 IU/kg)+(GnRH-a(10 ${\mu}$g), GnRH-a(10 ${\mu}$g/kg)+PG$F^2$(500 ng/kg) 및 GnRH-a(10 ${\mu}$g/kg)+pimozide(1-5 mg/kg)에서 89% 이상의 높은 부화율을 보였다. 본 연구에서 사용된 성성숙 호르몬과 자극물질로 처리된 모든 암컷에서 배란이 유도되었지만, HCG+GnRH-a+dopamine과 GnRH-a+PG$F_2$+indometacin 처리군에서는 배란이 억제되었다. 이들의 결과는 산란시기에 여러 가지 성성숙 호르몬과 관련된 호르몬과 성성숙 억제물질(GRIF)이 분비된다는 점을 시사한다.

Ginsenoside Rh2 upregulates long noncoding RNA STXBP5-AS1 to sponge microRNA-4425 in suppressing breast cancer cell proliferation

  • Background: Ginsenoside Rh2, a major saponin derivative in ginseng extract, is recognized for its anti-cancer activities. Compared to coding genes, studies on long noncoding RNAs (lncRNAs) and microRNAs (miRNAs) that are regulated by Rh2 in cancer cells, especially on competitive endogenous RNA (ceRNA) are sparse. Methods: LncRNAs whose promoter DNA methylation level was significantly altered by Rh2 were screened from methylation array data. The effect of STXBP5-AS1, miR-4425, and RNF217 on the proliferation and apoptosis of MCF-7 breast cancer cells was monitored in the presence of Rh2 after deregulating the corresponding gene. The ceRNA relationship between STXBP5-AS1 and miR-4425 was examined by measuring the luciferase activity of a recombinant luciferase/STXBP5-AS1 plasmid construct in the presence of mimic miR-4425. Results: Inhibition of STXBP5-AS1 decreased apoptosis but stimulated growth of the MCF-7 cells, suggesting tumor-suppressive activity of the lncRNA. MiR-4425 was identified to have a binding site on STXBP5-AS1 and proven to be downregulated by STXBP5-AS1 as well as by Rh2. In contrast to STXBP5-AS1, miR-4425 showed pro-proliferation activity by inducing a decrease in apoptosis but increased growth of the MCF-7 cells. MiR-4425 decreased luciferase activity from the luciferase/STXBP5-AS1 construct by 26%. Screening the target genes of miR-4425 and Rh2 revealed that Rh2, STXBP5-AS1, and miR-4425 consistently regulated tumor suppressor RNF217 at both the RNA and protein level. Conclusion: LncRNA STXBP5-AS1 is upregulated by Rh2 via promoter hypomethylation and acts as a ceRNA, sponging the oncogenic miR-4425. Therefore, Rh2 controls the STXBP5-AS1/miR-4425/RNF217 axis to suppress breast cancer cell growth.

Autocrine Regulation of Gonadotropin-releasing Hormone (GnRH) Operates at Multiple Control levels of GnRH Gene Expression in GT1-1 Neuronal Cells

  • We previously found that a potent gonadotropin-releasing hormone (GnRH) agonist, buserelin, decreases GnRH promoter activity together with GnRH mRNA level, providing evidence for an autoregulatory mechanism operating at the level of GnRH gene transcription in immortalized GT1-1 neuronal cells. To examine whether agonist-induced decrease in GnRH mRNA level requires the continuous presence of buserelin, we performed a pulse-chase experiment of buserelin treatment. Short-term exposure (15 min) of GT1-1 neuronal cells to buserelin ($10{\mu}M$) was able to decrease GnRH mRNA levels when determined 24 h later. When GT1-1 cells were treated with buserelin ( $10{\mu}M$) for 30 min and then incubated for 1, 3, 6, 12, 24, and 48 h after buserelin removal, a significant decrease in GnRH mRNA levels was observed after the 12 h incubation period. These data indicate that inhibitory signaling upon buserelin treatment may occur rapidly, but requires a long time (at least 12 h) to significantly decrease the GnRH mRNA level. To examine the possible involvement of de novo synthesis and/or mRNA stability in buserelin-induced decrease in GnRH gene expression, actinomycin D ($5{\mu}m/ml$), a potent RNA synthesis blocker, was co-treated with buserelin. Actinomycin D alone failed to alter basal GnRH mRNA Revel, but blocked the buserelin-induced decrease in GnRH mRNA level at 12 h of post-treatment. These data suggest that buserelin may exert its inhibitory action by altering the stability of GnRH mRNA. Moreover, a polvsomal RNA separation by sucrose gradient centrifugation demonstrated that buserelin decreased the translational efficiency of the transcribed GnRH mRNA. Taken together, these results clearly indicate that GnRH agonist buserelin acts as an inhibitory signal at multiple levels such as transcription mRNA stability, and translation.

유방촬영용 X선관 target/filter 조합에 따른 MTF영상평가에 관한 고찰 (Evaluation of MTF Image by Target/Filter Combined of X-ray Tube Using Mammography)

  • 인체의 유방은 조직 간의 엑스선 흡수차가 작기 때문에 엑스선관 Target/Filter의 적절한 조합으로 피사체의 대 조도를 높이는 일이 중요하다. 최근 장비들은 다양한 유방두께와 유선조직 밀도에 따라서 Target/Filter의 조합을 변화시켜 환자의 피폭 선량 경감 및 진단에 알맞는 화질을 얻고 있다. 본 실험은 Target/Filter의 조합에 따라 해상력 챠트를 이용하여 MTF 영상의 평가와 유방 팬톰 영상을 평가하여 다음과 같은 결론을 얻었다. 1. 아크릴 두께(2 cm, 3 cm, 4 cm)의 조합(Mo/Mo, Mo/Rh. Mo/Al, Rh/Rh, Rh/Al)에 따라 고유원자번호에 의해 엑스선 에너지에서 큰 차이를 보였다. 2. MTF 곡선에서 선예도를 나타내는 MTF 0.5에서의 Lp/mm 값은 아크릴 2 cm일 때 Mo/Mo은 2.4 Lp/mm, 4 cm일 때 Mo/Rh은 2.63 Lp/mm, 6 cm일 때 Rh/Rh은 2.9 Lp/mm로 높게 나타났다. 3. 분해능을 나타내는 MTF 값은 아크릴 2 cm일 때 Mo/Mo은 6.0 Lp/mm, 4cm일 때 Rh/Al은 4.60 Lp/mm, 6 cm일 때 Rh/Al은 6.03 Lp/mm로 MTF 값이 높게 나타났다. 4. 시각적 식별이 가능한 2.5 Lp/mm에서 MTF 값은 2 cm일 때 Mo/Mo은 0.48 Lp/mm, 4 cm일 때 Mo/Rh은 0.53 Lp/mm, 6 cm일 때 Rh/Rh은 0.59 Lp/mm로 높게 나타났다. 5. 유방 팬톰 영상평가에서는 Mo/Mo일 때 12점, Mo/Rh일 때 11점, Rh/Rh일 때 10.5점, Mo/Al일 때 10점, Rh/Al일 때 9점 순으로 나타났다.
