Effects of the Inclusion of Kimchi Lactic Acid Bacterial Culture in Extruded Pellets on the Growth, Body Composition and Immune Response of Juvenile Olive Flounder Paralichthys olivaceus (김치 유산균 배양물의 부상용 배합사료내 첨가시 넙치(Paralichthys olivaceus) 치어의 성장, 체조성 및 면역성 반응에 미치는 효과)

  • This study was performed to determine effects of the supplementation of Kimchi lactic acid bacterial culture in extruded pellets (EP) on the growth, body composition, blood chemistry and immune response of olive flounder Paralichthys olivaceus. Four hundred eighty individuals averaging 16.1 g were randomly distributed into 12, 180 L flow-through tanks (forty fish per tank). Four concentrations of Kimchi lactic acid bacterial culture (KL) were prepared: Control (0%), 0.1%, 0.2% and 0.5%. Three concentrations (0.1%, 0.2% and 0.5%) of Kimchi lactic acid bacterial culture were each diluted to 10% of EP weight and then fully absorbed by EP for 10 minutes. Each diet was fed to triplicate groups of fish. Fish were hand-fed to apparent satiation twice a day for 8 weeks. At the end of the 8-week feeding trial, the plasma lysozyme and bacterial activities of fish were determined. In addition, the cumulative mortality of fish was monitored for 8 days after their artificial infection with Edwardsiella tarda. The weight gain, specific growth rate, feed efficiency ratio, protein efficiency ratio, protein retention, hepatosomatic index and condition factor of fish were not affected by dietary supplementation with KL. None of the proximate composition, plasma parameters, lysozyme or bactericidal activities of fish was affected by dietary supplementation with KL. However, the cumulative mortalities of fish fed EP containing 0.1% and 0.5% Kimchi lactic acid bacterial culture were relatively low compared to that of fish fed the control diet. In conclusion, dietary supplementation with KL did not effectively improve growth, feed utilization, body composition, plasma chemistry, lysozyme, bactericidal activities or immune response of olive flounder after E. tarda infection under these experimental conditions.

Simultaneous Determination of Hesperidin and Glycyrrhizin in Pyungwi-san by HPLC/DAD (HPLC-DAD를 이용한 평위산 중의 Hesperidin 및 Glycyrrhizin의 동시분석법 확립)

  • A high performance liquid chromatographic (HPLC) method for the simultaneous determination of hesperidin and glycyrrhizin was established for the quality control of traditional herbal medicinal preparation, Pyungwi-san (PWS). Separation and quantification were successfully achieved with a Waters XTerra RP18 column ($5{\mu}m$, 4.6 mm I.D. ${\times}$ 150 mm) by gradient elution of a mixture of acetonitrile and water containing 0.03% phosphoric acid (pH 2.03) at a flow rate of 1.0 ml/min. The diode-array UV/vis detector (DAD) was used for the detection and the wavelength for quantification was set at 230 nm. The presence of hesperidin and glycyrrhizin in this extract was ascertained by retention time, spiking with each authentic standard and UV spectrum. All four compounds showed good linearity $(r^2>0.995)$ in a relatively wide concentration ranges. The R.S.D. for intra-day and inter-day precision was less than 7.0% and the limits of detection (LOD) were less than 60 ng. The mean recovery of each compound was 99.0-105.6% with R.S.D. values less than 4.0%. This method was successfully applied to the determination of contents of hesperidin and glycyrrhizin in three commercial products of PWS. These results suggest that the developed HPLC method is simple, effective and could be readily utilized as a quality control method for commercial PWS products.

The Effect of Plant Coverage on the Constructed Wetlands Performance and Development and Management of Macrophyte Communities (식생피도가 인공습지의 질소 및 인 처리효율에 미치는 영향과 습지식물의 조성 및 관리)

  • The field scale experiment was performed to examine the effect of plant coverage on the constructed wetland performance and recommend the optimum development and management of macrophyte communities. Four sets (each set of 0.88 ha) of wetland (0.8 ha) and pond (0.08 ha) systems were used. Water flowing into the Seokmoon estuarine reservoir from the Dangjin stream was pumped into wetland system. Water depth was maintained at 0.3 ${\sim}$ 0.5 m and hydraulic retention time was managed to about 2 ${\sim}$ 5 days; emergent plants were allowed to grow in the wetlands. After three growing seasons of the construction of wetlands, plant coverage was about 90%, even with no plantation, from bare soil surfaces at the initial stage. During the start up period of constructed wetlands, lower water levels should be maintained to avoid flooding newly plants, if wetland plants are to be started from germinating seeds. Effluent T-N concentration in low plant coverage wetland was higher in winter than high plant coverage wetland, whereas no T-P effluent concentration and removal efficiency difference was observed within 15% plant coverage. Dead vegetation affected nitrogen removal during winter because it is a source of organic carbon which is an essential parameter in denitrification. Biomass harvesting is not a realistic management option for most constructed wetland systems because it could only slightly increase the removal rate and provide a minor nitrogen removal pathway due to lack of organic carbon.

  • Liquid Chromatographic behavior of Ni(II) in Isonitrosoethylacetoacetate Imine(IEAA-NR), Ni(IEAA-NH)(IEAA-NR)(R = H, CH_3, C-2H_5, n-C_4H_9, C_6H_5-CH_2) chelates were investigated by reversed-phase HPLC on Micropak MCH-5 column using methanol/water as mobile phase. The optimum conditions for the separation of Ni(IEAA-NH)(IEAA-NR) chelates were examined with respect to the effect of the flow rate, sample solvent, mobile phase strength and column temperature. It was fo$und that metal chelates were properly eluted in an acceptable range of capacity factor value(0{\le}logk'{\le}1). The dependence of the logarithm of capacity factor (k') on the volume fraction of water in the binary mobile phase as well as on the liquid-liquid extraction distribution ratio (Dc) in methanol-water/n-alkane extraction system showed the good linearties, and the dependence of the logarithm of capacity factor (k') on the column temperature and on the enthalpy exhibited the good linearties, and the compensation temperature ({\beta}) from the slope was 773.47{\circ}K. It was suggested that the retention of metal chelates was largely affected by the hydrophobic effect.

Removal of Mixed Cd, Cr, Cu, Ni and Zn by Hibiscus canabinas (Hibiscus canabinas를 이용한 Cd, Cr, Cu, Ni 및 Zn의 제거)

  • Kenaf plants were hydroponically grown in reactor containing toxic metals as Cd, Cu, Cr, Ni and Zn to examine the ability to take up heavy metal. The plants were fertilized using a nutrient solution, which was appropriately adjusted to optimum pH, DO and conductivity. For n hydraulic retention time of 8 days, Cr, Cd, Cu, Ni and Zn were removed up to 90.5, 80.5, 66.1%, 71.1% and 79.4%, and reduced from 2.34 to 0.54 mg $L^{-1}$, 3.37 to 1.07 mg $L^{-1}$, 4.92 to 3.19 mg $L^{-1}$, 6.31 to 4.41 mg $L^{-1}$ and 6.27 to 2.09 mg $L^{-1}$. Especially, accumulation rate of Cr, Cd, Cu, Ni and Zn in the plant were measured up to 347.32, 275.39, 157.52, 50.48 and 211.01 mg DW kg $L^{-1}d^{-1}$, respectively. We considered that Kenaf plants removed Cr, Cd and Zn more effectively than other toxic metals applied.

Field Applicability of Low Temperature Thermal Desorption Equipment through Environmental Impact Analysis of Remediated Soil and Exhaust Gas (정화토양 및 배출가스의 환경적 특성 분석을 통한 저온열탈착장치의 현장 적용성 평가)

  • Geochemical and ecological properties of remediated soil and gas exhausted from a low-temperature thermal desorption (LTTD) process were analyzed to assess the environmental impact of LTTD treatment. Soil characteristics were examined with regard to the chemical (EC, CEC, and organic matter) and the ecological (dehydrogenase activity, germination rate of Brassica juncea, and growth of Eisenia andrei) properties. The exhaust gases were analyzed based on the Air Quality Act in Korea as well as volatile organic compounds (VOCs) and mixed odor. Level of organic Organic matter of the soil treated by LTTD process was slightly decreased compared to that of the original soil because the heating temperature ($200^{\circ}C$) and retention time (less than 15 minutes) were neither high nor long enough for the oxidation of organic matter. The LTTD process results in reducing TPH of the contaminated soil from $5,133{\pm}508$ mg/kg to $272{\pm}107$ mg/kg while preserving soil properties. Analysis results of the exhaust gases from the LTTD process satisfied discharge standard of Air Quality Law in Korea. Concentration of VOCs including acetaldehyde, propionaldehyde, butyraldehyde and valeraldehyde in circulation gas volatilized from contaminated soil were effectively reduced in the regenerative thermal oxidizer and all satisfied the legal standards. Showing ecologically improved properties of contaminated soil after LTTD process and environmentally tolerable impact of the exhaust gas, LTTD treatment of TPH-contaminated soil is an environmentally acceptable technology.

Effect of Dietary Supplementation of Sodium Salt of Isobutyric Acid on Ruminal Fermentation and Nutrient Utilization in a Wheat Straw Based Low Protein Diet Fed to Crossbred Cattle

  • The effect of dietary supplementation of sodium salt of isobutyric acid in low protein (10% CP) wheat straw based diet on nutrient utilization and rumen fermentation was studied in ruminally fistulated male crossbred cattle. The study included a 7 day metabolism and a 3 day rumen fermentation trials. The cattle were distributed into two equal groups of 4 each. The animals of control group were fed a basal diet consisting of wheat straw, concentrate mixture and green maize fodder in 40:40:20 proportion whereas branched chain volatile fatty acid (BCFA) supplemented group received a basal diet + isobutyric acid at 0.75 percent of basal diet. The duration of study was 36 days. The feed intake between experimental groups did not differ significantly and the average total DMI (% BW) was 2.01 and $2.28kg\;day^{-1}$ in control and BCFA supplemented diets. The dietary supplementation of BCFA improved (p<0.05) the DM, OM, NDF and cellulose digestibility by 4.46, 6.63, 10.57 and 11.31 per cent over those fed control diet. The total N retention on BCFA supplementation was improved (p<0.01) due to decreased (p<0.05) urinary N excretion. The concentrations of ruminal total N was 37.07 and $34.77mg\;100ml^{-1}$ in control and BCFA fed groups, respectively. Dietary supplementation BCFA significantly (p<0.01) reduced the ruminal ammonia N concentration as compared to control and the mean values ($mg\;100ml^{-1}$) were 13.18 and 9.42 in control and BCFA fed groups. The total VFA concentration was higher (p<0.01) in BCFA supplemented group (101.14 mM) than the control (93.05 mM). Among the VFAs, the molar proportion of acetate was higher (p<0.01) in BCFA supplemented group (71.07 mM) as compared to control (64.98 mM). However, the concentration of propionate and butyrate remained unchanged. Amino acids composition of bacterial hydrolysates was similar in both the groups. Ruminal outflow rate of liquid digesta was higher (p<0.01) in BCFA fed group ($67.56l\;day^{-1}$) than control ($52.73l\;day^{-1}$). It is concluded that the dietary supplementation of Na-salt of isobutyric acid in low protein diet improved the nutrient utilization and ruminal fermentation characteristics.

A Study on the Treatment of Ammonia-Nitrogen in the Septic Tank Effluent Using Biological Fluidized Bed (생물학적(生物學的) 유동층(流動層)을 이용(利用)한 정화조유출수(淨化槽流出水)의 암모니아성(性) 질소제거(窒素除去)에 관한 연구(研究))

  • This paper is a basic study of the experimental results for the treatment of ammonia-nitrogen in the septic tank effluent. The substrates in this experiment are actual septic tank effluent and synthetic waste-water which is similar to septic tank effluent containing a considerable amount of nitrogenous component. Experiments were conducted for organic removal and nitrification using various recycle ratio and hydraulic retention time at each stage. The results obtained show that organic removal rate was above 80% in the 1st and 2nd stage, but as nitrification process was proceeded, above 90% of ammonia-nitrogen was removed in the 3rd and 4th stage. In these cases, the recycle ratio and HRT were found 30 and 7 hrs respectively. In the relation of $NH_4{^+}-N$ removal to $NO_3{^-}-N$ formation in the synthetic waste-water and septic tank effluent, when $1mg/{\ell}$ of $NH_4{^+}-N$ was removed, $NO_3{^-}-N$ formations were $0.95mg/{\ell}$ and $0.82mg/{\ell}$ respectively. And kinetics of nitrification using Biological Fluidized Bed was discussed also.

Effects of Levels of Feed Intake and Inclusion of Corn on Rumen Environment, Nutrient Digestibility, Methane Emission and Energy and Protein Utilization by Goats Fed Alfalfa Pellets

  • The effect of high and low level of feed intakes on nutrient digestibility, nutrient losses through methane, energy and protein utilization by goats fed on alfalfa (Medicago sativa L.) pellets based diets was investigated in this study. Twelve castrated Japanese goats were employed in two subsequent digestion and metabolism trials. The goats were divided into three groups, offered three diets. Diet 1 consisted of 100% alfalfa pellet, Diet 2 was 70% alfalfa pellet and 30% corn, and Diet 3 was 40% alfalfa pellet and 60% corn. The two intake levels were high (1.6 times) and low (0.9 times) the maintenance requirement of total digestible nutrients (TON). Rumen ammonia nitrogen ($NH_3$-N) level of Diet 1 was lower (p<0.001) compared to Diets 2 and 3, but the values were always above the critical level (I50 mg/liter), The pH values of rumen liquor ranged from 6.02 to 7.30. Apparent digestibility of nutrient components did not show differences (p>0.05) between the two intake levels but inclusion of corn significantly altered the nutrient digestibility. Diet 3 had highest (p<0.001) dry matter (DM), organic matter (OM), ether extract (EE) and nitrogen fee extract (NFE) digestibility followed by the Diet 2 and Diet 1. The crude protein (CP) digestibility values among the three diets were in a narrow range (70.1 to 70.8%). Crude fiber (CF) digestibility for Diet 3 was slight higher (p>0.05) than that for other two diets. When alfalfa was replaced by corn, there were highly significant (p<0.001) increases in DM, OM, EE and NFE apparent digestibility and a slight increase in the CF digestibility (p>0.05). There were no differences (p>0.05) in energy losses as methane ($CH_4$) and heat production among the diets but energy loss through urine was higher for the Diet 1. The total energy loss as $CH_4$ and heat production were higher for the high intake level but the energy loss as $CH_4$ per gram DM intake were same (0.305 kcal/g) between the high and low intake level. Retained energy (RE) was higher for Diet 3 and Diet 2. Nitrogen (N) losses through feces and urine were higher (p<0.001) for Diet 1. Consequently, N retention was lower (p>0.05) for Diet 1 and higher in Diets 3 and 2. It is concluded that inclusion of corn with alfalfa increased the metabolizable energy (ME) and RE, and retained N through reducing the energy and N losses. The high level of intake reduced the rate of nutrient losses through feces and urine.

UV Photodegradation of Chlorinated VOCs: Removal Efficiency and Products (염소계 VOCs의 UV 광분해 연구: 제거율 및 부산물)

  • In this study, 4 gases containing typical chlorinated volatile organic compounds (VOCs) were treated by ultraviolet (UV) irradiation. The typical chlorinated VOCs are dichloromethane (DCM), trichloromethane (TCM), carbon tetrachloride (CTC) and trichloroethylene (TCE). The removal efficiency (RE) and the products of chlorinated VOCs by UV irradiation are investigated. At this time, 2 types of background gas (air and nitrogen) were used to figure out the RE by photooxidation and photolysis. The specification of UV-lamp used in this study was low-pressure mercury lamp emitting wavelength of 185~254 nm. The experimental conditions were set as initial VOC concentration of $180{\pm}10ppm$, empty bed retention time (EBRT) of 53 s, temperature of $23{\pm}2^{\circ}C$ and relative humidity of $65{\pm}5%$. In the photolysis condition with nitrogen ($N_2$) as background gas, the averaged RE of the 4 types of chlorinated VOCs was about 24% higher than that with photooxidation; and the REs of VOCs except CTC were confirmed as >99%. The composition of off-gases after UV photooxidation in air was investigated and several intermediates from DCM, TCM and TCE were detected by GC/MS. Among them, phosgene which is a toxics was detected as an intermediate of TCM. In addition, the concentration of carbon dioxide ($CO_2$) in the off-gases was measured to calculate the mineralization rate (MR). With the photooxidation, TCE showed the highest RE (>99%) while MR was the lowest (17%); and the MR of DCM was the highest (86%). In addition, particulate matters (PM) in the off-gases was also detected and high concentrated $PM_{10}$ ($21,580{\mu}g{\cdot}m^{-3}$) and $PM_{2.5}$ ($6,346{\mu}g{\cdot}m^{-3}$) were detected in TCE off-gas. More than 99% of the chlorinated VOCs could be removed using UV254-185 nm lamp, while it is necessary to conduct further studies on the production and treatment of secondary pollutants.
