Species Composition and Seasonal Variation of Fish Assemblage in the Coastal Water off Gadeok-do, Korea -1. fishes Collected by a Small Otter Trawl- (가덕도 주변 해역 어류의 종조성과 계절 변동 -1. 소형 기선저인망에 의해 채집된 어류-)

  • The species composition and seasonal variation of fish assemblage in the coastal water off Gadeok-do, Korea were studied using the monthly trawled samlpes through a year of 1998. During the study period, 110 species of fishes from 57 families were collected. Repomucenus valenciennei, Thryssa kammalensis, Leiognathus nuchalis and Zoarces gilli dominated the fish assemblage throughout the year, and were responsible for $ 57.6{\%}$ and $37.2{\%}$ in the number of individuals and biomass, respectively. Fish species of secondary importance in abundance were Sillago japonica, Apogon lineatus, Pholis fangi, Engraulis japonicus, Thryssa adelae, Pho;is nebulosa, Conger myriaster, Liparis tanakai, Acentrogobius pflaumii, Limanda yokohamae, Chaeturichthys hexanema, Erisphex pottii and Cunoglossus abbreviatus. Higher numbers of species were collected in spring and autumn than in summer and winter. Abundance was high in spring and autumn, and low in summer and winter, Species diversity indices showed that the fish assemblage was more diverse in autumn than any other seasons.

Anti-Viral Hemorrhagic Septicemia Virus (VHSV) Activity of 3-Methyl Catechol (바이러스성출혈성패혈증 바이러스 감염에 대한 3-Methyl Catechol의 항바이러스성 활성)

  • Viral hemorrhagic septicemia virus (VHSV) is a fish pathogen responsible for causing enormous economic loss to the aquaculture industry not only in Korea but worldwide. Thus, it is necessary to identify natural compounds that can be used to control the spread of VHSV. In this study, the anti-VHSV activities of five catechol derivatives, i.e., catechol, pyrogallol, 3-methyl catechol, veratrole, and 3-methyl veratrole-extracted from green tea-were assessed. The antiviral activities of these derivatives were found to be dependent on their structure, i.e., the hydroxyl or methoxyl group and their substituent groups-on the benzene ring. Catechol, pyrogallol, and 3-methyl catechol exhibited relatively high 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities than veratrole, and 3-methyl veratrole. Moreover, 3-methyl catechol harboring a methyl substituent group increased the viability of the virus-infected cells and resulted in a 2.86 log reduction in the gene copies of VHSV N (per mL) in real-time PCR analysis. In conclusion, the catechol derivatives harboring hydroxyl groups in their benzene ring exhibited higher antioxidant activities than those harboring the methoxyl groups. However, catechol derivatives with a methyl group at the 3'-position of the benzene ring exhibited higher antiviral activity than those harboring a hydroxyl group. To our knowledge, this is the first study to evaluate the relationship between the structure and the anti-VHSV activity of catechol derivatives.

Anti-Inflammatory Effect of Hexane Fraction from Eisenia bicyclis on Lipopolysaccharides-Treated RAW 264.7 Cells (LPS로 유도된 RAW 264.7 대식세포에 대한 대황(Eisenia bicyclis) 헥산 분획물의 항염증 효과)

  • Eisenia bicyclis is known to have secondary metabolites exhibiting various biological activities. In a preliminary study, the n-hexane fraction obtained from the ethanolic extract of E. bicyclis showed higher anti-inflammatory activity than the ethyl acetate and butyl alcohol fractions based on the inhibition of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 cells. Using this fraction (E. bicyclis hexane fraction, EHF), we investigated the molecular mechanisms underlying its anti-inflammatory effect in LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to 50 ㎍/mL EHF significantly inhibited NO and prostaglandin E2 production as well as their responsible enzyme proteins and mRNAs, in a dose-dependent manner (P<0.05). Similarly, EHF markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α as well as their mRNA levels. Nuclear translocation of nuclear factor-kappa B (NF-κB) was strongly suppressed by EHF treatment. EHF significantly reduced the phosphorylation of mitogen-activated protein kinases and phosphatidylinositol 3-kinase/Akt in LPS-stimulated cells. Moreover, EHF reduced ear edema in phorbol myristate acetate (PMA)-induced mice. These results indicate that EHF contains potent anti-inflammatory compounds, which may be used as a dietary supplement for the prevention of inflammatory diseases.

Analysis on the fishing conditions of tuna purse seiner belonging to the Korea-Ghana joint venture in the Atlantic Ocean (대서양 한-가나 합작법인 소속 다랑어 선망어선의 조업실태 분석)

  • The research was analyzed the catch data of the five years (2016-2020) for six joint venture company tuna purse seiners in the Atlantic Ocean, with the aim of suggesting improvement measures for responsible and sustainable fishing according to changes of recommendation by International Commission for the Conservation of Atlantic Tuna (ICCAT) on the tropical tunas. In the last five years, the average catch of six tuna purse seiner gradually increased to 7,745 tons, 8,364 tons and 9,053 tons from 2016 to 2018, but decreased to 7,761 tons in 2019 and 6,214 tons in 2020. The reason for the decrease in fishing volume in 2019 and 2020 was the fluctuation of the formation of the cold water zone (22-23℃), and the total ICCAT convention area of FAD closure in January and February due to the expansion of the FAD closure area and poor free school catching during two months period, respectively. The analysis on fishing area showed that the percentage of fishing in the high sea was about 85% although the FAD closure area included the EEZ zone in coastal countries; the rise of the fishing license in coastal countries is also believed to be a factor. In order to overcome such situations and improve catching volume, it will be possible if excellent manpower is secured, school fishing is expanded, and the production of high value-added catch (purse seine special: PS).

Evaluation of a novel TaqMan probe-based real-time polymerase chain reaction (PCR) assay for detection and quantitation of red sea bream iridovirus

  • The red sea bream iridovirus (RSIV) belonging to genus Megalocytivirus is responsible for red sea bream iridoviral disease (RSIVD) in marine and freshwater fishes. Although several diagnostic assays for RSIV have been developed, diagnostic sensitivity (DSe) and specificity (DSp) of real-time polymerase chain reaction (PCR) assays are not yet evaluated. In this study, we developed a TaqMan probe-based real-time PCR method and evaluated its DSe and DSp. To detect RSIV, the probe and primers were designed based on consensus sequences of the major capsid protein (MCP) genes from megalocytiviruses including RSIV, infectious spleen and kidney necrosis virus (ISKNV), and turbot reddish body iridovirus (TRBIV). The probe and primers were shown to be specific for RSIV, ISKNV, and TRBIV-types megalocytiviruses. A 95% limit of detection (LOD95%) was determined to be 5.3 viral genome copies/µL of plasmid DNA containing the MCP gene from RSIV. The DSe and DSp of the developed real-time PCR assay for field samples (n = 112) were compared with those of conventional PCR assays and found to be 100% and 95.2%, respectively. The quantitative results for SYBR Green and TaqMan probe-based real-time PCR were not significantly different. The TaqMan probe-based real-time PCR assay for RSIV may be used as an appropriate diagnostic tool for qualitative and quantitative analysis.

Single and Multi-Strain Probiotics Supplementation in Commercially Prominent Finfish Aquaculture: Review of the Current Knowledge

  • The Nile tilapia Oreochromis niloticus, Atlantic salmon Salmo salar, rainbow trout Oncorhynchus mykiss, olive flounder Paralichthys olivaceus, common carp Cyprinus carpio, grass carp Ctenopharyngodon idella and rohu carp Labeo rohita are farmed commercially worldwide. Production of these important finfishes is rapidly expanding, and intensive culture practices can lead to stress in fish, often reducing resistance to infectious diseases. Antibiotics and other drugs are routinely used for the treatment of diseases and sometimes applied preventatively to combat microbial pathogens. This strategy is responsible for the emergence and spread of antimicrobial resistance, mass killing of environmental/beneficial bacteria, and residual effects in humans. As an alternative, the administration of probiotics has gained acceptance for disease control in aquaculture. Probiotics have been found to improve growth, feed utilization, immunological status, disease resistance, and to promote transcriptomic profiles and internal microbial balance of host organisms. The present review discusses the effects of single and multi-strain probiotics on growth, immunity, heamato-biochemical parameters, and disease resistance of the above-mentioned finfishes. The application and outcome of probiotics in the field or open pond system, gaps in existing knowledge, and issues worthy of further research are also highlighted.

Galectin-1 from redlip mullet Liza haematocheilia: identification, immune responses, and functional characterization as pattern recognition receptors (PRRs) in host immune defense system

  • Galectins, a family of ß-galactoside-binding lectins, have emerged as soluble mediators in infected cells and pattern recognition receptors (PRRs) responsible for evoking and regulating innate immunity. The present study aimed to evaluate the role of galectin-1 in the host immune response of redlip mullet (Liza haematocheilia). We established a cDNA database for redlip mullet, and the cDNA sequence of galectin-1 (LhGal-1) was characterized. In silico analysis was performed, and the spatial and temporal expression patterns in gills and blood in response to lipopolysaccharide polyinosinic:polycytidylic acid, and Lactococcus garvieae were estimated via quantitative real-time PCR. Functional assays were conducted using recombinant protein to investigate carbohydrate binding, bacterial binding, and bacterial agglutination activity. LhGal-1 was composed of 135 amino acids. Conserved motifs (H-NPR, -N- and -W-E-R) within the carbohydrate recognition domain were found in LhGal-1. The tissue distribution revealed that the healthy stomach expressed high levels of LhGal-1. The temporal monitoring of LhGal-1 mRNA expression in the gill and blood showed its significant upregulation in response to immune challenges with different stimulants. rLhGal-1 exhibited binding activity in response to carbohydrates and bacteria. Moreover, the agglutination of rLhGal-1 against Escherichia coli was observed. Collectively, our findings suggest that LhGal-1 may function as a PRR in redlip mullet. Furthermore, LhGal-1 can be considered a significant gene to play a protective role in redlip mullet immune system.

Root Causes and Characteristics of Occupational Incidents by Cause Investigation - Focusing on Maintenance and Repair Work - (사고 원인조사를 통한 농작업 안전사고 특성 연구 - 유지, 보수 작업을 중심으로 -)

  • This study aims to identify the types of safety accidents that occur during maintenance and repair operations and analyze the root causes. We used the logic tree diagramming to extract the root causes of 65 safety incidents specifically related to maintenance and repair work out of a total of 1,024 agricultural safety incidents that occurred between 2007 and 2020 collected by investigation with interviews. The extracted root causes were analyzed by categorizing them into six different safety system classifications. The results of the analysis and suggestions are as follows. Incidents related to maintenance and repair work, which can lead to irreversible safety hazards, have occurred frequently. These incidents were found to be occurring due to the overlapping of various safety system errors such as personal protective system and safety operation procedure. In the short term, it is necessary to emphasize compliance with the use of personal protective equipment, and enhancement of maintenance training. In the long term, it is necessary to establish a legal distinction for maintenance and repair work and clarify the responsible parties. Introducing a maintenance system is also crucial to prevent occupational injuries during maintenance and repair in agriculture.

Improvements in the Environmental Impact Assessment on Seawater and Sediment Qualities for Coastal Dredging Projects (연안준설 사업에 따른 해양 수질 및 퇴적물 영향평가 개선 방안)

  • Since the early 2000s, demand for coastal dredging projects have been significantly increased, and the dredged sediments have showed the continuing marked increases through the multiple projects with other coastal development and constructions. As significant or potential degradation of marine environment has been mounting, we checked the current situation of marine environmental impact assessment through marine water and sediment qualities in relation with dredging projects of the sea area utilization consultation statements submitted in 2011. While analysis percentages of the general items were usually higher, harmful components such as metals revealed wide variation of analysis percentages. In the event of analysis of metals, the pre-treatment process (full digestion) and analysis method were not properly implemented in accordance with the guidelines for preparation of consultation statements. Although not specified in the guidelines, verification procedures (tests of recovery efficiency and detection limit) to secure the reliability were almost ignored. As a result, most of developers did conduct poor marine environmental impact assessment on coastal dredging and related projects. We suggested that the responsible government authority should establish new detailed guidelines on the sea area utilization consultation for more strict evaluation and diagnosis of marine environment and distinctly request the developer to obey the guidelines by complementing the system of the sea area utilization consultation.

Factors Influencing the National Agricultural Advisory Service in Uganda: Domestic and International Context (우간다 농촌지도사업의 국내외적인 영향 요인 고찰)

  • In Uganda, the National Agricultural Advisory Service (NAADS) is responsible for agricultural advisory and extension services of the country. NAADS is a semi-autonomous public agency under the Ministry of Agriculture, Animal Industry and Fisheries (MAAIF) accounted for 45% of the MAAIF total budget. It aims to have a decentralized farmer owned/controlled agricultural advisory service system with increasing participation of the private sector. NAADS is greatly influenced by the extension framework of Neuchatel Group comprising representatives from the cooperation agencies and Official Development Assistance (ODA) donor countries. At the initial stage of the program, only 7 districts promoted the initiative and eventually spread out all over the country afterwards. These initiatives greatly contribute to the agricultural development and increase the farmers' income by 42-53%. However, due to the political intervention, the original goal was manipulated and criticized with briberies and corruption. Recently, the government of Uganda is considering a way to unify the agricultural advisory and extension services under the government programs.
