  • The cometaboic biodegradation conditionso f trichloroethylene(TCE) by Burkholderia cepacia G4 were optimized using response surface analysis. The experimental sets of phenol concentration temperature and pH were designed using central composite experimental design. The optimal conditions of phenol concentration temperature and pH were determined to be 0.91 ppm 21.5$^{\circ}C$ and 7.65 respectively by the Ridge analysis of the contour plot for TCE biodegradation rates. The TCE biodegradation rate could be enhanced up to 2.43 nmol.mg protein$.$min by response surface methodology.

Optimization of Tri-enzyme Extraction Procedures for the Microbiological Assay of Folate in Red Kidney Bean and Roasted Peanut Using Response Surface Methodology

  • Total folate content was determined by microbiological assay using Lactobacillus casei spp. rhamnosis (ATCC 7469) with a 96-well microplate technique. Using roasted peanut and red kidney beans as representative legume samples, response surface methodology (RSM) was supplied to optimize the trienzyme procedures for the determination of folate in legumes. After response surface regression (RSREG), the second-order polynomial equation was fitted to the experimental data. Ridge analysis showed that the optimal digestion times were <2 hr for $Pronase^{(R)}$ and $\alpha$-amylase, and <5 hr for conjugase to obtain maximal folate values for legume samples. This study confirms that established digestion times for cereal products (AOAC Method 2004.05) of 3 for protease and 2 hr for $\alpha$-amylase are applicable to legumes. Conjugase treatment can be reduced to 5 from 16 hr and the conjugase level to 5 from 20 mg per sample, providing significant cost saving.

Experimental analysis and modeling of steel fiber reinforced SCC using central composite design

  • The emerging technology of self compacting concrete, fiber reinforcement together reduces vibration and substitute conventional reinforcement which help in improving the economic efficiency of the construction. The objective of this work is to find the regression model to determine the response surface of mix proportioning Steel Fiber Reinforced Self Compacting Concrete (SFSCC) using statistical investigation. A total of 30 mixtures were designed and analyzed based on Design of Experiment (DOE). The fresh properties of SCC and mechanical properties of concrete were studied using Response Surface Methodology (RSM). The results were analyzed by limited proportion of fly ash, fiber, volume combination ratio of two steel fibers with aspect ratio of 50/35: 60/30 and super plasticizer (SP) dosage. The center composite designs (CCD) have selected to produce the response in quadratic equation. The model responses included in the primary stage were flowing ability, filling ability, passing ability and segregation index whereas in harden stage of concrete, compressive strength, split tensile strength and flexural strength at 28 days were tested. In this paper, the regression model and the response surface plots have been discussed, and optimal results were found for all the responses.

  • Cylindrical fine-ceramics, $Al_{2}O_{3}$, was lapped on its outer surface by vibrational lapping unit manufactured in the laboratory. Cylindrical lapping of fine-ceramics is necessarily be characterized and optimized because its process as other finishing methods is time-spending and, so, inefficient one, and because it is very complicated and random process affected by numerous factors in itself and in its environment. In this study, an efficient experimental approach, experimental design method, was used to analyze characteristics of the cylindrical lapping of fine-ceramics, $Al_{2}O_{3}$, and response surface methodology(RSM) to find out the optimal variables combination for the maximum improvement of surface roughness($R_a$). From the final surface roughness point of view in the given lapping conditions, a stationary point or optimal lapping conditions as well as the possible maximum improvement of surface roughness($R_a$) was predicted.

Optimization of the Processing Conditions for Heated Garlic Juice by Response Surface Methodology

  • This study was designed to determine the optimum conditions of heating temperature, heating time and dilution rates for producing heated garlic juice by using central composite design of response surface methodology. Garlic 3Department of Biosystems Engineering, Chungbuk National University, Cheongju 361-763, Korea was heated using a high temperature and pressure treatment apparatus. Total soluble solid contents ranged from 4.4 at $130^{\circ}C$ 3 hr and 6 fold dilution to 5.89 at $115^{\circ}C$ for 2 hr and 8 fold dilution. The highest total acidity was 0.55% at $120^{\circ}C$ for 3 hr and 2 fold dilution. The pH ranged from 4.01 at $130^{\circ}C$, 3 hr and 6 fold dilution to 5.85 at $120^{\circ}C$, 1 hr and 6 fold dilution. From the results of statistical analysis on the sensory evaluation the predicted optimum processing conditions for best color, taste, flavor and overall acceptance were $119.41^{\circ}C$, 3.11 hr, 5.85 fold dilution, $118.23^{\circ}C$, 3.57 hr, 3.15 fold dilution, $120.54^{\circ}C$, 3.47 hr, 5.01 fold dilution and $119.54^{\circ}C$, 3.18 hr, 5.66 fold dilution, respectively. The application of response surface methodology for preparing heated garlic juice processing showed a good correlation with high significance.

Response Surface Methodology를 이용한 숭어(Mugil cephalus) 반염건품의 개발 (Development of Salted Semi-dried Common Gray Mullet Mugil cephalus using Response Surface Methodology)

  • This study examined the optimal salting drying method and processing conditions (salt concentration, curing time, dry temperature, and drying time) for preparing salted semi-dried common gray mullet (SSD-CGM) Mugil cephalus based on the moisture content, salinity, and overall acceptance using response surface methodology (RSM). The moisture content, salinity, and overall acceptance of SSD-CGM prepared with different salting methods revealed that dry salting was the optimal salting method for preparing high-quality SSD-CGM. The optimal drying method for preparing high-quality SSD-CGM based on the drying velocity and sensory color was hot air-blast drying. The results of the RSM program indicated that the optimal independent variables ($X_1$, salt concentration; $X_2$, curing time; $X_3$, dry temperature; $X_4$, drying time) based on the dependent variables ($Y_1$, moisture content; $Y_2$, salinity; $Y_3$, overall acceptance) for high-quality SSD-CGM were 5.6% for $X_1$, 2.7 h for $X_2$, $47.0^{\circ}C$ for $X_3$, and 8.5 h for $X_4$ for uncoded values. The predicted values of $Y_1$, $Y_2$, and $Y_3$ for SSD-CGM prepared under optimal conditions were 54.4%, 4.2%, and 6.3, respectively, while the experimental values were $55.2{\pm}1.0%$, $4.1{\pm}0.3%$ and $6.7{\pm}0.8$. The actual and predicted values did not differ.

Media Optimization for Laccase Production by Trichoderma harzianum ZF-2 Using Response Surface Methodology

  • Trichoderma harzianum ZF-2 producing laccase was isolated from decaying samples from Shandong, China, and showed dye decolorization activities. The objective of this study was to optimize its culture conditions using a statistical analysis of its laccase production. The interactions between different fermentation parameters for laccase production were characterized using a Plackett-Burman design and the response surface methodology. The different media components were initially optimized using the conventional one-factor-at-a-time method and an orthogonal test design, and a Plackett-Burman experiment was then performed to evaluate the effects on laccase production. Wheat straw powder, soybean meal, and $CuSO_4$ were all found to have a significant influence on laccase production, and the optimal concentrations of these three factors were then sequentially investigated using the response surface methodology with a central composite design. The resulting optimal medium components for laccase production were determined as follows: wheat straw powder 7.63 g/l, soybean meal 23.07 g/l, $(NH_4)_2SO_4$ 1 g/l, $CuSO_4$ 0.51 g/l, Tween-20 1 g/l, $MgSO_4$ 1 g/l, and $KH_2PO_4$ 0.6 g/l. Using this optimized fermentation method, the yield of laccase was increased 59.68 times to 67.258 U/ml compared with the laccase production with an unoptimized medium. This is the first report on the statistical optimization of laccase production by Trichoderma harzianum ZF-2.

반응표면분석법을 이용한 쌀 단백질 초고압 추출조건 최적화 (Optimization of the High-Pressure Condition for Rice Protein Extracting Using Response Surface Methodology (RSM))

  • The purpose of this study was to optimize the rice protein extracted using a response surface methodology. The experiment was designed based on a CCD (Central Composite Design), and the independent variables were the high pressure (X1, 0-400 MPa) and processing time (X2, 0-10 minutes). The results of the extraction content (Y1), residue content (Y2), and recovery yield (Y3) were fitted to a response surface methodology model (R2= 0.92, 0.92, and 0.93, respectively). Increasing the pressure and processing time has a positive effect on the extraction content (Y1), residue content (Y2), and recovery yield (Y3). Therefore, these high-pressure conditions (independent variables) can significantly affect the improvement in rice protein extraction efficiency. Thus, the optimal conditions of X1 and X2 were 400 MPa and 10 min., respectively. Under these optimal conditions, the predicted values of Y1, Y2, and Y3 were 62.93, 57.53 mg/g, and 91.76%, respectively.

  • The optimization of the addition ratios of Houttuynia cordata, Saururus chinensis, and Polygonatum odoratum var. pluriflorum hot water extract to Houttuynia cordata beverage was evaluated by applying response surface methodology. The quality characteristics of a selected best product were investigated. Sweet, savory, and fishy taste, and overall acceptability were optimized by response surface methodology using a central composite design. The concentration of effect of Houttuynia cordata, Saururus chinensis, and Polygonatum odoratum var. pluriflorum hot water extract was modeled using an equation. Hot water extracts of Houttuynia cordata ($1.75{\sim}1.89%$), Saururus chinensis ($1.03{\sim}1.04%$), and Polygonatum odoratum var. pluriflorum ($2.17{\sim}2.31%$) represented the optimal concentration conditions to obtain the best taste. The physical and chemical characteristics of the beverage manufactured at optimized conditions were pH 4.18, soluble solids 0.41 $^{\circ}Brix$, total acidity 0.21%, color value ($L^*$ value 40.08, $a^*$ value 4.53, and $b^*$ value 10.69), total polyphenol 54.63 mg/L, and electron donating ability 26.98%.

Optimization of Alcalase for Krill Byproduct Hydrolysis and Antioxidative Activities by Response Surface Methodology

  • Krill byproduct was hydrolyzed with Alcalase 2.4L to produce functional ingredients for high antioxidative activities against 1,1-dimethyl-2-picryl-hydrazyl (DPPH) radical and Fe. The objective of this study was to investigate the optimum condition for degree of hydrolysis and antioxidative activity of enzymatic hydrolysate produced with the commercial Alcalase using response surface methodology (RSM) with a central composite rotatable design (CCRD). The ranges of independent variables were pH 7.6~10.4 for initial pH and $50.9{\sim}79.1^{\circ}C$ for hydrolysis temperature and their dependent variables were degree of hydrolysis, Brix, amount of phenolic compounds, DPPH-scavenging activity and Fe-chelating activity. RSM with CCRD was well designed to investigate the optimum condition for functional ingredients with high antioxidative activities using Alcalase 2.4L because of their high $R^2$ values of the range of 0.93~0.99 except the $R^2$ value of 0.50 for the amount of total phenolic compounds. The optimum hydrolysis conditions were pH 9.5 and $62^{\circ}C$ for degree of hydrolysis (DH) and pH 9.1 and $64^{\circ}C$ for DPPH-scavenging activity by response surface methodology. The yield of DH and DPPH-scavenging activity were $14.1{\pm}0.5%$ and $10.5{\pm}0.2%$, respectively. It is advantageous to determine the optimum hydrolysis conditions of krill and its by-products for the creation of different kinds of food products, as well as to increase the usage of marine protein sources.
