On-line Monitoring of IPTG Induction for Recombinant Protein Production Using an Automatic pH Control Signal

  • The response of IPTG induction was investigated through the monitoring of the alkali consumption rate and buffer capacity during the cultivation of recombinant E. coli BL21 (DE3) harboring the plasmid pRSET-LacZ under the control of lac promoter. The rate of alkali consumption increased along with cell growth, but declined suddenly after approximately 0.2 h of IPTG induction. The buffer capacity also declined after 0.9 h of IPTG induction. The profile of buffer capacity seems to correlate with the level of acetate production. The IPTG response was monitored only when introduced into the mid-exponential phase of bacterial cell growth. The minimum concentration of IPTG for induction, which was found out to be 0.1 mM, can also be monitored on-line and in-situ. Therefore, the on-line monitoring of alkali consumption rate and buffer capacity can be an indicator of the metabolic shift initiated by IPTG supplement, as well as for the physiological state of cell growth.

  • A Direct injection -mixing total -flow -control sprayer was developed and evaluated . The system provided precise application rates and minimized operator exposure to chemicals as well as providing a possibility for recycling container so f unused chemicals that can causes environmental contamination. Chemicals were metered and injected proportionally to the diluent flow rate to provide constant concentrations. The main diluent flow was varied in response to changes in travel speed. Experimental variables of the sprayer were the control interval, the sensitivity of flow regulating valve, the tolerance of control object and the sensitivity of the injection pump system. The optimal performance of the flow control system was with an average response time of 8.5 sec at an absolute steady state of error of 0.067 L/min (0.8% of flow rate). The average response time of the injection rate was -0.53 sec and the coefficient of variation (CV) of concentration was 3.2%.

  • In this work, we investigate the global stability analysis of a viral infection model with antibody immune response. The incidence rate is given by a general function of the populations of the uninfected target cells, infected cells and free viruses. The model has been incorporated with two types of intracellular distributed time delays to describe the time required for viral contacting an uninfected cell and releasing new infectious viruses. We have established a set of conditions on the general incidence rate function and determined two threshold parameters $R_0$ (the basic infection reproduction number) and $R_1$ (the antibody immune response activation number) which are sufficient to determine the global dynamics of the model. The global asymptotic stability of the equilibria of the model has been proven by using Lyapunov theory and applying LaSalle's invariance principle.

반응표면 기법을 이용한 생물반응조 표면포기기 최적설계 (Optimum Design of Surface Aerator Using Response Surface Method)

  • In this study, we optimized the shape of the surface aerator that will be installed in a biological reactor using the response surface method. Response surfaces of mass flow rate, impeller torque, mass flow rate per impeller torque are generated and used to track the optimum shape of the aerator. MOGA(Multi-Objective Genetic Algorithm)method is adopted to find the optimum results. By increasing the mass flow rate per impeller torque, increase of oxygen supply efficiency to a reactor is anticipated. To verify the usability of the surface aerator, PIV measurements on flow fields inside a scale-downed biological reactor model are carried out.

반응표면분석에 의한 단감식초 제조조건의 모니터링 (Monitoring for the Fermentation Conditions of Sweet Persimmon Vinegar using Response Surface Methodology)

  • To utilize deteriorated sweet persimmon effectively, response surface methodology(RSM) was used to determine the optimal vinegar fermentation conditions and monitored by a divided two stage fermentation. The optimum conditions for maximum alcohol content were obtained when the first stage (alcohol fermentation) was carried out with an initial sugar concentration of 18.5$^{\circ}$Brix, agitation rate of 140.8 rpm, fermentation time of 127.6 hr. When sugar concentration was 14$^{\circ}$Brix maximum alcohol content(7.1%) was predicted at fermentation conditions of 160 rpm in agitation rate, 140hr in fermentation time. The optimum conditions for maximum acidity were obtaiend when second stage(vinegar fermentation)was carried out 249.5 rpm in agitation rate, 148.8 hr in fermentation time. Predicted values at the optimum conditions were similar to experimental values.

성인 정신장애의 청각자극에 따른 생리학적 반응 측정에 대한 체계적 고찰 (Automatic Nerve Activity to Physiologic Response in Adult With Psychiatric Disorder: A Systemic Review)

  • 목적 : 성인 정신장애를 대상으로 청각자극에 대한 생리학적 반응을 측정한 연구에서 사용된 청각자극의 종류와 측정도구를 알아보고 대상군의 자율신경계 반응특성을 제시하고자 한다. 연구방법 : 연구방법으로는 체계적 고찰을 사용하였다. 사용한 데이터베이스는 PubMed이었다. 검색어는 "auditory stimulation, auditory startle, electromyogram, skin conductance, heart rate, psychiatric disorder, anxiety disorder, schizophrenia, depression"이었다. 최종적으로 분석에 사용된 논문은 8개이었으며, 근거의 질적 수준은 모두 수준II이었다. 분석 내용은 대상군의 특성, 청각자극의 종류, 측정도구, 정신 장애의 청각자극에 대한 생리학적 반응특성이었다. 결과 : 1. 연구 대상군은 불안장애와 정신분열장애를 대상으로 한 연구가 각각 4개이었다. 2. 생리학적 반응 유발을 위해 청각자극만 제공한 연구가 6개, 청각자극과 함께 시각자극을 포함한 연구가 2개이었다. 3. 분석대상 연구 모두에서 근전도(electromyograph)를 사용하였으며, 근전도와 함께 피부전기전도(skin conductance)를 측정한 연구가 2개, 심박수(heart rate)를 측정한 연구가 2개이었다. 4. 정신분열장애와 불안장애(외상후 스트레스장애, 강박장애)는 일반인과 다른 생리학적 반응을 보였다. 결론 : 생리학적 반응을 측정하기 위한 연구에서 사용한 자극은 청각자극으로 단순음을 제시하였고, 근전도는 안륜근에 전극을 붙여 생리학적 반응을 측정하였다. 정신장애군은 청각자극에 대해 큰 반응, 짧은 잠재기, 지연된 습관화 등의 특징을 보였다. 앞으로 정신장애군의 감각 특성에 대하여 생리학적 측정을 통한 연구가 이루어져야 하겠다.

The Exercise Capacity and Cardiovascular Factors in Patients with Exaggerated Blood Pressure Response during Treadmill Exercise Testing

  • Exaggerated blood pressure response during exercise has been found to increase the risk of future hypertension, left ventricular hypertrophy, cerebrovascular stroke, and CVD (cardiovascular disease) death. The aim of this study was to evaluate exercise capacity, cardiovascular factors in exaggerated blood pressure response during treadmill exercise testing. For research subjects, 72 subjects (normal blood response: 49 subjects, exaggerated blood response: 23 subjects) who received treadmill exercise test at J General Hospital were selected in this study. Exaggerated SBP (systolic blood pressure) response was defined as an SBP of 210 mmHg or greater during a maximal treadmill exercise test. The group with an exaggerated SBP response showed significantly higher values for RPP (rate pressure product) compared with the group with a normal SBP response. Subjects with METs (metabolic equivalents) had lower exaggerated SBP response than normal SBP response group. Subjects with recovery SBP had delayed exaggerated SBP response than normal SBP response group. Exaggerated SBP response to exercise is negative correlation with METs.

전신병기(Extensive stage) 소세포 폐암 환자에서 항암 화학요법 성적 : VPP/CAV 및 Carboplatin/Etoposide(CE) 복합 화학요법의 치료효과 (Effect Of VPP/CAV Alternating Chemotherapy Versus Carboplatin/Etoposide(CE) Chemotherapy For Extensive Stage Small Cell Lung Cancer)

  • 연구배경 : 저자들은 전신병기 소세포 폐암 환자중 VPP/CAV 교대요법을 받은 환자군과 CE요법을 받은 환자군에 대하여 치료반응과 생존기간 그리고 부작용 등을 평가 하여 전신 병기 소세포 폐암 환자의 초치료로 CE요법이 유용한가에 대하여 알아보고자 하였다. 방법 : VPP/CAV교대요법을 받은 환자들은 cisplatin 60mg/$m^2$를 제 1일에 정주하였고 etoposide 100mg/$m^2$를 제 1일에서 제 3일까지 정주한 후, 3주 후에 다시 cyclophosphamide 1000mg/$m^2$, adriamycin 40mg/$m^2$, vincristine 1.4mg/$m^2$를 정주하였고 매 3주마다 반복하였다. CE요법을 받은 환자들은 carboplatin 325mg/$m^2$를 제 1일에 정주하였고, etoposide 100mg/$m^2$를 제 1일에서 제 3일까지 정주하여 매 3주마다 반복하였다. 양군 모두에서 필요한 경우에는 고식척인 방사선 시행하였다. 결과 : 전체 반응률은 61.4% 이었고 중앙 생존기간은 10.8개월이었다. VPP/CAV교대요법군의 경우 반응률은 54.5%, 중앙 생존기간은 9.5개월이었고, CE요법군의 경우 반응률은 68.2%, 중앙 생존기간은 11개월이었으나 양군간에 통계학적인 차이는 없었으며 부작용은 비슷하였으나, CE요법 군에서 빈혈이 더 많았다. 결론 : CE요법은 VPP/CAV교대요법과 동등한 반응률을 보였으며 부작용도 수용할만하여 전신병기 소세포 폐암환자들에서 초치료 약제로 적절한 것으로 판단된다.

전수층 무응답 편향보정 추정법에 관한 연구 (A study on non-response bias adjusted estimation for take-all stratum)

  • 사업체조사에서는 흔히 수정절사법이 사용되며 이 방법을 사용함으로써 표본의 수를 줄이면서도 추정의 정확성을 향상 시킬 수 있다. 그러나 전수층의 무응답률은 크게 높아지고 있으며 예비표본을 이용한 표본대체가 불가능하기 때문에 전수층에서 발생한 무응답은 추정의 정확성을 크게 떨어뜨리고 있다. 특히 무응답이 관심변수에 영향을 받는 경우에는 편향이 발생할 가능성이 매우 높기 때문에 이를 적절히 처리하는 것은 매우 중요하다. 본 연구에서는 전수층에서 발생한 무응답을 적절히 처리하는 방법의 하나로 편향보정 추정법을 제안하였다. 특히 Chung과 Shin(2020)에서 제안한 편향보정 추정량을 전수층 편향보정에 적용하였으며 전수층이라는 특수한 경우에 맞는 새로운 추정 방법을 제안하였다. 또한 모의실험을 통해 제안된 방법의 우수성을 살펴보았으며 실제 자료 분석을 실시하여 본 논문에서 제안한 방법의 우수성을 확인하였다.
