Proteomics 기술의 개발 및 응용 (Development and Applications of Proteomics Technology)

  • Proteomics research includes identification and quantitation of single protein and/or protein complex, profiling of protein expression changes in response to biological perturbations, characterization of protein functions and interactions, and elucidation the linkage between proteins and diseases. In this review paper, recent developments in the basic technologies involved in the proteomics research such as 2-dimensional PAGE and mass spectrometry are discussed. Also, the application areas of proteomics technology such as protein expression mapping and cell map proteomics are introduced with the focus on new drug development.

  • Effect of IDT structures on the frequency response of the SAW device are investigated. Various types of IDTs, such as MSC(multistrip-coupler), double split, reflector, and ground line. are designed and patterned on the widely used Y-Z cut LN$(LiNbO_3)$ substrate. Center frequencies, band widths, and insertion losses are measured and compared. Invalidity of the conventional model based on the impulse response model is also discussed.

Further Characterization of Activin A-induced IgA Response in Murine B Lymphocytes

  • We have recently shown that activin A, a member of TGF-$\beta$ superfamily, stimulates mouse B cells to express IgA isotype but other isotypes. In the present study, we further characterized effects of activin A on B cell growth and IgA expression. We found that activin A did not have effect on LPS-stimulated cell viability. In parallel, CFSE staining analysis revealed that activin A did not alter cell division. An increase of IgA secretion by activin A was completely abrogated by anti-activin A Ab but not by anti-TGF$\beta$1 Ab. In the same conditions, no other isotypes are significantly affected by each antibody treatment. Finally, activin A, as similar to TGF-$\beta$1, increased IgA secretion by mesenteric lymph node cells. These results suggest that activin A can specifically stimulate IgA response, independent of TGF-$\beta$ in the gut.

Optoelectronics based on 2D semiconductor heterostructures

  • Van der Waals (vdW) heterostructures built from two-dimensional layered materials provide an unprecedented opportunity in designing new material systems because the lack of dangling bonds on the vdW surfaces enables the creation of high-quality heterointerfaces without the constraint of atomically precise commensurability. In particular, the ability to build artificial heterostructures, combined with the recent advent of transition metal dichalcogenides, allows the fabrication of unique semiconductor heterostructures in an ultimate thickness limit for fundamental studies as well as novel device applications. In this talk, we will present the characterization of the electronic and optoelectronic properties of atomically thin p-n junctions consisting of vertically stacked WSe2 and MoS2 monolayers. We observed gate-tunable diode-like current rectification and a photovoltaic response across the p-n interface. Unlike conventional bulk p-n junctions, the tunneling-assisted interlayer recombination of the majority carriers is responsible for the tenability of the charge transport and the photovoltaic response. Furthermore, we will discuss the enhanced optoelectronic characteristics in graphene-sandwiched vdW p-n junctions.

Proteomic characterization of differentially expressed proteins associated with no stress in retinal ganglion cells

  • Proteomic analyses of differentially expressed proteins in rat retinal ganglion cells (RGC-5) following S-nitrosoglutathione (GSNO), an NO donor, treatment were conducted. Of the approximately 314 protein spots that were detected, 19 were differentially expressed in response to treatment with GSNO. Of these, 14 proteins were up-regulated and 5 were down- regulated. Notably, an increase in GAPDH expression following GSNO treatment was detected in RGC-5 cells through Western blotting as well as proteomics. The increased GAPDH expression in response to GSNO treatment was accompanied by an increase in Herc6 protein, an E3 ubiquitin ligase. Moreover, GSNO treatment resulted in the translocation of GADPH from the cytosol to the nucleus and its subsequent accumulation. These results suggest that NO stress-induced apoptosis may be associated with the nuclear translocation and accumulation of GAPDH in RGC-5 cells.

탄소나노튜브 복합 소재를 이용한 스트레인 센서 (Strain Sensors Using Carbon Nanotube Composites)

  • To address the need for new intelligent sensing of systems, this study presents a novel strain sensor based on piezoresistivity of carbon nanotube (CNT) and its nanocomposites. Fabrication and characterization of the carbon nanocomposite material are discussed and an electrical model of the CNT strain sensor was derived based on electrochemical impedance spectroscopy analysis and strain testing. The dynamic response of the sensor on a vibrating beam was simulated using numerical analysis and it was compared with experimental test. The simulation showed good agreement with the strain response of the actual sensor.

피코초 분해능의 시간 상관 단광자 계수 장치 구성 및 동작 특성 (Construction and Performance Characterization of Time-correlated Single Photon Counting System having Picosecond Resolution)

  • 모드록킹된 피코초 레이저, 고속전자장치, 전자관형 광증배관 등을 사용하여, 피코초 분해능을 갖는 시간상관 단광자 계수 장치 및 시분해 스펙트럼 측정 장치를 제작하였다. 기기감음함수는 레이저의 펄스모양, 고속전자장치의 timing jitter 및 walk, 광증배관과 증폭기의 특성에 민감함을 보여주었다. 광학계의 분산등을 보정하여 25 ps의 반치폭을 갖는 기기감응함수를 얻었으며, 이와 같은 결과는 이 장치를 사용할 경우 deconvolution을 통하여 10 ps 이하의 분해능으로 피코초에서 마이크로초의 넓은 범위에 걸쳐서 여기상태 소멸시간의 측정이 가능함을 보여준다.

  • AlN thin films were deposited on Si(100) and $SiO_2$/Si substrates using R.F. magnetron sputtering system. The effect of various deposition conditions on the crystal orientation of AlN films was investigated to obtain a highly (002)-oriented films. SAW filters were fabricated using AlN films with various thicknesses and their frequency response characterizations were measured. Experimental results showed that the (002)-orientation and surface roughness of AlN films played a crucial role of determining the frequency response of AlN SAW devices.

Third Harmonic Generation of Shear Horizontal Guided Waves Propagation in Plate-like Structures

  • The use of nonlinear ultrasonics wave has been accepted as a promising tool for monitoring material states related to microstructural changes, as it has improved sensitivity compared to conventional non-destructive testing approaches. In this paper, third harmonic generation of shear horizontal guided waves propagating in an isotropic plate is investigated using the perturbation method and modal analysis approach. An experimental procedure is proposed to detect the third harmonics of shear horizontal guided waves by electromagnetic transducers. The strongly nonlinear response of shear horizontal guided waves is measured. The accumulative growth of relative acoustic nonlinear response with an increase of propagation distance is detected in this investigation. The experimental results agree with the theoretical prediction, and thus providing another indication of the feasibility of using higher harmonic generation of electromagnetic shear horizontal guided waves for material characterization.
