Variation of Antifreeze Proteins during Cold Acclimation among Winter Cereals and Their Relationship with Freezing Resistance

  • Freezing-resistant plants can survive subzero temperatures by withstanding extracellular ice formation. During cold acclimation, their leaves accumulate antifreeze proteins (AFPs) that are secreted into the apoplast and have the ability to modify the normal growth of ice crystals. Three barley, two wheat and two rye cultivars were grown under two different temperature regimes (20/16$^{\circ}C$ and 5/2$^{\circ}C$, day/night). Apoplastic proteins from winter cereals were separated by SDS-PAGE and detected with antisera to AFPs from winter rye. Apoplastic proteins accumulated to much higher levels in cold-acclimated (CA) leaves compared with nonacclimated (NA) ones in winter cereals. After cold acclimation, the protein concentration of apoplastic extracts increased significantly from 0.088 $mgmL^{-1}$ to 0.448 $mgmL^{-1}$, with about 5-fold increment. Also, the apoplastic protein content per gram leaf fresh weight in CA leaves ranged from 31 $\mu\textrm{g}$ $(gFW)^{-1}$ to 120 $\mu\textrm{g}$ $(gFW)^{-1}$ with an averaged value of 77 $\mu\textrm{g}$ $(gFW)^{-1}$, and coefficients of variation of 54.9%. The CA leaves in Musketeer (a Canadian winter rye cultivar) showed the greatest AFPs and antifreeze activity followed by 'Geurumil' (a Korean winter wheat cultivar), and 'Dongbori l' (Korean facultative barley cultivar). The proteins secreted into the wheat leaf apoplast at CA condition were more numerous than those observed in winter rye, where two $\beta$-1,3-glucanase-like proteins (GLPs), two chitinase-like proteins (CLPs) and two thaumatin-like proteins (TLPs) accumulated during cold acclimation. The proteins in barley leaf apoplast at CA conditions were a little different from those in wheat leaves. The AFPs were various among and within species. More freezing-resistant cultivars had more clear and numerous bands than less freezing-resistant ones. The high determination coefficient ($R^2$ =91 %) between freezing resistance and AFPs per gram leaf fresh weight indicated that the amount of AFPs was highly related to freezing resistance in winter cereal crops.

Characteristics of Polycrystalline β-SiC Films Deposited by LPCVD with Different Doping Concentration

  • The physical and electrical properties of polycrystalline $\beta$-SiC were studied according to different nitrogen doping concentration. Nitrogen-doped SiC films were deposited by LPCVD(1ow pressure chemical vapor deposition) at $900^{\circ}C$ and 2 torr using $100\%\;H_2SiCl_2$ (35 sccm) and $5 \%\;C_2H_2$ in $H_2$(180 sccm) as the Si and C precursors, and $1\%\;NH_3$ in $H_2$(20-100 sccm) as the dopant source gas. The resistivity of SiC films decreased from $1.466{\Omega}{\cdot}cm$ with $NH_3$ of 20 sccm to $0.0358{\Omega}{\cdot}cm$ with 100 sccm. The surface roughness and crystalline structure of $\beta$-SiC did not depend upon the dopant concentration. The average surface roughness for each sample 19-21 nm and the average surface grain size is 165 nm. The peaks of SiC(111), SiC(220), SiC(311) and SiC(222) appeared in polycrystalline $\beta$-SiC films deposited on $Si/SiO_2$ substrate in XRD(X-ray diffraction) analysis. Resistance of nitrogen-doped SiC films decreased with increasing temperature. The variation of resistance ratio is much bigger in low doping, but the linearity of temperature dependent resistance variation is better in high doping. In case of SiC films deposited with 20 sccm and 100 sccm of $1\%\;NH_3$, the average of TCR(temperature coefficient of resistance) is -3456.1 ppm/$^{\circ}C$ and -1171.5 ppm/$^{\circ}C$, respectively.

Population Structure and Race Variation of the Rice Blast Fungus

  • Worldwide, rice blast, caused by Magnaporthe grisea (Hebert) Barr. (anamorph, Pyricularia grisea Sacc.), is one of the most economically devastating crop diseases. Management of rice blast through the breeding of blast-resistant varieties has had only limited xuccess due to the frequent breakdown of resistance under field conditions (Bonman etal., 1992; Correa-Victoria and Zeigler, 1991; Kiyosawa, 1982). The frequent variation of race in pathogen populations has been proposed as the principal mechanism involved in the loss of resistance (Ou, 1980). Although it is generally accepted that race change in M. grisea occurs in nature, the degree of its variability has been a controversial subject. A number of studies have reported the appearance of new races at extremely high rates (Giatgong and Frederiksen, 1968; Ou and Ayad, 1968; Ou et al., 1970; Ou et al., 1971). Various potential mechanisms, including heterokaryosis (Suzuki, 1965), parasexual recombination (Genovesi and Magill, 1976), and aneuploidy (Kameswar Row et al., 1985; Ou, 1980), have been proposed to explain frequent race changes. In contrast, other studies have shown that although race change could occur, its frequency was much lower than that predicted by earlier studies (Bonman et al., 1987; Latterell and Rossi, 1986; Marchetti et al., 1976). Although questions about the frequency of race changes in M. grisea remain unanswered, the application of molecular genetic tools to study the fungus, ranging from its genes controlling host specificity to its population sturctures and dynamics, have begun to provide new insights into the potential mechanisms underlying race variation. In this review we aim to provide an overview on (a) the molecular basis of host specificity of M. grisea, (b) the population structure and dynamics of rice pathogens, and (c) the nature and mechanisms of genetic changes underpinning virulence variation in M. grisea.

Numerical and Experimental Studies on the Fluidic Characteristics and Performance of Liner-type Microtube

  • Purpose: Methods: Three-dimensional CFD modeling was conducted to analyze the flow structure and discharge flow rate corresponding to the variation in the geometry of the flow channel in a microtube. Additionally, experiments were carried out, and the discharge flow rate was measured at various inlet pressures and inclination angles of the microtube. Results: The quantitative data of velocity distribution and discharge flow rate were obtained. As the width and length of the microtip increased, the discharge flow rate decreased significantly because of the increase in the loss of pressure along the microtube. As the depth of the microtip increased, the flow rate also increased because of the reduction in the flow resistance. However, in this analysis, the variation in the angle of the microtip did not influence the flow rate. From the experimental results, it was observed that the flow rate increased linearly with the increase in the inlet pressure, and the effects of the inclination angle were not clearly observed in those test cases. The values of the flow rate obtained from the experiments were significantly lower than that obtained from the CFD analysis. This is because of the distortion of the shape of the flow path inside the microtube during the fabrication process. The distortion of the flow path might decrease the flow cross-sectional area, and it would increase the flow resistance inside the microtube. The variation in the flow rate corresponding to the variation in the inlet pressure showed similar trends. Conclusions: Therefore, the results of the numerical analysis obtained from this study can be efficiently utilized for optimizing the shape of the microtip inside a microtube.

삽수(揷穗)의 수분(水分) 통도저항(通導抵抗) (Water Transport Resistances of Cutting)

  • 삽목후(揷木後) 발근(発根)하기 까지에 있어 발근용역(発根容易)한 수종(樹種)과 곤란(困難)한 수종(樹種)간의 통수저항(通水抵抗)이 어떻게 변화(变化)하고 있는가를 삽수(揷穗)의 부의별(部位別)로 측정(測定)하였다. 1. 삽수(揷穗)의 전체(全体) 저항(抵抗)(RT)은 수종(樹種)에 의해 크게 달랐으며 발근용역(発根容易)한 수종(樹種) (Euonymus japonica. Viburnum Awabuki)에서는 저항(抵抗)의 증대(増大)가 완만하였지만, 발근곤란(発根困難)한 수종(樹種)(Quercus glauca, Pasania edulis, Rhaphiolepis umbellata Mak var integerreima)에 있어서는 삽목후(揷木後) 시일(時日)이 지남에 따라 급격(急激)히 증가하였다. 2. 잎을 제거한 경전체(茎全体)의 저항(抵抗)(RS)은 발근용역(発根容易)한 수종(樹種)에 있어서는 삽목후(揷木後) 시일(時日)이 지남에 따라 급격(急激)히 증가하였으며 발근곤란(発根困難)한 수종(樹種)의 경전체(茎全体)의 저항(抵抗) (RS)은 거의 증가하지 않았다. 3. 절단면부위(切断面部位)의 경의 저항(抵抗)(RC)은 발근용역(発根容易)한 수종(樹種)에서는 RC는 급격(急激)히 증가하고 있지만 발근곤란(発根困難)한 수종(樹種)에서는 거의 증가(増加)하지 않았고, 잎을 제거한 경전체(茎全体)의 저항(抵抗)(RS) 절단면부위(切断面部位)의 경(茎)의 저항(抵抗)(RC)의 변화(变化)에 기인하고 있었다. 4. 발근곤란(発根困難)한 수종(樹種)에 있어서는 엽(葉)의 엽액부(葉腋部)의 저항(抵抗)(RL)이 가장 컸으며, 삽수(揷穗)의 전체(全体) 저항(抵抗)(RT)의 대부분(大部分)을 RL가 점하고 있었다.

QTLs Identification and Confiirmation of Field Resistance to Leaf Blast in Temperate japonica Rice (Oryza sativa L.)

  • Field resistance is defined as the resistance that allows effective control of a parasite under natural field condition and is durable when exposed to new races of that parasite. To identify the genes for field resistance to rice blast, quantitative trait loci (QTLs) conferring the resistance for races and blast nursery screening in japonica rice cultivars were detected and mapped using SSR markers. QTL analysis was carried out in 190 RILs population from the cross between Suweon365 (moderately resistant) and Chucheong (highly susceptible). Twelve QTLs against nine blast races inoculated were detected on chromosomes 1, 2, 4, 6, 7, 11 and 12. They explained from 5.1% to 34.9% of total phenotypic variation. Eight QTLs against blast nursery screening in four regions for three years were detected on chromosomes 1, 2, 4, 11 and 12. The phenotypic variation explained by each QTL ranged from 4.3% to 37.7%. Three chromosome segment substitution lines (CSSLs) of $BC_2F_6$ by backcross method were developed to transfer the QTLs into the susceptible cultivar Chucheong as a recurrent parent. A CSSL4-1 containing two QTLs qLB6.2 and qLB7 against blast races showed to the reaction of 6 to 7 at blast nursery in two regions for two years. The CSSL4-2 and CSSL93 containing QTLs, qLB11.2 and qLB12.1 of the resistance against leaf blast in blast nursery screening, respectively, had enhanced the resistance for blast nursery screening across two regions and in two years.

운전 온도에 따른 열전발전 모듈의 전기적 내부 저항 변화에 대한 연구 (Study on the Variation of Electrical Internal Resistance for Thermoelectric Generator Module with Operating Temperature)

  • An analysis model considered the manufacturing factors and the pellet size has been developed in order to predict the performance characteristics of thermoelectric modules as generators. Since the electrical internal resistance has a significant role in the performance of thermoelectric modules, the variations of electrical internal resistance with operating temperature are experimentally measured. The modified electrical internal resistance calculated from an experimental correlation is applied to the analysis model. To verify the modified analysis model, the output voltage, output current and output power are compared with experimental results for the operating temperature conditions of $T_h=85^{\circ}C$ and ${\Delta}T=40^{\circ}C$. The modified analysis shows a good agreement with the experimental results in terms of the output voltage, current, and power.

Probabilistic seismic and fire assessment of an existing reinforced concrete building and retrofit design

  • In this paper, a probability-based procedure to evaluate the performance of existing RC structures exposed to seismic and fire actions is presented. The procedure is demonstrated with reference to an existing old school building, located in Italy. The vulnerability assessment of the building highlights deficiencies under both static and seismic loads. Retrofit operations are designed to achieve the seismic safety. The idea of the work consists in assessing the performance of the existing and retrofitted building in terms of both the seismic and fire resistance. The seismic retrofit and fire resistance upgrading follow different paths, depending on the specific configuration of the building. A good seismic retrofit does not entail an improving of the fire resistance and vice versa. The goal of the current work is to study the variation of response due to the uncertainties considered in records/fire curves selection and to carry out the assessment of the studied RC structure by obtaining fragility curves under the effect of different records/temperature. The results show the fragility curves before and after retrofit operations and both in terms of seismic performance and fire resistance performance, measuring the percent improving for the different limit states.

  • The interfacial thermal resistance of pristine and defective carbon nanotubes (CNTs) embedded in low-density polyethylene matrix is studied in this paper. Interface thermal resistance in nanosystems is one of the most important factors that lead to the large variation in thermal conductivities in literature and the novelty of this paper lies in the estimation of the interfacial thermal resistance for defective nanotubes-systems. Thermal properties of CNT nanostructures are estimated using molecular dynamics (MD) simulations and the simulations were carried out for various temperatures by rescaling the velocities of carbon atoms in the nanotube. This paper also deals with the mesoscale thermal conductivities of composite systems, using effective medium theories by considering the size effect in the form of interfacial thermal resistance and also using the conventional micromechanical methods like Hashin-Shtrikman bounds and Wakashima-Tsukamoto estimates.

바이러스 외피단백질 유전자로 형질전환된 연초 식물체의 TMV 저항성 발현 및 유전자 안정성

  • Tobacco plants(Nicotiana tabacum cv. NC82) transformed with TMV CP cDNA were self-fertilized until 8th generation (R$_{8}$), and the transgenic plants from 6th to 8th generation were analized for their resistance to tobacco mosaic virus(TMV) and stability of the gene expression. The 6th generation of the plants(R$_{6}$) showed high resistance(81-91 %) to TMV at eight weeks after artificial inoculation with the virus. The transgenic cell line 601 was the most prominant in the expression of resistance. 98 % of the plants showed no symptom without any agronomic phynotepe variation when they were inoculated with the virus in a experimental field. However, 2% of the plants were revealed as delay type of symptom with mild mosaic on a few leaves. The viral resistance in greenhouse tests of the 7th generation (R$_{7}$) was 54-64%, and the number of delay type plants were increased than that of 6th generation plants. In the 8th generation, 81 % of the plants was complete resistant to the virus. The TMV CP cDNA of the transgenic plants of each generation was also confirmed by genomic PCR, and there was no systemic viral multiplication in the resistant plants. It suggests that the viral resistance and gene expression of the transgenic plants might be stable through the generations.ons.s.
