Optimal Conditions for the Distribution of Cryoprotectant into the Intact Fish Muscle of Oncorhynchus mykiss during Freeze/Thaw Cycling

  • Conditions for sufficient and rapid distribution of a cryoprotectant (sorbitol solution) into intact fish muscle (Oncorhynchus mykiss) were studied as changing in the residual Ca2+ ATPase activity during freeze/thaw cycling. Chunks of the fish muscle were immersed in 4 concentrations of sorbitol solutions ($20\%$, $30\%$, $45\%$, and $60\%$) by a shaker mechanism at 5$^${circ}C. Whole immersion samples (W) showed a higher value of the residual Ca2+ ATPase activity than those in the untreated controls (C), except in the treated controls (TC), while less effect of immersion concentration could be found. Comparing the extent of penetration of sorbitol into the surface layer to inner layer of immersed fish chunks, outer portion samples achieved excellent cryoprotection with $100\%$ of the residual ATPase activity values or more. For the inner portion samples, $30\%$ and $45\%$ sorbitol solution treatments indicated a higher ATPase activity than $60\%$ treatment. At high concentrations, mass transfer rates during osmotic dehydration might berapid and it causes faster surface drying by dewatering at surface solute layer. Periodically immersed and relaxed samples, W (5-3-1), led to good cryoprotection effect: W (5-3-1) indicated high residual Ca2+ ATPase activity values and the residual ATPase activity values excess $100\%$ in immersion of $30\%$ and $45\%$ sorbitol solutions.

Monitoring of Residual Veterinary Drugs in Incheon Fishery Products Distribution (인천 유통 수산물 중 잔류동물용의약품 조사)

  • Objectives: This study was carried out to provide safe seafood to the public through simultaneous analysis rapid inspections for residual veterinary drugs in 150 fishery products in 2016. Methods: Quinolones (9), Sulfonamides (14), Penicillins (2), Cephalosporins (3), Tetracyclines (4), Macrolides (4), Lincosamides (2), Pleuromutilin (1), Phenicols (4), benzylpyrimidines (2), Others (1) Malachite green (2), and Melamine (1) were analyzed for 49 species using by HPLC and HPLC-MSMS. Results: There were six unsuitable samples, five among fish and one for crustaceans. Within the standard detection were five fish. The antibiotics found were Quinolones (50%), Tetracyclines (33%) and Penicillins (17%). Conclusions: As a result of the experiment, there were six unsuitable samples, five for fish and one for crustaceans. The most prevalent antibiotics were Quinolones. The quinolone antibiotics are highly persistent in fish tissues, so they require more time than the withdrawal period for other common veterinary drugs. Careful attention is required when they are used in fish farms.

Aerobic Liquid Fermentation of Residual Food Waste by Thermophilic Bacteria (고온세균을 이용한 남은 음식물의 호기적 액상발효)

  • For the probiotic feed production from residual food waste, aerobic liquid fermentation was conducted by thermophilic bacteria. 11 Strains of bacteria were isolated from several soil sources and residual food waste. Screening was carried by shaking incubator for the separation of thermophilic strain at $55^{\circ}C$. The isolated strains were tested for enzyme activities such as ${\alpha}$-amylase and protease. 6 Bacterial strains were chosen and were adapted by repeated fermentation processes in food waste substrate. The viable cell count of them at final fermentation stages were shown as $3-7{\times}10^9/ml$ in 2L-jar fermenter. Among them B3, B6 showed higher enzyme activity. By the mixed fermentation of B3, B6 and Bacillus stearothermophilus, the highest viable cell count reached to $1.4{\times}10^{10}/ml$ in 8 hours.

Studies on the Contents of Occuring Sulfur Dioxide in Herbal Medicines distributed at Market (유통 한약재의 이산화황 함유량에 관한 연구)

  • This study was investigated the residual contents of sulfur dioxide on the 373 kinds of herbal medicine distributed from Korea, China, and Japan. A modified Monier-Williams method was described for the determination of $SO_{2}$ contents in herbal medicines. The residual contents of $SO_{2}$ were not detected at 221 products$(59.8{\%})$ in total 373 products. Regardless of region, $SO_{2}$ contents were not found at Farfarae Flos, Zizyphi Spinosi Semen, Castanea crenata, and Strychni Ignatii Semen. But it's found at Asparagi Radix, Codonopsitis Radix, Lilii Bulbus, and Kaempferiae Rhizoma of every region collected the samples. Also, $SO_{2}$ contents were not detected at the herbal medicines which collected cultural fields of dometic. $SO_{2}$ contents ranged 11$\~$3990 mg/kg(mean 152 mg/kg) at domestic samples, 11$\~$3440 mg/kg(mean 603 mg/kg) at imported samples. After treated with water wash and hot water extraction, the reduction rates of sulfur dioxide were appeared with $24.4{\%}\~68.7{\%}\;and\;83.8{\%}\~100.0{\%},$ respectively. These data will be used to establish a criteron of residual sulfur dioxide in herbal medicines.

  • The major kernels$(7{\sim}10\;mesh)$ of naked barley were pearled to give an average yield for each pearling of about 5% flour, with 70% of the naked barley left as residual kernel. The contents of protein, fat and ash were in the highest in the fraction of 1,2 and 3, respectively. These fractions contained 1, 4 times of protein, 3.16 times of fat and 3.08 times of ash more than those values of original kernel. Residual kernels contained 62% of protein, 38% of fat and 35% of ash in the original kernel. Among minerals, the content of Ca, Na, Fe and Mn were in the highest in fraction 1 ; P, Mg and Zn in fraction 2 ; and K and Cu in fraction 3. Concentrations of these minerals were the lowest in the residual kernel. Magnesium showed the deepest concentration gradient, while iron was evenly distributed within the kernel.

  • The increasing use of plastics in food packaging materials has led to the issue of food-packaging mutual interactions from residues in the plastics. Plastic films are commonly printed by using solvent-based ink to decorate the packaged food for consumer attention. However, the residual solvents can not be completely removed and they can migrate into the contained food which lead to undesirable off-flavors. Partitioning (Kp) of printing ink solvents was studied in two types of plastic films with different chemical structure and polarity. At $25^{\circ}C$, Kp of toluene is higher than that of isopropanol in PP, but isopropanol showed higher Kp value than toluene in EVOH. This showed that polarity had a significant effect on the partitioning of printing ink solvents into the plastic films. Printing solvents had a higher affinity to the polymer with similar polarity than it did to the different one.

  • The purpose of this study is the establishment of scientific processes for making food safety policies. Thus, we investigated pesticide residue level of the agricultural commodities from market, and performed risk assessment. Fifteen agricultural items are chosen based on the frequency of Korean consumption. The samples were collected from 9 cities where populations are more than one million. Total 283 active ingredients were monitoring ( total sample number =232). Single-analysis of target pesticides was for three kinds of possible growth regulators and the multicomponent analysis was for 280 kinds of pesticides, a total of 283 species were selected to perform the pesticide residues. Before monitoring the analytes, the improvements of the analytical methods were done by method validations under the CODEX analytical method development guidelines and can produce metrics that represent the international standards applied in accordance with the guidelines. In addition to residual pesticides detected during monitoring we compare the ADI to EDI values using detected result and dietary consumption data which is extracted from annual market basket survey. The 163 samples were non-detected in the total 232 samples so it means that every agricultural commodity will residual pesticides-free in 70.3%. The detected residual pesticides showed for a total of 69 cases (29.7%). Two of samples violate Korean MRL (0.9%). The ratio of EDI compared to ADI resulted in only from 0.00087 to 0.902%. In result, we can assume that all detected residual pesticides are very safe level and current policies of Korean pesticides control may be working.

Characterization of Natural Gardenia Color with Systhetic Color (천연치자색소의 합성색소와의 특성 비교)

  • In order to replance systhetic colors by natural colors as food additive, properties of Gardenia yellow color and Gardenia blue color were compared with Food yellow No. 4 and Food blue No. 1. Color differeance between Food yellow No. 4 and Gardenia yellow color was 7.55. Thermal stability of Food yellow No. 4 was above 99%. On the other hand, in case of Gardenia yellow color, showed adove 90% of residual color units in 8$0^{\circ}C$$\times$30min and 10$0^{\circ}C$$\times$30min at pH 7.0 but 75% in 121$^{\circ}C$$\times$15min. Difference of light stability between Food yellow No. 4 and gardenia yellow color was about 18%. Addition of ascorbic acid was increased about 6% in light stability. Color difference between Food blue No. 2 and Gardenia blue color was 107. Thermal stability of Food blue No. 2 was above 99%. But Gardenia blue color showed 92% of residual color units in 8$0^{\circ}C$$\times$30min and 10$0^{\circ}C$$\times$30min at pH 7.0 but 90% in 121$^{\circ}C$$\times$15min. Difference of light stability between Food blue No. 4 and Gardenia blue color was about 8%. Addition of -tocopherol was increased about 4% in light stability of Gardenia blue color.

  • The residual sulfur dioxide in the five herbal medicines dried with burning bituminous coal for 9 hours showed from 339 (Glycyrrhizae Radix) to 1,138 mg/kg (Zingiberis Rhizoma) measured right after drying. The residual sulfur dioxide, from 70.1 to 95.5%, was remained in the above five matrices even after 30 days passed. The marker components concentration in above five herbal medicines like glycyrrhizic acid, betaine et al., were almost same after drying in oven and with burning bituminous coal.

  • The presence and content of residual hexane in the olive oils were studied. Total 41 olive oils of imported and domestic brands, which were labeled as extra virgin and refined (mixed), were collected from the market. For analysis, electronic nose and headspace SPME-GC/MS were used. Electronic nose equipped with 12 metal oxide sensors was used for the discrimination of odor pattern of olive oils against the different concentrations of hexane. From the results, it is assumed that the contents of residual hexane in the collected olive oils were below 5 ppm. For Qualitative and quantitative analysis of hexane, polydimethylsiloxane (PDMS) fiber was employed for SPME-GC/MS. In the results, the peak of residual hexane was detected in 8 samples from 41 olive oils. But the detected level was no more than 1 ppm that is under the regulation limit (5 ppm) by Korea Food Additive Code.
