Effect of Dietary Supplementation with Vitamin and Mineral Complexes on Estrus Rate, Pregnancy Rate, Gestation Length, and Birth Weight of Calves in Multiparous or Primiparous Korean Native Cows (한우 경산우 및 미경산우에 비타민과 미네랄 복합제의 급여가 발정발현율, 임신율, 임신 기간 및 송아지 체중에 미치는 효과)

  • The increase in the total number of cows on farms, which breed Korean Native Cattle (KNC), is associated with many problems. In particular, the services per conception and calving interval have increased. In this study, we examined the effect of dietary supplementation with vitamin and mineral complex on the estrus rate, pregnancy rate, gestation length, and birth weight of KNC calves. Multiparous or primiparous KNC were divided into 3 groups with 40 heads per group. Experimental group 1 was administered a consisting of 35,000 IU vitamin A, 100 IU vitamin E, 200 mg $\beta$-carotene, 200 mg Zn methionine, and 1.5 mg Se. Experimental group 2 was administered a vitamin complex (100 g/day) consisting of 100 IU vitamin E and 200 mg $\beta$-carotene. In the case of multiparous KNC, the estrus detection rate in the control group was 90.0% and those in the experimental groups were 75.0% to 95.0%. However, the first-service pregnancy rate after parturition in the control group was 41.2%, which was significantly lower than that in the experimental groups (71.0% to 76.7%; p<0.05). The average duration of pregnancy in the group supplemented with the vitamin complex was days, which was similar to that in the case of the control group. The birth weight of calves from cows fed with vitamin complex was 25.3 to 27.0 kg, which was similar to that in the case of the control group (25.2 to 26.0 kg). In the case of primiparous KNC, no differences in the estrus rate, pregnancy rate, gestation length, or the birth weight of calves were noted between the groups. Thus, dietary supplementation vitamin and mineral complex have no effect on the reproductive efficiency in primiparous cows, but the pregnancy rate was observed to have increased in multiparous KNC with these supplements.

Analysis of sex ratio on bovine in vitro fertilized embryos using sex determination kit treated sperm (성분리 키트가 처리된 소정자를 이용한 체외수정란의 배양과 성분리 효율)

  • It has been claimed that artificial insemination (AI) of cows with frozen-thawed semen treated with commercially produced kits, Wholemom (in favour of female gender) increases the birth chance of calves with desired sex ratio by approximately 85% without decrease of pregnancy rates. Hence, this study was conducted to investigate the efficacy of wholemom kits as combined with frozen-thawed bovine semen during in vitro fertilization on the in vitro fertilization and developmental efficiency and sex ratios such as some reproductive parameters in bovine. For this, 1,737 oocytes were in vitro fertilized and developed. Agglutination effects on bovine after treatment of Wholemom kit were observed by time passage and dose respectively. To determine sex of embryos, Bovine embryo Y-specific gene primers(ConEY) and Bovine specific universal primer(ConBV) were used as multiple PCR method. Fertilization rate of wholemom-treated group was significantly lower than its of control group[66.9% (1,156/1,737) in Wholemom-treated group; 75.0% (610/813) in control group]. However, developmental rate after fertilization of both wholemom-treated and control groups were not significantly different [26.1% (404/1,156) in Wholemom-treated group; 27.4% (224/610) in control group]. Sex ratio of in vitro fertilized embryo with frozen-thawed semen treated with wholemom kit was determined by multi PCR. Female ratio in wholemom-treated group [85.4% (173/201)] was significantly higher than its of control group [47.2% (66/141)]. In conclusion, wholemom treatments of semen used in the in vitro fertilization and development of bovine oocytes provided increase in female ratio with decrease of fertilization rate.

  • A brief general discussion of the nature and function of biosensors is presented. While the primary motivator for biosensor development has been the health-care industries, recent research efforts have spread to problems in agriculture and biological production systems. To illustrate some of the research from our laboratory, three example biosensors and their corresponding applications are presented. The first of these is an immunosensor for measurement of the hormone progesterone during milking as a method to improve reproductive management of dairy herds. The second example is an enzyme sensor for measurement of urea in milk as a menas to determine the efficiency of conversion of input protein to milk protein and, thus, improve nutritional management of dairy herds. The third example is a DNA sensor using polymerase chain reaction to detect pathogenic bacteria in the wash water of fresh and minimally processed fruits and vegetables. The potential for application of biosensors in agriculture, agrobiotechnology, food processing, and environmental monitoring has barely been realized.

Changes of plasma progesterone concentrations during the gestation and peripatum period, and return to estrus after parturition in Korean native goats (한국재래산양(韓國在來山羊)의 임신(姙娠) 및 분만전후(分娩前後)의 혈장(血漿) progesterone 농도변화(濃度變化)와 분만후(分娩後) 발정재귀시기(發情再歸時期))

  • The purpose of present study was to investigate the progesterone concentrations throughout gestation and peripartum period, and the return to the first estrus postpartum for improvement of reproductive efficiency in Korean native goats. The average length of gestation was 148 days(range : 144~154 days) and the average number of live births was 2 kids(range : 1~5 kids) in 12 Korean native goats. Progesterone concentrations were measured in blood samples taken from 12 goats every 5 days during gestation period. Plasma progesterone concentrations were 0.10 ng/ml at Day 0 of pregnancy and increased gradually until Day 20(6.58 ng/ml). Then they decreased slightly from Day 30 to 40(range : 4.32~4.82 ng/ml), increased again after Day 40 and remained thereafter until Day 140(range : 4.32~10.36 ng/ml). The progesterone levels declined sharply to basal levels at parturitum. Plasma progesterone concentrations during the pestpartum were 6.98 ng/ml at 10 days, 4.86 ng/ml at 6 days 3.18 ng/ml at 2 days before parturition, and 0.10 ng/ml at parturition, respectively. The basal levels were maintained until the first estrus postpartum. The mean intervals from parturition to the first estrus postpartum on the basis of progesterone determination and estrus detection were $100{\pm}64(mean{\pm}S.D.)$ days in 7 Korean native goats.

Analysis of Mineral Contents in Tissues and Bone Mineral Density of Rats (흰쥐의 조직 중 무기질 함량과 골밀도 분석)

  • The purpose of present study was to analyze mineral contents in various tissues and investigate theirs relation with bone mineral density (BMD) in rats. Fifteen Sprague-Dawley rats were fed standard diet for 4 weeks. Body weight gain, feed intake, and feed efficiency ratio were 41.00 g/week, 171.15 g/week, and 0.24 respectively. Among 12 minerals in serum, Ca is the highest with 6.86 mg/dl. Serum Mg, Se, and Cu were 2.52 mg/dl, 0.23 mg/dl and 0.22 mg/dl respectively. Mg contents in liver, spleen, and kidney were $246.36\;{\mu}/g$, $105.01\;{\mu}/g$, and $273.38\;{\mu}/g$ respectively. Tibia contents of Ca, Mg, Zn, Fe and V were 194.91 mg/g, 23.10 mg/g, 0.60 mg/g, 0.35 mg/g and 0.14 mg/g respectively. BMDs of right tibia and spine were 122.04 mg/cm and $153.61\;mg/cm^2$. There were significantly positive correlations between tibia BMD and Se (p<0.05), tibia BMD and V (p<0.01), spinal BMD and V(p<0.05), respectively. It's expected that these results are used as a reference data in following study to elucidate physiological function of minerals.

Effects of Cryo-extenders for Spermatozoa Sorted by Percoll on In Vitro Fertility of in Miniature Pigs (Percoll 분리된 미니돼지 정액의 체외 수정 능력에 있어서 동결보존액의 영향)

  • The objective of this study was to evaluated the efficiency on sperm cryosurvival and ability of in vitro fertilization using Triladyl and Lactose Egg-Yolk(LEY) as extenders for cryopreservation of separated sperm by 65% percoll in miniature pig. Sperm viability was measured with SYBR-14/PI double stained sperm by flow cytometry. Ability on embryo cleavage rate and blastocyst development were observed by in vitro fertilization after frozen-thawing of sperm separated by 65% percoll. The experimental groups were designed that separated sperm by 65% percoll with Triladyl (ST) or LEY(SL) and unseparated sperm with Triladyl(UT) or LEY(UL) for cryopreservation. As a results, the viability was significantly(p<0.05) higher in ST(55.1%), SL(63.1%), UL(58.8%) than UT(38.2%) group. Sperm viability in SL(63.1%) group was significantly(p<0.05) higher than other experimental groups. On the other hand, embryo cleavage rate was significantly(p<0.05) higher in ST(79.1%), SL(83.2) than UT(74.1) and UL(75.7%) groups at 96h after in vitro fertilization. Blastocyst development was also significantly(p<0.05) higher in ST(21.5%), SL(20.9%) than UT(17.0%) and UL(18.8%) groups. In conclusion, cryopreservation of miniature boar sperm separated by 65% percoll were beneficial to viability and capacity on in vitro fertilization.

Possible Production of Transgenic Chicken by Transferring Foreign Genes and Germ Cells (외부유전자의 전이에 의한 배아세포와 트란스젠닉 가금 생산의 가능성)

  • In recent years, numerous researches have been carried out in author's laboratory to develop several kinds of methods for producing transgened chicken, leaving a lot of new findings. Some of them are very useful to search for new approaches necessary to improve the efficiency of hatchability and the survival rate of developing trasgened embryos. The results obtained hitherto might be summarized as follows: (1) foreign gene(Lac Z/ Miw Z) introduced into blastodermal cells of developing embryos was successfully transferred to embryos, leading to the production of primordial germ cells(PGCs) carrying foreign DNA. However, hatched hickens failed to show the incorporation of introduced gene into the gonads. (2) When foreign gene was introduced into germinal crescent region (GCR), the gene was also efficiently incorporated into germ cells, resulting in the production of transgened chickens(offspring) which produced fruther offspring having foreign gene in the gonads. In this case, 2nd and 3rd generations of chickens were obtained through the reproduction of transgened birds. (3) In another way, the gene was injected into blood vessels of developing embryos at stage 13∼15, creating PGCs having foreign gene, and produced some transgened chickens. In this work, the PGCs were transfered between embryos, resulting in the production of transgenic chickens. (4) in these experiments, PGCs were effectively employed for producing transgenic birds, developing some kinds of chimeric chickens from homo- or hetero-sexual transfer of the PGCs from embryos. This means that the gonads from donor PGCs developed in some degree to the stage of hatching. However, these gonads showed slightly abnormal tissues similar to ovotestis like organs through histological examination. (5) Avian Leukosis Virus(ALV) induced B cell line(DT40) successfully carried foreign genes into chicken embryos, suggesting the possibility of the cells as a vector in this field of study in the future. Treatment of Agricultural By-Products by Vermicomposting-Effects of Mixture Ratio of Apple Pomace and Nightsoil Sludge on the Growth of Earthworm(Eisenia Foetida) and the Chemical Composition of Worm Casts (Vermicompostiong에 의한 농산부산물 처리 -사과박과 분뇨 슬러지의 혼합비율이 지렁이의 생육과 분립의 화학적 조성에 미치는 영향)

  • In order to investigate the possibility of treating the different mixture ratios of apple pomace and nightsoil sludge by vermicomposting was performed and the stability of worm casts, and availability of worm casts as plant growth media was evaluated by the analysis of chemical composition. The results were obtained as follows ; 1. The value of cast production at 100% apple pomace treatment was higher than those of other mixture ratios of apple pomace and nightsoil sludge. 2. The highest values of increasing rate and reproductive efficiency were obtained at mixture ratios of 60% apple pomace with 40% nightsoil sludge. 3. The value of increasing rate at 100% nightsoil sludge treatment was lower that those of other mixture ratios of apple pomace and nightsoil sludge. 4. Organic matter, available inorganic nutrients and stability of worm casts showed high values in all treatments. It means that worm casts are an excellent plant growth media source with a high chemical composition and their stability.

Characterization of Vaginal Microbiota Associated with Pregnancy Outcomes of Artificial Insemination in Dairy Cows

  • The profitability of the dairy and beef industries is largely affected by the actually achieved reproductive efficiency. Although a large proportion of cows worldwide are bred by artificial insemination (AI) services, many potential factors affecting the outcome of pregnancy by AI remain to be addressed. In the present study, we investigated the vaginal microbiota by high-throughput sequencing of 16S rRNA gene and analyzed their association with differential pregnancy outcomes (i.e., pregnant vs. nonpregnant) of multiple AI services in dairy cows. Sequencing of the V3-V4 region totally produced 512,046 high-quality sequences that were computationally clustered into 2,584 operational taxonomic units (OTUs). All OTUs were taxonomically assigned to 10 bacterial phyla. There were statistically significant differences among the three AI service times (T1, T2 and T3) with respect to the Shannon index and number of observed OTUs (p < 0.05). Bray-Curtis distance-based PCoA analysis also revealed that T2 group could be significantly distinguished from T1 and T3. However, no significant difference between the pregnant and nonpregnant cows was found in confidence regarding both alpha diversity and beta diversity. These results could help us better understand the possible influence of vaginal microbial community on pregnancy outcomes of AI service in cows.

Effect of Methyl-Beta-Cyclodextrin (MBCD) on Cryopreservation of Bull Spermatozoa (소 정액의 동결 보존시 Methyl-Beta-Cyclodextrin (MBCD)의 영향)

  • The objective of this study was to evaluate efficiency of methyl-beta-cyclodextrin (MBCD) in the sperm preservation of bull. For this study, the freezing of diluted semen were added with Triladyl containing 20% egg-yolk and/or 0, 1, 5, 10 and 20 mM MBCD before freezing process. Analysis of viability in frozen-thawed sperm was estimated by SYBR14/PI double stain, hypoosmotic swelling test(HOST) and acrosome damage with FITC-PNA, and mitochondria activation with Rhodamin123 by flow-cytometry. The sperm viability was significantly in 0 mM and 5 mM concentrations of MBCD than other groups (p<0.05). However, the HOST was significantly lower at 20 mM concentration of MBCD than other concentrations (p<0.05). In addition, acrosome damage and mitochondria activation rates were significantly lower at 20 mM concentration of MBCD than other groups (p<0.05). In conclusion, the viability of sperm was not significantly different among concentrations of MBCD 0, 5 and 10 mM, but MBCD 20 mM was significantly lower than other groups. In addition, as concentrations of MBCD was high, HOST, acrosome damage and mitochondria activation rates had a negative effect in bull sperm.
