  • In recent years, distributed computing environments have been radically changing to a structure of global, heterogeneous, federative and wide-area systems. This structure's environments consist of a let of objects which are implemented on telecommunication network to provide a wide range of services. Furthermore, all of objects existing on the earth have the duplicated characteristics according to how to categorize their own names or properties. But, the existing naming or trading mechanism has not supported the binding services of duplicated objects, because of deficiency of independent location service. Also, if the duplicated objects which is existing on different nodes provide the same service, it is possible to distribute the client requests considering each system's load. For this reason, we designed and implemented a new model that can not only support the location management of replication objects, but also provide the dynamic binding service of objects located in a system with minimum overload for maintaining load balancing among nodes in wide-area object computing environments. Our model is functionally divided into two parts; one part is to obtain an unique object handle of replicated objects with same property as a naming and trading service, and the other is to search one or more contact addresses by a location service using a given object handle. From a given model mentioned above, we present the procedures for the integrated binding mechanism in design phase, that is, Naming/Trading Service and Location Service. And then, we described in details the architecture of components for Integrated Binding Service implemented. Finally, we showed our implement environment and executing result of our model.

  • A Multiplayer Online Game(MOG) is a game capable of supporting hundreds or thousands of players and is mostly played using the Internet. P2P(peer-to-peer) architectures for MOGs can potentially achieve high scalability, low cost, and good performance. The basic idea of many P2P-based games is to distribute the game state among peers and along with it processing, network, and storage tasks. In a primary-copy based replication scheme where any update to the object has to be first performed on the primary copy, this means distributing primary copies of objects among peers. Most multiplayer games use a primary-copy model in order to provide strong consistency control over an object. Games consist of various types of actions that have different levels of sensitivity and can be categorized according to their consistency requirements. With the appropriate consistency level of each action type within a game, this paper allows developers to choose the right trade-off between performance and consistency. The performance for P2P game architecture with the primary-copy model is evaluated through simulation experiments and analysis.

Chemosensitization of Human Ovarian Carcinoma Cells by a Recombinant Adenoviral Vector Containing L-plastin Promoter Fused to Cytosine Deaminase Transcription Unit

  • We have demonstrated previously on a replication incompetent recombinant adenoviral vector, AdLPCD, in which the expression of cytosine deaminase (CD) gene is driven by the tumor-specific L-plastin promoter. The object of this study was to evaluate the efficacy of AdLPCD together with 5-fluorocytosine (5-FC) in suppression of the growth of established human tumor cells of ovary, Consistent with the knowledge that infection of OVCAR-3 cells with AdLPCD resulted in expression of a functional intracellular CD enzyme capable of converting 5-FC to 5-fluorouracil (5-FU) (Chung and Deisseroth, 2004), statistically significant differences in cytotoxicity were observed when AdLPCD infected cells were also exposed to 5-FC for 6 days (p=0.05), 9 days (p<0.0005) and 12 days (p<0.005), compared to 5-FC exposure alone, These results indicate that the CD gene delivered by adenoviral vector could efficiently sensitize OVCAR-3, otherwise non-toxic 5-FC. On the other hand, SKOV-3 cells, an ovarian carcinoma cell line, were more resistant to the CD/5-FC strategy compared with OVCAR-3 cells under the same condition. The results of present study suggest that the replacement of 5-FU with CD/5-FC in combination chemotherapy would be less toxic and much greater cytotoxicity than the conventional combination chemotherapy in some patients.

  • A robot system was developed to pick desired colony. This robot system consisted of an image acquisition process which acquires the image information of colony, an illumination device which irradiates the object once when it gets the image of it, a picking head, a replicating head, a bed for fixing well-plates, and a sterilization device of sterilizing pin stained with colony. Picking pins were washed in an ultrasonic wave washing for takes ten seconds. Picking pins were dipped for sterilizing in alcohol for ten seconds. The time for resterilizing picking pins in a heater was five seconds. This performance test resulted 100% success rate for both the colony picking and the colony replication process. Considering the procedure in which 8 pins were smeared by colony, picked on a well plate, and sterilized as one cycle, the system could carry out one cycle in 110 seconds. It took about 138 minutes to pick around 600 colonies.

A tendency of herbal medicines on anti-viral effect (항바이러스 효과에 응용되는 한약의 연구 동향)

  • In order to find antiviral effect against Human immunodeficiency virus(HIV), Herpes simplex virus type I(HSV-1) and II(HSV-2) from herb medicines, publicated 29 paters on anti-viral effect of herbal medicines and a convenient virus-induced cytopathic effect (CEP) inhibition assay was introduced. The major virus on experiment are HIV, Hepatitis B virus and HSV-1,2. Those of other studies showed inhibition of infected virus DNA replication and screening test of herbal medicines. More than 15 extractions were prepared by pure water boiling from herbal medicines, and their toxicity of infected cell and anti-viral activities were evaluated. Among them, the major part of herbal medicines showed cell stability compared with the contrast. Cytotoxic concentration (CC) of the $H_2O$ extracts of Padoo against HIV was <4.0, Hyungbangpaedoksan against HIV was 9.3, Whangyonhaedoktang against HIV-1 and HSV-2 was 15.3. These are high level cytotoxic concentration compared with the contrast. But antiviral effect was unable to figure out for selective $index(SI)=CC_{50}/EC_{50}$. The other herbal medicines were unable to showed potent anti-HIV and anti-HSV activity. The antiviral activation using herbs in this thesis have unlimited objects, to select research object will help to show the direction of antiviral drug development that have less side effect and more excellent efficiency.

Recombinant Adenoviral Vector Containing Tumor-Specific L-Plastin Promoter Fused to Cytosine Deaminase Gene as a Transcription Unit: Generation and Functional Test

  • The expression of therapeutic transgenes in recombinant adenoviral vectors is a major cause of toxicity in dividing cancer cells as well as non dividing normal cells. To solve the problem of toxicity to normal cells, we have reported on a recombinant adenoviral vector system (AdLP-) in which the expression of the transgene is directed by the tumor-specific L-plastin promoter (LP) (Chung et al., 1999). The object of this study was to generate a recombinant adenoviral vector system which would generate tumor cell specific expression of cytosine deaminase (CD) gene. We report the construction of a replication-incompetent adenoviral vector in which CD is driven by the L-plastin promoter (AdLPCD). Infection of 293 cells by AdLPCD generated the functional CD protein as measured by HPLC analysis for the conversion of 5-Fluorocy-tosine (5-FC) to 5-Fluorouracil (5-FU). HPLC analysis in conjunction with counting radioactivity for [6-$^3$H]-5FC and [6-$^3$H]-5FU demonstrated vector dose-dependent conversion of 5-FC to 5-FU in AdLPCD infected ovarian cancer cells. The results from present and previous studies(Peng et al., 2001; Akbulut et al., 2003) suggest that the use of the AdLPCD/5-FC system may be of value in the treatment of cancer including microscopic ovarian cancer in the peritoneal cavity.

A Distributed Real-Time Concurrency Control Scheme using Transaction the Rise of Priority (트랜잭션 우선 순위 상승을 이용한 분산 실시간 병행수행제어 기법)

  • As real-time database systems are extended to the distributed computing environment, the need to apply the existing real-time concurrency control schemes to the distributed computing environment has been made. In this paper we propose an efficient concurrency control scheme for distributed real-time database system. Our proposed scheme guarantees a transaction to commit at its maximum, reduces the restart of a transaction that is on the prepared commit phase, and minimizes the time of the lock holding. This is because it raises the priority of the transaction that is on the prepared commit phase in the distributed real-time computing environment. In addition, it reduces the waiting time of a transaction that owns borrowed data and improves the performance of the system, as a result of lending the data that the transaction with the raised priority holds. We compare the proposed scheme with DO2PL_PA(Distributed Optimistic Two-Phase Locking) and MIRROR(Managing Isolation in Replicated Real-time Object Repositories) protocol in terms of the arrival rate of transactions, the size of transactions, the write probability of transactions, and the replication degree of data in a firm-deadline real-time database system based on two-phase commit protocol. It is shown through the performance evaluation that our scheme outperforms the existing schemes.

Efficient Peer-to-Peer Lookup in Multi-hop Wireless Networks

  • In recent years the popularity of multi-hop wireless networks has been growing. Its flexible topology and abundant routing path enables many types of applications. However, the lack of a centralized controller often makes it difficult to design a reliable service in multi-hop wireless networks. While packet routing has been the center of attention for decades, recent research focuses on data discovery such as file sharing in multi-hop wireless networks. Although there are many peer-to-peer lookup (P2P-lookup) schemes for wired networks, they have inherent limitations for multi-hop wireless networks. First, a wired P2P-lookup builds a search structure on the overlay network and disregards the underlying topology. Second, the performance guarantee often relies on specific topology models such as random graphs, which do not apply to multi-hop wireless networks. Past studies on wireless P2P-lookup either combined existing solutions with known routing algorithms or proposed tree-based routing, which is prone to traffic congestion. In this paper, we present two wireless P2P-lookup schemes that strictly build a topology-dependent structure. We first propose the Ring Interval Graph Search (RIGS) that constructs a DHT only through direct connections between the nodes. We then propose the ValleyWalk, a loosely-structured scheme that requires simple local hints for query routing. Packet-level simulations showed that RIGS can find the target with near-shortest search length and ValleyWalk can find the target with near-shortest search length when there is at least 5% object replication. We also provide an analytic bound on the search length of ValleyWalk.

  • The mobile agent has very various performance according to the element of communication number of times between hosts, quantity of transmission data agent's size, network state etc. specially, migration method is caused much effect in whole performance of distributed system. Most existing migration methods have simplicity structure that it moves doing to accumulate continuously result after achieving task by visiting host in the fixed order. Therefore, in case there are situation such as fault, obstacle, and service absence etc. This can be inefficient due to mobile agent increased network required time. In this paper, we design and implementation Multicast Migration Model for minimizing network required time by solving this problems. Multicast Migration Model includes components such as mobile agent including call module and naming agent, which provides object replication information and distributed server's location transparence. And we evaluate and compare with existing migration method applying prototype system to verify implemented migration model.

The Basis of Different Sensitivities of Ovarian Cancer Cells to the Recombinant Adenoviral Vector System Containing a Tumor-Specific L-plastin Promoter and E. coli Cytosine Deaminase Gene as a Transcription Unit

  • We have reported previously on a replication incompetent recombinant adenoviral vector, AdLPCD, in which the expression of cytosine deaminase gene (CD) is driven by the tumor-specific L-plastin promoter. AdLPCD vector had been evaluated for its efficacy of chemosensitization of ovarian cancer cells to 5-FC. In spite of the fact that ovarian cancer cells, i.e., OVCAR-3 and SK-OV-3, are capable for adenoviral transduction judged by LacZ reporter gene analysis, two cell lines demonstrated quite different sensitivities toward AdLPCD/5-FC system. In OVCAR-3 cells, infection of AdLPCD followed by exposure to 5-FC resulted in the suppression of cell growth with statistical significance. On the other hand, SK-OV-3 cells were more resistant to the CD/5-FC strategy compared with OVCAR-3 cells under the same condition. The object of study was to investigate factors that would determine the sensitivity to AdLPCD/5-FC. We evaluated conversion rate of 5-FC to 5-FU after infection of AdLPCD by HPLC analysis, $IC_{50}$ of 5-FU, the expression level of integrin receptors i.e., ${\alpha}v{\beta}3$ and ${\alpha}v{\beta}5$, and status of p53 in OVCAR-3 and SK-OV-3 cells. The results indicated that OVCAR-3 cells have few favorable features compared with SK-OV-3 cells to be more effective to the AdLPCD/5-FC strategy; higher level of ${\alpha}v{\beta}5$ integrin, higher rate of conversion of 5-FC into 5-FC, and lower $IC_{50}$ of 5-FU. The results suggest that the replacement of 5-FU with CD/5-FC in combination chemotherapy would be less toxic and much greater cytotoxicity than the conventional combination chemotherapy in some patients.
