Qualitative and Quantitative Analysis of Microbial Community Structure in the Sequencing Batch Reactor for Enriching ANAMMOX Consortium (연속회분식 반응기를 이용한 혐기성 암모늄 산화균 농후배양에서의 정성 및 정량적 미생물 군집구조 분석)

  • Enrichment of anaerobic ammonium oxidation (ANAMMOX) bacteria is the essential step for operating full-scale ANAMMOX bioreactor because adding a significant amount of seeding sludge is required to stabilize the ANAMMOX reactor. In this study, the enrichment of ANAMMOX bacteria from an activated sludge using sequencing batch reactor was conducted and verified by analyzing changes in the microbial community structure. ANAMMOX bacteria were successfully enriched for 70 days and the substrate removal efficiencies showed 98.5% and 90.7% for $NH_4\;^+$ and $NO_2\;^-$ in the activity test, respectively. The phylogenetic trees of Planctomycetes phylum showed that the diverse microbial community structure of an activated sludge was remarkably simplified after the enrichment. All 36 clones, obtained after the enrichment, were affiliated with ANAMMOX bacteria of Candidatus Brocadia (36%) and Candidatus Anammoxoglobus (64%) genera. The quantification using real-time quantitative PCR (RTQ-PCR) revea ed that the 16S rDNA concentration of ANAMMOX bacteria was 74.8% compared to the granular ANAMMOX sludge obtained from an upflow ANAMMOX sludge bed reactor which had been operated for more than one year. The results of molecular analysis supported that the enriched sludge could be used as a seeding sludge for a full-scale ANAMMOX bioreactor.

Empirical Study on Applicability of Phosphorus Recovery Process in Wastewater Treatment Plant (하수처리시설에서 인 회수공정의 도입 가능성에 대한 실증적 검토)

  • In this study, we have made the annual total phosphorus (TP) mass balance diagrams for I wastewater treatment plant by utilizing the data of flow rate and TP of each process and tried to choose the optimum unit process empirically for phosphorus recovery. For the applicability evaluation, we have suggested several quantitative indices of flow rate, TP concentration, TP load and SS. Based on the analyses of TP mass balance, it became clear for reducing TP load of the wastewater treatment plant that it is efficient to recover phosphorus from the side stream in which the amount of flow rate is just 1/16, but TP concentration and load are 78 and 4.8 times larger than those of the influent of the plant. After the detailed applicability evaluation for the side stream, it could be concluded that the unit process of waste activated sludge thickener supernatant or dehydration filtrate are appropriate. Meanwhile, we did fundamental experiments utilizing the dewatering filtrate with TP concentration of 141.5 mg/L. After adjusting pH 10 and $Ca^{2+}$ concentration 250, 500, 1000 mg/L, it was stirred slowly. As a result, the $PO_4-P$ and TP removal efficiencies were above 95 percent; the results of the experiment imply the applicability of phosphorus recovery process in a wastewater treatment plant strongly.

  • Bad sitting postures are known to cause for a variety of diseases or physical deformation. However, it is not easy to fit right sitting posture for long periods of time. Therefore, methods of distinguishing and inducing good sitting posture have been constantly proposed. Proposed methods were image processing, using pressure sensor attached to the chair, and using the IMU (Internal Measurement Unit). The method of using IMU has advantages of simple hardware configuration and free of various constraints in measurement. In this paper, we researched on distinguishing sitting postures with a small amount of data using just one IMU. Feature extraction method was used to find data which contribution is the least for classification. Machine learning algorithms were used to find the best position to classify and we found best machine learning algorithm. Used feature extraction method was PCA(Principal Component Analysis). Used Machine learning models were five : SVM(Support Vector Machine), KNN(K Nearest Neighbor), K-means (K-means Algorithm) GMM (Gaussian Mixture Model), and HMM (Hidden Marcov Model). As a result of research, back neck is suitable position for classification because classification rate of it was highest in every model. It was confirmed that Yaw data which is one of the IMU data has the smallest contribution to classification rate using PCA and there was no changes in classification rate after removal it. SVM, KNN are suitable for classification because their classification rate are higher than the others.

A Study on the Loss of Tooth Substance and Surface Changes following Root Planing (치근면 활택술후 치질삭제와 표면형태변화에 관한 연구)

  • The purpose of this study was to evaluate in vitro effects of the loss of tooth substance and root surface changes following root planing with various periodontal instruments. The 39 extracted human teeth due to severe periodontal disease were included. The total 50 root surfaces of 30 teeth were root planed with following instruments : Group 1, with Gracey curette, Group 2, with ultrasonic scaler, Group 3, with rotating root planing bur, Group 4, with Gracey curette plus rubber cup polishing, and Group 5, with ultrasonic scaler plus rubber cup polishing. Following root planing, the amount of tooth substance loss was evaluated by measuring the weight of the removed tooth substance and then 5 specimens ($5{\times}5{\times}2mm$) were randomly selected from the each group for roughness measurement. Root planed areas of each specimen were subjected to five measurements using the Profilometer and an average surface roughness values(Ra) for each group was obtained. Statistical difference for roughness values of each group was analyzed using oneway ANOVA and student t-test. For scanning electron microscopic(SEM) examination of root surface changes following root planing, 15 root surfaces of remaining 9 teeth were root planed and 3 specimens were randomly selected. The mean loss of tooth substance removed was Group 1, $7.0{\pm}1.09mg$, Group 2, $1.3{\pm}1.00mg$, Group 3, $5.8{\pm}1.72mg$, Group 4, $8.7{\pm}1.34mg$, and Group 5, $4.5{\pm}1.68mg$ following root palning, respectively. These results indicate that curette is effective instrument in the respect of diseased root substance removal. The average surface roughness values are following results : Group 1 and Group 4 were the smoothest surface ($Ra=0.34{\pm}0.06{\mu}m$, $Ra=0.34{\pm}0.04{\mu}m$, respectively) and Group 2 was the roughest surface ($Ra=2.09{\pm}0.06{\mu}m$). Statistical analysis of roughness values demonstrated a highly significant difference (P<0.05) between each experimental groups. However, no statistically significant difference in roughness values were observed between the Group 1 and Group 4. The results in this study suggest that curette and/or polishing procedure should be done after root planing with ultrasonic scaler and caution should be used with dia-mond-coated bur during routine root planing procedure.

Effect of Suprabulge Clasp and Infrabulge Clasp on the Mobility of Abutment Teeth for Distal Extension Removable Partial Dentures (유리단 국소의치에서 Suprabulge Clasp와 Infrabulge Clasp가 지대치 동요에 미치는 영향에 관한 연구)

  • Distal-extension removable partial dentures have long been implicated in the increase in mobility and the destruction of the supporting structures of the primary abutment teeth. Various clasping systems have traditionally been used to retain distal extension removable partial dentures, and other designs have been proposed to minimize torquing forces on the abutment teeth. Most recent studies investigating the effects of removable partial dentures on abutment teeth have been performed in it laboratory setting. Results obtained from in vitro research have given dentists insight into removable partial denture design, but laboratory test model cannot be constructed that simulates actual functional or parafunctiona1 movements and forces. The purpose of this study was to clinically evaluate the degree of tooth mobility produced by two clasping systems (suprabulge type and infrabulge type) used for distal extension removable partial dentures. Akers clasp and R.P.I. system were selected for the evaluation, and four patients required a distal extension removable partial denture on the mandibular arch were selected for participation in the study. Two partial dentures were constructed in the same condition expect the design of clasp. All abutments in the study were mandibular first or second premolars. Measurements of mobility were made with a research tool designed by $M\"{u}hlemann$. This instrument, periodontometer, measures tooth mobility in the mouth by means of a dial gauge accurated to 0.01mm when the tooth is stressed with a force meter. Lingual and buccal deflection of abutment tooth was measured using buccal and lingual pressure. The amount of force applied was 500gm. Tooth mobility tests were made at four key stages; 1. Before insertion of the first removable partial denture, baseline mobility was establsihed. 2. After wearing of the first prosthesis, measurement was made at weekly intervals for 4 weeks. 3. The removable partial denture was then taken from the patient, and tooth mobility was measured again at weekly intervals until the patient's established baseline mobility had returned. 4. The second prosthesis of different clasp design was worn for a month and evaluated in the same manner as the first. The sequence of placement of clasping system was alternated between patients. The following results were obtained from this study; 1. The mobility of abutment tooth increased during the initial stage of wear and returned to baseline mobility after removal of removable partial dentures. 2. The mobility of abutment tooth showed no difference between Akers clasp and I-bar clasp during the 4-week test period. 3. All teeth tested showed greater mobility toward the buccal than the lingual direction.

Effect of Sludge Digestion on Removal Efficiency of Heavy Metals from Sewage Sludge Using Thiobacillus thiooxidans MET (Thiobacillus thiooxidans MET를 이용한 중금속 제거 효율에 미치는 슬러지 성상의 영향)

  • The effect of sludge digestion on the leaching efficiency of heavy metals from sludge by bioleaching with Thiobacillus thiooxidans MET was investigated. The used sludges were a non- and anaerobically digested. The leaching efficiency of heavy metals was strongly dependent on the pH of the sludge solution rather than the sludge condition and stolid concentration. The lower the pH the more heavy metal was leached from 3.0 of pH. The sequent orders of leaching heavy metals were Zn, Cu, and Cr. Although the buffering capacity of non-digested sludge was similar to anaerobically digested sludge, the pH decrease rate of the anaerobically digested sludge solution was faster than that of the non-digested sludge solution due to the higher sulfur oxidation rate of T. thiooxidans MET in the anaerobically digested sludge. The amount of leached heavy metals from the anaerobically digested sludge showed higher than that of non-digested sludge at the same pH value. This result might be caused by the difference of the insoluble metal types, which were contained in the sludge. An increase in sludge solids concentration decreased the leaching efficiency of heavy metals in the range of solids concentration 10 g/L to 70 g/L. The optimum ratio of S° to sludge stolid was 0.1 in both the sludge. The bioleaching process of heavy metals with T. thiooxidans MET showed the disinfecting effect over 90% as well as the reduction effect in sludge weight of 20%.

  • There are several kinds of hazardous materials in incinerator flue gas, such as particulate matter, acid gas, heavy metal, dioxin, etc. The activated carbon adsorption is considered as one of the methods removing dioxin from flue gas. Without any additional equipment and facilities, the activated carbon was mixed with lime and sprayed in the semi-drying reactor of an incinerator and filtered in the bag filter, and its efficiency of removing hazardous organic material was investigated. 1,2-dichlorobenzene (o-DCB) was used as a precursor material of dioxin and the effects of the activated carbon amount, the operating temperature of the reactor, and the atomizer r.p.m were measured and analyzed. Experimental results showed that the optimum outlet temperature of the reactor was $145^{\circ}C$ considering the performance of the bag filter, and the adsorption performance improved with the increase of the atomizer r.p.m. Also the performance of removing o-DCB in the bag filter is higher than of the semi-drying reactor.

Seasonal Ground Water Table Changes Following Forest Harvesting in Small Headwater Riparian Areas (산지계류 수변지역에서 산림벌채 후 지하수위의 계절 변화)

  • This study addressed the influence of forest harvesting on seasonal water table dynamics in small headwater riparian areas. Four treatments including potential Best Management Practices(BMPs) for ephemeral and intermittent streams were implemented(BMP1, BMP2, clearcut and reference). Water table measurements were obtained at bi-monthly intervals for 3 years including one year of pre- and two years of post-harvest observations. Overall, water table responses affected largely by rainfall amount. In addition, significant increases in water table levels following harvesting occurred throughout the two post-harvest years. Water table levels increased up to 28.2cm in the clearcut treatment during 2008 and up to 54.2cm in BMP2 during 2009. However, increase in water table elevation was not directly related to basal area removal despite considerable differences in basal area removed between BMP2 and clearcut treatments. Water table rises were apparent in that water table were more elevated during dry season(June through November) than during wet season(December through May). These seasonal fluctuations were presumably driven by changes in evapotranspiration caused by differences in leaf area of overstory canopy and understory following harvest.

  • Changes in runoff and soil erosion at slightly hilly erosive plots with pear trees over a three-year period were monitored under two distinct types of weed treatment by herbides : (1) pre-emergence herbicide with glyphosate; (2) post-emergence herbicide with paraquat. The numbers of rainfall events from June to Nov for three years of experimental periods were approximately 50 times in the plots having 5.5%to 10.2%slope at an altitude of 125 m. The steady-state infiltration rate was generally increased in the bare plot from which all weeds were removed while it was decreased in the herbicide treated plots and control. The runoffs from the control plot during the experimental periods were always less than those from plots of the herbicide-treated and the bare. The runoff under the same rainfall intensity was decreased in the order of bare, glyphosate, paraquat, and control. This results indicated that the removal time of weed by the different types of herbicides might influenced the runoff rate. For the first two years of the experimental periods, loss of fine fraction was much greater than that of coarse fraction while soil loss was correlated neither with total rainfall nor amount of runoff. The soil erosion rate under the same rainfall intensity was increased in the order of control, glyphosate, paraquat, and bare plot. However, there were not much differences in the soil loss for all plots under a relatively lower rainfall intensity less than 30 mm $day^{-1}$, resulting in rainfall intensity was important factor on soil erosion.

Differences in Population Density of 3 Rodent Species Between Natural Restored and Red Pine Silvicultured Forests after Forest fire (산불피해 후 자연복원과 소나무 조림을 실시한 지역에서 설치류 3종의 개체군 밀도 차이)

  • This study was conducted to clarify the differences in rodents population densities between natural restored and red pine silvicultured forests after forest fire in Samcheok, Gangwon Province, Korea from March to December 2008. One ha size of 3 study plots were set up in each natural restored and silvicultured stand. We trapped the small rodents during 4 consecutive nights every 2 months in each stand. Understory coverage and number of shrub stems were higher in silvicultured stand than in natural restored stand. Coverage of overstory, suboverstory and midstory, number of tree stem, woody seedling stems and dead wood, and amount of coarse woody debris were higher in natural restored stand than in silvicultured stand. Six hundred eighty eight individuals of four species, such as Apodemus agrarius, A. peninsulae, Eothenomys regulus and Tamias sibiricus were captured in our study. Number of captured small rodents were higher in natural restored stand than in silvicultured stand. Also, species compositions were differed in both stands. The captured number of A. agrarius and A. peninsulae were most highest in April and December. E. regulus were shown higher number of captured in April and June, and T. sibiricus were in June and October. Removal of coarse woody debris and silvicultural practice would not be good for the inhabitation of small rodents. For the conservation of small rodents diversity, management of understory and canopy would be needed in forest fired area.
