Evaluation on Anti-oxidant Activity and Anti-inflammatory Effects for the New Formulation of Gamisoyosan (가미소요산의 새로운 제형에 대한 항산화 활성 및 항염증 효능평가)

  • Objectives : Gamisoyosan (GMS) is a useful prescription for treating insomnia, dysmenorrhea and infertility induced by a stress. Also, GMS has been used traditionally to improve systemic circulation and biological energy production. The purpose of this study was to assess the anti-oxidant activity and anti-inflammatory effects of Gamisoyosan Formulation (Soft extract, GMS-SE). Methods : The biological activities such as anti-oxidant and anti-inflammatory effects were measured through cell line-based in vitro assay. We investigated the anti-oxidant properties of GMS-SE on the 1,1-diphenyl-2-picryhydrazyl (DPPH) radical, contents of total flavonoid and polyphenol. GMS-SE compared to butyl hydroxy anizole (BHA). Furthermore, based on this result the anti-inflammatory effects of GMS-SE have verified by mechanism from LPS- treated Raw264.7 macrophages. Results : The anti-oxidant activities of GMS-SE increased markedly, in a dose-dependent manner. The GMS-SE showed significant scavenging activity (GMS-SE $500{\mu}g/m{\ell}$ : $32.77{\pm}1.65%$, GMS-SE $1000{\mu}g/m{\ell}$ : $45.06{\pm}1.04%$ and BHA $100{\mu}g/m{\ell}$ : $39.25{\pm}2.41%$ for DPPH assay). and, The total phenolic compound and flavonoids contents of GMS-SE were $73.93{\pm}6.87{\mu}g/mg$ and $698.75{\pm}6.78{\mu}g/mg$. GMS-SE which is LPS has diminished in the LPS-induced release of inflammatory mediators (NO, iNOS, COX2 and PGE2) and pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$) from the RAW264.7 macrophages. Moreover, GMS-SE inhibited the activation of phosphorylation of p38 and ERK MAPKs by induced LPS. Conclusion : The present results indicate that GMS-SE has an anti-oxidant and anti-inflammatory properties, therefore may be beneficial in diseases which related to oxidative stress-mediated inflammatory disorders.

Effect of Baclofen on the Cholinergic Nerve Stimulation in Isolated Rat Detrusor (흰쥐의 적출배뇨근에서 baclofen의 콜린성신경 억제작용)

  • This study aimed to investigate the mechanism of action of baclofen on the detrusor muscle isolated from rat. Rats (Sprague-Dawley) were sacrificed by decapitation and exsanguination. Horizontal muscle strips of $2mm{\times}15mm$ were prepared for isometric myography in isolated muscle chamber bubbled with 95% / 5%-$O_2$ / $CO_2$ at $37^{\circ}C$, and the pH was maintained at 7.4. Detrusor strips contracted responding to the electrical field stimulation (EFS) by 2 Hz, 20 msec, monophasic square wave of 60 VDC. The initial peak of EFS-Induced contraction was tended to be suppresed by ${\alpha},{\beta}$-methylene-adenosine 5'-triphosphate (mATP), a partial agonist of purinergic receptor, and baclofen, a $GABA_B$ receptor agonist (statistically nonsignificant). The late sustained contraction by EFS was suppressed significantly (p < 0.05) by additions of atropione, a cholinergic muscarinic receptor antagonist and baclofen. The adenosine 5'-triphosphate-induced contraction was completely abolished by mA TP but not by baclofen. In the presence of atropine, the subsequent addition of acetylcholine could not contract the muscle strips: but the addition of acetylcholine in the presence of baclofen evoked a contraction to a remarkable extent. These results suggest that in the condition of present study, the cholinergic innervation may play a more important role than the purinergic one, and baclofen suppresses the contractility of rat detrusor by the stimulation of the $GABA_B$ receptors to inhibit the release of neurotransmitter from the cholinergic nerve ending.

Technology and Exploitation : Limitation of Capitalist Technological Development (과학기술과 착취 : 자본주도형 기술 개발의 한계)

  • This article attempts to deal with the problem that science and technology function as a mechanism to oppress and exploit humans rather than to release humans from labor. To explore this subject, it is necessary to consider the difference between the theory of labor value and the theory of 'technology value'. In addition, it is also important to refer to Marx's critical view of the 'capitalist' use of technology. Above all, Marx' concepts of relative surplus value and special surplus value, and his analysis of organic composition of capital are still valid in explaining that development of technology tightens control over workers and intensity of labor, and worsens instability of employment. Reflection of the limitations of capitalist development of technology is also important for realization of its usefulness. Industry 4.0 in Germany therefore deserves to be noticed as a good example because it shows a different way from extreme capitalist exploitation. The model suggests also some points that shouldn't be overlooked, when we try to actualize the tremendous slogan of the current fourth industrial revolution as real innovation and progress in human life. In this matter, the most important point is the possibility of technological development that doesn't oppose workers' interests.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

Baicalein induces cell death in Human Lung Carcinoma A549 Cells: Role of Apoptosis and Autophagy pathway (인체폐암 A549 세포에서 Baicalein에 의한 세포사멸 유도: Apoptosis와 Autophagy 경로의 역할)

  • Baicalein is one of the main flavonoids derived from roots of Scutellaria baicalensis Georgi, a traditional Oriental medicine. Although baicalein has high antitumor effect on several human carcinomas, the mechanism responsible for this property is not unclear. In this study, the data revealed that baicale-ininduced growth inhibition was associated with the induction of apoptosis connecting with cytochrome c release, down-regulation of anti-apoptotic Bcl-xl and increased the percentage of cells with a loss of mitochondria membrane permeabilization. Baicalein also induced the proteolytic activation of caspases and cleavage of PARP; however, blockage of caspases activation by z-VAD-fmk inhibited baicalein-induced apoptosis. In addition, baicalein enhanced the formation of autophagosomes and up-regulated LC3-II/LC3-I ratio. Interestingly, the pretreatment of bafilomycin A1 recovered baicalein-induced cell death suggesting that autophagy by baicalein roles as protective autophagy. Taken together, our results indicated that this flavonoid induces apoptosis and cell protective autophagy. These data means combination treatment with baicalein and autophagy inhibitor might be a promising anticancer drug.

Application of Molecular Diagnostics Technology in the Development of a Companion Diagnostics for Malignant Solid Tumors (악성 고형암의 항암제 동반진단 기술에서 분자진단기술의 적용)

  • Unlike benign tumors, malignant tumors are capable of metastasis, easy to relapse, poor survival, and low quality of life. In Korea, here is a tendency to treat the tumors collectively according to the General Principles of Cancer Chemotherapy(GPCC) of the Health Insurance Review & Assessment Service (HIRA). But recently, companion diagnostics(CDx) is recommended rather than unilateral medication because biomarker-based molecular diagnostics is possible to predict the drug response of patients before drug treatment. Not only domestic but also overseas Food and Drug Administratio (FDA) recommends the development of the CDx system at the stage of drug development to ensure the responsiveness and safety of medicines. In this study, I focused on the necessity of CDx development direction as well as CDx development status through literature review. Furthermore I also discussed CDx types according to the molecular diagnostic technology such as immunohistochemistry (IHC), polymerase chain reaction (PCR), in situ hybridization (ISH), and next-generation sequencing (NGS) not only in the approved CDx but also in the developing one by US FDA. And I suggested the technology issue of CDx development process such as a selection of molecular diagnostics at the time of release, a clear understanding of the CDx mechanism, and a convergence of drug with CDx development. The necessity of social insurance system also was proposed for CDx development.

Review of Research on Chloride-Induced Stress Corrosion Cracking of Dry Storage Canisters in the United States (미국의 건식저장 캐니스터에서의 CISCC 연구에 대한 검토)

  • It is important to study how to manage dry storage casks of spent nuclear fuels (SNF), because wet storage spaces for SNF will shortly be at full capacity in the Republic of Korea. The US has operated a dry storage cask system for several decades, and has carried out significant studies into how to successfully manage dry storage cask for SNF. This type of expertise and experience is currently lacking in the Republic of Korea. The degradation of dry casks is an important issue that must be considered. In particular, chloride-induced stress corrosion cracking (CISCC) is known to lead to the release of radioisotopes from canisters. The U.S. Department of Energy, U.S. Nuclear Regulatory Commission, and the Electric Power Research Institute have undertaken research into the CISCC mechanism. In addition, Sandia National Laboratories (SNL) has extensively researched CISCC and how to manage it in dry storage canisters. In this review paper, the probabilistic model proposed by the SNL is analyzed and, based on this model, US-based CISCC research is reviewed in detail. This paper will inform the management of dry cask storage of SNF from light water reactors in austenite stainless steel canisters in the Republic of Korea.

Radio-sensitivity of Human Colorectal Cancer Cell is Regulated by Potassium Cyanate (Potassium cyanate에 의해 조절되는 사람 대장암 세포의 방사선 감수성 변화)

  • Potassium cyanate (KCN) is an inorganic reagent and can induce the post-translation carbamylation of proteins. The carbamylated reaction in the body is involved in cell death in various diseases. According the results in our previous study, KCN enhances the radiosensitivity of human colorectal cancer cell line, HCT 116 cells. However, it was not enough to confirm the mechanism that KCN works in these cells. To determinated the mechanisms of KCN in the cells with increased radiosensitivity, HCT 116 cells were treated KCN with low-dose gamma-radiation. And then, we examined alteration of the cell cycle, cell proliferation, cytokine level and the activation of cell signaling protein. As a result, cell cycle arrest and cell death were induced by the activation of caspase-3 and PARP in the irradiated cells with KCN treatment. These changes of the irradiated cell with KCN treatment were induced by the release of $TNF-{\alpha}$ via $NF-{\kappa}B$ activation. In conclusions, enhanced radio-sensitivity mediated by KCN induced cell death and it occurs by $NF-{\kappa}B$-dependent $TNF-{\alpha}$ production.

Detrimental effects of lipopolysaccharides on maturation of bovine oocytes

  • Objective: Gram-negative bacteria lipopolysaccharide (LPS) has been reported to be associated with uterine impairment, embryonic resorption, ovarian dysfunction, and follicle retardation. Here, we aimed to investigate the toxic effects of LPS on the maturation ability and parthenogenetic developmental competence of bovine oocytes. Methods: First, we developed an in vitro model to study the response of bovine cumulusoocyte complexes (COCs) to LPS stress. After incubating germinal vesicle COCs in $10{\mu}g/mL$ of LPS, we analyzed the following three aspects: the expression levels of the LPS receptor toll-like receptor 4 (TLR4) in COCs, activities of intracellular signaling protein p38 mitogen-activated protein kinase (p38 MAPK) and nuclear factor-kappa B (NF-${\kappa}B$); and the concentrations of interleukin (IL)-$1{\beta}$, tumor necrosis factor (TNF)-${\alpha}$, and IL-6. Furthermore, we determined the effects of LPS on the maturation ability and parthenogenetic developmental competence of bovine oocytes. Results: The results revealed that LPS treatment significantly elevated TLR4 mRNA and protein expression levels in COCs. Exposure of COCs to LPS also resulted in a marked increase in activity of the intracellular signaling protein p-p38 MAPK and NF-${\kappa}B$. Furthermore, oocytes cultured in maturation medium containing LPS had significantly higher concentrations of the proinflammatory cytokines IL-$1{\beta}$, TNF-${\alpha}$, and IL-6. LPS exposure significantly decreased the first polar body extrusion rate. The cytoplasmic maturation, characterized by polar body extrusion and distribution of peripheral cortical granules, was significantly impaired in LPS-treated oocytes. Moreover, LPS exposure significantly increased intracellular reactive oxygen species levels and the relative mRNA abundance of the antioxidants thioredoxin (Trx), Trx2, and peroxiredoxin 1 in oocytes. Moreover, the early apoptotic rate and the release of cytochrome C were significantly increased in response to LPS. The cleavage, morula, and blastocyst formation rates were significantly lower in parthenogenetically activated oocytes exposed to LPS, while the incidence of apoptotic nuclei in blastocysts was significantly increased. Conclusion: Together, these results provide an underlying mechanism by which LPS impairs maturation potential in bovine oocytes.

Numerical Study of Combustion Characteristics by Pressure and Oxygen Concentration in Counter-Flow Diffusion Flame Model (대향류 확산 화염 모델에서의 압력 및 산소분율에 따른 연소 특성 변화에 관한 수치해석 연구)

  • As the seriousness and necessity of responding to climate change and reducing carbon emissions increases, countries around the world are continuing their efforts to reduce greenhouse gases. Among various efforts, research on CCUS, capturing and utilizing carbon dioxide generated when using carbon-based fuels, is actively being conducted. Studies on pressurized oxy-fuel combustion (POFC) that can be used with CCUS are also being conducted by many researchers. The purpose of this study is to analyze basic information related to the flame structure and pollutant emissions of pressurized oxy-fuel combustion. For this, a counter-flow diffusion flame model was used to analyze the combustion characteristics according to pressure and oxygen concentration. As the pressure increased, the flame temperature increased and the flame thickness decreased due to a reaction rate improvement caused by the activation of the chemical reaction. As oxygen concentration increased, both the flame temperature and the flame thickness increased due to an improvement to the reaction rate and diffusion because of a change in oxidizer momentum. Analyzing the related heat release reaction by dividing it into three sections as the oxygen concentration increased showed that the chemical reaction from the oxidizer side was subdivided into two regions according to the mixture fraction. In addition, the emission index of NO classified according to the NO formation mechanism was analyzed. The formation trend of NO according to each analysis condition was presented.
