Factors of Intention to take Hormone Replacement therapy in korean Menopausal Women (폐경기 여성의 호르몬 대체요법 선택의도에 영향을 미치는 요인)

  • Purpose: To identify factors influencing women's intention to take hormone replacement therapy(HRT). Method: A cross-sectional survey design was utilized. A total of 116 married women aged 40 to 60 were recruited from women's groups in communities. They completed a structured questionnaire containing demographic characteristics, Orientation to Life Questionnaire, Menopausal Symptom Checklist, Health Belief of Korean Adult, Sexrole Idea of Kim, Dong-il, chronic illnesses, and a single item measuring marital satisfaction. Result: Thirteen percents of the women were currently taking HRT and 28.4% had intention to take HRT. Education, sense of coherence, and sex role attitude were related to women's susceptibility to menopause. Hysterectomy status and the level of sense of coherence explained women's perception of seriousness regarding menopause. In addition, chronic health conditions accounted for the variances of benefits and barriers to HRT. Factors influencing women's intention to take HRT were education, menopausal symptoms, and perceived benefits of HRT. Conclusion: Women's chronic health conditions, psychosocial characteristics as well as menopausal discomforts were associated with women's choice regarding HRT. Other factors related to decision making process of women's health seeking behaviors need to be explored.

Changes in Stress Hormone, Lymphocytes, and Neutrophils Related to Examination and Social Support in University Students (시험과 사회적 지지에 따른 대학생의 스트레스 호르몬, 림프구 및 호중구의 변화)

  • Purpose: The purpose of the study was to identify levels of anxiety, stress hormone, lymphocytes, and neutrophils of the nursing students before an examination and to examine effects of social support on those variables related to taking examinations. Methods: Thirty eight healthy nursing students participated. They completed the questionnaire including state anxiety, test anxiety (VAS scale) and social support two weeks before the exam and again just before the exam. Simultaneously, a venous sample for ACTH, cortisol, lymphocytes & neutrophils count was drawn by a trained nurse. Descriptive statistics, paired t-test, student t-test, and Pearson's correlation with SPSS/WIN 14.0 were used to analyze the data. Results: Test anxiety scores and cortisol level on the exam day showed a significant increment compared with those on the non-exam day. The participants with higher levels of total social support scores showed significantly lower state anxiety on the exam day than those with lower levels of total social support scores. Conclusion: These data indicate a possible alteration in cortisol responsiveness to academic stress in nursing students. Social support would play an important role in modulation of academic stress.

  • The parathyroid hormone related protein (PTHrP) is the most common causative peptide of humoral hypercalcemia of malignancy. In contrast, the serum level of parathyroid hormone (PTH) is low to undetectable in the majority of patients with malignancy associated hypercalcemia. Few cases exist in which the production and secretion of PTH by malignant nonparathyroid tumors have been authenticated. To our knowledge, there is very rare case in which a nonparathyroid tumor expressed simultaneously both the PTH and PTHrP. We report a case of squamous cell carcinoma of the lung with hypercalcemia which presented with simultaneous elevation of serum PTH and PTHrP. Severe hypercalcemia (serum calcium, 7.5 mEq/L) was found in a 65-year-old man who had a squamous cell carcinoma of the lung without any bony metastasis and detectable parathyroid abnormalities on isotope scintigraphy. The serum level of intact parathyroid hormone (PTH) con centration was markedly elevated as measured in two site radioimmunoreactive PTH assays (intact PTH 150 pg/mL ; normal 9~55). The serum level of a PTHrP was also increased as measured in C-terminal region specific radioimmunoassay (PTHrP 99.1 pmol/L; normal 13.8~55.3). There are no evidences of coincidental primary hyperparathyroidism in parathyroid MIBI scan and other imaging studies including neck ultrasonography and computed tomography. These results suggest that simultaneous elevation of serum PTH and PTHrP in this patient can be caused by production of both PTHrP and PTH in other nonparathyroid lesions such as squamous cell carcinoma.

Circadian rhythm of melatonin secretion and growth-related gene expression in the tiger puffer Takifugu rubripes

  • Somatostatin (SS) and growth hormone-releasing hormone (GHRH) are primary factors regulating growth hormone (GH) secretion in the pituitary. To date, it remains unknown how this rhythm is controlled endogenously, although there must be coordination of circadian manners. Melatonin was the main regulator in biological rhythms, and its secretion has fluctuation by photic information. But relationship between melatonin and growth-related genes (ghrh and ss) is unclear. We investigated circadian rhythms of melatonin secretion, ghrh and ss expressions, and correlation between melatonin with growth-related genes in tiger puffer Takifugu rubripes. The melatonin secretion showed nocturnal rhythms under light and dark (LD) conditions. In constant light (LL) condition, melatonin secretion has similar patterns with LD conditions. ss1 mRNA was high during scotophase under LD conditions. But ss1 rhythms disappeared in LL conditions. Ghrh appeared opposite expression compared with melatonin levels or ss1 expression under LD and LL. In the results of the melatonin injection, ghrh and ss1 showed no significant expression compared with control groups. These results suggested that melatonin and growth-related genes have daily or circadian rhythms in the tiger puffer. Further, we need to know mechanisms of each ss and ghrh gene regulation.

Effects of Amydae Carapax on Bone Metabolism in Ovariectomized Rats (난소 절제 흰쥐의 골대사에 미치는 별갑의 영향)

  • Objectives : This study was undertaken to investigate the effects of Amydae Carapax (AC) on parameter related to bone metabolism in ovariectomized rats. Methods : We measured alkaline phosphatase activity and contents of estrogen, calcium, hydroxyproline, osteocalcin, calcitonin and parathyroid hormone after the ovariectomized rats were treated with AC for 30 days. Results : Serum estrogen, calcium and calcitonin contents in ovariectomized rats significantly decreased, but increased after AC treatment. [Significant increase of serum alkaline phosphatase activity, parathyroid hormone activity and osteocalcin content in ovariectomized rats was remarkably decreased by AC treatment. Increase of urinary calcium and hydroxyproline content in ovariectomized rats was decreased by AC treatment.] Conclusions : These results shows that AC has the ability to counteract abnormal calcium metabolic processes due to sex hormone inequality, promoting bone absorption and inhibiting bone formation.

Effects of Rhizoma Davalliae on Bone Metabolism in Ovariectomized Rats (난소 절제 흰쥐의 골대사에 미치는 골쇄보(骨碎補)의 영향)

  • Objectives : This study was undertaken to study the action mechanism of Rhizoma Davalliae (RD) at parameter related to regulating bone density. Methods : We measured alkaline phosphatase activity and the contents : calcium, hydroxyproline, osteocalcin, calcitonin, and parathyroid hormone after the ovariectomized rats had been treated with RD for 30 days. Results : The following changes occurred: First, the serum calcium content and the calcitonin content, which decreased in ovariectomized rats, increased with RD treatment. Second, the serum alkaline phosphatase activity, the parathyroid hormone ativity, the osteocalcin content, the urinary calcium content, and the hydroxyprolin content-which increased in ovariectomized rats-decreased with RD treatment. Conclusions : These results show that RD treatment can recover abnormal calcium metabolic process by sex hormone inequality, promoting bone formation and inhibiting bone absorption.

Three-Dimensional Structure Prediction of Follicle-Stimulating Hormone Receptor Transmembrane Domain by Homology Modelling

  • The follicle stimulating hormone receptor (FSHR) is a glycoprotein hormone, that belongs to the GPCR superfamily. FSHR plays a major role in reproduction. The aberrant activation of FHS receptor leads to infertility and several reproductive disorders. The recently recognized roles of the FSHR in diverse extragonadal tissues is also closely related to Alzheimer's disease and cancers. Analysing the structural characteristics of the receptor is important in understanding the pathophysiology of diseases associated with the receptor. In this present study, homology modelling of FSHR-TM domain was developed using four different templates. Totally 20 models were developed using single template-based approach and selected three based on the validation of RC plot, RMSD, ProSA, QMEAN and ERRAT values. The developed models would be useful for further research on the structural characteristics and binding characteristics of the FSHR-TM domain.

The Effects of 12-Weeks in a Combined Exercise Program on the Self-Reliance Fitness and Growth Hormone in the Obese Elderly Women (12주 복합운동이 비만 여성노인의 자립생활체력과 성장호르몬에 미치는 영향)

  • The purpose of this study was to observe self-reliance fitness and growth hormones in obese elderly women. Twenty-four females over the age of 65 years, who did not exercise regularly, were chosen as test subjects. All subjects were evaluated pre- and post-test. The results of this study were as follows; Results related to the experimental group revealed that there were statistically significant differences between the mean scores pre- and post-test in self-reliance fitness. Results related to the experimental group revealed that there were statistically significant differences between the mean scores pre- and post-test in growth hormone. Overall, participation in a 12-week combined exercise program had a positive impact on the self-reliance fitness and growth hormone in the obese elderly women. Based on the results of this study, obese elderly women are encouraged to participate in a combined exercises program on a regular basis.

The Cellular Localization of GnRH and LHR in Aged Female Mice

  • Gonadotropin releasing hormone (GnRH) centrally plays a role in control of the hypothalamic-pituitary-gonadal axis-related hormone secretions in the reproductive neuroendocrine system. In addition, hormone receptors like luteinizing hormone receptor (LHR) are important element for hormones to take effect in target organ. However, ageing-dependent changes in terms of the distribution of GnRH neurons in the brain and LHR expression in the acyclic ovary have not been fully understood yet. Therefore, we comparatively investigated those ageing-dependent changes using young (1-5 months), middle (11-14 months) and old (21-27 months) aged female mice. Whereas a number of GnRH positive fibers and neurons with monopolar or bipolar morphology were abundantly observed in the brain of the young and middle aged mice, a few GnRH positive neurons with multiple dendrites were observed in the old aged mice. In addition, acyclic ovary without repeated development and degeneration of the follicles was shown in the old aged mice than others. LHR expression was localized in theca cells, granulosa cell, corpora lutea and atretic follicle in the ovaries from young and middle aged mice, in contrast, old aged mice had few positive LHR expression on the follicles due to acyclic ovary. However, the whole protein level of LHR was higher in the ovary of old aged mice than others. These results are expected to be used as an important basis on the relationship between GnRH and LHR in old aged animals as well as in further research for reproduction failure.

Transcriptome and Small RNAome Analyses Reveal the Association of pre-harvest Sprouting and Heat Stress Response in Rice (Oryza sativa L.)

  • Pre-harvest sprouting (PHS) in rice (Oryza sativa L.) is one of the main problems associated with seed dormancy. PHS causes yield loss and reduction of grain quality under unpredictable humid conditions at the ripening stage, thus affecting the economic value of the rice crop. To resolve this issue, it is important to understand the molecular mechanism underlying seed dormancy in rice. Recent studies have shown that seed dormancy is affected by a large number of genes associated with plant hormones. However, the effect of heat stress on seed dormancy and plant hormones is not well understood. In this study, we compared the PHS rate as well as the transcriptome and small RNAome of the seed embryo and endosperm of two different accessions of rice, PHS-susceptible rice (low dormancy) and PHS-resistant rice (high dormancy) under three different maturation stages. We identified and verified the candidate genes associated with seed dormancy and heat stress-related responses in rice using quantitative real-time PCR. We newly discovered hormone-related genes, heat shock protein-related genes, and miRNAs potentially involved in PHS. These findings provide a foundation for understanding the dynamics of transcriptome and small RNAome of hormone- and heat stress-related genes, which affect PHS during seed maturation.
