NAD(P)H-quinone oxidoreductase-1 silencing modulates cytoprotection related protein expression in cisplatin cytotoxicity

  • NAD(P)H-quinone oxidoreductase-1 (NQO1) is a down-stream target gene of nuclear factor erythroid 2-related factor 2 (Nrf2), and performs diverse biological functions. Recently, NQO1 is recognized as an effective gene for the cytotoxic inserts with its diverse biological functions, which is focused on antioxidant properties. The aim of present study was to assess the impact of NQO1 knockdown on cytoprotection-related protein expression in cisplatin cytotoxicity by using small interfering (si) RNA targeted on NQO1 gene. Cytotoxicity of cisplatin on ACHN cells was assessed in a dose- and time-dependent manner after siScramble or siNQO1 treatment. After cisplatin treatment, cells were subjected to cell viability assay, western-blot analysis, and immunofluorescence study. The cell viability was decreased in the siNQO1 cells (50%) than the siScramble cells (70%) after 24 h of cisplatin ($20{\mu}M$) treatment. Moreover, cytoprotection-related protein expressions were markedly suppressed in the siNQO1 cells after cisplatin treatment. The expression of Nrf2 and Klotho were decreased by 20% and 40%, respectively, of that in siScramble cells. Nrf2 and Klotho activation were also decreased in cisplatin treated siNQO1 cells, confirmed by cytoplasm-tonuclear translocation. Our findings demonstrate that the increased cisplatin-induced cytotoxicity was accompanied by suppressed Nrf2 activation and Klotho expression in siNQO1 cells.

  • ICT 기술의 발전에 힘입어 다양한 모바일 어플리케이션이 속속들이 등장하고 이들이 개인의 삶 속에 깊이 파고든 시점, 영화산업의 새로운 성장판로를 모색하기 위하여 본 연구는 영화 콘텐츠 관련 어플리케이션의 성장 방안을 논의하였다. 실증연구를 통하여 영화 콘텐츠 관련 어플리케이션이 갖춰야 할 핵심요인을 탐색하기 위하여 본 연구에서는 정보시스템 성공 모델을 활용하여 각 품질요인이 영화 소비자의 영화에 대한 태도와 관람의도에 미치는 영향을 알아보았다. 연구 결과, 영화 콘텐츠 관련 어플리케이션의 품질요인 중 가장 중요한 요인은 정보 품질요인이었으며, 이어 서비스 품질이 차선으로 중요한 요인임이 도출되었다. 본 연구에서는 이와 같은 연구 결과를 바탕으로 국내 영화 산업의 발전을 위해 영화 콘텐츠 관련 어플리케이션들이 개선되어야할 방안을 제시하여 새로운 유통 채널로서의 영화 콘텐츠 관련 어플리케이션의 방향성을 제안하였다.

아동의 우울에 영향을 미치는 어머니 관련변인에 관한 연구 (The Study on the Effect of the Maternal Parenting Stress on the Children Depression)

  • The purpose of this research is to delineate the effect of maternal variables of the children's depression. Such variables as the mother's age, her education level and maternal parenting stress have been chosen for the analysis. The sample subject were 659 pairs of fifth/sixth grade of elementary school and first/second grade of middle school and their mothers. The data were analyzed frequency, percentage, factor analysis, Cronbach's alpha, ANOVA, Pearson's correlation, and stepwise multiple regression analysis. The major findings of the research are as follows : First, the children's depression is significantly related to mother's education level but the children's depression is not significantly related to mother's age. Second, parenting stress related to temperament, relationship and learning expectation is significantly related to children's depression. Third, the result of step multiple regression analysis on the effects of the maternal variables to the children's depression indicates that maternal parenting stress related to temperament, relationship and mother education level are the significant contributing factor. All in all, the maternal variables account for 7% of the children's depression score.

대인관계성향, 남성성유형에 따른 외모관련소비행동 연구 - 외모관심도의 매개효과 분석 - (Appearance-Related Consumption Behavior according to Interpersonal Relations and Masculinity - Mediated effect of Appearance Concern -)

  • This study identifies the influences of appearance-related consumption behavior according to interpersonal relations and masculinity examined through amediated effect of appearance concerns based on a structural study method. Five hypotheses were established to verify the relationships of interpersonal relations, masculinity and appearance-related consumption behavior. Questionnaires were administered to 201 males in their 20s to 50s living in Daegu South Korea. The SPSS 20.0 package utilized for data analysis included frequency analysis, factor analysis, correlation analysis, regression analysis and Cronbach's ${\alpha}$. This study utilized an Amos 21.0 program, a Confirmative Factor Analysis(CFA)and a Structural Equation Modeling(SEM). The results of the study are as follows: First, the appearance concern showed a partial mediation effect between measurement variables that verified the causal relationship of the structural model. Second, interpersonal relations showed a positive influence on masculinity. Third, masculinity showed a positive influence on the appearance concern. Fourth, masculinity had a positive influence on appearance-related consumption behavior. Fifth, interpersonal relations did not have a significant influence on appearance-related consumption behavior. Sixth, the appearance concern showed a positive influence on appearance-related consumption behavior. The findings of this study can influence a market segmentation strategy by predicting future emotional and new consumption markets strategies for male's appearance-related product.

학령기 행동문제의 위험요인과 보호요인으로서의 또래관계 (Peer Relationships as Risk and Protective Factors in the Behavior Problems of Elementary School Children)

  • This study examined relationships between peer variables and externalizing behavior problems of 290 4th and 5th grade children in elementary school. The children completed the Friendship Quality Scale and the peer nomination questionnaire. Parents and teachers rated behavior problems of children using the Connors' Rating Scale. Results indicated that rejected children had more behavior problems than popular, average, or neglected children. Socio-emotional supports from a close friend and satisfaction with the friend were negatively related to behavior problems. Behavior problems were mainly predicted by low peer acceptance, and behavior problems of rejected children were negatively and strongly related to socio-emotional supports of a close friend. In conclusion, peer rejection was a risk factor and a friend's support was a protective factor for externalizing behavior problems in 4th and 5th graders.

Japanism 디자인의 소비자 감성 연구 (A Study on the Consumer Sensibility of Japanism Design)

  • The purpose of this study was to investigate the characteristic and sensibility of Japanism fashion designs which represented by Japanese designers and Western designers. The stimulus were 29 pictures of contemporary fashion designs which represented the Japanism style fashion designs from fashion collections. The data were analyzed by Cluster analysis, Factor analysis, Multidimensional Scaling Method and Regression Analysis. The specific objectives were as follows ; 1) As result of design analysis, Japanism fashion sensibility is unique and good-looking. 2) As result of the factor analysis. 4 factors which are Attractiveness, Attention, Maturity and Hardness and softness. 3) According to sensibility positioning, The Japanism fashion design was classified by Decorative-Simple, Hard-Soft. 4) As result of the Regression Analysis, The preference of Japanism fashion design was related to attractive factor. 5) As result of the Regression Analysis. The buying desirable of Japanism fashion design was related to attractive, attentive and mature factor.

  • The acute toxicity of recombinant human epidermal growth factor, DWP-401 was evaluated in ICR mice of both sexes. Six groups of mice were administered orally or subcutaneously with 0, 0.125, 0.25, 0.5, 1 and 2 mg/kg of DWP-401. Abnormal clinical signs related to the compound were not observed, and no deaths occurred. Gross findings of necropsy revealed no evidence of specific toxicity related to DWP-401. LD$_{50}$ values for both male and female mice were evaluated to be over 2 mg/kg, which is approximately 2, 000 fold of presumed clinical dose.e.

환경지향적 소비자 행동에 관한 연구 -제주시 주부를 대상으로- (A study on the Environment Oriented Consumer Behavior of Housewives -As Related to Wives in Cheju City-)

  • The purpose of this study is to investigate かe factors related to the environment-oriented consumer behavior, and to analyze the effects of environmental knowledge, perception of environmental problems, and environment-oriented attitude on the environment-oriented consumer behavior. The data used in this study were collected through questionnaires on 535 wives living in Cheju City. The data were analvzed by using SPSS WIN program. The summaries of this study are as follows. Using behaviors was influenced by age and family income, but buying and disposing behavior are not influenced by socio-economic variables of wives. Environmental knowledge only has effects on disposing behaviors, and environment-oriented attitude behavior has effects on buying, using, and disposing behavior. But the perception of environmental problems hasn't effects on them. Wives' environment-oriented behavior are more effected by environment-oriented attitude than perception of environmental problems. In buying behavior, environment-oriented attitude is the most influential factor, and the reference group is the second influential factor. But in disposing behavior, the reference group is the most influential factor, and environment-oriented attitude is かe second influential factor.

취학전 아동의 학습준비도와 성취동기와의 관계 -아동용 성취동기 검사(나롱이)의 타당화- (The Relationship of Children's Achievement Motivation to Children's School Readiness)

  • The purpose of this research was to examine the concurrent validity of the Nalongyee children's achievement motivation test by studying its relationship to a children's school readiness test. The subjects were 335 preschool children (171 urban and 164 rural) selected from 8 public and 7 private kindergartens located in two metropolitan areas (Seoul and Pusan) and 5 rural counties in Kyungsang Nam Do. Instruments included the Nalongyee Children's Achievement Motivation Test by the author and the School Readiness "Lest by Unhai Rhee. Data were analyzed by Pearson r and Z-test. Readiness was positively related to the total score and sub-areas of achievement motivation (.10-.43). Sub-areas of the achievement motivation test were related to each of the 4 factors of the readiness test. The correlations ranged from .00-.35 with the personal-social response factor, .00-.26 with the associative vocabulary factor, .05-.31 with the number concept factor, and .03-.37 with the perception factor. Significant differences in correlations were found between urban and rural areas for self-confidence, interest in learning, interest in kindergarten and physical competence.

아파트 옥외공간 특성화에 대한 거주자의 인지특성에 관한 연구 (A Study on Resident's Environmental Cognition of Distinctive Outdoor Space in Apartment Estates)

  • This study was to evaluate residents' environmental cognition, satisfaction and needs of distinctive outdoor space in apartment housing estates. Distinctive outdoor space of 75 apartment estates throughout the country were investigated. And the residents of 16 apartment estates were asked how they cognize disticntive outdoor space qualities, and what activities they prefer in apartment's outdoor space. Thre important factors were extracted in residents' cognition of distinctive outdoor space qualities. First factor was naturalness of the greenery, second factor was outdoor activities especially related to fitness and exercise, and third factor was design of street furniture. Residents' satisfaction degree of distinctive outdoor space tended to increase as high as the occupied housing area. Places with water, tree shades, pedestrian way, greenery with flowers, playlots, fitness places and entrances of the estates were evaluated high in the order named. And places full with greenery, places related to fitness and exercisem, playlots were prefered by the residents.
