A Time to Fast, a Time to Feast: The Crosstalk between Metabolism and the Circadian Clock

  • The cyclic environmental conditions brought about by the 24 h rotation of the earth have allowed the evolution of endogenous circadian clocks that control the temporal alignment of behaviour and physiology, including the uptake and processing of nutrients. Both metabolic and circadian regulatory systems are built upon a complex feedback network connecting centres of the central nervous system and different peripheral tissues. Emerging evidence suggests that circadian clock function is closely linked to metabolic homeostasis and that rhythm disruption can contribute to the development of metabolic disease. At the same time, metabolic processes feed back into the circadian clock, affecting clock gene expression and timing of behaviour. In this review, we summarize the experimental evidence for this bimodal interaction, with a focus on the molecular mechanisms mediating this exchange, and outline the implications for clock-based and metabolic diseases.

  • As a part of the studies on the regulatory mechanism of gene expression by gibbrellic acid, the effects of $GA_3$ on the protein biosynthesis and phosphorylation in maize seedlings were investigated in the presence of actinomycin D. The activities of protein biosynthesis and phosphorylation in germinating seeds treated with $GA_3$ were greater than those of the control at the 3-day point after germination. It is assumed that the enhancement of protein biosynthesis by $GA_3$ in the presence of actinomycin D is due to the effects of $GA_3$ on the translational processes in which protein is produced from the mRNA synthesized previously.

  • cDNAs for long- and short-chain acyl-CoA oxidases in fatty acid $\beta$-oxidation were isolated and were characterized their enzymatical and molecular properties. Both oxidases were exclusively localized in glyoxysomes, indicating that glyoxysomes can completely metabolize fatty acids to acyl-CoA by their cooperative action. In order to clarify the regulatory mechanisms underlying degradation of storage oil, we tried to obtain glyoxysome-deficient mutants of Arabidopsis. We screened 2,4-dichlorophenoxybutyric acid (2,4-DB) mutants of Arabidopsis which have defects in glyoxysomal fatty acid $\beta$-oxidation. Four mutants can be classified as carrying alleles at three independent loci, which we designated pedl, ped2, and ped3, respectively (where ped stands for peroxisome defective). The characteristics of these ped mutants are described.

Interaction of Nonreceptor Tyrosine-Kinase Fer and p120 Catenin Is Involved in Neuronal Polarization

  • The neuronal cytoskeleton is essential for establishment of neuronal polarity, but mechanisms controlling generation of polarity in the cytoskeleton are poorly understood. The nonreceptor tyrosine kinase, Fer, has been shown to bind to microtubules and to interact with several actin-regulatory proteins. Furthermore, Fer binds p120 catenin and has been shown to regulate cadherin function by modulating cadherin-${\beta}$-catenin interaction. Here we show involvement of Fer in neuronal polarization and neurite development. Fer is concentrated in growth cones together with cadherin, ${\beta}$-catenin, and cortactin in stage 2 hippocampal neurons. Inhibition of Fer-p120 catenin interaction with a cell-permeable inhibitory peptide (FerP) increases neurite branching. In addition, the peptide significantly delays conversion of one of several dendrites into an axon in early stage hippocampal neurons. FerP-treated growth cones also exhibit modified localization of the microtubule and actin cytoskeleton. Together, this indicates that the Fer-p120 interaction is required for normal neuronal polarization and neurite development.

  • In neurons and some endocrine cells, $Ca^{2+}$ plays a pivotal role as the final signal for rapid stimulus-evoked release of neurotransmitters and hormones. In contrast, intracellular signals other than $Ca^{2+}$, such as protein A and C, were mor emphasized the exocytosis in non-neuronal cells. We questioned if the regulatory mechanisms of exocytosis are fundamentally different between cell types.(omitted)

  • In this study, an application of crack morphology variables in the Leak-Before-Break(LBB) evaluation for nuclear piping systems is investigated, including influences on the leakage crack size and crack instability loads. The crack surface roughness and the number of flow turns as a function of the crack opening displacement are applied to LBB evaluations for KSNP pressurizer surge line, for which fatigue and stress corrosion cracking are considered as failure mechanisms. As a result, there would be a significant impact on safety margins to acceptance criteria for the surge line if crack morphology variables are applied additionally to the current regulatory guide without re-analyses for justification of safety factors being applied on the leakage crack size and piping loads for evaluations.

  • Candida albicans possess cyanide-resistant respiratory pathway, which is mediated by alternative oxidase. The activity of alternative oxidase has been found to be dependent on several regulatory mechanisms. In order to investigate the influence of D-erythroascorbic acid on respiration of C. albicans, the respiratory activity of the cells was measured with oxygen monitor. ALO1 is known to encode D-arabinono-1,4-lactone oxidase that catalyses the final step of D-erythroascorbic acid biosynthesis in C. albicans.(omitted)

  • Our previous studies showed that the relaxation defect of diabetic heart was due to the changes in the expressional levels of SR $Ca^{2+}$-ATPase and PLB. In the diabetic heart contractile abnormalities were also observed, and one of the mechanisms for these changes could include alterations in the expression and/or activity levels of various $Ca^{2+}$ regulatory proteins involving cardiac contraction.(omitted)

  • Monocyte interaction with fibronectin (FN) mediates specific cell surface receptors and results in cell attachment and differentiation. Several cell-mediated activities for various fragments of FN have been documented. To investigate the regulatory mechanisms of monocyte differentiation by cell binding domains of FN and their receptors, cell attachment-, cell migration-, and its respective inhibition assay were carried out. Monocyte recognizes 38-kDa domain distinctively from its recognition of 85-kDa domain, and the heparin-binding site of the 38-kDa fragment is not involved in monocyte adhesion. Based on these experimental results, it can be suggested that monocvte/macrophase interacts with at least two different sites in FN, which is critical step in cell adhesion and (or) migration.
