The Effect of Growth Regulators and Light Quality on the Changes in Protein Pattern of Callus from Intergeneric Protoplast Fusion between Nicotiana tabacum and Solanum nigrum (Nicotiana tabacum과 Solanum nigrum의 속간 원형질체 융합에서 유도된 캘러스의 단백질 양태변화에 미치는 생장조절제 및 광선의 효과)

  • The effect of growth regulators (NAA, BA and $ extrm{GA}_3$) and light (blue, red and far-red) on the changes in total protein and thylakoid membrane protein pattern of callus from intergeneric protoplast fusion between Nicotiana tabacum and Solanum nigrw were investigated. When the callus were irradiated with different wavelengths of light, blue and red light accelerated the synthesis of total proteins and thylakoid membrane proteins. Particularly, red light led to an increase in the protein synthesis compared to blue light. When the callus were subjected to various combinations of growth regulators, NAA+$ extrm{GA}_3$ and NAA+BA treatments induced remarkable increase of total proteins and thylakoid membrane proteins accumulation, particularly in the combination of NAA+$ extrm{GA}_3$. NAA.$ extrm{GA}_3$ treatment with irradiation of red ligh showed highest value in the accumulation of total proteins and thylakoid membrane proteins. We conclude that simultaneous application of red light and NAA+$ extrm{GA}_3$ treatment may induce synergistic effect in the synthesis of total proteins and thylakoid membrane Proteins.

In vitro Shoot Propagation Derived from Stem and Shoot Tip in Hovenia dulcis var. koreana Nakai by Plnat Growth Regulators and Light Resources (식물생장조절제 및 광원처리에 따른 헛개나무 줄기와 경정유래 신초의 기내증식)

  • This study was conducted to examine effects of plant growth regulators and light resources on the formation of multiple shoot and plant regeneration of Hovenia dulcis var. koreana Nakai. Stem and shoot tip were cultured on MS medium or WPM supplemented with various plant growth regulators. At the single treatment, the highest shoot formation was obtained when stem explants were cultured on WPM supplemented with kinetin $1.0mg{\cdot}L^{-1}$. MS medium containing NAA 0.1 and TDZ $0.1mg{\cdot}L^{-1}$ gave the best results for shoot induction rate and shoot growth in combination treatments. Of the BAP and kinetin tested, BAP $0.5mg{\cdot}L^{-1}$ on WPM was found to be more effective for shoot growth from shoot tip. Under white fluorescent light treatment, shoot growth was much higher than blue, red LED treatments. Root induction from in vitro growth of plantlet was the best on WPM supplemented with $1.0mg{\cdot}L^{-1}$ IBA. The results suggest that selection of plant growth regulators and light resources could be important factor to achieve an efficient in vitro growth.

  • Proteins are involved in promoting or controlling virtually every event on which our lives depend. Proteins are synthesized in cytosol and in the endoplasmic reticulum where their synthesis machinery are tightly controlled. However, not all of newly synthesized proteins are survived and conduct their essential functions to maintain cell's lives. It was reported that one-third of synthesized proteins are rapidly destroyed by proteasome under the most physiological conditions. full-length translated proteins, which survived, must undergo proper folding and assemble process. Some proteins are spontaneously folded while others require molecular chaperones and folding enzymes to be properly folded. Molecular chaperones are ubiquitously present within the subcellular organelles and from bacteria to animals and plants. Among those members of Hsp70 family have been extensively studied and their regulators have been discovered in the last decade. Here, a brief overview is presented for functional mechanism of Hsp70 homologues and the roles of their regulators. Since biological function of Hsp70 family other than chaperonic function are expending the review would give basic understanding of partnership between Hsp70 family and their regulators.

Effects of Plant Growth regulators on Rapid in vitro Propagation of Camptotheca acuminata from Axillary Buds

  • An efficient method for the rapid micropropagation of Camptotheca acuminata from axillary buds was established by application of various plant growth regulators. Among various cytokinins, $0.5mg\;L^{-1}$ BA showed the best performance on shoot multiplication, number average multiple shoots up to 10.8. The propagated shoot cuttings in vitro were elongated on NN basal medium without plant growth regulators. The secondary multiple shoots were induced at the site of initially induced buds. Rooting was induced directly near the base of the shoot on half-strength NN medium containing $0.5mg\;L^{-1}$ of IBA, whereas high concentration of $1.0mgL^{-1}$ IBA could induce callus at the base of the shoot. The camptothecin content, anticancer compound of the micropropagated plants was contained in various tissues. Camptothecin contents were 1.8 and $2.5mg\;g^{-1}$ dry weight in stems from propagated in vitro and mother plant, respectively. This result may be used to develop strategies for large-scale propagation of elite C. acuminata trees.

Effect of Plant Growth Regulators in Minimizing Low Temperature Stress in Rice(Oryza sativa L.) (식물생장조절제(植物生長調節劑) 처리(處理)가 벼(Oryza sativa L.) 냉해경감(冷害輕減)에 미치는 영향(影響))

  • This experiment was conducted at Andong county cropping season to investigate rice response and consequent changes in the physiological activities and agronomic characters of rice as affected by growth regulator treatments, and to determine how low temperature controls some plant growth mechanisms. Plant growth regulators, particularly ABA, applied to 15 DAT regardless of concentration, gave higher tar plant height and tiller number after 10days of low temperature treatment than did the untreated plant. However, the differences in plant height and tiller number among treatments diminished to plant growth. Chlorophyll content seemed to be enhanced by growth regulators under low temperature condition. The 10-day treatment of low temperature decreased chlorophyll content by more than 17% compared with the untreated control. The oxidizing activity of roots decreased sharply to 52.2a after 10 days in low temperature condition. On the other hand, ABA at concentration of $10^{-4}M$, highly attains oxiding activity of roots. Generally, plant growth regulators applied at 15 DAT under low temperature reduced grain yield more than did the untreated control which can be attributed to decreased yield components. When growth regulators were applied to booting stage, the tiller number did not show significant difference, whereas the significant difference in culm length at harvest was observed in all treatment as a result of different concentration of growth regulators. The growth regulators generally exhibited a significant effect on yield component except on panicle length. Grain yield of low temperature treated plot at booting stage was significantly influenced by application of growth regulators. This was due to decreased number of spikelet and low filled spikelet percentage. However, TIBA treatment at booting stage showed no significant differences in grain yield.

Effect of Pollination Method on Fruit Setting and Quality of Oriental Melon(Cucumis melo L. var, makuwa Makino) (착과방법이 참외의 착과 및 품질에 미치는 영향)

  • A study was conducted on three pollination methods on oriental melon(sageageol-ggul) grafting with pumpkin(seongjutozoa) for the labor-saving and to improve fruit set. Fruit weight, flesh thickness and fruit setting rate of oriental melon were greater in growth regulators treatment than those of pollinated by bees. Sugar content and hardness of fruits pollinated by bees were higher than those of by growth regulators. From the last ten days of the February to the first ten days of the March, fruit setting rate was $95\%$ in fruit setting growth regulators, whereas it was $46\%$ and $45\%$ in pollinated by honey and bumble bee, respectively. After the middle of March, the percentage of fruit setting was >$98\%$ in all the pollination methods. The cultivation under plastic houses of oriental melon, suitable fruiting time far the pollination by bees was decided after middle days of the March. Chromaticity and especially the value of 'a' of fruit of oriental melon pollinated by bees were higher than those of growth regulators. The percentage of fermented fruits of bee pollinated and growth regulators treated was $6.7\~9.1\%\;and\;28.1\%$, respectively. The weight of 100 seeds of bees pollinated were higher than that of growth regulators. The more increased the weight of 100 seeds the less appeared the rate of fermented fruits. The percentage of marketable fruits of the honey and bumble bee pollinated and that of growth regulators treated was $82\%,\;80.3\%\;and\;62.5\%$, respectively. The decreasing rate of fruit weight during storage of bees pollinated was less than those of growth regulators. In these results, the introduction of honey bee and bumble bee for the pollination of oriental melon was able to labor-saving of fruit set and increase of fruit quality.

  • Effect of plant growth regulators and end-product on the enzyme activities in cotyledons of Vigna angularis during germination was investigated by measuring the changes of $\alpha-amylase$ activities in attached and detached cotyledons applied growth regulators and sugars. The higher levels of $\alpha-amylase$ in detached cotyledons than those in cotyledons attached to the embryonic axis were due to both faster synthesis and slower degradation of the enzyme in the detached cotyledons than in the attached cotyledons. Levels of $\alpha-amylase$ activity were reduced by high concentrations of glucose and sucrose, and it is suggested that this effect was caused mostly by osmotic stress and partly by end-product repression. In detached cotyledons exogenously supplied $GA_3,$ IAA, kinetin, or their combinations has a small promotive effect on the developmental patterns of $\alpha-amylase$ activity ABA and uniconazole both prevented the synthesis of $\alpha-amylase$. Glucose inhibition of enzyme activity was partly reversed by the application of $GA_3,$ and CAMP. $GA_3,$ and cAMP seemed to act through a similar mechanism. The addition of inhibitors of protein and RNA synthesis largely prevented the increase of enzyme activity in the presence or absence of exogenous $GA_3,$. The pretreatment experiments with canavanine indicated that the earlier the time of addition was, the lower the amylase activity was.

Enzymatic Characteristics of Biosynthesis and Degradation of Poly-$\beta$-hydroxybutyrate of Alcaligenes latus

  • The enzymatic characteristics of Alcaligenes latus were investigated by measuring the variations of various enzyme activities related to biosynthesis and degradation of poly-${\beta}$-hydroxybutyrate (PHB) during cultivation. All PHB biosynthetic enzymes, ${\beta}$-ketothiolase, acetoacetyl-CoA reductase, and PHB synthase, were activated gradually at the PHB accumulation stage, and the PHB synthase showed the highest value among three enzymes. This indicates that the rate of PHB biosynthesis is mainly controlled by either ${\beta}$-ketothiolase or acetoacetyl-CoA reductase rather than PHB synthase. The enzymatic activities related to the degradation of PHB were also measured, and the degradation of PHB was controlled by the activity of PHB depolymerase. The effect of supplements of metabolic regulators, citrate and tyrosine, was also investigated, and the activity of glucose-6-phosphate dehydrogenase was increased by metabolic regulators, especially by tyrosine. The activities of ${\beta}$-ketothiolase and acetoacetyl-CoA reductase were also activated by citrate and tyrosine, while the activity of PHB depolymerase was depressed. The increased rate and yield of PHB biosynthesis by metabolic regulators may be due to the increment of acetyl-CoA concentration either by the repression of the TCA cycle by citrate through product inhibition or by the activation of sucrose metabolism by the supplemented tyrosine.

Role of Regulators of G-Protein Signaling 4 in $Ca^{2+}$ Signaling in Mouse Pancreatic Acinar Cells

  • Regulators of G-protein signaling (RGS) proteins are regulators of $Ca^{2+}$ signaling that accelerate the GTPase activity of the G-protein ${\alpha}$ -subunit. RGS1, RGS2, RGS4, and RGS16 are expressed in the pancreas, and RGS2 regulates G-protein coupled receptor (GPCR)-induced $Ca^{2+}$ oscillations. However, the role of RGS4 in $Ca^{2+}$ signaling in pancreatic acinar cells is unknown. In this study, we investigated the mechanism of GPCR-induced $Ca^{2+}$ signaling in pancreatic acinar cells derived from $RGS4^{-/-}$ mice. $RGS4^{-/-}$ acinar cells showed an enhanced stimulus intensity response to a muscarinic receptor agonist in pancreatic acinar cells. Moreover, deletion of RGS4 increased the frequency of $Ca^{2+}$ oscillations. $RGS4^{-/-}$ cells also showed increased expression of sarco/endoplasmic reticulum $Ca^{2+}$ ATPase type 2. However, there were no significant alterations, such as $Ca^{2+}$ signaling in treated high dose of agonist and its related amylase secretion activity, in acinar cells from $RGS4^{-/-}$ mice. These results indicate that RGS4 protein regulates $Ca^{2+}$ signaling in mouse pancreatic acinar cells.

PQ Control of Micro Grid Inverters with Axial Voltage Regulators

  • This paper presents a PQ control strategy for micro grid inverters with axial voltage regulators. The inverter works in the voltage-controlled mode and can help improve the terminal power quality. The inverter has two axial voltage regulators. The 1st regulator involves the output voltage amplitude and output impedance, while the 2nd regulator controls the output frequency. The inverter system is equivalent to a controllable voltage source with a controllable inner output impedance. The basic PQ control for micro grid inverters is easy to accomplish. The output active and reactive powers can be decoupled well by controlling the two axial voltages. The 1st axial voltage regulator controls the reactive power, while the 2nd regulator controls the active power. The paper analyses the axial voltage regulation mechanism, and evaluates the PQ decoupling effect mathematically. The effectiveness of the proposed control strategy is validated by simulation and experimental results.
