국내 주요 가공식품에 대한 위해순위 결정 (Determining of Risk Ranking for Processed Foods in Korea)

  • 우리나라 식품공전상에 수록된 식품군 및 식품유형에 대한 위해순위(risk ranking) 결정은 식품안전관련 위생관리 우선순위 적용 등에 있어 중요한 수단으로 작용할 수 있다. 하지만 세계적인 관심의 증대에도 불구하고 아직까지 국내에서는 체계적으로 수행된 적이 없었다. 본 연구는 식품공전에 포함되어 있는 101개의 식품유형에 대하여 상대적인 위해 순위를 결정하였다. 상대위해도는 노출평가, 심각성평가 및 섭취량 추정부분으로 구성된 위해평가모델을 이용하였다. 추정결과, 즉석섭취식품(RTE)이 가장 높은 위해수준을 나타내었고, 다음이 어육가공품, 빵류 등의 순으로 나타났다. 이러한 식품공전상의 식품유형에 대한 과학적인 위해순위 결과는 식품안전에 있어 가장 많은 영향을 미치는 식품과 이들 식품에 대한 관리목표를 설정하는데 크게 활용될 수 있을 것이다.

Evaluation of measurement uncertainty for quantitative determination of chlorite and chlorate in fresh-cut vegetables using ion chromatography

  • This study aimed to evaluate the measurement uncertainty for the quantitative determination of chlorite and chlorate in ready-to-eat fresh-cut vegetables using ion chromatography with a hydroxide-selective column. One gram of the homogenized sample in deionized water was sonicated and centrifuged at 8,500 rpm. The supernatant was purified by passing it through a Sep-Pak tC18 cartridge, followed by chromatographic determination using a Dionex IonPac AS27 column. The linearity of the calibration curves, recovery, repeatability, and reproducibility of the method were satisfactory. The method detection limit was estimated to be approximately 0.5 mg/kg. Each uncertainty component was evaluated separately, and the combined and expanded uncertainty values were calculated at the 95% confidence level. The measured concentrations for 3 mg/kg of chlorite and chlorate standard materials were $3.18{\pm}0.32$ and $3.10{\pm}0.42mg/kg$, respectively. These results confirmed the reliability of the developed method for measuring the two chlorine-based oxyanions in fresh-cut vegetables.

식품 유래 대장균군의 발색 배지 분리 효율 비교 (Comparative Evaluation of Selective Chromogenic Media for Coliforms Bacteria Isolated from Food)

  • 식품에서 대장균군을 효율적으로 분리하기 위해 대장균군 선택배지 5종: Chromocult Chromocult coliform agar (Merck), Hicrome coliform agar (Sigma), CHROMagar ECC, Brilliance E.coli/coliform medium (OXOID), endo agar (Merck)을 선별하여 식품 분리 대장균군 83종 및 표준균주 21종에 대해서 민감도와 특이도를 분석한 결과 Chromocult coliform agar와 HICrome coliform agar에서 94%도 민감도가 가장 높았으며, Brilliance E.coli/coliform medium는 93%, CHROMagar ECC는 92%, ENDO agar는 74%의 민감도를 나타냈다. Chromocult coliform agar와 HICrome coliform agar는 대장균군의 회수율도 높았다. 그러므로 Chromocult coliform agar와 HICrome coliform agar는 대장균군을 분리하는데 가장 효율적인 배지로 생각된다.

볶음밥의 Bacillus cereus 위해 수준 및 위해 관리를 위한 모니터링 기준 설정 (Prevalence of Bacillus cereus from Fried Rice Dishes and Monitoring Guidelines for Risk Management)

  • Contamination levels of aerobic colony counts, coliforms and pathogenic bacteria were tested in fried rice dishes to monitor quality for risk management. The prevalence of Bacillus cereus in dishes from 8 Chinese-style restaurants and 2 institutional foodservices was 10%, and the bacteria's contamination levels was 3.47 log CFU/g. Echerichia coli, Staphylococcus aureus, and Salmonella were not detected in any of the 10 samples. However, for their aerobic colony counts and numbers of isolated coliforms, the samples were 30% and 70% over the microbial criteria, respectively, for ready-to-eat foods presented in the Korean Food Code. This suggests that fried rice dishes, although cooked with oil at high temperature, require special care. For the prediction of the growth curve of B. cereus spp. in the fried rice samples, an experiment design of 3 storage temperatures ($7^{\circ}C$, $35^{\circ}C$, $57^{\circ}C$) x 5 storage times (0 h, 2 h, 4 h, 6 h, 24 h) was applied. The sample exposed to $35^{\circ}C$ showed no B. cereus spp. at 0 h; however, there was a tendency of slow growth (1.0 log CFU/g) after 4 hours of storage and then faster growth at 6 h (3.7 log CFU/g) and 12 h (4.7 log CFU/g), showing a growth rate of 0.56 log CFU/g/hr. These results indicate that fried rice, despite being heat-treated, can become heavily contaminated with B. cereus spp. when held over 2 hours at room temperature. However, the samples stored at $7^{\circ}C$ and $57^{\circ}C$ over 24 hours were not contaminated with B. cereus. Based on these results, management guidelines for controlling B. cereus are suggested.

Microbial Contamination by Bacillus cereus, Clostridium perfringens, and Enterobacter sakazakii in Sunsik

  • The powdered cereal sunsik is a partially thermal-processed product that required safety evaluations for food-borne pathogens. Thirty-six sunsik products from Korean markets were collected and analyzed for contamination by total viable cell counts, coliforms, Escherichia coli, and the spore-forming Clostridium perfringens and Bacillus cereus. Enterobacter sakazakii, as a newly emerging pathogen, was also analyzed. Approximately 28% of sunsik were contaminated at 5 log CFU/g for total viable counts. Coliforms and E. coli were detected in 33 and 4% of the samples, respectively. The spore-forming B. cereus was found in 42% of the samples at a maximal level of 3 log CFU/g on average. About 6% the samples were contaminated with Cl. perfringens at an average level of 15 CFU/g. Forty-five % of sunsik contained E. sakazakii, at levels from 0.007 to over 1.1 cell/g by MPN method. In addition, one sunsik product for infants and children showed over 3 log CFU/g for both B. cereus and E. sakazaki. Therefore, concern should be placed on controlling for microbial hazards such as B. cereus and E. sakazakii in sunsik, particularly for the products fed to infants under 6 months of age.

냉장 중 품질변화 측정 지표 개발을 위한 냉장고 형태별(개방형 및 도어형) 저장 중 주요 냉장 식품의 품질변화 측정 (Comparative Assessment of Quality Changes in Refrigerated Foods Stored in Open-type and Door-type Refrigerators: Towards Developing Quality Indicators)

  • This study was conducted to investigate the impact of refrigerator type and temperature fluctuations on the quality changes of refrigerated foods. Phycochemical and sensory quality, as well as microbial growth, were measured for various foods stored in open- or door-type refrigerators set at 5℃ during storage. The average temperatures recorded were 11.50±1.14℃ in an open refrigerator, and 6.34±0.97℃ in a closed refrigerator. The average surface temperatures of the food items were 9.60±1.20 and 6.00±0.80℃ for open and closed refrigerators, respectively. Significant changes in color and appearance quality were observed in lettuce, mackerel, ground beef, and cut pineapples when stored in open refrigerators. Ready-to-Eat foods such as gimbap and sandwiches exhibited higher levels of microbiological proliferation when stored in open refrigerators compared to closed refrigerators. Processed foods, such as sterilized milk and packaged tofu, did not show significant differences in quality among various types of refrigerators. The installation of refrigerator doors can effectively minimize temperature fluctuations caused by external factors, thereby reducing variations in food quality. These findings provide essential insights into the quality changes associated with the implementation of refrigerator doors, serving as fundamental data for ensuring optimal food preservation.

Microbiological Quality and Safety Assessment of Commercial Ready-to-Eat Side Dishes Sold in Gyeonggi-do

  • 2019년 경기도내 전통시장, 대형마트, 반찬전문점에서 수거한 반찬류의 미생물 품질을 조사하였다. 반찬류 108건의 식중독 원인균을 검사하였고, 그 중 75건에 대해서는 구매 장소별, 조리 방법별 위생세균 검사를 진행하였다. 14건(12.9%)에서 Bacillus cereus가 검출되었으며, 나머지 94건에서는 식중독 원인균이 검출되지 않았다. 위생세균 검사에서 일반세균의 평균 검출량(범위)은 전통시장이 5.8 log CFU/g (3.0-8.2 log CFU/g), 대형마트는 4.3 log CFU/g (2.3-7.8 log CFU/g), 반찬전문점에서는 3.8 log CFU/g (0.0-6.9 log CFU/g)로 나타났으며, 구입 장소에 따른 유의적인 차이가 있었다(Ρ <0.05). 전통시장의 일반세균수와 대장균군은 대형마트, 반찬점과 통계적으로 유의한 차이가 있었고, 생채류, 나물류, 볶음류, 젓갈류, 조림류 순으로 일반세균수와 대장균군이 높게 검출되었다. 콩나물 무침의 보관온도별 일반세균수의 변화는 냉장보관(4℃)에서는 72시간 경과에서도 큰 변화가 없었으나, 상온보관(20℃) 및 고온보관(35℃) 시 구입 후 각각 9시간, 6시간 경과 시 부패의 가능성이 제기되어 구입 즉시 냉장보관 할 것을 권장하며, 제품을 판매하는 시설에서도 냉장보관을 하여야할 것이다.

Antimicrobial Activity of Oleanolic Acid on Listeria monocytogenes under Sublethal Stresses of NaCl and pH

  • The objective of this study was to evaluate the inhibition of Listeria monocytogenes growth by oleanolic acid under sublethal stresses of NaCl and pH. L. monocytogenes ATCC15313 (6 log CFU/mL) was inoculated in microplate wells containing brain heart infusion (BHI) broth supplemented with oleanolic acid in various amounts (0, 0.25, 0.5, 1.0, 1.5, 2.0, and $4.0\;{\mu}g/mL$), and different pHs (5 and 7) and NaCl concentrations (0, 3, and 6%), followed by incubation under accelerated storage condition ($37^{\circ}C$, 48 h). The optical density (OD) of the samples was measured at 0, 6, 12, 24, and 48 h at 600 nm. After the lag phase duration was observed at the early stage of incubation, the OD values of L. monocytogenes significantly increased (p<0.05) in BHI broth formulated with 0 and 3% of NaCl during accelerated storage at pH 5 and 7. However, the growth of L. monocytogenes in 6% NaCl and at less than $0.5\;{\mu}g/mL$ of oleanolic acid had no growth at pH 5 and only gradual growth at pH 7. Moreover, L. monocytogenes generally had lower OD values as the concentrations of oleanolic acid increased. As expected, the OD values of L. monocytogenes were generally higher (p<0.05) at pH 7 than at pH 5. These results indicate that oleanolic acid should be useful in inhibiting the growth of L. monocytogenes.

Evaluation of Microbiological, Physicochemical, and Sensory Properties of Galbi-jjim Prepared by Sous-vide and Cookchill Method at Different Temperatures

  • The aim of this study was to evaluate the physico-chemical, sensory, and microbiological properties of ready-to-eat Korean traditional seasoned beef ribs ("galbi-jjim") prepared by sous-vide/cookchill technology during storage at three different temperatures (4, 10, and $20^{\circ}C$). Beef short ribs marinated in soy sauce for 24 h at $3^{\circ}C$ were packed with vegetables under vacuum. Vacuum-packed beef ribs mixed with vegetables were heated at $90^{\circ}C$ for 90 min in a water bath, and then immediately chilled below $3^{\circ}C$ within 120 min in an ice slurry. Physicochemical (pH, water activity, TBARS, $L^*a^*b^*$ color, and texture profile), sensory (appearance, odor, flavor, texture, and acceptance) and microbiological (Coliform, Escherichia coli, food-borne pathogenic bacteria) properties of the samples were determined during storage at different temperatures. Results showed that pH, $a_w$, and sensory evaluation of products were not affected in any consistent way as a function of either storage duration or temperature. Coliform, E. coli and food-borne pathogens were not detected during storage at any temperature. However, TBARS significantly increased during storage period (p<0.05). Based on TBARS values, SV/CC "galbi-jjim" can be stored for 15 d, 12 d and 1 d at 4, 10 and $20^{\circ}C$, respectively.

유통중인 파프리카, 딸기 및 토마토의 생물학적 위해요소 분포 조사 (Biological Hazard Analysis of Paprikas, Strawberries and Tomatoes in the Markets)

  • 생활수준의 향상으로 건강과 웰빙에 대한 관심이 늘면서 즉석섭취(ready-to-eat) 농산물의 수요가 증가하고 있다. 이들 농산물중 파프리카, 딸기, 토마토 등은 우리나라의 대표적인 수출 농산물로서 안전성 확보가 중요하다. 본 연구는 대전시내 2개 백화점과 2개 대형할인점에서 유통 중인 파프리카, 딸기 및 토마토를 대상으로 총호기성균, 대장균군, 식중독 유발 세균 및 기생충의 오염 수준을 배지 배양법, multiplex PCR, 상업용키트 및 현미경으로 조사하였으며, 또한 세척방법별 세균 감소율을 평가하였다. 유통중인 파프리카, 딸기 및 토마토의 평균 총호기성균 수는 $1.3{\times}10^4{\sim}1.8{\times}10^5$CFU/g, 평균 대장균군의 수는 $1.4{\times}10^3{\sim}9.6{\times}10^3$CFU/g 범위이었으며, 세균 오염 정도는 매장별로 유의한 차이를 보이지 않았다. 총호기성균 및 대장균군의 오염 정도는 딸기에서 가장 높았으며, 딸기에서 미확인 기생충 충란이 발견되었다. 조사한 농산물에서 가장 흔히 분리되는 세균은 Enterobacter cloacae 이었으며, 식중독 유발 세균중 Staphylococcus aureus가 딸기에서 발견 되었으나 그 외의 식중독 유발 세균은 분리되지 않았다. 딸기를 대상으로 세척방법별 세균 감소율을 조사한 결과 수돗물로 5회이상 세척시 일반세균의 80%이상을 제거할 수 있었으며 야채용 세제를 사용하거나 초음파 세척시 세균 감소율이 증가하였다. 이상의 결과로 보아, 파프리카, 딸기 및 토마토의 세균 및 기생충의 오염 정도를 조사한 결과 딸기에서 가장 많은 생물학적 오염을 보였으며, 이들 농산물들의 생물학적 오염 정도는 구입 매장별로 유의한 차이를 보이지 않았다.
