Cytoprotective effects of eupatilin, a novel antioxidative flavone, in oxidative stress- induced gastric mucosal cell damage

  • Alcohol, Helicobacter pylori, stress and NSAIDs-activated neutrophils all produce reactive oxygen species (ROS), which play an important role in gastric mucosal damage. Eupatilin is an active component of Artemisia asiatica possessing cytoprotective effect. The effect of eupatilin on the production of ROS and cellular damage in AGS and ECV304 cells were evaluated to prove the cytoprotective action against the above mentioned gastric mucosal cell damages. A Small Antenna of High Radiation Efficiency Employing a Ground Radiator (그라운드 방사체를 활용한 고효율의 소형 안테나)

  • This paper introduces the method to utilize a terminal ground as a radiator only using reactive components without an antenna structure. Characteristics of the proposed antenna is compared with that of the meander IFA on the same ground plane($40{\times}20mm^2$) for the bluetooth band. From simulation and measurement data, it is found that the proposed antenna using only capacitors provides the highest radiation efficiency. This is because of that the higher inductance reduces radiation resistance of a ground and the capacitor has a lower loss resistance comparing to that of the IFA or the inductor. In spite of the high radiation efficiency, the area ($5{\times}2.5mm^2$) of the proposed antenna is less than half of the area ($12{\times}2.5mm^2$) of the IFA.

  • Conventional average power theory has been used to design and control active power filters But compensating reference currents of active power filters calculated by conventional average power theory are definitively influenced by three phase source voltage conditions such as unbalance or distortion. This paper presents a new average power algorithm for active power filters which can detect symmetrically equally active or fundamental reactive currents in each phase based on decomposition of fundamental reactive component and harmonics under unbalanced power conditions. The effectiveness of the proposed algorithm is demonstrated by MATLAB/SIMULINK simulation and experimental results for a three wire distribution system with 15% unbalanced source voltages.

Antioxidant Activity of the Salt Marsh Plant Corydalis heterocarpa (염주괴불주머니 (Corydalis heterocarpa) 의 항산화 활성)

  • The antioxidant activities of two crude extracts ($CH_2Cl_2$ and MeOH) and their solvent fractions (n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions) from Corydalis heterocarpa were determined by evaluating authentic $ONOO^-$ and $ONOO^-$ generated from SIN-1 (3-morpholinsydnonimine) in vitro as well as by measuring the degree of occurrence of intracellular reactive oxygen species (ROS) and nitric oxide (NO). Scavenging activities of solvent fractions on authentic $ONOO^-$ increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions, while those on $ONOO^-$ generated from SIN-1 increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions. In addition, all solvent fractions effectively inhibited the intracellular ROS and NO levels. The n-BuOH fraction especially exhibited the strongest ROS scavenging effect. Further purification of n-BuOH fraction led to the isolation of cnidimoside A, which presented the potent ROS scavenging effect at $10\;{\mu}M$. From these results, extracts of C. heterocarpa and its component, cnidimoside A, were predicted to be potentially useful as ingredients for protecting against oxidation.

Functional characterization of $P_{2X}/P_{2Y}$ receptor in isolated swine renal artery

  • To understand the role of $PM_{2X}/P_{2Y}$ receptor in cortex region of kidney and renal artery, molecular and functional analysis of $PM_{2X}/P_{2Y}$ receptor by pharmacophysiological skill in conventional swine tissues were performed. In functional analysis of $P_{2Y}$ receptor for vascular relaxation, 2-methylthio adenosine triphosphate, a strong agonist of $P_{2Y}$ receptor, induced relaxation of noradrenaline (NA)-precontracted renal artery in a dose-dependent manner. Strikingly, relaxative effect of ATP, 2-msATP, agonists of $P_{2Y}$ receptor, abolished by treatment of reactive blue 2, a putative $P_{2Y}$ receptor antagonist. In contrast, no significant differences of gene encoding $PM_{2X}/P_{2Y}$ and protein expression in immortalized suprachiasmatic nucleus from brain, primary isolated vascular smooth muscle cells from renal artery of pigs and HEK293 from human embryonic kidney under with/without adenosine triphosphate were observed. Taken together, the relationship between molecular and functional characteristic of $PM_{2X}/P_{2Y}$ receptors in conventional pig should be considered that they are another important factor which regulate the kidney function in swine. Based on this study, we propose the purinergic receptor as well as adrenergic and cholinergic receptors is an essential component of the renal homeostasis.

  • Protein O-GlcNAcylation, dictated by cellular UDP-N-acetylglucosamine (UDP-GlcNAc) levels, plays a crucial role in posttranslational modifications. The enzyme GlcNAc kinase (NAGK, E.C. 2.7.1.59) catalyzes the formation of GlcNAc-6-phosphate, which is a major substrate for the biosynthesis of UDP-GlcNAc. Recent studies have revealed the expression of NAGK in different types of cells especially in neuronal dendrites. Here, by immunocytochemistry (ICC) and immunonucleochemistry (INC) of cultured rat hippocampal neurons, HEK293T and GT1-7 cells, we have showed that NAGK immuno-reactive punctae being present in the nucleoplasm colocalized with small nuclear ribonucleoprotein-associated protein N (snRNPN) and p54NRB, which are speckle and paraspeckle markers, respectively. Furthermore, NAGK IR cluster was also found to be colocalized with GTF2H5 (general transcription factor IIH, polypeptide 5) immuno reactive punctae. In addition, relative localization to the ring of nuclear lamin matrix and to GlcNAc, which is highly enriched in nuclear pore complexes, showed that NAGK surrounds the nucleus at the cytoplasmic face of the nuclear outer membrane. By in situ proximity ligation assay (PLA) we confirmed the colocalization of NAGK with snRNPN in the nucleus and in dendrites, while we also verified the interactions of NAGK with p54NRB, and with GTF2H5 in the nucleus. These associations between NAGK with speckle, paraspeckle and general transcription factor suggest its regulatory roles in gene expression.

Construction of a Network Model to Reveal Genes Related to Salt Tolerance in Chinese Cabbage (배추 염 저항성 관련 유전자의 네트워크 모델 구축)

  • Abiotic stress conditions such as cold, drought, and salinity trigger physiological and morphological changes and yield loss in plants. Hence, plants adapt to adverse environments by developing tolerance through complex regulation of genes related to various metabolic processes. This study was conducted to construct a coexpression network for multidirectional analysis of salt-stress response genes in Brassica rapa (Chinese cabbage). To construct the coexpression network, we collected KBGP-24K microarray data from the B. rapa EST and microarray database (BrEMD) and performed time-based expression analyses of B. rapa plants. The constructed coexpression network model showed 1,853 nodes, 5,740 edges, and 142 connected components (correlation coefficient > 0.85). On the basis of the significantly expressed genes in the network, we concluded that the development of salt tolerance is closely related to the activation of $Na^+$ transport by reactive oxygen species signaling and the accumulation of proline in Chinese cabbage.

Inhibitory Effect of Capsaicin against Carcinogen-induced Oxidative Damage in Rats

  • Capsaicin (trans-8-methyl-N-vanillyl-6-nonenarnide), a major pungent component of hot pepper, is known to exert antioxidative properties. In this study, we investigated the protective effects of capsaicin against chemical carcinogen-induced oxidative damage in rats. Male Sprague Dawley rats weighting 230~250 g were treated with chemical carcinogens such as 2-nitropropane (2NP) or n-methyl-N'-nitro-N-nitrosoguanidine (MNNG) after (or before) the administration of capsaicin at doses of 0.5, 1,5 mg/kg. The level of lipid peroxidation in rat liver was estimated by measuring the amounts of thiobarbituric acid reactive substances. The degree of oxidative DNA damage was evacuated by measuring a DNA adduct, 8-hydroxydeoxyguanosine (8-OHdG), in urine. Antioxidative activities of capsaicin and its metabolites in vitro were determined by the measurement of DPPH (1,1-diphenyl-2-picrylhydrazyl), a radical quencher. Significant inhibition of 2-NP induced lipid peroxidation was observed in the liver of the rat when treated with capsaicin. MNNG-induced urinary excretion of 8-OHdG was decreased by capsaicin treatment. Capsaicin and its metabolites inhibited net only the formation of free radicals, but also lipid peroxidation in vitro. Our results show that capsaicin may function as a free radical scavenger against chemical carcinogen-induced oxidative cellular damage in vivo. The observed antioxidative activities of capsaicin may play an important role in the process of chemoprevention.

An efficient shear deformation theory with stretching effect for bending stress analysis of laminated composite plates

  • The focus of this paper is to develop an analytical approach based on an efficient shear deformation theory with stretching effect for bending stress analysis of cross-ply laminated composite plates subjected to transverse parabolic load and line load by using a new kinematic model, in which the axial displacements involve an undetermined integral component in order to reduce the number of unknowns and a sinusoidal function in terms of the thickness coordinate to include the effect of transverse shear deformation. The present theory contains only five unknowns and satisfies the zero shear stress conditions on the top and bottom surfaces of the plate without using any shear correction factors. The governing differential equations and its boundary conditions are derived by employing the static version of principle of virtual work. Closed-form solutions for simply supported cross-ply laminated plates are obtained applying Navier's solution technique, and the numerical case studies are compared with the theoretical results to verify the utility of the proposed model. Lastly, it can be seen that the present outlined theory is more accurate and useful than some higher-order shear deformation theories developed previously to study the static flexure of laminated composite plates.

Relationship between Nutrients Intakes, Dietary Quality, and hs-CRP in Korea Metabolic Syndrome Patients - The 2015 Korea National Health and Nutrition Examination Survey - (한국 성인 남녀 대사증후군 집단의 영양소 섭취와 식사의 질 및 hs-CRP와 관련성 - 국민건강영양조사(2015년) 자료를 활용하여 -)

  • Metabolic syndrome is a risk factor for cardiovascular and type 2 diabetes. This study was conducted to examine the relevance between nutrition intake, meal quality, and high-sensitivity C-reactive protein in Koreans with metabolic syndrome. The 2,536 subjects, aged 19~64, who participated in 2015 National Nutrition Survey were included in this study. The 24-hour recall method was employed to analyze nutrition intake and dietary quality. Subjects were grouped into either the non-metabolic syndrome group (n=1,938) or the metabolic syndrome group (n=598). Total males and females were divided into 3 groups according to the high-sensitivity C-reactive protein (hs-CRP) level to study its relationship to metabolic syndrome and its components, including odds ratio (OR) and confidence interval (CI). Results showed the homeostasis model assessment of insulin resistance (HOMA-IR) value was higher in the metabolic syndrome group (3.37) than non-metabolic syndrome group (1.57) (p<0.001). In the Index of Nutrition Quality, males in the non-metabolic syndrome group showed higher niacin (p<0.05) than males in metabolic syndrome group. Females in the non-metabolic syndrome group had higher vitamin $B_1$ (p<0.01), vitamin $B_2$ (p<0.001), niacin (p<0.05), calcium (p<0.001), and phosphate (p<0.01). Female in the high hs-CRP group showed high OR in blood glucose component (OR 2.488, 95% CI: 1.269~4.879) and metabolic syndrome risk (OR 2.856, 95% CI: 1.292~6.314). Females in the middle hs-CRP group had high triglycerides component (OR 2.956, 95% CI: 1.920~4.551), compared to the low hs-CRP group. The study showed females with higher hs-CRP had a higher risk of metabolic syndrome.
