Dissection of rat hippocampus using high-definition stereoscopic microscope system is demonstrated. Many surgical operation or animal anatomies are anatomized with stereoscopic microscope. With stereoscopic microscope system, the environments of operation is important factor to dissector. Lights, resolution of acquired stereo images, reflected on the liquid, colors and eye fatigue are fatal factors to dissector. We reduced reflections with two incident angle of lights at 45 degree and used a complementary color at the basement and reform the stereoscopic microsystem. Dissector has felt more comfortable after compensation and operation time is more continuous.
Capsaicin cream has been used to attenuate the pain associated with diabetic neuropathy, rheum-atoid arthritis, osteoarthritis and postherpetic neuralgia. But its common side effect, local irritation, limits the use of it and there is still a need for a new analgesic devoid of this side effect. This study was conducted to compare the local irritant effect of DA-5018, a new capsaicin derivative, with that of capsaicin in various animal models and human beings. Capsaicin, applied topically to the mouse ear, produced dose-dependent increase of ear volume and the frequency of ear scratching behavior in mice. Neither ear volume nor scratching behavior was affected by DA-5018. In eye wiping test of rat, DA-5018 was 10 times less irritant than capsaicin. Capsaicin administered intradermally into the rat paw elicited paw lick/lift response with a potency which was three times that of DA-5018. Zostrix-HP (0.075% capsaicin cream), but not DA-50180.3% cream, increased ear volume of rat and induced thermal hyperalgesia in normal and carrageenan inflamed paws. Six day-treatment of Zostrix-HP failed to develop tolerance against this thermal hyperalgesia. In human beings, Zostrix-HP produced burning sensation and itching in more than 90% of volunteers involved and its maximum irritant effect was significantly higher than that of DA-5018 cream. These results suggest that local irritation and burning sensation produced by DA-5018 is much less than capsaicin.
Hydrocepbalus may be an acquired or a congenital condition. We have studied the macroscopic and microscopic changes in the hydrocephalus of an inbred Sprague-Dawley rat at postnatal week 8. The animal suspected with the hydrocephalus showed clinical syndromes such as depression, severe ataxia, eye abnormalities, dome-shaped head, and persistent fontanelle. With the postmortem examination, the suspected animal was clearly revealed as a severe internal hydrocephalus. In this animal, severe ventriculomegaly was limited to the third and lateral ventricles, and cortical thining was most apparent in the parieto-occipital region. With the routine histological examination, brain tissue showed aqueductal obstruction, thinning of the cerebral cortex, severe ependymal damage, subependymal edema, damage of choroid plexus of fourth ventricle, enlarged cortical vessels, and expanded ventricles. Aqueductal obstruction was observed with the appearance of simple stenosis at the level of rostral colliculus. Subsequently, the other structures of brain such as septal nucleus, caudate nucleus, and hippocampus etc. were abnormally reconstructed by hydrocephalus. This study suggests that the hydrocephalus can be taken place by primary aqueductal obstruction and this type of hydrocephalus is classified as uncommunicating type. Though the mechanism of aqueductal obstruction is not clear, the morphological studies of this case may be helpful for the further study of hydrocephalus.
Xiaoshuan Zaizao Wan (XZW) has been used in China to improve hemiplegia, deviation of eye and mouth, and dysphasia due to cerebral thrombosis. To characterize pharmacological actions of XZW, we evaluated its effects on neuronal cell damage induced in primary cultured rat cortical cells by various oxidative insults, glutamate or N-methyl-D-aspartate (NMDA), and $\beta$-amyloid fragment ($A_{\beta(25-35)}$). XZW was found to inhibit the oxidative neuronal damage induced by $H_2O_2$, xanthine/xanthine oxidase, or $Fe^{2+}$/ascorbic acid. It also attenuated the excitotoxic damage induced by glutamate or NMDA. The NMDA-induced neurotoxicity was more effectively inhibited than the glutamate-induced toxicity. In addition, we found that XZW protected neurons against the $A_{\beta(25-35)}$-induced toxicity. Moreover; XZW exhibited dramatic inhibition of lipid peroxidation in rat brain homogenates and mild 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Taken together; these results demonstrate that XZW exerts neuroprotective effects against oxidative, excitotoxic, or $A_{\beta(25-35)}$-induced neuronal damage. These findings may provide pharmacological basis for its clinical usage treating the sequelae caused by cerebral thrombosis. Furthermore, XZW may exert beneficial effects on Alzheimer's disease and other oxidative stress-related neurodegenerative disorders.

Purpose: Gestational nutrition has an impact on the growth and development of the fetus. Choline (C) and docosahexaenoic acid (DHA) are important and essential nutrients for humans that play a role in the structural integrity of the membranes as well as signalling. C is used in the synthesis of phosphatidylcholine, and cell membranes are highly enriched with DHA. The dietary intake of C or DHA during pregnancy directly influences fetal development. Currently, there is no evidence to prove the effectiveness of the combined dietary supplementation of both C and DHA during gestation on developmental outcomes in the offspring. Methods: The current study was designed to assess the physical, sensory, and motor development of rat pups born to mothers supplemented with C and/or DHA during the entire gestational period. Pregnant rat dams were divided into the following five groups: Normal control (NC), Saline control (SC), Choline (C), DHA, and Choline+DHA (C+DHA). The NC dams did not receive any supplementation during the entire gestation period. The experimental groups were supplemented with Saline, C, and/or DHA, respectively, during the entire gestation (E0 to delivery). Results: Rat pups (n = 6/group) exposed to combined C and DHA showed significant improvement in birth weight, fur development, eye-opening as well as weight gain on the 7th, 14th, and 21st postnatal day and pinnae detachment (assessed from birth to postnatal day 21) when compared with age-matched NC, SC or C or DHA pups. Further, significant reflex responses were observed in visual placing and bar holding of pups exposed to both C and DHA, whereas the differences in surface righting, negative geotaxis, and grasping reflexes were not significant between the groups. Conclusion: Gestational supplementation of both C and DHA rather than either of them alone is better in enhancing developmental outcomes in rat pups.
The present investigation was performed to evaluate the homeostatic regulation of sleep architecture by the ethanol extract of Korea red ginseng (KRG), since the available data were often controversial. In addition, it was also interested in whether the sleep-wake stages were differently affected by low and high doses of KRG. Each adult Wistar male rat was implanted with a transmitter for recording EEG and activity via telemetry. After one week of surgery, polygraphic signs of undisturbed sleep-wake activities were recorded for 12 h (between 9:00 am and 9:00 pm) after KRG administration. KRG (10 and 100 mg/kg) increased non-rapid eye movement (NREM) sleep as well as total sleep. The total percentages of wakefulness were decreased comparably. KRG (10 mg/kg) decreased the power density of the ${\delta}-wave$ (0.75-4.5 Hz) and increased ${\alpha}-wave$ (8.0-13.0 Hz) in the NREM and rapid eye movement (REM) sleep. KRG also decreased ${\delta}-wave$ power density in wake time. However, KRG (100 mg/kg) increased ${\delta}-wave$ and decreased ${\theta}-wave$ (5.0-9.0 Hz) power density in wake time, while showed little effect on the power density in NREM and REM sleep. In conclusion, low and high doses of KRG increase spontaneous sleep and NREM sleep and differently regulate the EEG spectra in REM and NREM sleep.
본 연구에서는 안구 영상을 위한 OCT용 손잡이 형 프로브를 개발하였다. 프로브는 휴대하기 간편한 손잡이 형태로 구현하였으며, 어댑터의 대물렌즈를 교체함으로써 각막(cornea)과 망막(retina) 모두 영상화 할 수 있도록 설계 하였다. 시스템의 성능검증을 위해 in vivo로 실험용 쥐의 눈 영상을 2D로 획득하였으며, 획득한 영상을 3D 영상으로 재구성하였다. 3D 영상은 쥐의 미세한 부분의 구조를 잘 나타내었으며, 움직임에 의한 흔들림 현상(Motion artifact)이 나타나지 않았으므로 개발된 OCT시스템의 이미지 디스플레이 속도는 생체 내 실험 (in vivo)에 적합함을 알 수 있었다.
The drug therapy for rheumatoid arthritis must be maintained constantly or for the whole life but is apt to induce the adverse effects in gastrointestinal or renal system. Therefor, newer methods are paid attention to reduce adverse effects. The thirty-two rat female separated into seven groups depending on the therapy or Freund's adjuvant applied: Normal group(n=8) not received anything, Positive control group(n=8) only received adjuvant, USC group(n=8) received continuous US, USP group(n=8) received pulsed US. The physical and radiological findings by thermal and non-thermal effects of ultrasound were evaluated in groups with continuous mode at 1 MHz, $0.5\;W/cm^2$ for 6 minutes and with pulsed mode (duty cycle 1:9) at 1 MHz, $0.5\;W/cm^2$ for 6 minutes. The result summarized followings. 1. Swelling of forepaw and hindpaw was significantly reduced in USP. 2. Arthritis indices in USP group were significantly reduced than those in PCG. 3. In naked eye and radiologic findings, swelling was significantly prevented in USP group but not in PCG. The change of swelling, arthritis index, gross feature in naked eye, radiologic finding were significantly improved in all groups except for USC group, taking care for using its continuous mode.
본 연구에서는 임실특산물인 쥐눈이콩및 쥐눈이식초콩의 혈당강하 및 인슐린 감수성에 대한 효과를 검정콩, 메주콩과 비교하여 조사하였다. 식이물질의 성분검사 결과 쥐눈이콩과 쥐눈이식초콩이 검정콩과 메주콩에 비하여 당뇨개선의 효과가 있는 여러 아미노산 및 식이섬유 함량이 유의적으로 높게 측정 되었다. 실험 을 위 해 평균체중 195.5$\pm$0.98 g의 숫컷쥐를 정상군(NC), 당뇨대조군(DI, DC), 검정콩식이군(DB), 메주콩식이군(DY), 쥐눈이콩식이군(DS), 쥐눈이식초콩식이군(DSS)으로 총 7개군으로 나누어 해당식이첨가 시료로 4주간 사육하였다. 당뇨유발을 위 해 STZ를 1회 (70 mg/kg/rat) 투여하였으며 그 결과 체중변화는 당뇨대조군(DC)에서 21.12 g감소함에 반하여 각각의 시료를 첨가식이한 당뇨실험군에서 특히 쥐눈이콩(DS)과 쥐눈이식초콩(DSS) 식이군은 높은 식이 이용 효율(FER)을 보여 각각 14.73$\pm$3.65 g 및 16.71$\pm$5.54 g의 몸무게 증가를 보였다. 음용수 및 뇨량의 측정 결과 역 시 당뇨실험군에서 유의적인 감소(p<0.05)를 보였다. 혈당량은 당뇨대조군(DC)에 비하여 검정콩(DH),메주콩(DY), 쥐눈이콩(DS), 쥐눈이식초콩(DSS) 식이군이 각각 17.9%, 16.9%, 10.35%, 19.54%씩 유의하게 감소하였으나 정상대조군(NC)보다는 높은 수치를 나타내었다. 또한 당뇨쥐의 인슐린 감수성에 대한 관찰결과 당뇨대조군 중 인슐린 무처지군(DC)에 비하여 모든 당뇨실험군에서 높은 값을 보였다. 결론적으로 임실산 쥐눈이콩과 쥐눈이식초콩은 당뇨쥐의 혈당강하 및 인슐린감수성에 대한 개선작용의 가능성이 있으므로 당뇨합병증 예방의 기능성 식품으로 기대된다.

[Purpose] Here, we aimed to determine the effect of Polygonum cuspidatum stem extract (PSE) on exorbital lacrimal gland-excised rat models and hyperosmotic stress-stimulated human conjunctival cells (HCCs). [Methods] Seven week old male Wistar rats were divided into six groups. Only the rats in the control group (NOR, n=5) did not undergo surgery. Three days after the surgery, the exorbital lacrimal gland-excised rats were randomly allocated to five groups: (1) vehicle-treated dry-eyed rats (DED, n=5); (2) PSE (10 mg/kg) treated DED rats (PSE-10, n=5); (3) PSE (100 mg/kg) treated DED rats (PSE-100, n=5); and (4) PSE (250 mg/kg) treated DED rats (PSE-250, n=5). In addition, the HCC line was co-treated with hyperosmolar media (528 mOsm) and PSE (1-100 μg/ml). [Results] PSE treatment restored the tear volume and goblet cell density by inhibiting severe corneal irregularities and damage. The treatment with PSE significantly attenuated the hyperosmolar stress-induced inflammation and cell death through the suppression of mRNA expression levels of Tumor necrosis factor-α (TNF-α), Interleukin-6 (IL-6), Interleukin-1β (IL-1β), and Interferon-γ (IFN-γ), and the expression of Bcl-2-associated X protein (Bax) as well as the activation of caspase-3 in vitro. [Conclusion] The inhibitory effects of PSE treatment on dry eye disease indicate the potential of nutritional intervention by PES against inflammatory diseases without adverse effects.
