비유에 대한 래복자 약침(藥鍼)이 고지방 식이로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향 (Effects of Herbal Acupuncture(Raphanus Sativus) at $BL_{20}$ on the Hyperlipemia Rats Induced by High Fat Diet)

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Raphanus sativus) at Bisu($BL_{20}$) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into high fat diet group(Control), high fat diet and saline in $BL_{20}$ treated group(Saline), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-1, 0.058mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-2, 0.029mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-3, 0.011mg/ml/g), Herbal acupuncture was bilaterally treated at 0.2cc per 4days, total 14 times in 56days. Results : Body weight and food efficiency were decreased in saline, RHA-1, RHA-2. Feed intake was increased in saline, RHA-1, RHA- 2, RHA- 3. The level of serum total cholesterol was decreased in saline, RHA-1. The level of serum HDL-cholesterol, LDL-cholesterol were not significant. The level of serum triglyceride, Total Bilirubin were decreased in saline, RHA-1, RHA-2, RHA-3.The level of serum AST was decreased in RHA-1. But the level of serum ALT was increased in RHA-1. The level of atherogenic index was decreased in saline, RHA-1. The level of HTR was increased in saline, RHA-1. Conclusions : Raphanus sativus-herbal acupuncture in Bisu($BL_{20}$) can control body weight, feed intake and feed efficency ratio, total cholesterol, total bilirubin, and prevention cardiovascular risk.

Comparison of Glucosinolate Contents in Leaves and Roots of Radish (Raphanus spp.)

  • Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.

알로자임에 의한 무 씨의 순수성 검증 (Determination of Seed Purity in Radish (Raphanus sativus L.) Using Allozyme)

  • 무(Raphanus sativus L.)는 세계적으로 중요한 작물 중의 하나이다. 십자화과 식물 종자 생산에서 원하지 않은 내교잡에 의한 종자 결실로 오염이 발생하므로 씨의 순수성 검증은 매우 중요하다. 재배종 진주 대평 무(R. sativus cv. Daepeng)와 백자 무(R. sativus cv. Backza)의 교잡 분석을 실시하였다. 알로자임으로 상업적으로 이용되는 잡종 제1세대($F_1$) 무에 있어서 씨의 순수성을 평가하였다. 웅성과 자성 양친 360개체에 27개 대립유전자좌위를 조사하였다. 특히 Par-1 ($aa{\times}bb$), Lap-1($aa{\times}bb$), Est-1 ($aa{\times}bb$)에서 명확한 잡종 밴드를 나타내었다. Est-1 대립유전자좌위에서 자성 배우체로부터 기원된 것이 15개체(8.3%)가 발견되었고, 웅성 배우체로부터 기원된 것이 26개체(14.4%)가 발견되었다. 또한 다양도 측면에서 양친 계통에 비해 잡종 계통에서 높은 유전적 다양도를 유지하고 있었다. 샤논의 정보지수(Shannon's index)를 이용한 표현형 다양도는 교잡 계통이 가장 높았다. 알로자임에 의한 무 계통의 교잡에 의한 종자 생성에서 씨에 대한 순수성 검증이 효과적으로 탐지되어 육종 연구에 기여할 수 있을 것으로 판단된다.

Antimicrobial Action of Raphanus raphanistrum subsp. sativus (radish) Extracts against Foodborne Bacteria Present in Various Milk Products: A Preliminary Study

  • Seeds and leaves of Raphanus raphanistrum subsp. sativus (radish) are known to contain "raphanin," which has the potential to inhibit pathogenesis associated with foodborne pathogenic bacteria and fungi. In this study, ethanol extracts from R. raphanistrum subsp. sativus (radish) powder was evaluated for antimicrobial action against 6 different foodborne pathogenic bacteria. The current study demonstrated the potential of R. raphanistrum subsp. sativus (radish) in inhibiting the growth of Salmonella enteritidis 110, Cronobacter sakazakii KCTC 2949, Bacillus cereus ATCC 10876, and Staphylococcus aureus ATCC 6538. However, these antimicrobial action were not observed against Listeria monocytogenes ATCC 51776 and Escherichia coli 23716. Hence, this study indicates that R. raphanistrum subsp. sativus (radish) could be used as a natural biopreservative with antimicrobial effects for improving food safety, and as a functional food in the commercial food industry.

신선초와 무시레기의 항산화 성분 및 활성 비교 (Antioxidant Activities of Angelica keiskei L. and dried leaves of Raphanus sativus L.)

  • 무시레기와 신선초의 항산화 성분 및 활성을 분석하여 비교하였다. 무시레기와 신선초 추출물의 polyphenol 함량은 각각 100 g당 $2.84{\pm}0.11g$, $2.88{\pm}0.02g$이었으며, flavonoid 함량은 $0.19{\pm}0.00g$, $0.38{\pm}0.01g$ 으로 확인되었다. 항산화 활성 측정 결과, 무시레기 및 신선초 추출물이 DPPH radical 소거활성에서 $IC_{50}$값이 $348.72{\pm}31.65{\mu}g/mL$, $183.37{\pm}48.17{\mu}g/mL$으로 나타났으며, $ABTS^+$ radical 소거활성은 17.8 %, 11.9 %의 활성을, SOD 유사활성에서는 63%, 58%의 활성을 가진 것으로 나타났다. 총 polyphenol 및 flavonoid 함량과 항산화 활성간의 상관관계에서 총 flavonoid 함량과 DPPH radical 소거능이 r=0.9804, $ABTS^+$ radical 소거활성은 r=-0.9863의 상관관계를 보였으며, DPPH radical 소거활성은 $ABTS^+$ radical 소거활성과 r=-0.9609, SOD 유사활성과는 r=0.4865로서 각각 유의한 부와 정의 상관관계를 나타내었다.

Comparison of Anti-Inflammation Effects of Specimens Before and After the Oil Extraction of Raphanus sativus L. Seed in RAW 264.7 Macrophage Activated by LPS

  • Raphanus sativus L. has been reported to have anti-inflammatory and anti-tumor activity. However, the anti-inflammatory effect and mechanism of action of the Raphanus sativus L. seeds (RSS) with or without oil are still unknown. This study was undertaken to investigate the in-vitro anti-inflammatory effect with or without oil in the RSS on RAW 264.7 cells stimulated by lipopolysaccharide (LPS). Results showed the suppressed LPS-induced secretion of pro-inflammatory mediators such as nitric oxide (NO), inflammatory cytokine (IL-6, TNF-α). Additionally, a decrease in protein expression of iNOS was observed, but nuclear translocation of NF-κB p65 was not inhibited. To elucidate the underlying mechanism of the anti-inflammatory effect of RSS, the involvement of mitogen-activated protein kinase (MAPK) signaling pathways was examined. We also found that RSS blocked LPS-induced phosphorylation of c-Jun N-terminal kinase/stress-activated protein kinase (JNK) signaling but did not affect the phosphorylation of p38 MAPK and extracellular signal-regulated kinase (ERK) 1/2. These results suggest that RSS may have potential as an anti-inflammatory agent through the inhibition of LPS-induced inflammatory cytokine production via regulation of the JNK pathway.

New Antimicrobial Activity from Korean Radish Seeds (Raphanus sativus L.)

  • To isolate antifungal substances from Korean radish (Raphanus Sativus L.) seeds, various purification techniques such as DE52 cellulose anion exchange, SP-Sephadex C-25 cation exchange, and Sephadex G-50 gel filtration chromatographies were used. The molecular masses of two purified R. sativus antifungal proteins (RAPs) were estimated to be about 6.1 kDa (RAP-1) and 6.2 kDa (RAP-2) by SDS-PAGE, and 5.8 kDa(RAP-1) and 6.2 kDa (RAP-2 by a gel filtration chromatography, respectively. Purified proteins RAP-1 and 2 clearly exhibited different growth inhibitory activities against other microorganisms like Candida albicans and Saccharomyces cerevisiae. Although they have similar molecular masses, both RAP-1 and 2 proteins are not identical because their microbial inhibitory actions were different. Therefore, RAP-1 could be a new antifungal protein when compared with the antifungal activities of 2S albumins, Rs-AFPs, Mj-AMPs, chitinase, glucanase, permatin, and ribosome inactivating proteins, all of which are anifungal proteins of plants.

Antisteroidogenic activity of Raphanus sativus seed extract in female albino mice

  • The defatted methanol extract of Raphanus sativus Linn. (Cruciferae) seed (MERS) was evaluated for its antisteroidogenic potential in mature female Swiss albino mice. The methanol extract at the doses of 100 and 200 mg/kg body weight significantly elevated the levels of cholesterol and ascorbic acid contents which serve as a precursor for the synthesis of steroid hormones in ovaries. The extract also significantly inhibited glucose-6-phosphate dehydrogenase and ${\Delta}^5-3{\beta}$-hydroxy steroid dehydrogenase, the two key enzymes involved in ovarian steroidogenesis. Hence the extract (MERS) exhibited significant antisteroidogenic activity.
