Synthesis of a New 4-(Pyridin-3-yl)pyrimidine Derivatives for Anticancer Activity

  • This study is focused on the synthesis of urea and amide derivatives particularly, since the amide moiety is an essential binding group at the binding site. Urea derivatives 3-7 and 13-14 were obtained by reaction of 2-aminopyrimidines and other amines with diverse isocyanates in pyridine as a solvent under reflux. The urea derivatives were obtained in low yield because of the highly electron deficient nature of the amino group of the 2-aminopyrimidine. Amide derivatives 8-10 were obtained in moderate yields by reaction of compound 1 with aryl chloride derivatives. Also, arylamine 11 was synthesized by Buchwald-Hartwig amination in moderate yields. Most of the compound did not show good activity against A375P melanoma cells, compared with Sorafenib as control compound.

  • Since it is troublesome to estimate adequate flow rates in four sections of SMB chromatography, a systematic determination of the flow rates has been suggested by using ketoprofen as a model chiral enantiomer. S-ketoprofen. less retained species, was separated from raffinate stream and the variation in its purity was dependent on the changes of the flow rate of section 4 ($Q_4$), the raffinate flow rate ($Q_{raf}$), and the feed flow rate ($Q_{feed}$) under a fixed switch­ing time t$^{\ast}$. When one parameter was changed at the given experimental condition, purities of product were changed and these phenomena has be well explained by the triangle theory.

  • The RAS family genes encode small GTP-binding cytoplasmic proteins. Activated KRAS engages multiple effector pathways, notably the RAF-mitogen-activated protein kinase, phosphoinositide-3-kinase (PI3K) and RalGDS pathways. In the clinical field, K-ras oncogene activation is frequently found in human cancers and thus may serve as a potential diagnostic marker for cancer cells in circulation. This mini-review aims to summarise information on Ras-induced oncogenesis and the clinical significance of K-ras.

S-ketoprofen enantiomer의 정제를 위한 SMB(Simulated Moving Bed) 크로마토그래피 장치의 모델링

  • We have performed batch chromatography experiments to calculate parameters which are used to design SMB chromatography system. Isotherms of S- and R-ketoprofen enantiomers were gained from small amount loading experiments in a column, and flow rate of four zones of SMB chromatography were calculated using mass balance equations. As a results, diameter and length of columns were 10mm and 600mm, and flow rate of each zone were as follows; $Q_{Feed}$=1.00ml/min ,$Q_{Elu}$=2.81ml/min ,$Q_{Raf}$=1.81ml/min ,$Q_{Ext}$=2.00ml/min , and $Q_{Rec}^{TMB}$=10.75ml/min , From the Darcy's equation, the pressure drop in whole columns of SMB chromatography was 128bar.

Induction of Melibiase in Yeast

  • Exposing yeast cells with a certain genotype to different inducers, the ability of the yeast cells (Saccharomyces cerevisiae) to obtain enhanced fermentation for carbohydrates was observed. Regardless of the preexposure to any substrate, the inherent character incapable of fermenting a certain carbohydrate was maintained, while utilization of carbohydrates by the cells with a certain gene markers was varied by the previous conditions where they were exposed. Galactose was the best inducer for the cells to elaborate melibiase, even the galactose was not utilized as a substrate. Preexposure to galactose seemed to be necessary for the cells to utilize galactose and melibiose. Galactose fermentation by GA cells was enhanced by the exposure of the cells to galactose, but not to melibiose, raffinose, sucrose or glucose. Delayed fermentation of sucrose by the cells exposed to glucose or melibiose, but not to galactose, was observed. Raffinose fermentation was obtained by the cells with either SU RAF or GA ME genes, but the enhanced fermentation of raffinose seemed to be dependent on which inducer the cells were exposed previously and enzymes induced by the inducer to break either one of the linkages of raffinose molecule, the alpha0galactosidic or the beta-fructo-furanosidic.

  • Computational modeling of signal transduction is currently attracting much attention as it can promote the understanding of complex signal transduction mechanisms. Although several mathematical models have been used to examine signaling pathways, little attention has been given to crosstalk mechanisms. In this study, an attempt was made to develop a computational model for the pathways involving growth-factor-mediated mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3'-kinase/protein kinase B (PI3K/Akt). In addition, the dynamics of the protein activities were analyzed based on a set of kinetic data. The simulation approach integrates the information on several levels and predicts systems behavior. The in-silico analysis conducted revealed that the Raf and Akt pathways act independently.

  • Polyphenoloxidase activity (PPO) in the leaves of Erechitites hieracifolia was estimated by Warburg's manometric method. The emzyme was most reactive toward chlorogenic acid followed by caffeic acid. Diethyldithiocarbamate and potassium cyanide were shown powerful inhibition rate to the polyphenoloxidase from the leaves of Erechitites hieracifolia. We confirmed antioxidant activity of the leaves of Erechitites hieracifolia by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method. Electrophorectic isoenzyme banding patterns of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) were observed by native PAGE. The correlation of PPO and antioxidant enzymes is not investigated yet. That is need to further study.

Design and Synthesis of 3-(3-Chloro-4-substituted phenyl)-4-(pyridin-4-yl)-1Hpyrazole- 1-carboxamide Derivatives and Their Antiproliferative Activity Against Melanoma Cell Line

  • Design and synthesis of new 3,4-diarylpyrazole-1-carboxamide derivatives are described. Their antiproliferative activity against A375 human melanoma cell line was tested and the effect of substituents on the diarylpyrazole scaffold was investigated. The pharmacological results indicated that most of the synthesized compounds showed moderate activity against A375, compared with Sorafenib. On the other hand, compounds Ia, Ie, IIb, and IIh were more potent than Sorafenib. In addition, compound IIa was equipotent to Sorafenib. Among all of these derivatives, compound IIb which has diethylamino and phenolic moieties showed the most potent antiproliferative activity against A375 human melanoma cell line. Virtual screening was carried out through docking of the most potent compound IIb into the domain of V600E-b-Raf and the binding mode was studied.

  • 해양활동의 급속한 확대로 해마다 발생하는 해양사고의 발생이 증가하고 있으며, 사상자 수도 비례적으로 증가하고 있다. 이러한 사고를 줄이기 위해서, 다양한 구조 요청 기기가 개발되고 제시되되어 왔으나 단일화 되지 못한 통신 방법, 구조 요청 과정에 대한 사용자의 인식 부족으로 실질적인 구조 요청 장치의 사용율이 지극히 낮다. 또한 해양의 특수성으로 인하여 제시되어온 기기를 이용하기 위해서는 큰 비용이 소요되었다. 이에 따라 실질적인 구조 가능성을 높이고 해양사고로 인한 사상자를 최소화하기 위해 본 연구의 목적은 실험평가와 경험적 공식을 바탕으로 일반 사용자가 손쉽게 접근 가능한 안드로이드 애플리케이션과 연결된 자동 구조 요청 장치를 개발한 것으로, 본 연구는 현장 및 실험실 시험의 결과에 근거하여, MOB 구출을 위한 자동 구조 요청 장치의 사양을 확정 검증하는 실용화 연구를 수행하였다.

Local and Global Attention Fusion Network For Facial Emotion Recognition (얼굴 감정 인식을 위한 로컬 및 글로벌 어텐션 퓨전 네트워크)

  • Deep learning methods and attention mechanisms have been incorporated to improve facial emotion recognition, which has recently attracted much attention. The fusion approaches have improved accuracy by combining various types of information. This research proposes a fusion network with self-attention and local attention mechanisms. It uses a multi-layer perceptron network. The network extracts distinguishing characteristics from facial images using pre-trained models on RAF-DB dataset. We outperform the other fusion methods on RAD-DB dataset with impressive results.
