Applications of Prescriptions Phlegm-Fluid Substances Chapter Depending on Symptoms in Dongeuibogam ("동의보감(東醫寶鑑)" "담음문(痰飮門)"의 병증(病症)에 따른 처방(處方) 고찰(考察))

  • Objectives : This study was performed to investigate 93 prescriptions which are all affiliated Phlegm-Fluid Substances chapter depending on Symptoms in Dongeuibogam. Methods : The following conclusions are reached through investigations on the applications of prescriptions which are all affiliated Phlegm-Fluid Substances chapter Depending on Symptoms in Dongeuibogam. Results : 1. Fluid detention disease article represents that Byukeum(癖飮), Hyuneum(懸飮) and Yueum(流飮) have the same treat criteria, in effect, there is not any problem left in case it is reached fluid detention disease is not 8 but 6. 2. Strong water-utilization herbs can be mainly used in treating Fluid detention disease, eliminating phlegm warm herbs are almost used two times more than eliminating phlegm cold herbs in eliminating phlegm disease. 3. Arisamatis Rhizoma is only used by itself in Chongmongseok-hwan, the others are used at the same time with Pinelliae Rhizoma. As a result of this, Arisamatis Rhizoma has an effects on eliminating Wind relative dusease but eliminating phlegm disease which can cause upper body phlegm disease. 4. Fritillariae Cirrhosae Bulbus and Tricosanthis Radix are able to get rid of obstinate phlegm-Fluid disease in eliminating phlegm cold herbs. 5. Gamisachil-tang and Gamieejin-tang which are located in throat chapter are more proper prescriptions to treat phlegm at throat than Gwache-san or Jeoljehwadam-hwan which is located in Phlegm-Fluid Substances chapter in Dongeuibogam. 6. Glycyrrhizae Radix is used 46 times(49%) at total 93 prescriptions in eliminating phlegm disease herbs. It seems to be needed more study whether Glycyrrhizae Radix can control the Phlegm-Fluid disease or not. Conclusions : The 93 prescriptions for eliminating Phlegm-Fluid Substances in Dongeuibogam are mainly composed of Sobanha-tang, Eejin-tang and Baeksang-hwan, etc.

Effects of Yuldahansotang(熱多寒少湯) water extract on Blood Pressure, Regional Cerebral Blood Flow and Pial Arterial Diameter (열다한소탕(熱多寒少湯)이 혈압(血壓), 국소뇌혈류량(局所腦血流量) 및 뇌연막동맥(腦軟膜動脈)에 미치는 영향(影響))

  • Yuldahansotang(YH) has been used in Sasang(四象) constitution medicine for many years as a therapeutic agent for cerebral disease. The effect of YH on the vascular system is not known. The purpose of this study was to determine the effect of YH on blood pressure, regional cerebral blood flow(rCBF) and pial arterial diameter of rats. 1. Blood pressure decreased by YH in rats. 2. rCBF was increased by YH in a dose-dependent manner. 3. Pretreatment with propranolol, methylene blue and indomethacin significantly inhibited YH induced increase in rCBF. 5. Blood pressure increased by Radix Puerariae(RP) and Radix Ligustici Tenuissimae(RLT) but Radix Scutellariae(RC) decreased blood pressure in rats. 6. rCBF was increased by RP and RLT in a dose-dependent manner but RC decreased low dosage, and RC increased high dosage. 7. Pial arterial diameter was increased by YH in a dose-dependent manner. 8. Pretreatment with propranolol significantly inhibited the increased in pial arterial diameter induced by YH. These results suggest that YH causes a diverse response of blood pressure, regional cerebral blood flow(rCBF) and pial arterial diameter. The increase in rCBF is also mediated by prostaglandins, cyclic GMP and adrenergic ${\beta}$ receptor and the increase in pial arteral diameter is mediated by adrenergic ${\beta}$ receptor.

  • From the study of acupuncture and external diseases on mammary diseases, the following conclusions are obtained. 1. Point Shanzhong, Shaoze and Rugen are often used for galactostasis in acupuncture. 2. Often used vessels for galactostasis in acupuncture are Stomach Meridian, Conception Vessel, Small Intestine Meridian, Bladder Meridian, Liver Meridian and Large Intestine Meridian in order. 3. The direct stimulations are used in external treatment for galactostasis, often accompanied by internal medical treatment, to promote the circulation of Qi and Blood. 4. Point Zuimup, Guangming of Gall bladder Meridian and Zusanli of Stomach Meridian is used for lactation pain in acupuncture. 5. Point Jianjing, Zusanli and Shanzhong are most often used for acuremastitis and breast cancer in acupuncture. 6. Often used vessels for acuremastitis and breast cancer in acupuncture are Stomach Meridian, Gall bladder Meridian, Conception Vessel, Small Intestine Meridian, Bladder Meridian, Pericardium Meridian in order. 7. The external treatment used for acuremastitis and breast cancer can be classed into its pathological period. When lactation is interrupted, Kumhwang Powder, Okro Powder, Robongbang, Allii Radix, Pinelliae Rhizoma, garlic moxibustion and Arusaenatus Rgizoma powder are used. When pus is piled up, Chunghwa Extracts, Dioscoreae Rhizoma, Leonuri Herba, Typa Latifolia, Taraxacum platycarpum, Phaseolus angularis, Manchong, Crvi Cornus latex, and Aucklandiae Radix are used. When pus flows down, Salviae Miltiorrhizae Radix Extract, nine-one pellet, Yongboo extract, eight-dampness extract, Tissue-regeneration Powder, Toad-net treatment, Kitten-hair, Hongseung pellet are used. 8. Stomach Meridian, Conception Vessel, Small Intestine Meridian, Gall Bladder Meridian, Bladder Meridian are most often used vessels for mammary diseases. From the conclusions above, It appears that the medical effect could be maximized by further studying and developing of the acupuncture and external treatment for mammary diseases according to the its nature, while accompanying internal medicine appropriately at the same time.

Effects of Herba, Radix, Fructus-xanthii Extract on the Immunities against infections Diseases and Tumors (창이초(蒼耳草)의 약용부위별(藥用部位別) 추출물이 항감염(抗感炎) 및 항종양(抗腫瘍) 면역반응(免疫反應)에 미치는 영향)

  • Herba Xanthii(HX), Radix Xantluii(RX) and Fructus Xanthii(FX) is one of the oriental medicine that has been used for the treatment of such infectious diseases and tumors. However, the mechanism of the drug is not investigated much. This study was done to know the effects of HX, RX and FX extract on the such innate immunities as phagocytic function and reactive radical formtions from phagocytes and the such acquired immunities as humoral and cell-mediated immunities. The followings are the results obtained from this study: 1. HX2 and FX1 groups increases the in vivo phagocytic activity of mononuclear phagocytes. 2. HXB, RXB, RXC, FXB and FXC groups increase the in vitro phagocytic activities. 3. RXB group stimulated the macrophages to produce nitric oxide in the presence of $interferon-{\gamma}$ $(IFN-{\gamma})$. 4. HX and RX whole groups increased the luminol-amplified reactive oxygen intermediate production in vivo. 5. HX whole and RX1, FX2 groups increased the lucigenin-amplified reactive oxygen intennediate production in vivo. 6. HXC group only increased the luminol-amplified reactive oxygen intermediate production in vitro. 7. HXB, FXB and FXC groups increased the lucigenin-amplified reactive oxygen intermediate production in vitro. 8. HX2, RX1 and FX whole groups increased the hemolysin formations from B cells. 9. HX, RX and FX whole groups significantly increased the rosette forming cells from the spleen. 10. HX, RX and FX whole groups significantly decreased the delayed-type hypersensitivity measured by footpad swelling. The above results demonstrate that HX, RX and FX has enhancing effects on innate immunity selectively and decreasing effects on delayed-type hypersensitivity of cell-mediated immunity according to medicinal part and diluted condition. This immunomodulating effects of HX, RX and FX might be responsible for the treatment of immune-mediated disorders.

  • Purpose : This study was designed to make a basis of in vivo or clinical study about colporrhea. Methods : This study is based on Yang's study, An investigation on the prescriptions of colporrhea(1999). We investigate the prescriptions in the Yang's study and examine the frequency of metioned single herb. Also we investigate the herb's classification. Effect of Glycyrrhizae Radix on the Immune Responses(I) - Immuno-regulatory Action of 50% Methanol Extract - (감초가 면역반응에 미치는 영향 (I) - 50% 메탄올 엑스의 면역조절작용 -)

  • These experiments were conducted to investigate the effects of Glycyrrhizae Radix extract(GR) on histamine synthesis, lymphocyte blastogenesis in C57BL/6J mice splenocytes, IL-1 production, $Ca^{2+}$ uptake by macrophage-like P388D$_{1}$ cells and plaque forming cell assay against SRBC. Histamine contents, lymphocyte blastogenesis, IL-1 activity, $Ca^{2+}$ uptake and plaque forming cell were determined by enzyme isotope method, [$^{3}$H]-thymidine incorporation, C3H/HeJ mouse thymocytes proliferation, the addition of 5 $\mu$Ci/ml $^{45}Ca^{2+}$ to P388D$_{1}$ cell suspension and assay to sheep red blood cell, respectively. Cytotoxicity, which was expressed as 50% mortality, was occurred by the addition of GR(10$^{-3}$g/ml). Histamine production in mouse spleen cell culture was significantly increased by 48 hour incubation added 0.25$\mu\textrm{g}$/ml of Con A. Con A-dependent T-lymphocyte proliferation was also enhanced by the addition of 0.25 $\mu\textrm{g}$/ml of Con A. GR depressed histamine contents at 10$^{-9}$~10$^{-4}$g/ml. and Con A (0.25 $\mu\textrm{g}$/ml) dependent T-lymphocyte proliferation at 10$^{-5}$~10$^{-4}$g/ml. IL-1 activity was significantly decreased by 10$^{-8}$~10$^{-4}$g/ml of GR. $Ca^{2+}$ uptake was not changed by GR, but antibody production markedly increased at 10.0~50.0 mg/kg of GR. From the above results, it is suggested that GR have immuno-regulatory action; GR decreased cell-mediated immune response and increased antibody production by B lymphocyte at high doses.

  • The matrix metalloproteinases (MMP) play an important role in tumor invasion, angiogenesis and inflammatory tissue destruction. Increased expression of MMP was observed in benign tissue hyperplasia and in atherosclerotic lesions. Invasive cancer cells utilize MMP to degrade the extracellular matrix and vascular basement membrane during metastasis, where MMP-2 has been implicated in the development and dissemination of malignancies. The present study attempted to examine the antiangiogenic activity of the medicinal herbs of Aspergillus usamii var. shirousamii-transformed Angelicae Gigantis Radix and Zizyphus jujube (tAgR and tZj) with respect to MMP-2 production and endothelial motility in phorbol 12-myristate 13-acetate (PMA)- or VEGF-exposed human umbilical vein endothelial cells (HUVEC). Nontoxic tAgR and tZj substantially suppressed PMA-induced MMP-2 secretion. In addition, $25{\mu}g/mL$ tAgR and tZj prevented vascular endothelial growth factor-stimulated endothelial cell transmigration and tube formation. The results reveal that tAgR and tZj dampened endothelial MMP-2 production leading to endothelial transmigration and tube formation. tAgR and tZj-mediated inhibition of endothelial MMP may boost a therapeutic efficacy during vascular angiogenesis.

Development of Ultrafine Angelica Powder-Added Syrup (초미세 당귀분말 첨가 시럽의 제조 기술 개발)

  • Angelicae gigantis Radix (dried root of Angelica gigas) including major bioactives such as decursin and decursinol angelate provides rich flavors and several healthy benefits. Recent studies have shown that ultrafine powders of herbal medicines provide better physical properties and biological activities. Thus, ultrafine Angelica powder was added into the oligosaccharide syrup to provide flavors and healthy benefits in this study. Angelicae gigantis Radix was pulverized into d(0.1) = 3.220, d(0.5) = 7.822, and d(0.9) = 7.817 $\mu$m respectively using an air-flow mill. The ultrafine Angelica powder was added into the oligosaccharide syrup process with different ratios of water to oligosaccharide syrup at 1:5, 1:8, 1:11, and 1:14. The physicochemical properties such as viscosity and bulk density were measured. The Stokes' law was applied to predict the sedimentation velocity of the added Angelica powder in the syrup. The Angelica syrup prepared in this experiment showed good stability since the Angelica particles precipitated down slowly. The ratio of water to oligosaccharide syrup at 1:11 showed the optimal preparation in terms of the stability and the viscosity. The ultrafine-sized herbal powders such as Angelicae gigantis Radix have potentials for various food and pharmaceutical applications.

Toxicological Evaluation of Saposhnikoviae Radix Water Extract and its Antihyperuricemic Potential

  • Although the dried root of Saposhnikovia divaricata (Turcz.) Schischk. (Umbelliferae) is a popular medicinal plant in East Asia, there has been no systemic toxicological evaluation of a water extract of Saposhnikoviae Radix (SRE). In this experiment, an oral acute and 13-week subchronic toxicological evaluations of SRE (500-5,000 mg/kg body weight) were performed in both sexes of Crl:CD(SD) rats. Based on the results from mortality, clinical signs, effects on body weight and organ weight, clinical biochemistry, hematology, urinalysis, and histopathology, significant acute, 4-week repeated dose range finding (DRF) and 13-week subchronic toxicity of SRE was not observed in either sex of rats; thus, the no observed adverse effect level (NOAEL) was 5,000 mg (kg/day). To identify anti-hyperuricemia potential of SRE, the suppressive effect of SRE was determined in mice challenged with potassium oxonate (PO; 250 mg/kg) via intraperitoneal injection for 8 days (each group; n = 7). SRE supplementation suppressed the uric acid level in urine through significant xanthine oxidase (XO) inhibitory activity. Kidney dysfunctions were observed in PO-challenged mice as evidenced by an increase in serum creatinine level. Whereas, SRE supplementation suppressed it in a dose-dependent manner. Collectively, SRE was safe up to 5,000 mg (kg/day) based on NOAEL found from acute and 13-week subchronic toxicological evaluations. SRE had anti-hyperuricemia effect and lowered the excessive level of uric acid, a potential factor for gout and kidney failure.

Systemic Analysis of Antibacterial and Pharmacological Functions of Scutellariae Radix (시스템 약리학적 분석에 의한 황금의 항균효과)

  • This study was performed to find antibacterial substances contained in Scutellariae Radix (SR) using a systems pharmacological analysis method and to establish an effective strategy for the prevention and treatment of infectious diseases. Analysis of the main active ingredients of SR was performed using Traditional Chinese Medicine Systems Pharmacology (TCMSP) Database and Analysis Platform. 36 active compounds were screened by the parameter values of Drug-Likeness (DL), Oral Bioavailability (OB), and Caco-2 permeability (Caco-2), which were based on the drug absorption, distribution, metabolism, and excretion indicators. The UniProt database was used to obtain information on 159 genes associated with active compounds. The main active compounds with antibacterial effects were wogonin, β-sitosterol, baicalein, acacetin and oroxylin-A. Target proteins associated with the antibacterial action were chemokine ligand 2, interleukin-6, tumor necrosis factor, caspase-8,9 and mitogen-activated protein kinase 14. In the future, systems pharmacological analysis of traditional medicine will be able to make it easy to find the important mechanism of action of active substances present in natural medicines and to optimize the efficacy of medicinal effects for combinations of major ingredients to help treat certain diseases.
