Protective Effect of Radix Trichosanthis Extracts on Cytotoxicity of Pancreatic ${\beta}-Cells$ by Cytokines (싸이토카인 유발 췌장 ${\beta}$세포 독성에 대한 천화분 추출물의 방어효과)

  • In this study, the preventive effects of Radix Trichosanthis extracts (RTE) against cytokine-induced ${\beta}-cell$ death were assessed. Cytokines generated by immune cells infiltrating pancreatic islets are crucial mediators of ${\beta}-cell$ destruction in insulin-dependent diabetes mellitus. The treatment of RIN cells with $interleukin-1{\beta}$ ($IL-1{\beta}$) and $interferon-{\gamma}$ ($IFN-{\gamma}$) resulted in a reduction of cell viability. RTE protected $IL-1{\beta}$ and $IFN-{\gamma}$-mediated viability reduction in a concentration-dependent manner. Incubation with RTE also induced a significant suppression of $IL-1{\beta}$ and $IFN-{\gamma}$-induced inducible nitric oxide synthase (iNOS) protein expression. The molecular mechanism by which RTE inhibited iNOS protein expression appeared to involve the inhibition of $NF{-\kappa}B$ activation. The $IL-1{\beta}$ and $IFN-{\gamma}$-stimulated RIN cells showed increases in $NF{-\kappa}B$ binding activityand $I{\kappa}B{\alpha}$ degradation in cytosol compared to unstimulated cells. However, pretreatment with RTE inhibited cytokines-induced $I{\kappa}B{\alpha}$ degradation and $NF{-\kappa}B$ activation in RINm5F cells. Furthermore, the protective effects of RTE were verified via protection of impairment in glucose-stimulated insulin secretions in $IL-1{\beta}$ and $IFN-{\gamma}$-treated islets.

Effects of Seman Armenicae and Radix Trichosanthis on the iNOS expression and superoxide formation in the RAW264.7 cells (행인 과루인 추출물이 마우스 대식세포주인 RAW264.7 세포주의 iNOS 발현 및 Superoxide 형성에 미치는 영향)

  • Macrophage play a major role in host defence against infection and tumor development and this activity is regulated through the production of several mediators. In particular, the production of NO by macrophages mediates killing or growth inhibition of tumor cells, bacteria, fungi and parasites. However, over-expression of iNOS by various stimuli, resulting in over-production of NO, contributes to the pathogenesis of septic shock and some inflammator and auto-immune disease. Therefore, it would be valuable to develop potent and selective inhibitors of for potential therapeutic use. Thus the agent that supprees the expression of iNOS mRNA or enzyme protein will be usefull for the prevention of various diseases. We are intersted in identifying selective inhibitiors of iNOS which might be useful intreating inflammatory human diseases. In summary, we have demenstrated that scopoletin, isolated from Seman Armenicae and Radix Trichosanthis the production of NO induced by $IFN-\gammer$ plus LPS in RAW 264.7 macrophages, The mechanism for the inhibition of NO production was due to suppression of the expression of iNOS mRNA or enzyme protein.

  • Objectives : This study was carried out to investigate the traditional diet therapies for the period of postpartum recovery and lactation and to develop modern postpartum recovery foods. Korean traditional medical treatment for postpartum care focused on treating postpartum obesity, edema, agalactia was studied. Methods : We investigated the documents recorded in the medical classics of "Hyangyakjipseongbang(鄕藥集 成方)", "Donguibogam(東醫寶鑑)" and "Bangyakhappyeon(方藥合編)" published in 1433, 1613 and 1885 respectively in the Joseon Dynasty. Results : First, Radix Angelicae Sinensis was most frequently used to treat the postpartum obesity, and a lot of nourishing herbs including Radix Paeoniae Alba, Radix Astragali, Radix Ginseng, Rhizoma Chuanxiong were also used. Second, Radix Glycyrrhizae, Rhizoma Atractylodis Macrocephalase, Pericarpium Citri Reticulatae, Rhizoma Zingiberis Recens were used to treat the postpartum edema. Third, Radix Rhapontici, Medulla Tetrapanacis, Fructus Trichosanthis etc. were used to treat the postpartum agalactia. Conclusions : Consequently, we hope this study on the prescriptions and herbs for gynecology in "Hyangyakjipseongbang(鄕藥集成方)", "Donguibogam(東醫寶鑑)" and "Bangyakhappyeon(方藥合編)" will provide knowledge for diet therapy model development of postpartum care of the modern times.

Study on the Antiinflammatory Effect of Gleditschiae Spina -Focussed on Takrisodokum and Kwaluubangtang- (조각자의 소염작용(消炎作用)에 대(對)한 실험적(實驗的) 연구(硏究) 부제(附題) : 탁이소독음(托裏消毒飮)과 과루우방탕(瓜蔞牛蒡湯)의 가감(加減)에 따른 효능비교(效能比較))

  • The inhibitory effects of Gleditschiae Spina, Cleditschia saponin, two complex prescriptions such as Takrisodokum and Kwaluubangtang, which contain Gleditschiae Spina, and their altered prescriptions on the activity of cyclooxygenase in vitro were investigated. The water extract of Gleditschiae Spina and Gleditschia saponin markedly inhibited the cyclo-oxygenase activity by 59% and 67%, respectively, at a concentration of $1mg/m{\ell}$, on the other hand, Takrisodokum and Kwaluubangtang showed 15% and 7% inhibition, respectively. The extracts with the substraction of Gleditschiae Spina and/or Platycodi Radix inhibited to a much smaller extent than Takrisodokum. However, in case of the extracts with the substraction of Gleditschiae Spina and/or Trichosanthis Semen, only one prescription, which does not contain Trichosanthis Semen, exhibited the same inhibition as Takrisodokum. From above results, Gleditschiae Spina was found to be most important for the effect of Takrisodokum and Kwaluubangtang on their antiinflammatory action. Instead of Gleditschiae Spina, another parts of this plant were added to both complex prescriptions and tested according to above methods, indicating that Gleditschiae Spina is more important than another parts for both prescriptions. When the administered dose of Gleditschiae Spina is increased(twofold and threefold) in both complex prescriptions, the inhibitory effect in Takrisodokum was diminished. however, the effect in Kwaluubangtang was increased.

Antibiotic Effect of Heat-Clearing Medicinals on Vaginal Microorganisms (청열약(淸熱藥)의 질내(膣內) 미생물(微生物)에 대(對) 항균효과(抗菌效果))

  • Purpose : This study was conducted to investigate the in vitro inhibitory effects of heat-clearing medicinal on common bacterias in gynecology. Methods : The heat-clearing medicinals ( Trichosanthis Radix, Sophorae Fructus, Phragmitis Rhizoma, Buddleiae Flos, Bambusae Folium, Anemarrhenae Rhizoma, Celosiae Semen, Gardeniae Fructus, Prunellae Spica, Sophorae Radix, Dictamni Radicis Cortex, Coptidis Rhizoma, Gentianae Scabrae Radix, Scutellariae Radix, Phellodendri Cortex) were used in this study. Staphylococcus aureus, MRSA, Candida albicans and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Streptococcus spp., Escherichia coli HB101, Lactobacillus gasseri were used for normal vaginal florae. We evaluated antibiotic effect by the optical density and the colony test. Results : The optical density and colony test showed that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex of herbs among heat-clearing medicinal had antimircobial effect. Celosiae Semen and Prunellae Spica had antimicrobial susceptibility and selective toxicity in MRSA. Scutellariae Radix and Phellodendri Cortex had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis. Conclusion : According to the above results, we could suggest that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex among heat-clearing medicinal be available to antimicrobial agent of vaginal pathogenic microbial species in vitro.

Antihistaminic Action of Medicinal Plants

  • The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiae fructus dose-dependently inhibited [$^3$H]mepyramine binding to H$_1$ receptor and histamine-induced contraction in guinea-pig brain homogenates and isolated guinea-pig ilea, respectively. Antihistaminic action of the hexane and ethylacetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus was more potent than their antimuscarinic action evaluated from the inhibition of [$^3$H]QNB binding and carbachol response. The ethylacetate and chloroform fractions and six known flavonoids from Scutellariae radix also inhibited histamine-induced contraction, but antihistaminic potencies of these fractions and compounds were almost identical with their antimuscarinic potencies. The hexane fractions of Mori cortex radicis and Evodiae fructus, as shown in ketotifen, inhibited selectively the increase of cutaneous vascular permeability induced by histamine. However, wogonin (SC-1) from Scutellariae radix was a nonselective inhibitor for the effect of histamine and serotonin on the vascular permeability. These results demonstrate that the hexane and ethylacetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H$_1$ receptor blocking activities.

  • The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiae fructus dosedependently inhibited [$^3$H] mepyramine binding to H$_1$, receptor in guinea-pig brain homogenates and histamine-induced contraction of isolated guinea-pig ileum. These results demonstrate that the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H$_1$receptor blocking activity.

Examine inhibitory effect against lipase activity in vitro. It was compared Xenical(Orlistat) with 56 kind of herb medicines. Cytotoxicity and Physiological Activity of SunbangHwalmyung-um (선방활명음의 항암 및 항산화효과 검증)

  • Objectives : The purpose of this research was physiological activities and investigate cytotoxicity of Sunbanghwalmyung-um extract. Methods : Physiological activity and a cytotoxicity were examined through the hot water and ethanol extracts from Sunbanghwalmyung-um and its ingredient. Results : The electron donating ability(EDA) was 66.9%, 71.3% in 1000 ppm water extract and ethanol extract. Above 90% from 1000 ppm ethanol extract showed a higher activities and it is Rhei Radix et Rhizoma, Lonicerae Flos, Gleditsiae Spina, Glycyrrhizae Radix, Myrrh. SOD-like activity was weak as 12.24%, 16.62% in 700 ppm. In water and ethanol extracts cytotoxicity were against G361, B16F10, MDA, A549, high cytotoxity over 70%. Rhei Radix et Rhizoma, Gleditsiae Spina, Trichosanthis Radix, Paeoniae Radix Rubra showed high cytotoxicity in water and ethanol extracts. Conclusions : We observed physiological activities and investigated cytotoxicity of Sunbanghwalmyung-um and its ingredients. The results also demonstrated in food or cosmetic industry.

Studies on the Antihypercholesterolemic Effects of Gamigwaruhaebaekwhanggum - Tang (가미과루해백황금탕의 항고지혈증 효과)

  • Gamigwaruhaebaekwhanggum-Tang (GGHWT) have been evaluated for antihyperlipidemic effects on experimental hyperlipidemic rats and mice induced by Triton WR-1339, com oil and high cholersterol-diet. Especially, GGHWT is formulated with Trichosanthis Fructus, Pinelliae Tuber, Aurantii Immaturus Fructus, Magnoliae Cortex, Allii Macrostemi Bulbuls, Cinnamomi Ramulus and Scutellariae Radix. Oral administration of GGHWT at 500 mg/kg/day for 3 days significantly inhibited the increase of serum triglyceride and LDL-cholesterol, liver triglyceride in hyperlipidemic rats induced by Triton WR-1339. And, GGHWT significantly inhibited the increase of serum triglyceride in hyperlipidemic rats induced by corn oil. Also, administration of GGHWT (500 mg/kg, once daily for 1 week, p.o.) prevented the increase of serum total cholesterol, triglyceride, LDL-cholesterol, liver total cholesterol and triglyceride in 1% cholesterol-diet fed mice. These results suggest that GGHWT is effective for the treatment of hyperlipidemia.
