함초 추출액을 첨가한 브라운 소스의 품질 특성 (Quality Characteristics of Salicornia herbacea L. Extract Added Brown Sauce)

  • This study performed physicochemical characteristics and sensory tests on brown sauce with different Salicornia herbacea L. (saltwort) content to find the best brown sauce recipe with saltwort. Moisture content increased with greater saltwort extract content. There was significant (p<0.001) pH difference with saltwort content. There was also significant (p<0.001) sweetness difference in the samples and sweetness increased with greater saltwort content. Viscosity measured at $20^{\circ}C$ and $60^{\circ}C$ was significantly (p<0.001) lower with greater saltwort content. The brightness of brown sauce containing saltwort extracts significantly (p<0.001) increased with greater saltwort content and its redness and yellowness usually decreased. SPS4 with the greatest saltwort content showed the highest value of 4.25% and DPPH radical removal increased significantly (p<0.001) with greater saltwort content. In the sensory test, the color value was lowest at 4.82 with BSL1 and the flavor value was lowest at 5.00 with BSL0, the control group. The taste and the flavor values were highest with BSL2 with 2% saltwort content. Overall acceptance was highest with BSL3, the brown sauce with 3% saltwort content, at 6.09. As a result, it was concluded that the brown sauce with 3% saltwort content was most suitable. Therefore, this study concluded that saltwort with various functions could be used for food and may be used to replace salt and have other functions in brown sauce, a popular sauce consumed in Korea.

Effects of Thermal Treatment on Antioxidant Activity in Yam (Dioscorea batatas DECNE.)

  • The aim of this study was to investigate the antioxidant activity of thermal treatment yam (Dioscorea batatas DECNE.) in Korea. Thermal treatment yam was extracted by different solvents including 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v). Then color property, total phenol content and antioxidant activity were analyzed. Yam possessed high $L^*$ value and $H\limits^{\circ}$ value, which were $54.92{\pm}2.18$ and $73.20{\pm}0.77$, respectively. Thermal treatment yam exhibited great antioxidant activity evaluated by ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power. Total phenol contents of various extracts from thermal treatment yam increased in the following order: 70% methanol extract ($63.53{\pm}0.33mg\;CAE/g$), 70% ethanol extract ($69.47{\pm}1.00mg\;CAE/g$) and CM extract ($97.49{\pm}0.66mg\;CAE/g$), respectively. The same trend was also could be found in antioxidant activity assays except for reducing power assay. These results implied that these extracts from thermal treatment yam might be useful to take a good part in prevention of human diseases and aging.

쥐오줌풀의 항산화 및 산화적 DNA 손상 억제 활성 (Antioxidative Activities and Inhibition Effects on Oxidative DNA Damage of Valeriana fauriei)

  • Background: Valeriana fauriei (Valerianaceae) has been used to as a traditional medicine to treat a variety of symptoms, including headache, insomnia, hypertension, and menstrual irregularity. However, the present study investigates the species' antioxidant activity and its inhibition of oxidative DNA damage, which have yet to be studied. Methods and Results: The antioxidant activity was assessed using radical scavenging assays with 1,1-diphenyl-2-picryl hydrazyl (DPPH) and, 2, 2'-azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) and a reducing power assay. The total phenol content was also analyzed, and phenolic compounds were detected using HPLC/UV, whereas the inhibitory effect of Valeriana fauriei on oxidative DNA damage was measured using ${\phi}-174$ RF I plasmid DNA cleavage assay. The DPPH and ABTS radical scavenging activity were $75.17{\pm}3.55%$ and $95.83{\pm}0.63%$, repectively, and the reducing power was $93.14{\pm}1.74$ at $200{\mu}g/m{\ell}$. The total phenol content was $10.24{\pm}0.04mg/g$, whereas chlorogenic acid, catechin, caffeic acid and epicatechin were identified using HPLC/UV, and the ${\phi}-174$ RF I plasmid DNA cleavage assay indicated that V. fauriei provided protection against oxidative damage. Conclusions: The results of the present study suggest that V. fauriei has powerful antioxidant activity that can provide protective effects against the oxidative DNA damage caused by free radicals. The species, therefore, provides a valuable resource for the development of natural pharmaceutical to treat aging, cancer, and degenerative diseases.

쪽의 항산화 및 항염증 활성에 대한 연구 (Study on Antioxidant and Anti-inflammatory Activities of Persicaria tinctoria)

  • Objectives : Persicaria tinctoria belongs to the Polygonaceae family and it has been used as the natural dye traditionally. Also, it is well known that the Persicaria tinctoria is used for treating the following symptoms such as fever, inflammation and edema. The purpose of this study is to investigate the effective source of antioxidants and anti-inflammatory agent from various parts of Persicaria tinctoria.Methods : We investigated the antioxidative and anti-inflammatory properties of the Persicaria tinctoria extracts. Antioxidant activities were measured by 1,1-diphenyl-2- picrylhydrazyl (DPPH), 2, 2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, Fe2+ chelating activity and Reducing power of Persicaria tinctoria extracts. And its inhibitory effect against oxidative DNA damage was evaluated in non-cellular system using φX-174 RF I plasmin DNA. The anti-inflammatory effect of Persicaria tinctoria was measured by using the inhibitory efficacy for the amount of nitric-oxide (NO) produced in LPS induced RAW264.7 cells.Results : The extracts from stem part showed better DPPH scavenging activity compared to those of the leaf and root extracts. Their IC50s were measured as 7.17, 144.40 and 165.07 ug/ml, respectively. These results were similar to that of ABTS radical scavenging assay and reducing power. Also, Persicaria tinctoria showed the protective effects of DNA damage against oxidative stress and anti-inflammatory effect by suppression of NO production in LPS induced RAW264.7 cells.Conclusions : These results showed that various parts of Persicaria tinctoria can be used as an effective source of antioxidants and anti-inflammatory agents via antioxidative activities and anti-inflammatory effect.

Anti-Diabetic and Anti-Inflammatory Effects of Green and Red Kohlrabi Cultivars (Brassica oleracea var. gongylodes)

  • The aim of the present study was to evaluate the anti-diabetic, anti-inflammatory, antioxidant potential, and total phenolic content (TPC) of green and red kohlrabi cultivars. Anti-diabetic and anti-inflammatory activities were evaluated via protein tyrosine phosphatase (PTP1B) and rat lens aldose reductase inhibitory assays and cell-based lipopolysaccharide (LPS)-induced nitric oxide (NO) inhibitory assays in RAW 264.7 murine macrophages. In addition, scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical, and peroxynitrite ($ONOO^-$) were used to evaluate antioxidant potential and TPC was selected to assess phytochemical characteristics. Between the two kohlrabi cultivars, red kohlrabi (RK) had two times more TPC than green kohlrabi (GK) and showed significant antioxidant effects in DPPH, ABTS, and $ONOO^-$ scavenging assays. Likewise, methanol (MeOH) extracts of RK and GK inhibited LPS-induced NO production in a dose dependent manner that was further clarified by suppression of iNOS and COX-2 protein production. The MeOH extracts of RK and GK exhibited potent inhibitory activities against PTP1B with the corresponding $IC_{50}$ values of $207{\pm}3.48$ and $287{\pm}3.22{\mu}g/mL$, respectively. Interestingly, the RK MeOH extract exhibited significantly stronger anti-inflammatory, anti-diabetic, and antioxidant effects than that of GK MeOH extract. As a result, our study establishes that RK extract with a higher TPC might be useful as a potent anti-diabetic, antioxidant, and anti-inflammatory agent.

A Simple $H\ddot{u}ckel$ Approach to Intramolecular Photocyclization Reaction of N-(2-Chlorobenzyl)-Pyridinium, N-(Benzyl)-2-Chloropyridinium, and N-(2-Chlorobenzyl)-2-Chloropyridinium Salts

  • We have calculated the ${\pi}$-electron density, atom self-polarizability, and free valence on each atom of N-(2-chlorobenzyl)-pyridinium, N-(benzyl)-2-chloropyridinium, and N-(2-chlorobenzyl)-2-chloropyridinium salts using a simple Huckel method in order to discuss their intramolecular photocyclization reaction in a qualitative method. Our calculation qualitatively predicts that photocyclization occurs through forming radicals as a reaction intermediate by breaking a C-Cl bond after photoexcitation into a triplet state via intersystem crossing from an initially excited singlet state. We noticed that this C-Cl bond breaking is aided by ${\pi}$-complex formation between a chlorine atom and the ${\pi}$ -electrons of the neighboring ring in the triplet state and a stronger ${\pi}$-complex bond makes C-Cl bond breaking, i.e., radical formation, much easier. A chlorine atom will form a stronger ${\pi}$ -complex bond to a benzyl ring of N-(benzyl)-2-chloropyridinium than a pyridinium ring of N-(2-chlorobenzyl)-pyridinium because the former can donate its ${\pi}$-electron more easily than the latter. The chlorine at position 15 of N-(2-chlorobenzyl)-2-chloropyridinium salt in the excited state also provides its ${\pi}$-electron to the benzyl ring. So this ${\pi}$-electron can increase the bond strength of the $\pi-complex.$ Therefore, the strength of ${\pi}$-complex follows the order of N-(2-chlorobenzyl)-2-chloropyridinium, N-(benzyl)-2-chloropyridinium, and N-(2-chlorobenzyl)-pyridinium salts and thus the radical formation rate. This provides us with an intramolecular photocyclization reaction rate of the same order as given above.

'후지' 사과즙을 첨가한 반죽 및 식빵의 품질 특성 (Quality and Antioxidant Properties of Bread Added with 'Fuji' Apple Juice)

  • In this study, bread was prepared using various amount of 'Fuji' apple juice, and quality and antioxidant properties investigated. Bread was prepared with water, butter, yeast, salt, sugar, wheat flour, skim milk powder and 10, 20 and 30% (w/w) 'Fuji' apple juice. Leavening ability and pH of dough were also investigated. Volume, pH, specific volume, baking-loss rate, chromaticity and texture were investigated as quality properties, whereas the total polyphenol content, ABTS and DPPH radical scavenging activity as the antioxidant properties of bread containing various amount of apple juice. The pH and leavening ability of dough were observed to decrease with increasing amount of apple juice. In the quality properties of bread, pH, volume, specific volume, and baking-loss rate decreased. Regarding chromaticity and texture of bread prepared with increasing amount of 'Fuji' apple juice, L (bright) values at crust and crumb of bread decreased, while hardness, chewiness and gumminess increased. Total polyphenol content increased with increasing amount of apple juice, while DPPH and ABTS radical scavenging activities showed no significant differences. Consequently, these results support the possible use of apple juice for baking foods in food industry, as addition of apple juice enhances the quality and antioxidant properties of bread.

제주지역 전통발효식품에서 분리한 유산균의 기능적 특성 (Functional Characteristics of Lactic Acid Bacteria Isolated From Jeju Island Traditional Fermented Food)

  • Jeju island is the largest island of South Korea, and has a relatively warm climate due to its geographical characteristics. These features have fostered development of various fermented foods distinct to Jeju island. Therefore, the functional activity of the lactic acid bacteria (LAB) isolated from the Jeju region's traditional fermented foods, including Kimchi and Jeotgal was characterized in this study. Fifteen strains were isolated from 6 types of fermented food. These strains include Enterococcus spp., Lactoplantibacillus spp., and Weisella spp. Several experiments were carried out to assess functional characteristics including acid resistance, bile resistance, hemolysis, DPPH and ABTS radical scavenging activity, and cholesterol-lowering activity. Three isolated strains (J-4, J-6 and J-10) exhibited high acid resistance, while the other 3 isolates (J-1, J-4 and J-6) showed high bile salt resistance. The strains displayed varied DPPH and ABTS radical scavenging activity: 71.14-86.42% among the first 3 strains, and 50.11-75.11% among the remaining 3 strains. Cholesterol-lowering activities were in the range 15.74-82.90% for all isolated strains. In conclusion, these studies suggest that isolated LAB strains from Jeju island traditional fermented foods possess the potential for broader application in the food industry.

괴화와 괴각 에틸아세테이트 분획물의 항산화 및 산화적 DNA 손상 억제 활성 (Antioxidative Activities and Inhibition of DNA Damage of Ethylacetate Fraction from Sophorae Flos and Sophorae fructus)

  • Objectives : In this study, we demonstrated the antioxidant activities and the inhibitory effect on oxidative DNA damage of ethyl acetate fractions extracted from Sophorae Flos and Sophorae fructus. Methods : Sophorae Flos and Sophorae fructus were extracted with methanol(MeOH) and divided to Petroleum ether, Ethyl acetate(EtOAC) and Water fraction. The antioxidant activities were conducted by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azine-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay, $Fe^{2+}$ chelating assay and Reducing power assay. The inhibitory effect of DNA damage were characterized on ${\varphi}$ X-174 RF I plasmid DNA cleavage assay. In addition, we analyzed the Total phenol contents and the Vitamin C contents of Sophorae Flos and Sophorae fructus. Results : The results of DPPH were 92.71% and 94.72%, ABTS were 87.16% and 62.44%, and $Fe^{2+}$ chelating were 95.81% and 85.11% at $200{\mu}g/m{\ell}$ of Sophorae Flos and Sophorae fructus respectively. The Sophorae Flos showed stronger effect than Sophorae fructus in Reducing Power assay. Total phenol content was 111.77 mg/g and 122.54 mg/g, and Vitamin C content was 2.59 mg/g and 3.03 mg/g. Also both Sophorae Flos and Sophorae fructus have inhibitory antioxidant effect on ${\varphi}$ X-174 RF I plasmid DNA cleavage assay. Conclusions : Over all, this study suggests that Sophorae Flos and Sophorae fructus can be used as not only effective antioxidant but also natural medicine.

세발나물 분말 첨가 국수의 품질 특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activities of Noodles added with Spergularia marina L. Griseb Powder)

  • This study evaluated the quality characteristics and antioxidant activities of Spergularia marina L. Griseb powder from halophyte plants added to noodles at ratios of 0%, 0.5%, 1.0%, 1.5%, and 2.0% in order to examine its applicability as a substitute for salt. For quality characteristics of noodles with Spergularia marina L. Griseb powder, pH increased (p<0.05) and salinity decreased (p<0.01) with higher added Spergularia marina L. Griseb powder content. Water absorption ratio decreased (p<0.01) while turbidity decreased (p<0.001) with greater addition of Spergularia marina L. Griseb powder. However, volume expansion ratio increased with greater addition of Spergularia marina L. Griseb powder, but there was no significant difference. For chromaticity, L value and a value decreased (p<0.001) while b value increased (p<0.001) with greater addition of Spergularia marina L. Griseb powder. For texture, hardness (p<0.001), gumminess (p<0.001), and cohesiveness (p<0.01) increased with greater amount of added Spergularia marina L. Griseb powder; however, there were no significant differences in adhesiveness, springiness, and chewiness according to the amount of added Spergularia marina L. Griseb powder. In the sensory evaluation, for color, the score was 5.55 points when 1.5% Spergularia marina L. Griseb powder was added, which was the best preference (p<0.001); for flavor, preference was best (5.64 points) when 2.0% Spergularia marina L. Griseb powder was added; for taste, preference was best (5.36 points) (p<0.001) when 2.0% Spergularia marina L. Griseb powder was added; for overall preference (overall quality) the score was 5.55 points when 1.0% Spergularia marina L. Griseb powder was added, which was the best preference (p<0.001). For antioxidant activities, antioxidant activities including total phenol content, total flavonoid content, and DPPH free radical scavenging activity increased (p<0.001) with the amount of added Spergularia marina L. Griseb powder. As a result of the above research, preference was the highest and quality was excellent overall when 1.0% Spergularia marina L. Griseb powder was added. Thus, addition of 1.0% Spergularia marina L. Griseb instead of salt would be desirable.
