Reducing effects of taurine on organ toxicity of paraquat

  • Paraquat(1.1'-dimethyl-4,4'-bipyridinium ion, PQ)는 전세계적으로 가장 많이 사용되고 있는 농약으로 자살 또는 실수로 마시는 경우 예외없이 폐독성으로 사망하게 된다. 하지만 아직까지 임상에 사용되어지고 있는 효과적인 독성 경감제는 전무한 실정이다. 현재까지 밝혀진 paraquat의 주된 독성기전은 NADPH Cytochrome P$_{450}$ reductase에 의해 산화,환원 반응을 거치는 동안 free radical을 생성하여 세포막에 지질과산화를 일으켜서 세포막의 기능상실과 cell death를 일으킨다고 보고되고 있다. 따라서 본 연구에서는 항산화작용이 뛰어난 아미노산인 taurine(TA)의 radical scarvenging 효과에 의한 PQ 독성경감효과를 in vivo에서 검색하였고, in vitro에서 TA의 PQ 독성경감 mechanism을 밝히고자 하였다. in vivo에서 PQ의 간독성(s-GOT,s-GPT), 신장독성(BUN, Creatinine), 폐 및 전신독성(ALP, MDA, G-6-phosphatase)의 정도를 혈액 및 조직균질액 중에서 검색함으로써 TA의 독성억제효과를 측정하였다.

Anti-Oxidation Property of Organo-Molybdenum Dialkyldithiophosphate (유용성 몰리부덴 화합물의 산화방지기능에 관한 연구)

  • ln this paper, the fuction of molybdenum dialkyl dithiophosphate (MoDTP) as an oxidation inhibitor of mineral oils was investigated and compared with 2,6-Di-tert-Butyl-4-Methylphenol (DBMP). Oxidation tests were conducted using an oxygen absorption apparatus. MoDTP showed anti-oxidation property, and length of induction time prolonged by increasing MoDTP concentration. However the induction time of DBMP was longer than those of MoDTP. The anti-oxidation property of MoDTP was found to be inferior to that of DBMP The capability of hydroperoxide decomposition ability with MoDTP was much greater than that with DBMP. However the rate constant of radical scavenging with MoDTP was much better than that with DBMP. It was found that the performance of MoDTP is exellent with respect to hydroperoxide decomposition but it is susceptible to chemical decomposition. From the fact that formation of phenol was observed when MoDTP was added to hexane solution of cumene hydroperoxide (CHPO), it is indicated that the decomposition of hydroperoxide with MoDTP occurs by means of ionic mechanism.

  • This tutorial review provides a general account of the electrochemical behavior of quinones and their various applications. Quinone electrochemistry has been investigated for a long time due to its complexity. A simple point of view is developed that considers the relative stability of the reduced quinone species and the values of the first and second reduction potentials. The 9-membered square scheme in buffered aqueous solutions is explained and semiquinone radical stability is discussed in this context. Quinone redox reaction has also been employed in various studies. Diverse examples are presented under three broad categories defined by the roles of quinone: molecular tool for physical chemistry, versatile electron mediator, and charge storage for energy conversion devices.

  • The Kinetics velocity for hydrolysis reaction of vanillylidene imine derivatives has been measured by ultra-violet ray spectrophotometer in 20wt% $dioxane-H_2O$ at $25^{\circ}C$. It was measured the reaction rate Constant of vanillylidene imine derivatives that can be applied widely following to pH-change at $25^{\circ}C$. Final products that hydrolyzed the vanillylidene imine certified in vanillin and aniline derivative, and the effect of substitution radical that has affected on hydrolysis reaction was largely promoted to reaction rate by electron attrating group in acidity and electron donoring group in basic. From the results of rate constant to hydrolysis reaction, substituent radical effect and final products. It has certified the hydrolysis reaction mechanism of vanillylidene imine derivatives.

Protective Effect of Copper against Pancreatic Insult in Streptozotocin-induced Diabetogenic Rat

  • The present study examined the alterations in endogenous oxy-radical scaverging system of pancreatic tissue associated with the dose of 45 mg/kg steptozotocin (STZ) alone or with various combinations. The activities of pancreatic Min-superoxide dismutase (SOD) and catalase were no apparent changes in the other groups except for the Cu(II) 4 mg/kg pretreated group. The presence of 4 mg/kg of Cu(II) with or without 125 mg /kg of diethylenetriaminepentaacetic acid (DTPA) markedly attenuated the fall in activity of Cu, Zn-Sod by STZ stress. In particular, STZ-induced superoxide generation was dramatically abolished by prior administration of Cu(II) 4 mg/kg. Conculsively, We suggested the possible involvement that copper may enhance the defence mechanism of pancreatic oxidative damage by STZ challenge.

Sonochemical Reaction Mechanism of a Polycyclic Aromatic Sulfur Hydrocarbon in Aqueous Phase

  • Hydroxybenzothiophenes, dihydroxy-benzothiophenes, and benzothiophenedione were identified as inter-mediates of benzothiophene (BT) exposed to ultrasonic irradiation. It is proposed that benzothiophene is oxidized by OH radical to sequentially for m hydroxybenzothiophenes, dihydroxybenzothiophenes, and benzothiophenedione. Benzothiophene is decomposed rapidly following pseudo-first-order kinetics in a first-order manner by ultrasonic irradiation in aqueous solution. The toxicity of sonochemically treated solutions was checked by E. coli and a less inhibition in bacterial respiration was observed from the 120-min treated benzothiophene sample than from the untreated benzothiophene sample. Also evolution of carbon dioxide and sulfite was observed during ultrasonic reaction. A pathway for ultrasonic decomposition of benzothiophene in aqueous solution is proposed.

  • Searching for new molecular radicals which are believed to play an important role as reaction intermediates in aromatic chain reactions, we have applied the technique of corona excited supersonic expansion employing a pinhole-type glass nozzle to obtain the vibronic spectrum from the corona discharge of precursor 3,5-difluorotoluene with a large amount of inert carrier gas helium. An analysis of the observed spectrum revealed that many vibronic bands are from other isomeric difluorobenzyl radicals generated in the jet by migration of the fluorine atom or methylene group to the adjacent position in the 3,5-difluorobenzyl radical. A possible mechanism was proposed for the formation of other isomers by using a bridged cyclic intermediate structure.

Aggregation of α-Synuclein Induced by Oxidized Catecholamines as a Potential Mechanism of Lewy Body

  • Lewy bodies (LBs) are neuronal inclusions that are closely related to Parkinson's disease (PD). The filamentous component of LB from patients with PD contains biochemically altered $\alpha$-synuclein. We have investigated the effect of the oxidized products of catecholamines on the modification of $\alpha$-synuclein. When $\alpha$-synuclein was incubated with the oxidized 3,4-dihydroxyphenylalanine (L-DOPA) or dopamine, the protein was induced to be aggregated. The oxidized catecholamine-mediated $\alpha$-synuclein aggregation was enhanced by copper ion. Radical scavengers, azide and N-acetyl cysteine significantly prevented the oxidized catecholamine-mediated $\alpha$-synuclein aggregation. The results suggest that free radical may play a role in $\alpha$-synuclein aggregation. Exposure of $\alpha$-synuclein to the oxidized products of catecholamines led to the formation of dityrosine. Antioxidant dipeptides carnosine, homocarnosine and anserine significantly protected $\alpha$-synuclein from the aggregation induced by the oxidized products of catecholamines.

Antioxidant and Anti-inflammatory Activities of Equisetum hyemale

  • Equisetum hyemale L. has been prescribed widely as a traditional medicine for the treatment of inflammatory diseases such as rheumatoid arthritis, conjunctivitis, pyelonephritis. In order to identification the mechanism, we examined an antioxidant and anti-inflammatory activity of 85% methanol extract of E. hyemale. In this study E. hyemale exhibited strong scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, superoxide radical, and nitric oxide. To elucidate the anti-inflammatory properties of E. hyemale, we investigated the inhibition effects of nitric oxide and IL-6 by E. hyemale in IFN-gamma and LPS-stimulated mouse peritoneal macrophages. E. hyemale suppressed nitric oxide, IL-6 production and iNOS expression dose-dependently without notable cytotoxic activity. These data suggest that E. hyemale might be useful in inflammatory diseases by inhibiting the free radicals and inflammatory mediators.

Protective Effects of Angelica tenuissima Nakai on Hepatotoxicity by Carbon Tetrachloride in Rats (사염화탄소에 의한 간손상에 미치는 고본의 보호작용)

  • Hepatoprotective activity of methanol extract of Angelica tenuissima Nakai on the $CCl_4$-induced hepatotoxicity was investigated. To elucidate the hepatoprotective activity and free radical scavenging effect, we examined alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin, total protein, cholesterol, malondialdehyde (MDA) levels in serum and activities of superoxide dismutase (SOD), catalase (CAT) in hepatic tissue as compared with those of carbon tetrachloride-induced rats. The action mechanism also has been estimated by quantative analysis of cytochrome P450 (CYP), NADPH-CYP reductase for phase I metabolism and glutathion (GSH), glutathion S-transferase (GST) level for phase II metabolsim. Treatment of Angelica tenuissima methanol extract significantly lowered the levels of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol, triglyceride, MDA, CAT were decreased, and SOD was activated. This result indicates that the hepatoprotective effect of Angelica tenuissima methanol extract on the CCl4-induced hepatotoxicity would be originated from reduction of the NADPH-CYP reductase, GSH and the enhancement of the activities of GST, CYP.
