Whitening effects of fermented Trigonotis radicans var. sericea with Lactobacillus brevis in α-MSH-stimulated B16F10 melanoma cells

  • This study was designed to compare the whitening effects of 60% ethanol extracts of Trigonotis radicans var. sericea (TR) and Lactobacillus brevis-fermented T. radicans var. sericea (FTR). Measurement of cytotoxicity in B16-F10 melanoma cells to confirm the whitening effect, FTR showed higher cell viability than TR. FTR showed inhibitory activity on melanin contents similar to the normal group at concentrations of 50 and 100 ㎍/mL. MITF expression was used to confirm the effect on melanogenesis-related protein expression. TR and FTR showed significant concentration-dependent decrease, and FTR showed lower expressions than the normal group at concentrations of 25, 50, and 100 ㎍/mL. Additionally, the mRNA expression of melanogenesis-related genes (MC1R, Rab27a, TGF-β1 and Myo5a) were measured by RT-qPCR to confirm the whitening effect. In MC1R expression at a concentration of 100 ㎍/mL in FTR showed effective inhibitory activities, and in TGF-β1 expression, TR and FTR both showed effective activities compared to normal groups even at low concentrations. Results of myo5a and Rab27a, a similar pattern was shown, and FTR showed effective inhibitory activities at a concentration of 100 ㎍/mL. As a result, FTR had higher whitening effects through bioconversion and is expected to be a good material for whitening functional cosmetics.

Deciphering the role of a membrane-targeting domain in assisting endosomal and autophagic membrane localization of a RavZ protein catalytic domain

  • The bacterial effector protein RavZ from a pathogen can impair autophagy in the host by delipidating the mammalian autophagy-related gene 8 (mATG8)-phosphatidylethanolamine (PE) on autophagic membranes. In RavZ, the membrane-targeting (MT) domain is an essential function. However, the molecular mechanism of this domain in regulating the intracellular localization of RavZ in cells is unclear. In this study, we found that the fusion of the green fluorescent protein (GFP) to the MT domain of RavZ (GFP-MT) resulted in localization primarily to the cytosol and nucleus, whereas the GFP-fused duplicated-MT domain (GFP-2xMT) localized to Rab5- or Rab7-positive endosomes. Similarly, GFP fusion to the catalytic domain (CA) of RavZ (GFP-CA) resulted in localization primarily to the cytosol and nucleus, even in autophagy-induced cells. However, by adding the MT domain to GFP-CA (GFP-CA-MT), the cooperation of MT and CA led to localization on the Rab5-positive endosomal membranes in a wortmannin-sensitive manner under nutrient-rich conditions, and to autophagic membranes in autophagy-induced cells. In autophagic membranes, GFP-CA-MT delipidated overexpressed or endogenous mATG8-PE. Furthermore, GFP-CA△α3-MT, an α3 helix deletion within the CA domain, failed to localize to the endosomal or autophagic membranes and could not delipidate overexpressed mATG8-PE. Thus, the CA or MT domain alone is insufficient for stable membrane localization in cells, but the cooperation of MT and CA leads to localization to the endosomal and autophagic membranes. In autophagic membranes, the CA domain can delipidate mATG8-PE without requiring substrate recognition mediated by LC3-interacting region (LIR) motifs.

Proteome analysis of developing mice diastema region

  • Different from humans, who have a continuous dentition of teeth, mice have only three molars and one incisor separated by a toothless region called the diastema in the hemi mandibular arch. Although tooth buds form in the embryonic diastema, they regress and do not develop into teeth. In this study, we evaluated the proteins that modulate the diastema formation through comparative analysis with molar-forming tissue by liquid chromatography-tandem mass spectroscopy (LC-MS/MS) proteome analysis. From the comparative and semi-quantitative proteome analysis, we identified 147 up- and 173 down-regulated proteins in the diastema compared to the molar forming proteins. Based on this proteome analysis, we selected and evaluated two candidate proteins, EMERIN and RAB7A, as diastema tissue specific markers. This study provides the first list of proteins that were detected in the mouse embryonic diastema region, which will be useful to understand the mechanisms of tooth development.

Early Preterm Infant Feeding Behaviors in Adolescent and Adult Mothers

  • Background: Adolescent mothers may find the transition to motherhood to be overwhelming when compared to adult mothers. The adaptiveness of mother and infant feeding behaviors may greatly influence the quality of adolescent mothers' feeding interactions when compared to adult mothers. Purpose: To examine the adaptiveness of early maternal and infant feeding behavior between adolescent mothers and adult mothers. Method: In this secondary analysis, six adolescent mothers (< 20 years old) and six adult mothers were videotaped while feeding. The mothers were observed 3 times: when the infants were in the hospital, 1 month old, and 4 months old. The videotapes were coded using four scales of the Parent-Child Early Relational Assessment (PCERA). The two maternal scales were Positive Affective Involvement and Sensitivity/Responsiveness (PAISR) and Regulation of Affect and Behavior (RAB). The two infant scales were Infant Positive Affect, Communication, and Social Skills (IPACS) and Infant Regulation of Affect and Behavior (IRAB). The dyads were videotaped just before hospital discharge and at one and four months corrected age. The Wilcoxon signed-rank test was used to examine the difference in the PCERA between the two groups. Results: Adolescent mothers had a significantly lower score on the PAISR before hospital discharge (p-value 0.005) and at one month corrected age (p-value 0.008) than adult mothers. Adolescent mothers also had a significantly lower score on the RAB before hospital discharge, 1 and 4 months corrected age than adult mothers. There were no significant differences in IPACS and IRAB of infants of adolescent and adult mothers. Conclusion: Clinicians need to be aware of the differences in sensitivity that occur.

Inhibition of Melanosome Transport by Inducing Exon Skipping in Melanophilin

  • Exon skipping is an efficient technique to inhibit specific gene expression induced by a short-sequence peptide nucleic acid (PNA). To date, there has been no study on the effects of PNA on skin pigmentation. In melanocytes, the tripartite complex is responsible for the transport of mature melanosomes from the nucleus to the dendrites. The tripartite complex is composed of Rab27a, Mlph (Melanophilin), and Myosin Va. Defects in the protein Mlph, a melanosome transport-related protein, are known to cause hypopigmentation. Our study shows that Olipass peptide nucleic acid (OPNA), a cell membrane-permeable PNA, targets exon skipping in the Mlph SHD domain, which is involved in Rab27a binding. Our findings demonstrate that OPNA induced exon skipping in melan-a cells, resulting in shortened Mlph mRNA, reduced Mlph protein levels, and melanosome aggregation, as observed by microscopy. Therefore, OPNA inhibits the expression of Mlph by inducing exon skipping within the gene. These results suggest that OPNA, which targets Mlph, may be a potential new whitening agent to inhibit melanosome movement.

LRRK2 and membrane trafficking: nexus of Parkinson's disease

  • Recent evidence from genetics, animal model systems and biochemical studies suggests that defects in membrane trafficking play an important part in the pathophysiology of Parkinson's disease (PD). Mutations in leucine-rich repeat kinase 2 (LRRK2) constitute the most frequent genetic cause of both familial and sporadic PD, and LRRK2 has been suggested as a druggable target for PD. Although the precise physiological function of LRRK2 remains largely unknown, mounting evidence suggests that LRRK2 controls membrane trafficking by interacting with key regulators of the endosomal-lysosomal pathway and synaptic recycling. In this review, we discuss the genetic, biochemical and functional links between LRRK2 and membrane trafficking. Understanding the mechanism by which LRRK2 influences such processes may contribute to the development of disease-modifying therapies for PD.

Complete genome sequence of Lactococcus lactis strain K_LL005, a xylose-utilizing bacterium isolated from grasshopper (Oxya chinensis sinuosa)

  • Lactococcus lactis is a fermentative lactic acid bacterium that is used extensively in food fermentations. The L. lactis strain K_LL005 was isolated from the grasshopper (Oxya chinensis sinuosa) gut in Korea. In this study, we reported the complete genome sequence of Lactococcus lactis K_LL005. The final complete genome assembly consist of one circular chromosome (2,375,093 bp) with an overall guanine + cytosine (G + C) content of 35.0%. Annotation results revealed 2,281 protein-coding sequences (CDSs), 19 rRNAs, and 68 tRNA genes. Lactococcus lactis K_LL005 has a gene encoding xylose metabolism such as xylR, xylA, and xylB (xylRAB).

B16F10 멜라닌 세포에서 신규 헥사펩타이드의 MITF 조절을 통한 멜라닌 생성 저해 효과 (Inhibitory Effects of Novel Hexapeptide on Melanogenesis by Regulating MITF in B16F10 Melanoma Cells)

  • 본 연구에서는 6 개의 아미노산으로 이루어진 헥사펩타이드(hexapeptide)의 미백 효능에 대해 수행하였다. 실험 결과 헥사펩타이드 처리에 의해 유의한 수준의 멜라닌 생성 저해가 관찰 되었고, 멜라닌 생성 과정에 관여하는 주요 효소인 tyrosinase의 활성이 농도 의존적으로 억제됨이 관찰 되었다. 멜라닌 생성 관련 인자들의 발현을 관찰 한 결과 tyrosinase (TYR), tyrosinase-related protein 1 (TYRP1) 및 이들의 상위 전사인자인 microphthalmia-associated transcription factor (MITF)의 발현이 헥사펩타이드 처리에 의해 유의한 수준으로 저해 되었다. 또한 헥사펩타이드 처리에 의해 MITF 발현을 조절하는 상위 전사인자인 cAMP-response element binding protein (CREB)의 인산화가 저해 되었고 MITF 인산화를 통해 프로테아좀 분해(proteasomal degradation)를 유도하는 extracellular signal-regulated kinase (ERK) 인산화가 증가 되었다. 이외에도, 멜라노좀의 세포 내 이동에 관여하는 복합체의 구성 인자들로 알려진 Rab27A, melanophilin, myosinVa의 발현도 헥사펩타이드에 의해 유의한 수준으로 저해 되었다. 이 결과를 통해, 본 연구의 헥사펩타이드는 멜라닌세포의 멜라닌 생성 관련 핵심 전사인자인 MITF의 발현 및 분해 조절을 통해 멜라닌 생성 억제 및 멜라노좀 이동과 같은 전반적인 멜라노좀 성숙 과정에 저해 효과를 나타내는 것으로 보인다. 헥사펩타이드의 이러한 미백 효능은 신규 미백 기능성 화장품 소재로 응용될 수 있을 것으로 기대된다.

농촌 지역사회노인의 만성질병수와 치매예방행위에 미치는 치매인식의 매개효과 (The Mediating Effect of Dementia Recognition on the Number of Chronic Diseases and Dementia Prevention Behaviors of Elders in Rural Communities)

  • Purpose: This study was a descriptive study to identify the factors affecting the dementia prevention behavior of elders in rural communities. Methods: The participants in this study were 125 elders aged 60 or older who lived in Eup or Myeon areas of P city. For data analysis, SPSS/WIN 22.0 was used to perform descriptive statistics, t-test, ANOVA, Pearson correlation, and linear multiple regression and mediated effects. Results: Scores for dementia recognition, dementia attitude and dementia prevention behavior averaged 5.6±2.50 points in the 0~11 range, 38.8±4.59 in 14~56 and 20.2±3.59 in 10~30 respectively. Dementia recognition (a), dementia attitude (b), dementia prevention behavior (c) and the number of chronic diseases of the elders (d) were positively or negatively correlated with each other (rab=.29, p<.01; rbc=.26, p<.01; rac=.36, p<.01; rad=-.29, p<.01; rcd=.19, p<.05). Factors affecting dementia prevention behavior were dementia recognition, dementia attitude, and degree of dementia interest. When the number of chronic diseases affects dementia prevention behavior, dementia recognition has a mediating effect. Conclusion: In order to prevent dementia among elders in rural areas, appropriate management of chronic diseases and provision of appropriate dementia-related education and information to enhance dementia recognition should be provided.

Mon1 Is Essential for Fungal Virulence and Stress Survival in Cryptococcus neoformans

  • Mon1 is a guanine nucleotide exchange factor subunit that activates the Ypt7 Rab GTPase and is essential for vacuole trafficking and autophagy in eukaryotic organisms. Here, we identified and characterized the function of Mon1, an ortholog of Saccharomyces cerevisiae Mon1, in a human fungal pathogen, Cryptococcus neoformans. Mutation in mon1 resulted in hypersensitivity to thermal stress. The mon1 deletion mutant exhibited increased sensitivity to cell wall and endoplasmic reticulum stress. However, the mon1 deletion mutant showed more resistance to the antifungal agent fluconazole. In vivo studies demonstrated that compared to the wild-type strain, the mon1 deletion mutant attenuated virulence in the Galleria mellonella insect model. Moreover, the mon1 deletion mutant was avirulent in the murine inhalation model. These results demonstrate that Mon1 plays a crucial role in stress survival and pathogenicity in C. neoformans.
