High performance liquid chromatography (HPLC) analysis was applied for determination of guanine plus cytosine (G + C) contents of DNA of Butyrivibrio fibrisolvens. By values of G + C contents, a reference strain and 20 wild strains of B. fibrisolvens were classified into at least two distinct subgroups, i.e. G + C contents of 18 strains were 38-40 mol% and those of 3 strains including the reference strain were 43-45 mol%. Clear relationships were not observed between G + C contents and biological properties of 21 strains of B. Fibrisolvens.
In order to clarify the cadmium(Cd) contents of normal deer, tissue samples including kidney, liver, muscle, spleen, heart, lung, rumen, abomasum, intestine, skin and bone were collected from 18 deer which were slaughtered in Korea. All samples were burned to ashes and analyzed for Cd contents by atomic absorption spectrophotometric method. The mean Cd contents in \mu\textrm{g}$/g wet matter for 18 deer were liver 0.013, kidney 0.286, muscle 0.010, spleen 0.001, heart 0.007, lung 0.005, men 0.019, abomasum 0.016, intestine 0.018, skin 0.016 and bone 0.312, respectively. Two samples in kidney, one sample in muscle out of 18 deer showed higher Cd contents than normal limit value of 0.50\mu\textrm{g}$/g for liver and kidney, 0.10\mu\textrm{g}$/g for muscle. In addition, three deer also showed higher Cd contents in one body tissue than normal limit value which reported foreign countries.

The quality including in situ degradability in the rumen of Holstein of the orchardgrass silage added with apple pomace was investigated in this study. The amount of apple pomace added in different treatments were 0, 20, 40 and 60% respectively. With higher amount of addition of apple pomace to orchardgrass, ADF, NDF and crude ash contents decreased significantly (p<0.05). Crude protein contents in the silages (11.8- 12.9%) were similar to that of 100% orchardgrass silage. Moisture contents increased according to the higher proportion of apple pomace in the silages. On the contrary to moisture content, pH was lower in 40-60% addition of apple pomace (3.7-3.9) than that of 100% orchardgrass silage (4.7). However the contents of lactic acid (1.7-2.5%), acetic acid (1.3- 1.7%) and total organic acid (2.9-4.2%) significantly increased according to higher levels of addition of apple pomace compared to the respective values of 100% orchardgrass silage (1.1%, 0.6% 1.7%). In siru disappearance rates of dry matter and NDF in the rumen were significantly higher at the stages of incubation after 24h in 40-60% addition of apple pomace than in 100% orchardgrass silage. No statistical differences were observed with quickly degradable fraction (a) and slowly degradable fraction (b) in the disappearance rates of dry matter and NDF. However, fractional rate of disappearance (c) and effective degradability (ED, k=0.08) for dry matter and NDF were significantly higher in 20-60% addition of apple pomace as 0.0076-0.0079 and 0.0099-0.0130. and 39.3-41.7% and 18.4- 20.6% respectively than the respective values of 0.0054 and 0.0064, and 36.8 and 16.5% of 100% orchardgrass silage. In this study, the effect of forage sources in the total mixed ration (TMR) on in vitro goat rumen fermentation was investigated. Rice straw (RS), Italian ryegrass (IRG), timothy (TIM), and alfalfa (ALF) were used as forage sources. Each forage source was mixed with a commercial goat concentrate diet in the ratio of 1:1. Total 4 TMR were prepared. Rumen simulated in vitro fermentation using goat rumen fluid collected from the slaughterhouse was conducted until 72th. For fermentation parameters, gas production (GP), volatile fatty acids (VFAs), and ammonia nitrogen (NH3-N) were examined. All assays were performed at 24th, 48th, and 72th h of incubation individually. Contents of crude protein and non-fibrous carbohydrate were greater in the order of RS < IRG < TIM < ALF. Significant treatment effects were found in valerate and NH3-N at 24th h of incubation (p<0.05). ALF showed the greatest contents of them and RS was the lowest. At 48th incubation, a significant effect was detected at GP (p<0.05) and RS was greater than others. However, GP of RS was lower than others at 72th. Significant effects on Total VFA, butyrate, and valerate productions were found at 72th h of incubation (p<0.05). ALF showed the greatest production. Methane production from all treatments was not significantly different for each incubation time (p>0.05). The present study provided primary information on how goat rumen fermentation responds to different nutrient contents and forage sources of TMR. And the information could be used for the design or optimizing economical diet formulation for goats.
Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.
To estimate the nutritional values and the optimum cooking method of the Cow's rumen-reticulum (tripe) which is a Korean traditional food, the nutrients of raw tissue of tripes, the changes in total N, ${\alpha}-amino$ N, calcium and phosphorus contents and nucleotides and their related compounds contents in soup stock prepared of tripe tissues according to various boiling time period with saucepot or pressure cooker and various ratios of the water to the tripe (wt/wt) were measured. No significant difference was observed in nutrients contents between rumen (1st stomach) and reticulum (2nd stomach) which contained 83% moisture, 0.4-0.5% ash, 3% fat, 13% protein, 50-56 mg% calcium, 75-76 mg% phosphorus. 75-77 mg% ${\alpha}-amino$ N on fresh weight basis. The results obtained show that the significant loss of nutrients observed after removing epitherial cell layer from tripe, and in aspect of the nutrients contents, the nucletides contents, and the sensory evaluation score in soup stock, the optimum cooking time period of tripe was 8 hours in boiling in saucepot and 1 or 2 hours in cooking in pressure cooker, and the ratio of the water to the tripe (wt/wt) was above ten.
The aim of this study is to examine feeding habits of the Korean water deer(Hydropotes inermis argyropus) from its rumen contents using a PCR-DGGE method. For this study, rumen contents were collected from water deer causalities by natural death or road-kill in two different sites(Cheorwon, Gangwon province and the Eastern part of Jeonnam province). DNA was extracted from rumen contents of a total of 44 individuals. Two primers, rbcLZ1aF(GC) and rbcL19bR, were used for PCR amplifications of ribulose-1,5-bisphosphate carboxylase large subunit (rbcL) gene. Among 44 samples, twenty-nine samples were successfully amplified by PCRs. The 29 PCR products of partial rbcL gene were applied for PCR-DGGE. Totally, six families of plants were detected from the diet analyses. Five families of plants were found in Cheorwon, Gangwon province, but only three families of plants were found in the Eastern part of Jeonnam province. The PCR-DGGE method will provide us with a potential tool to study feeding habits of ungulates including water deer, even though our results failed to identify the prey plants at the level of species.

The objectives of this study was to evaluate the degradability and digestibility of crude protein (CP), rumen undegradable protein (RUP), and individual amino acids (AA) on six by-product feedstuffs (BPF) (rice bran, RB; wheat bran, WB; corn gluten feed, CGF; tofu residue, TR; spent mushroom substrate from Pleurotus ostreatus, SMSP; brewers grain, BG) as ruminants feed. Three Hanwoo steers (40 months old, $520{\pm}20.20kg$ of body weight) fitted with a permanent rumen cannula and T-shaped duodenal cannula were used to examine of the BPF using in situ nylon bag and mobile bag technique. The bran CGF (19.2%) and food-processing residue BG (19.7%) had the highest CP contents than other feeds. The RUP value of bran RB (39.7%) and food-processing residues SMSP (81.1%) were higher than other feeds. The intestinal digestion of CP was higher in bran RB (44.2%) and food-processing residues BG (40.5%) than other feeds. In addition, intestinal digestion of Met was higher in bran RB (55.7%) and food-processing residues BG (44.0%) than other feeds. Overall, these results suggest that RB and BG might be useful as main raw ingredients in feed for ruminants. Our results can be used as baseline data for ruminant ration formulation.

In order to evaluate the feed value of rice straw as influenced by ripening stage, and to determine the effects of chemical treatments and ensiling on its feeding value, in situ studies using a rumen fistulated Korean cow nylon bag technique, and digestion trials with sheep were conducted. Experiments were conducted at the Experimental Farms, Woosuk University, Wanju in 1998. The results obtained are summarized as follows; 1. The contents of crude protein and crude fat were decreased(P<0.05), while those of crude fiber and crude ash were increased as the ripening of rice straw progressed. The content of NDF was not affected by the ripening stage. After the yellow stage hemicellulose was remarkably decreased while cellulose and lignin were increased. 2. Degradation of rice straw NDF in the rumen was most rapid when at the heading stage, but degradation of straws after the milky stage were similar each other. Degradation of rice straw ADF, on the other hand, did not show any difference with advancing ripening stage. In Conclusion, the change of chemical composition and degradation rate of rice straw in the rumen under the different ripening stage, it can be concluded that the lignification of rice straw was accelerated after the heading stage.

This study was conducted to examine the effect of corn (Zea mays L.) - soybean (Glycine max L.) silage prepared by intercropping method on the nutritive value of the silage, in vitro rumen fermentation characteristics, dry matter degradability, as well as milk yield and milk composition of dairy cows. In a couple of experiments intercropped corn-soybean silage (CSBS) was compared with corn silage (CS) and/or Italian ryegrass hay (IRG). Numerically, CSBS had higher crude protein, ether extract, and lactic acid contents compared to CS. In vitro rumen fermentation analysis demonstrated that up to a 24-h incubation period, both CS and CSBS showed higher total gas production, ammonia N concentration, and dry matter degradability compared to IRG (p<0.05). The investigation on animals was conducted in a commercial dairy farm located in Gyeongju, South Korea, employing 42 Holstein cows that were divided into 2 group treatments: CS and CSBS in a completely randomized design. Although no significant difference was observed in milk yield, animals fed on CSBS showed significantly higher milk protein (p<0.05) and milk fat content (p<0.01), compared to animals fed on CS. Taken together, our findings indicate that corn-soybean silage that is cultivated, harvested, and prepared through intercropping can improve the protein content of the silage, and can also enhance in vitro rumen fermentation, dry matter degradability, and performance of dairy cattle.
