Bioconversion of Pinoresinol Diglucoside from Glucose Using Resting and Freeze-Dried Phomopsis sp. XP-8 Cells

  • Phomopsis sp. XP-8 (an endophytic fungus) was previously found to produce pinoresinol diglucoside (PDG), a major antihypertensive compound of Tu-Chung (the bark of Eucommia ulmoides Oliv.), which is widely used in Chinese traditional medicines. In the present study, two bioconversion systems were developed for the production of PDG in Tris-HCl buffer containing glucose and Phomopsis sp. XP-8 cells (both resting and freeze-dried). When other factors remained unchanged, the bioconversion time, glucose concentration, cell ages, cell dosage, pH, temperature, and stirring speed influenced PDG production in a similar and decreasing manner after an initial increase with increasing levels for each factor. Considering the simultaneous change of various factors, the optimal conditions for PDG production were established as 70 g/l cells (8-day-old), 14 g/l glucose, $28^{\circ}C$, pH 7.5, and 180 rpm for systems employing resting cells, and 3.87 g/l cells, 14.67 g/l glucose, $28^{\circ}C$, pH 7.5, and 180 rpm for systems employing freeze-dried cells. The systems employing freeze-dried cells showed lower peak PDG production ($110.28{\mu}g/l$), but at a much shorter time (12.65 h) compared with resting cells (23.62 mg/l, 91.5 h). The specific PDG production levels were 1.92 and $24{\mu}g$ per gram cells per gram glucose for freeze-dried cells and resting cells, respectively. Both systems indicated a new and potentially efficient way to produce PDG independent of microbial cell growth.

A Comparative Study of Sea Trials and Production Processes for Propulsion Type Working Boats with a Tuna Purse Seiner (다랑어 선망어선 탑재용 보조 작업선의 추진기 형태 변화에 대한 제작과정 및 해상시운전 비교 연구)

  • In Korea, much research and development have occurred to enhance the technological competitiveness of tuna purse seining fisheries. Due to these efforts, fishing efficiency has been improved with the development of radar, sonar and global positioning systems for fish detection and revisions to the hull forms of tuna purse seiners. However, for skiff boats, net boats and speed boats, which are auxiliary working boats mounted on tuna purse seiners, technology has lagged behind relative to the modernization of the main vessel. In this study, the hull of an existing propeller-based net boat with steel wire net to protect tuna was changed to the hull of a water jet propulsion vehicle to reduce resistance and improve maneuverability. As a result, a prototype of a water jet propulsion option was produced according to the aluminum structure strength standards specified by the Ministry of Oceans and Fisheries, and safety was confirmed by performing a drop test. Moreover, through a sea trial test, an existing net boat was shown to have a speed of 12.0knots and a towing force of 2,545 kgf at 2,500 RPM. The prototype had a speed of 26.7 knots and a towing force of 2,011 kgf at 3,200 RPM, which satisfied the towing capacity standards of auxiliary working boats mounted on tuna purse seiners.

A Study on the Cutting Optimal Power Requirements of Fast Growing Trees by Circular Saw (원형톱에 의한 속성수 절단 적정 소요동력 산정에 관한 연구)

  • In this study, Italy poplar(Populus euramericana) was selected for test specimen to measure cutting power when it harvested. The experiment has been controlled as three levels of feed rate (0.41, 1.25 and 2.5 m/s), sawing speed (800, 1,000 and 1,200 rpm), and the five levels of root collar diameter (50, 70, 90 and 110, 130 mm). The harvested volume after 3 years (root collar diameter 50 mm) was 10.5 tons, which falls short of the target amount of biomass is 20~30 ton/ha. In addition, the biomass amount of diameter 90 and 110 mm which reached the target amount were estimated to be 23.5 and 32.5 ton/ha respectively. As a result of experiment, it was found out that power of 128.2 and 175.8 W are consumed in case of cutting with the feed rate of 0.41m/s and minimum sawing speed (800 rpm) respectively. With the working area of 0.3 ha/h, it is considered to present working capacities of 16.5 and 22.8 ton/h respectively. The power consumed at the feed rate of 1.25 m/s is estimated to be 113.8 and 153.7W respectively and working capacity in a working area of 1 ha/h is estimated to be 23.5 and 32.5 ton/h. The power consumed at the feed rate of 2.5 m/s is estimated to be 119.8 and 166.9 W respectively and working capacity in a working area of 2 ha/h is estimated to be 47.0 and 65.5 ton/ha respectively. Therefore, the power source of harvest machine at the feed rate of 1.25, 2.50 m/s and sawing speed of 800 rpm shall be selected as it can process the target amount of estimated biomass.

A Determination Model of the Data Transmission-Interval for Collecting Vehicular Information at WAVE-technology driven Highway by Simulation Method (모의실험을 이용한 WAVE기반 고속도로 차량정보 전송간격 결정 모델 연구)

  • This paper deals with the transmission interval of vehicle data in smart highway where WAVE (Wireless Access for Vehicular Environments) systems have been installed for advanced road infrastructure. The vehicle data could be collected at every second, which is containing location information of the vehicle as well the vehicle speed, RPM, fuel consuming and safety data. The safety data such as DTC code, can be collected through OBD-II. These vehicle data can be used for valuable contents for processing and providing traffic information. In this paper, we propose a model to decide the collection interval of vehicle information in real time environment. This model can change the transmission interval along with special and time-variant traffic condition based on the 32 scenarios using microscopic traffic simulator, VISSIM. We have reviewed the transmission interval, communication transmission quantity and communication interval, tried to confirm about communication possibility and BPS, etc for each scenario. As results, in 2-lane from 1km highway segment, most appropriate transmission interval is 2 times over spatial basic segment considering to communication specification. In the future, if a variety of wireless technologies on the road is introduced, this paper considering not only traffic condition but also wireless network specification will be utilized the high value.

Tool-Setup Monitoring of High Speed Precision Machining Tool

  • Recently the monitoring system of tool setting in high speed precision machining center is required for manufacturing products that have highly complex and small shape, high precision and high function. It is very important to reduce time to setup tool in order to improve the machining precision and the productivity and to protect the breakage of cutting tool as the shape of product is smaller and more complex. Generally, the combination of errors that geometrical clamping error of fixing tool at the spindle of machining tool and the asynchronized error of driving mechanism causes that the run-out of tool reaches to 3$^{\sim}$20 times of the thickness of cutting chip. And also the run-out is occurred by the misalignment between axis of tool shank and axis of spindle and spindle bearing in high speed rotation. Generally, high speed machining is considered when the rotating speed is more than 8,000 rpm. At that time, the life time of tool is reduced to about 50% and the roughness of machining surface is worse as the run-out is increased to 10 micron. The life time of tool could be increased by making monitoring of tool-setup easy, quick and precise in high speed machining tool. This means the consumption of tool is much more reduced. And also it reduces the manufacturing cost and increases the productivity by reducing the tool-setup time of operator. In this study, in order to establish the concept of tool-setup monitoring the measuring method of the geometrical error of tool system is studied when the spindle is stopped. And also the measuring method of run-out, dynamic error of tool system, is studied when the spindle is rotated in 8,000${\sim}$60,000 rpm. The dynamic phenomena of tool-setup are analyzed by implementing the monitoring system of rotating tool system and the non-contact measuring system of micro displacement in high speed.

  • Solution processed Resistive random access memory (ReRAM)은 간단한 공정 과정, 고집적도, 저렴한 가격, 대면적화 플라즈마 데미지 최소화 등의 장점으로 차세대 비휘발성 메모리로 써 많은 관심을 받고 있으며, 주로 high-k 물질인 HfOx, TiOx, ZnO 가 이용 된다. IGZO와 ZTO와 같은 산화물 반도체는 높은 이동도, 대면적화, 넓은 밴드갭으로 인하여 투명한 장점으로 LCDs (Liquid crystal displays)에 이용 가능하며, 최근에는 IGZO와 ZTO에서 Resistive Switching (RS) 특성을 확인한 논문이 보고되면서 IGZO와 ZTO를 ReRAM의 switching medium와 TFT의 active material로써 동시에 활용하는 것에 많은 관심을 받고 있다. 이와 같은 산화물 반도체는 flat panel display 회로에 TFT와 ReRAM의 active layer로써 집적가능 하며 systems-on-panels (SOP)에 적용 가능하다. 하지만 IGZO 보다는 ZTO가 In과 Ga을 포함하지 않기 때문에 저렴하다. 그러므로 IGZO를 대신하는 물질로 ZTO가 각광 받고 있다. 본 실험에서는 ZTO film에 Al을 doping하여 메모리 특성을 평가하였다. 실험 방법으로는 p-type Si에 습식산화를 통하여 SiO2를 300 nm 성장시킨 기판을 사용하였다. 그리고 Electron beam evaporator를 이용하여 Ti를 10 nm, Pt를 100 nm 증착 한다. 용액은 Zn와 Tin의 비율을 1:1로 고정한 후 Al의 비율을 0, 0.1, 0.2의 비율로 용액을 각각 제작하였다. 이 용액을 이용하여 Pt 위에 spin coating방법을 이용하여 1000 rpm 10초, 6000 rpm 30초의 조건으로 AZTO (Al-ZnO-Tin-Oxide) 박막을 증착한 뒤, solvent 및 불순물 제거를 위하여 $250^{\circ}C$의 온도로 30분 동안 열처리를 진행하였다. 이후 Electron beam evaporator를 이용하여 top electrode인 Ti를 100 nm 증착하였다. 제작된 메모리의 전기적 특성은 HP 4156B semiconductor parameter analyzer를 이용하여 측정하였다. 측정 결과, AZTO (0:1:1, 0.1:1:1, 0.2:1:1)를 이용하여 제작한 ReRAM에서 RS특성을 얻었으며 104 s이상의 신뢰성 있는 data retention특성을 확인하였다. 그리고 Al의 비율이 증가할수록 on/off ratio가 증가하고 endurance 특성이 향상되는 것을 확인하였다. 결론적으로 Al을 doping함으로써 ZTO film의 메모리 특성을 향상 시켰으며 AZTO film을 메모리와 트랜지스터의 active layer로써 활용 가능할 것으로 기대된다.

Experimental Study on Performance Characteristics of Air Driven Scroll Expander (공기구동 스크롤 팽창기 성능특성에 관한 실험적 연구)

  • The performance of a scroll expander is the most important factor for the efficiency of small scale Organic Rankine cycle waste heat power generation systems. In this research, a scroll compressor was purchased and operated in reverse to function as a scroll expander. With air as a working fluid, a series of performance test were conducted on this expander by varying the inlet and outlet pressure. Secondly, We have tested through 2000 to 3500 rpm rotational speed to find the maximum power and efficiency of the expander. And last, It was observed in the initial experiments that the design of the expander's orbiting scroll wrap partially blocked the fluid intake which may have caused unnecessary flow resistance. To verify this theory, a small part of the scroll wrap was removed and the performance test was redone. It was observed that the lower back pressure assure the higher efficiency and power of expander and the rotational speed that shows maximum adiabetic efficiency of scroll expander is 69% at 2500 rpm. And by modified wrap of the scroll, we could get volume flow rate for 13% to 19% and power for 5% to 18% increased. But the maximum efficiency of the modified scroll was decreased 8%.

  • Research and development of LP EGR system for the performance improvement and emission reduction on diesel engine is proceeding at a good pace. LP EGR system seems to be helpful method to further reduce$NO_x$ emissions while maintaining PM emissions at a low level because the boost pressure is unchanged while varying EGR rate. This study is experimentally conducted on a 2.0L common rail DI engine at the medium load condition (2000 rpm, BMEP 1.0 MPa, boost pressure 181.3 kPa) that difficult to use large amount of EGR gas because of deteriorations of performance and fuel consumption. And we investigated the characteristics of performance and fuel consumption while varying EGR systems. The overall results using LP EGR system equipped with ETC identified benefits on reduction of PM and improvement of fuel consumption and thermal efficiency while keep the $NO_x$ level compared to HP EGR and LP EGR with back pressure valve.

Preparative Isolation of Ginseng Saponin from Panax ginseng Root Using High-speed Countercurrent Chromatography (High-speed countercurrent chromatography를 이용한 인삼 saponin의 대량 분리 농축)

  • Ginseng saponin was isolated from panax ginseng root using high-speed countercurrent chromatography (HSCCC). Preliminary studies were performed to optimize physical properties of two-phase solvent system and operating parameters including rotation speed of column, elution mode of mobile phase, and flow rate. Two-phase solvent system for isolation of ginseng saponins was composed of chloroform, water, and methanol as blending solvent. Chloroform-aqueous methanol (4:6) systems with various concentration of methanol in water were evaluated for retention of stationary phase in column. Retention of stationary phase decreased with increasing flow rate in tail-to-head elution mode using upper phase as mobile phase and head-to-tail elution mode using lower phase as mobile phase. Latter mode produced high retention at flow rate of 5 mL/min. Optimum conditions for isolation of saponin were chloroform/methanol/water (40/39/21) solvent system; mobile phase, of lower organic layer, flow rate, of 5 mL/min, head to tail elution mode, rotation speed, of 800 rpm, and sample injection, of $200{\mu}L$, Recovery yield of ginseng saponin from panax ginseng root extract by HSCCC was 63.6%, and the purity of HSCCC fractions was verified by TLC.

Application of Scale-Up Criterion of Constant Oxygen Mass Transfer Coefficient ($k_La$) for Production of Itaconic Acid in a 50 L Pilot-Scale Fermentor by Fungal Cells of Aspergillus terreus

  • The scale-up criterion of constant oxygen mass transfer coefficient ($k_La$) was applied for the production of itaconic acid (IA) in a 50 L pilot-scale fermentor by the fungal cells of Aspergillus terreus. Various operating conditions were examined to collect as many $k_La$ data as possible by adjusting the stirring speed and aeration rate in both 5 L and 50 L fermentor systems. In the fermentations performed with the 5 L fermentor, the highest IA production was obtained under the operating conditions of 200 rpm and 1.5 vvm. Accordingly, we intended to find out parallel agitation and aeration rates in the 50 L fermentor system, under which the $k_La$ value measured was almost identical to that ($0.02sec^{-1}$) of the 5 L system. The conditions of 180 rpm and 0.5 vvm in the 50 L system turned out to be optimal for providing almost the same volumetric amount of dissolved oxygen (DO) into the fermentor, without causing shear damage to the producing cells due to excessive agitation. Practically identical fermentation physiologies were observed in both fermentations performed under those respective operating conditions, as demonstrated by nearly the same values of volumetric ($Q_p$) and specific ($q_p$) IA production rates, IA production yield ($Y_{p/s}$), and specific growth rate (${\mu}$). Specifically, the negligible difference of the specific growth rate (${\mu}$) between the two cultures (i.e., $0.029h^{-1}$ vs. $0.031h^{-1}$) was notable, considering the fact that ${\mu}$ normally has a significant influence on $q_p$ in the biosynthesis of secondary metabolites such as itaconic acid.
