Terpenoids and Phenolics from Geum japonicum (뱀무로부터 테르페노이드 및 페놀성 성분의 분리)

  • Twenty-five compounds were isolated from the methanolic extract of Geum japonicum (Rosaceae), and their structures were identified as eleven triterpenoids [ursolic acid 3-acetate (2), cecropiacic acid 3-methyl ester (3), pomolic acid 3-acetate (5), ursonic acid (6), ursolic acid (7), pomolic acid (8), corosolic acid (9), euscaphic acid (11), arjunic acid (16), tormentic acid (18), 23-hydroxytormentic acid (21)], two saponins [rosamultin (22) and kaji-ichigoside $F_1$ (23)], two megastigmanes [blumenol A (14) and (+)-dehydrovomifoliol (15)], three flavonoids [apigenin (13), isoquercitrin (17) and tiliroside (24)], two ellagic acid derivatives [3,3'-di-O-methylellagic acid (12) and ducheside B (25)] and five others [eugenol (1), emodin (4), vanillic acid (10), gallic aldehyde (19), salidroside (20)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. This is the first report of the eleven compounds, 2~6, 10, 15, 16, 20, 23, and 25 from the genus Geum, as well as the first report of apigenin (13) and 3,3'-di-O-methylellagic acid (12) from G. japonicum. The antioxidant properties of 22 isolates (1~11, 14, 16~25) were evaluated by the intracellular reactive oxygen species (ROS) radical scavenging using 2',7'-dichlorodihydrofluorescein diacetate (DCF-DA) assay. Among them, isoquercitrin (17) showed significant scavenging activity, and gallic aldehyde (19) and ducheside B (25) showed weak scavenging activity.

Chemical Characterization and Utilization of $19{\alpha}-Hydroxyursane-type$ Triterpenoids in Rubus species (Rubus속 식물에 존재하는 $19{\alpha}-Hydroxyursane-type$ Triterpenoid의 특성과 이용)

  • The plant Rubus species (Rosaceae) mainly contains $19{\alpha}-hydroxyurane-type$ triterpenoids $(19{\alpha}-HUT)$ as bioactive components. Available functional food includes blackberry (the fruit of thornless Rubus sp.), red raspberry (R. idaeus) and black raspberry (R. occidentalis). However, the fruit of R. coreanus, which is used in Korea as a functional food, substitutes black raspberry. Rubi Fructus, which has been traditionally used as an oriental medicinal drug, designates only unripe fruit of R. coreanus but not its ripe fruit which indicates that it needs high content of $19{\alpha}-HUT$ as a crude drug. Throughout our experiment, we found that ripe fruits contain very little amount of $19{\alpha}-HUT$ when compared to unripe fruits. In addition, various and rich $19{\alpha}-HUT$ has been reported from Rubus species. The most common structure of $19{\alpha}-HUT$ of Rubus species, euscaphic acid or tormentic acid with $3{\alpha}-OH$ or $3{\beta}-OH$, respectively, can be glycosylated or oxidized to produce a number of $19{\alpha}-HUTs $as euscaphic acid and tormentic acid derivatives and even esterified to form dimeric triterpenoids. In this review, the bioactivity and biosynthetic pathway and chemical characterization of $19{\alpha}-HUTs$ found in Rubus species are discussed.

The Molecular Phylogenetic Study of Filipendula (Rosaceae) (터리풀속(Filipendula)의 분자계통학적연구)

  • 터리풀속(Filipendula)은 장미과(Rosaceae), 장미아과(Rosoideae)에 속하는 다년생 초본이며, 북반구 온대지역의 산지지역에 서식하며 15-20여 종이 보고되어 있고, 이 중 10여종이 한국, 중국, 일본, 타이완 등의 동아시아 지역에 분포한다. 본 연구의 목적은 DNA 염기서열 자료를 이용하여 터리풀속(Filipendula)내 종들간의 계통관계를 규명하기 위하여 본 연구를 수행하였다. 이를 위해서 11종 29개체의 터리풀속(Filipendula)샘플과 외군인 산딸기나무속(Rubus)에 속하는 3종 5개체의 샘플을 이용하였다. 추가로 Genbank에서 3속 10종 18개의 염기서열을 다운받아 비교분석에 이용하였다. 계통연구를 위하여 엽록체에 존재하는 atpF-atpH, psbK-psbI, psbA-trnH, matK, rbcL, 5개 마커와 핵에 존재하는 ITS, 총 6개 마커의 염기서열을 생산하였다. 총 52개의 샘플에 대하여 엽록체유전체 5개 마커지역은 염기서열 길이가 3,485bp였고 핵 ITS지역은 631bp였으며, 이들을 합한 염기서열 길이는 4,116bp였다. 계통분석결과, 터리풀속(Filipendula)은 단계통군을 이루었다. F. occidentalis와 F. vulgaris가 기저분류군을 이루었고 이들은 각각의 아속에 해당한다. 그리고 나머지 종들은 모두 하나의 단계통군을 이루었다. 위의 결과들은 1961년 시미즈가 본 속을 Hypogyna아속, Filipendula아속, Ulmaria아속으로 나눈 분류시스템과 일치한다. 나아가 분자계통수에서 Ulmaria아속은 크게 4개의 subclade로 구분되었다. 먼저 subclade I에는 F. vestita, F. kiraishiensis, F. tsuguwoi, F. multijiuga, F. purpurea 등 5개 종으로 구성되었다. Subclade II는 F. ulmaria 한 종으로만 구성되었다. Subclade III에는 F. glaberrima, F. koreana, F. formosa, F.camtschatica 로 구성되었으며 subclade III에는 한국에 서식하는 3종이 포함되었다. Subclade IV에는 F. rubra, F. angustiloba, F. palmata, F. intermedia 4종으로 구성되었다. 이번연구에서는 Ulmaria아속내에 4개의 subclade가 존재함이 처음으로 확인되었다.

Taxonomy of tribe Neillieae (Rosaceae): Neillia (나도국수나무족(장미과)의 분류: 나도국수나무속)

  • Neillieae was traditionally recognized as a small tribe in Rosaceae, which consists of Neillia, Stephanandra, and Physocarpus. Recently, Stephanandra was merged into Neillia based on molecular phylogenetic analyses, meaning that Neillieae now contains Neillia and Physocarpus. The genus Neillia is distinguished from Physocarpus by ovate to lanceolate leaves with acuminate to caudate apices, racemose or paniculate inflorescences, and a unicarpellate (rarely bicarpellate) gynoecium. Plants of Neillia are distributed from the Himalayas across China and Korea to Japan in the east, and south to Indonesia. This study provides a taxonomic treatment of Neillia based on a morphological examination of herbarium specimens, including types, and field observations, as the second part of the taxonomic revision of the tribe Neillieae. A summary of the phylogeny of Neillia, keys to the species, nomenclatural reviews, descriptions, distribution maps, and lists of specimens examined are provided. Twelve species with ten varieties are recognized in Neillia. A lectotype was designated here for the following species: N. sinensis f. glanduligera and N. thyrsiflora.

DNA barcoding analysis of Rosase Multiflorae Fructus and its adulterants (영실(營實)과 그 위품의 유전자 감별)

  • Objectives : Rosae Multiflorae Fructus is a traditional medicine derived from the fruit of Rosa multiflora Thunb. a member of the Rosaceae family. Even though it has a single origin, the possibility of adulterants has always existed. In fact, we had discovered suspicious commercial samples of Rosae Multiflorae Fructus, imported from China. Methods : To define the taxonomic origin of Rosae Multiflorae Fructus and its adulterants, DNA barcode analysis of the internal transcribed spacer, trnL-F intergenic spacer, and psbA-trnH sequences was carried out. These DNA barcode sequences from the correct origin of Rosae Multiflorae Fructus were analyzed and compared with those of other samples from genus Rosa used as medicinal herbs. Results : The analyses of the three DNA barcode sequences efficiently distinguished Rosae Multiflorae Fructus from six other species in genus Rosa and also separated each species used in this study. According to the DNA barcoding results, none of the suspicious commercial samples were Rosae Multiflorae Fructus. RMF09 was identified as Rosa acicularis, whereas RMF10 and RMF11 were identified as Rosa davurica and Rosa rugosa, respectively. These results corroborated the existence of adulterants of Rosae Multiflorae Fructus. Conclusions : Our research provides useful information that could be used as a criterion for distinguishing between Rosae Multiflorae Fructus and its adulterants. These results will help in the prevention of adulteration and also suggest effective methods for verifying the origin of commercial herbal medicines derived from genus Rosa.

Studies on the Vegetation around the Industrial Complexes (사대공단(四大工團) 주변(周邊)의 식생(植生)에 대(對)한 연구(硏究))

  • The present study was conducted during the period July 18-August 11, 1981, at the four industrial complexes(Pohang, Ulsan, Onsan, and Yeochon) using the naked eye method. The study included the survey of plant species growing within a 2km from the area of pollution source, selection of plants tolerant to air pollution and effects of pollution on their growth. The results obtained are as follows; 1) Vascular plants surveyed as to industrial complexes consist of 90 families, 6 subfamilies, 213 genera and 290 species. Major plant families and species for each area are as follows: 1) Pohang area: 54 families, 102 genera and 126 species Leguminosae(16) Graminae(11), Compositae(9) Rosaceae(9) 2) Ulsan area: 61 families, 131 genera, and 158 species Compositae(17) Gramineae(16) Leguminosae(15) Rosaceae(9) Polygonaceae(9) Solanaceae(6) 3) Onsan area: 46 families, 98 genera, and 158 species Compositae(12) Gramineae(12) Leguminosae(12) Rosaceae(6) Polygonaceae(6) 4) Yeochon: 71 families, 150 genera, and 188 species Rosaceae(15) Leguminosae(15) Compositae(14) Graminese(14) Polygonaceae(8) Salicaceae(6) 2. Table 1 shows tolerant plants to air pollution in this study. 3. The following plants may be recommended as planting species at the industrial complex area. Leguminosae: Amorpha fruticosa L.   • The plants medicinal resources of Mt. Eb-aon were investifeted 18times from JuLy, 1988 to August, 1989. In orther to analyze the vegeta-tion of Eb-am mountain area, medical wild plants structure and distr-ibution. Medical wild Plants of Eb-am mountain consisted of 100 familis, 337 species in alL. The resources of important herb drugs were, Graminea, Liliaceae, Molaceae, Polygonaceae, Ranunculaceae, Brassicaceae, Rosaceae, Drupaceae, Fabaecae, Apiaceae, Labiatae, Solanaceae, Scrophulariaceae, Rubiaceae, CampanuLaceae, ComposLtae, The herb drufs were Comparatively more than in other mountains in our country.

  • Aegiceradienol (28-noroleana-12, 17-dien-$3{\beta}$-ol) was isolated from the roasted Sanguisorbae radix and its structure was elucidated by various spectroscopic techniques. Aegiceradienol was an artifact produced by heat treatment of the herbal drug. Its content was increase as the time of heat treatment increased.

  • Fruits of Rubus coreanum (Korean name: Bog-bun-ja) have been used in oriental traditional medicine as the remedies for impotence, pollution, premature ejaculation and frequency of urination etc. It is known to have phenolic compounds as an astringent. By means of chromatographic methods, four hydrolyzable tannins were isolated from the fruits of R. coreanum. The structures of these compounds were established as gallic acid, 2,3(S)-HHDP-D-glucopyranose, sanguiin H-4 and sanguiin H-6 on the basis of physicochemical and spectroscopic evidences.

  • Four ursane type triterpenes have been isolated from the methanolic extract of the aerial parts of Agrimonia pilosa Ledeb. (Rosaceae) through repeated silica gel and reverse-phase C-18 column chromatography. Their chemical structures were elucidated as ursolic acid (1), pomolic acid (2), tormentic acid (3), and corosolic acid (4) on the basis of their MS, $^1H-$, and $^{13}C-NMR$ spectral data. Compounds 2 and 4 were isolated from the genes of Agrimonia for the first time.
