Antioxidants in Leaves of Rosa rugosa (해당화(Rosa rugosa) 잎의 항산화물질)

  • To search for useful antioxidants from plant materials, we investigated the antioxidative activity in the methanol extracts of 30 Chinese medicinal plants using DPPH method. The highest activity $(RC_{50}:\;12\;{\mu}g)$ was showed in the methanol extract of Rosa rugosa, followed by Potentilla fruticosa $(14\;{\mu}g)$, P. fragarioides $(16\;{\mu}g)$. And Geum aleppicum $(18\;{\mu}g)$. From the leaves of R. rugosa, two antioxidative compounds were isolated by a bioassay guided purification and identified as isoquercitrin and ${\beta}-glucogallin$ on the basis of $^1H-,\;^{13}C-\;NMR$, and FAB-MS data. ${\beta}-Glucogallin$ is the first report in this plant. The DPPH radical scavenging activity of ${\beta}-glucogallin\;(RC_{50}:\;8.5\;{\mu}g)$ was more effective than those of ${\alpha}-tocopherol\;(12\;{\mu}g)$ and BHA $(14\;{\mu}g)$.

Biological activities of Rosa multiflora Ethanol Extract as Cosmetic Material

  • The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity as a functional ingredient for cosmetic products have not yet been studied. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. The antioxidant activities were determined by DPPH. Additionally, the contents of total phenols and flavonoids were analyzed. Also, the phenolic compounds were detected using HPLC. The melanogenesis regulatory effect was evaluated using melanin content and cellular tyrosinase activity in B16F10 melanoma cells. The elastase inhibitory activity assay was performed for anti-wrinkle effect. The antimicrobial activity was assessed using the disc diffusion assay. The DPPH radical scavenging ability, denoted by the $SC_{50}$ value was found to be $123.1{\mu}g/ml$, whereas that of positive control (ascorbic acid) was $27.5{\mu}g/mL$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In addition, astragalin and gallic acid were identified in the extract. Also, the ethanol extract significantly inhibited ${\alpha}$-MSH-induced melanogenesis in B16F10 cells. For anti-wrinkle effect, elastase inhibition activity of the ethanol extract was 53.2% at a concentration of $100{\mu}g/ml$. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 0.5 - 5%, and no significant activity was noted against C. albicans. Therefore, the ethanol extract of Rosa multiflora can be used effectively for development of functional cosmetic materials.

  • The antimicrobial activity of 70% ethanol extracts from Rosa multiflora Thunberg fruit against Helicobacter pylori was examined. The inhibitory activity of Rosa multiflora Thunberg fruit extracts against H. pylori was determined to clear a zone of 14 mm with 70% ethanol extracts. Purification of inhibitory compounds was carried on Sephadex LH-20 and $C_{18}$ cartridge column chromatography using a gradient procedure, with increasing ethanol ($0{\rightarrow}100%$) in $H_2O$. The chemical structure of the purified inhibitory compounds on H. pylori was identified to be protocatechuic acid, chlorogenic acid and quercetin by FAB-MS, NMR and IR spectrum.

Anti-Inflammatory Effect of Rosa rugosa Flower Extract in Lipopolysaccharide-Stimulated RAW264.7 Macrophages

  • Rosa rugosa Thunb, a deciduous shrub of the genus Rosa, has been widely used to treat stomach aches, diarrhoea, pain, and chronic inflammatory disease in eastern Asia. In recent years, our research team has extensively studied the Rosa rugosa flower extract, and specifically undertook pharmacological experiments which have optimized the extraction process. Our methods have yielded a standard extract enriched in phenolic compounds, named PRE. Herein, we expand our efforts and evaluated the anti-inflammatory activity of PRE on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophages. PRE significantly inhibited production of nitric oxide (NO), prostaglandin $E_2(PGE_2)$, tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and interleukin $1{\beta}$ (IL-$1{\beta}$), as well as expression of their synthesizing enzymes, inducible nitric oxide synthase (iNOS) and cyclooxygenase2 (COX-2). Furthermore, PRE inhibited activity of mitogen-activated protein kinases (MAPK) as well as nuclear factor-kappa B (NF-${\kappa}B$) signaling pathway. Our findings are the first to explain the anti-inflammatory mechanism by PRE in LPS-stimulated macrophages. Given these results, we propose that PRE has therapeutic potential in the prevention of inflammatory disorders.

Effects of Aqua-acupuncture with Rosa rugosae Radix on Diabetic Rats induced by Streptozotocin (매괴화근 약침(藥鍼)이 streptozotocin으로 유발시킨 당뇨(糖尿)에 미치는 영향(影響))

  • This experiment was performed in order to investigate the effect of Rosa rugosae Radix on hyperglycemia and diabetes in streptozotocin-induced rats. In this study, the experimental rats were divided into three groups: Control group, Group-A(Aqua-acupuncture group) and Group-B(Orally administ:ered group). Under the same condition, three groups were injected 50mg/kg of streptozotocin into abdomen for 3days. And then, Control group was administered water, Group-A was administered Rosa rugosae Radix aqua-acupuncture to S36, B20 and B21. Group-B was administered Rosa rugosae Radix water extract for 14days. Then kidney weight, serum glucose levels, BUN levels, creatinine levels and Langerhans islets were measured on 7th and 14th days. The obtained results are summarized as follows: 1. The kidney weight of control group was heavier than those of group-A and group-Bon 7th day. 2. The levels of serum glucose were decreased in group-A and group-B at l20minutes 14th day as compared with control group. 3. The levels of BUN were decreased in group-A and group-B as compared with control group. 4. In histological observations, Langerhans islets of control group showed disrupted or condensed nudei, and intercellular spaces between acini were enlarged. In the group-A and group-B, histological damages of the acini and Lagerhans islets were recovered as compared with control group. These results suggest that Rosa rugosae Radix can applied for the treatment of diabetes melitus.

Effects of Rosa multiflora and Rosa multiflora Complex on Lipid Content in Rats Fed a High-Fat·High-Cholesterol Diet (찔레꽃 및 찔레꽃 복합물 추출물이 고지방·고콜레스테롤 식이 흰쥐의 지질 함량 개선에 미치는 영향)

  • This study was conducted to investigate the effects of water extract from Rosa multiflora and Rosa multiflora complex on lipid metabolism in rats fed a high cholesterol diet. Experimental rats were divided into four groups, which were composed of normal diet group (N group), high cholesterol diet group (HF group), high cholesterol and 5% Rosa multiflora diet group (HR group), and high cholesterol and Rosa multiflora complex diet group (HRC group). Rosa multiflora complex is composed of Duchesnea chrysantha, Salvia plebeia R. BR., and Sasa borealis. The serum triglyceride (TG) content of the HF group was significantly higher than that of the N group, whereas that of the HRC group was significantly lower. Serum high-density lipoprotein-cholesterol contents of the HR and HRC groups were significantly higher than that of the HF group. The serum total cholesterol and low-density lipoprotein- cholesterol contents of the HF group were significantly higher than those of the N group, and especially the atherogenic index of the HRC group was significantly reduced compared to the HF group. The liver TG and total cholesterol contents of the HF group were significantly increased compared to the normal diet group, while those of the HR and HRC groups were significantly decreased compared to the HF group. Fecal total lipid and total cholesterol contents of the HR and HRC groups were significantly increased compared to the HF group. Uridine 5'-diphospho (UDP)-glucuronyl transferase activity of the HR and HRC groups were increased compared to the HF group. Total bile acid contents of the HR and HRC groups were increased compared to the HF group, and that of the HRC group was significantly increased compared to the HF groups. These results suggest that Rosa multiflora supplementation has powerful health benefits due to UDP-glucuronyl transferase activity, bile acid, and lipid metabolism.

Anti-HIV-1 Protease Activity and in Vivo Anti-lipid Peroxidative Effect on Rosa davurica (생열귀나무의 항 HIV-1 protease 작용과 생체내 과산화지질생성 저해효과)

  • Anti-human immunodeficiency virus (HIV) type I protease (PR) and anti-lipid peroxidation effects on Rosa davurica were investigated. Of the various parts tested from R. davurica, the water extracts of stem and leaves inhibited the HIV-1 PR activity by more than 45% at a concentration of $100\;{mu}g/mL$. Hyperoside from the percarp of title plant showed 25% inhibition on HIV-1 PR at $200\;{mu}M$. The methanol extract of the root of R. davurica reduced the level of lipid peroxides induced by bromobenzene in vivo.

  • A tannin compound has been isolated from the underground part of Rosa rugosa Thunb. and its structure has been established to be methyl gallate $3-O-{\beta}-D-(6'-O-galloyl)-glucopyranoside(1)$ on the basis of chemical and spectroscopic evidences. This compound was the first example in this plant.

First Report of Die-Back on Rose (Rosa hybrida) Caused by Lasiodiplodia pseudotheobromae in Korea (Lasiodiplodia pseudotheobromae에 의한 장미 가지썩음병의 발생 보고)

  • In 2015, symptoms of die-back on Rosa hybrida were observed in Taean, Korea. The aims of this study were to determine the cause of die-back on Rosa hybrida and characterize the pathogen. The fungal isolates were obtained and used for pathogenicity test, morphological and molecular analyses. The pathogenicity test on healthy branches of Rosa hybrida produced die-back, as the original symptoms. For the morphological study, the isolates were inoculated onto potato dextrose agar and incubated for 7 days at $25^{\circ}C$. The colonies grew up quickly and turned white to gray in color. Conidia were observed under an optical microscope. The features of conidia were ellipsoidal, grayish brown in color, $20-31{\times}11-17{\mu}m$ in size and had one septum. Molecular analyses of the ITS region, TEF and TUB genes were conducted to confirm the identity of the pathogen. The phylogenetic tree of the multi-gene sequences indicated that the causal agent was Lasiodiplodia pseudotheobromae. This study is the first report of die-back caused by Lasiodiplodia pseudotheobromae on Rose (Rosa hybrida).

Genetic Relationship of Genus Rosa Germplasm and Genetic Diversity of Rosa rugosa in Korea (장미속 유전자원의 유전적 관계와 국내 해당화의 유전적 다양성)

  • Fifty-nine accessions of 23 species in genus Rosa were collected, and 15 accessions of Rosa rugosa were collected throughout 10 regions of Korea. Their genetic relationship was investigated by using morphological analysis and RAPD marker. The morphological analysis was measured for 7 quantitative and quantified for 4 qualitative traits. RAPD analysis obtained a total of 959 polymorphic bands by using twenty primers. Morphological analysis classified most according to the rose section system except for several species. The cluster analysis of genus Rosa based on RAPD data could identify the subgenus $Platyrhodon$ and $Eurosa$. The subgenus $Eurosa$ separated five sections; $Gallicanae$, $Cinnamomeae$, $Pimpinellifoliae$, $Synstylae$ and $Caninae$. Correlation analysis between morphological and RAPD analysis showed low significance ($r$ = 0.35). The accessions of R. rugosa belonged to the section $Cinnamomeae$ clustered into three groups at genetic distance ranging from 0.28 on the base of RAPD analysis. In conclusion, the genetic relationship of the genus Rosa was consistent to the previously reported rose section system, and domestic collections of $R.$ $rugosa$ were separated from 3 groups on the base of RAPD marker.
